Abstract:An efficient route to synthesize 4-chloro-7H-pyrrolo[2,3-d]pyrimidine (6) and 2,4-dichloro-7H-pyrrolo[2,3-d]pyrimidine (10) starting from diethyl malonate in five steps with the total yields of 45.8% and 44.8% was disclosed in the presented paper. The chemical structures of the objective products as well as the related intermediates were confirmed.

Abstract:A convenient two-step, one-pot synthesis of 4-chloro-2-(trichloromethyl)pyrimidines starting from 2-(trichloromethyl)-1,3-diazabutadienes is described. These nitrogen heterocycles were prepared by a sequential acylation/intramolecular cyclization reaction between 2-(trichloromethyl)-1,3-diazabutadienes and acyl chlorides in the presence of triethylamine followed by treatment with POCl3. This is the first report for the synthesis of this type of 4-chloro-2-(trichloromethyl)pyrimidine derivatives and serves as a source for a wide variety of other substituted pyrimidines by nucleophilic substitution reactions.

Abstract:Herein, synthesis of a new series of pyrano[2,3- d ]pyrimidine scaffolds is presented. Ethyl 6-amino-4-(4-chlorophenyl)-5-cyano-2-methyl-4 H -pyran-3-carboxylate has been reacted with formamide, formic acid, urea, thiourea, semicarbazide, and thiosemicarbazide to give six new pyrano[2,3- d ]pyrimidine derivatives in moderate to high yields (48–78%). Acylation and intramolecular cyclization of pyran with carbon disulfide, ethyl cyanoacetate, acetic anhydride, or chloroacetyl chloride has resulted in formation of the corresponding desired bicyclic pyrimidine derivatives. The chloromethyl derivative was subjected to nucleophilic substitution by its reaction with morpholine, piperidine or aniline resulting in the corresponding 2-substituted pyrimidine derivatives in good yields.

Abstract:8-Bromo-7-chloro[1,2,4]triazolo[4,3-c]pyrimidines were prepared by bromine-mediated oxidative cyclization of the aldehyde-derived hydrazones. The structure of one such product was unambiguously confirmed by single-crystal X-ray analysis. While the C-5 unsubstituted 1,2,4-triazolo[4,3-c]pyrimidine chemotype is extremely susceptible to ring isomerization at ambient conditions, the C-5-substituted analogues were found to be quite stable, permitting isolation in pure form. Nevertheless, they can still be converted into the 1,2,4-triazolo[1,5-c]pyrimidines by base-or acid-promoted Dimroth rearrangement. The presence of halogen functionalities on the pyrimidine nucleus renders the products useful as versatile synthetic intermediates for the easy diversification, as evidenced by palladium-catalyzed Kumada cross-couplings and Buchwald-Hartwig amination, as well as direct aromatic substitution.

Abstract:Reactions of easily accessible chloropyrimidinyl hydrazones with bromine were investigated. Oxidative cyclization followed by concomitant bromination led to the formation of [1,2,4]triazolo[4,3-c]pyrimidines, which then underwent the Dimroth rearrangement to afford highly functionalized 2-substituted [1,2,4]triazolo[1,5-c]pyrimidines.

Abstract:Previously synthesized ethyl 2-benzamido-4-(1,4-benzodioxan-2-yl)thiophene-3-carboxylates were cyclized to 2-aryl-3-aminothieno[2,3- d ]pyrimidin-4(1 H )-ones by the action of hydrazine hydrate. The condensation of ethyl 2-amino-4-(1,4-benzodioxan-2-yl)thiophene-3-carboxylate with chloroacetyl chloride gave the corresponding N-substituted 2-chloroacetamide which reacted with piperidine and morpholine to produce 2-aminoacetamides, and the latter were also subjected to cyclization with hydrazine hydrate to obtain 3-aminothienopyrimidin-4-one derivatives. The synthesized compounds were tested for antihypoxic activity.

Abstract:Canadian Journal of Chemistry, Ahead of Print. We report the synthesis of a series of substituted 2-hydroxy-pyrido[1,2-a]pyrimidin-4-ones through a condensation of 2-aminopyridines and diethyl malonate. This method is contrasted with three reported general procedures for the preparation of these compounds, revealing that simple, "neat" synthesis conditions are suitable for a number of derivatives, including halogenated compounds suitable for further functionalization. Environmental, safety, and economic factors were considered in exploring this effective and robust synthetic method.
