Abstract:The venerable Friedel-Crafts reaction appends alkyl or acyl groups to aromatic rings through alkyl or acyl cation equivalents typically generated by Lewis acids. We show that phenyl cation equivalents, generated from otherwise unreactive aryl fluorides, allow extension of the Friedel-Crafts reaction to intramolecular aryl couplings. The enabling feature of this reaction is the exchange of carbon-fluorine for silicon-fluorine bond enthalpies; the reaction is activated by an intermediate silyl cation. Catalytic quantities of protons or silyl cations paired with weakly coordinating carborane counterions initiate the reactions, after which proton transfer in the final aromatization step regenerates the active silyl cation species by protodesilylation of a quaternary silane. The methodology allows the high-yield formation of a range of tailored polycyclic aromatic hydrocarbons and graphene fragments.

Abstract:All-cis-multifluorinated cycloalkanes exhibit intriguing electronic properties. In particular, they display extremely high dipole moments perpendicular to the aliphatic ring, making them highly desired motifs in material science. Very few such motifs have been prepared, as their syntheses require multistep sequences from diastereoselectively prefunctionalized precursors. Herein we report a synthetic strategy to access these valuable materials via the rhodium-cyclic (alkyl)(amino)carbene (CAAC)-catalyzed hydrogenation of readily available fluorinated arenes in hexane. This route enables the scalable single-step preparation of an abundance of multisubstituted and multifluorinated cycloalkanes, including all-cis-1,2,3,4,5,6-hexafluorocyclohexane as well as cis-configured fluorinated aliphatic heterocycles.

Abstract:Difluoroenoxysilane, a commonly used difluoroallylating reagent, has attracted considerable attention in recent years. However, its application in the annulation reaction for the construction of fluorinated heterocyclic compounds remains relatively limited. Presented here is the Bronsted acid-catalyzed efficient formal [4 + 2] annulation of difluoroenoxysilanes with alpha-cyano chalcones. The developed protocol demonstrates tolerance to various substituents under mild reaction conditions, providing a reliable approach to construct gem-difluoro-3,4-dihydro-2H-pyrans in good to excellent yields with high diastereoselectivities.

Abstract:Sulfur functional groups are ubiquitous in molecules used in the pharmaceutical and agrochemical industries, and within these collections sulfones hold a prominent position. The double aza-analogues of sulfones, sulfondiimines, offer significant potential in discovery chemistry but to date their applications have been limited by the lack of convenient synthetic routes. The existing methods mainly rely on imination of low-valent-sulfur intermediates, or the combination of pre-formed organometallic reagents and electrophilic S(VI)-functionalities. Herein, we describe a Friedel-Crafts-type reaction of sulfondiimidoyl fluorides with (hetero)aryls. This new SuFEx reactivity benefits from broad functional group tolerance, mild reaction conditions, and does not require the use of pre-formed organometallic reagents. The efficient use of unprotected indoles and pyrroles, as well as furan, thiophene and carbocyclic aromatics, further demonstrates the advantages of these reactions. We show that the reactivity of the sulfondiimidoyl fluorides can be tuned by switching the N-substituents, allowing an expansion of the range of coupling partners. The utility of the transformation is exemplified by the synthesis of the sulfondiimine analogue of the HIV-I reverse transcriptase-inhibitor L-737,126.

Abstract:Since the initial report, the Friedel–Crafts reaction has become a powerful tool to functionalize arenes. Nevertheless, the use of nitrogen heterocycles as electrophiles in Friedel‐Crafts reactions has been less explored. Here, we show a Friedel–Crafts‐like reaction of electron‐rich arenes with quinazolin‐4(3H)‐ones, enabling late‐stage C2–H arylation of quinazolin‐4(3H)‐ones via triflic anhydride (Tf2O) activation. A series of substrates can be efficiently coupled under mild reaction conditions, affording C(sp3)‐C(sp2) coupling product 2‐aryl dihydroquinazolinones that can be further converted into the corresponding quinazolinone in the presence of base. This methodology offers efficient access to 2‐aryl quinazolin‐4(3H)‐ones and exhibits good functional group compatibility and site selectivity. Mechanistic investigations reveal the formation of highly electrophilic iminium intermediates upon Tf2O activation of quinazolin‐4(3H)‐ones, which serve as the key reactive species, enabling the Friedel‐Crafts reaction to proceed efficiently.

Abstract:Palladium-catalyzed CF bond formation via directed CH activation has recently emerged as a significant method to introduce fluorine atoms into organic molecules in an atom- and step-economical path. At the interface between fluorine chemistry and synthetic organic chemistry, the development of direct fluorinations of the CH bond remains sluggish, due to the major challenge of reductive elimination to afford the CF bond. One strategy is to oxidize palladium(II) species to more electron-deficient palladium(IV or III) fluorides to promote the CF bond forming reductive elimination, which opens up palladium-catalyzed CH fluorination as a powerful transformation. This focused review gives a brief summary of the mere handful of examples, including C(sp(2))F and C(sp(3))F bond formations, in this field so far.
Defluorinative C−O Coupling between Trifluoromethylarenes and Alcohols via Copper Photoredox Catalysis

Abstract:Fluorine-containing compounds have shown unparalleled impacts in the realm of functional molecules, and the ability to prepare novel structures has been crucial in unlocking new properties for pharmaceutical and materials science. Herein, we report a copper-catalyzed, photoinduced defluorinative C‒O coupling between trifluoromethylarenes and alcohols. This method allows for direct access to a wide selection of difluorobenzylether (ArCF2OR) molecules, including a com-pound displaying liquid crystal behavior. Through slight modification of the protocol, we were able to generate ArCF2I products, another class of synthetically useful fluorine-bearing molecules. Comprehensive mechanistic investigations first suggested ArCF2I as a reservoir to steadily supply the key ArCF2• radical species. Furthermore, experimental evi-dence supported a mechanism consisting of two collaborative cycles: C‒F activation operated by a homoleptic Cu(I) co-ordinated by two bisphosphine ligands as the photocatalyst and C‒O coupling promoted by a Cu(I) ligated by a single bisphosphine ligand. The critical roles of the two salt additives, lithium iodide and zinc acetate, in orchestrating the two cycles were also elucidated. This dual-role copper catalyst demonstrates the power of base metal photoredox catalysis in achieving both substrate activation and chemical bond formation via a single catalytic system.

Abstract:The construction of C(sp 3 )–C(sp 3 ) bonds is pivotal in organic synthesis; however, traditional methods involving alkyl halides are often limited by substrate tolerance and bond dissociation energies, particularly with alkyl fluorides. Herein, we report a silylboronate-mediated cross-coupling strategy that circumvents these challenges, enabling the efficient formation of C(sp 3 )–C(sp 3 ) bonds between alkyl fluorides and aryl alkanes under mild conditions. Various alkyl fluorides have also been effectively utilized, demonstrating the versatility and broad applicability of this approach. The use of diglyme is critical for this transformation which encapsulates potassium cations and enhances the reaction efficiency. Conventional alkyl halides, including chlorides, bromides, and iodides, are also suitable for this transformation. Density functional theory (DFT) calculations were conducted on the silylboronate-mediated coupling reactions for the first time. Interestingly, while experimental results suggest a radical mechanism, DFT calculations indicate a preference for an ionic pathway.

Abstract:Cross‐coupling reactions for constructing C‐N bonds represent a pivotal advancement in chemical science. Traditional methodologies, including nucleophilic aromatic substitution (SNAr) and transition metal‐catalyzed cross‐couplings, have limitations concerning aryl scope, reliance on toxic and costly transition‐metal catalysts, and issues related to atom economy and waste generation from ligands and additives. In this work, we introduce a novel method for aminating neutral, electron‐rich, and electron‐deficient aryl halides, eliminating the need for transition metals. Our approach involves the activation of aryl halides using solvated electrons generated from granulated lithium and sonication. This serves as a sustainable source of reducing power, facilitating the efficient formation of C‐N bonds under near ambient conditions. Competitive selectivity studies between halide and ester functionalities were explored. Reaction scope and conducted mechanistic studies support the proposed radical‐nucleophilic substitution (SRN1) mechanism for the reaction. Notably, the developed reaction has a highly competitive reductive dehalogenation pathway during the C‐N coupling reaction, and this mechanistic divergency was thoroughly explored. This work not only broadens the scope of C‐N coupling reactions which typically employs aryl bromides and iodides and rarely aryl fluorides which is also equally abundant, but also introduces a new way to do C‐N coupling reactions using solvated electrons.

Abstract:A metal‐free one‐pot synthesis route has been documented for the production of 1,1‐unsymmetrical diaryl alkanes via reductive Friedel−Crafts alkylation of arenes with carbonyl compounds in 1,1,1,3,3,3,‐hexafluoroisopropanol (HFIP) solvent. This approach exhibits a wide substrate scope accommodating diverse functionalities. The transformation of carbonyl compounds into alkylated products is facilitated by dimethylchlorosilane (Me2SiClH) reduction, succeeded via nucleophilic attack of arenes. The established protocol demonstrates a promising strategy for converting indoline‐2,3‐dione into α‐aryl oxindoles with high yields. The reaction involves reduction with Me2SiClH, followed by the attack of arenes in the presence of TfOH catalyst. The devised protocol offers scalability and remarkable tolerance towards various functional groups. Control experiments provide insights into the reaction mechanism, indicating an ionic pathway and highlighting the crucial roles of HFIP and Me2SiClH. Furthermore, the method is adaptable for late‐stage functionalization of numerous natural products and pharmaceuticals such as Sesamol, Thymol, Paracetamol, Menthol, and Nerol. Significantly, the technique has proven effective in the efficient synthesis of the anticoagulant drug Phenprocoumon and the rodent control agent Coumatetralyl.
