Abstract:Under mild conditions, a series of alpha,alpha-difluoro-beta-ketoamides were synthesized with Selectfluor (R) as the F+ source in the presence of K2CO3 in the mixed-solvent of water and PEG-400 (V-H2O : VPEG-400 = 3 : 1), without additional phase-transfer catalyst. Abstract:Electrochemical 1,2-hydroxydifluoromethylation and C-H difluoromethylation of acrylamides were developed by using CF(2)HSO(2)NHNHBoc as the source of the CF2H group. These electricity-powered oxidative alkene functionalization reactions do not need transition-metal catalysts or chemical oxidants. The reaction outcome, 1,2-difuntionalization or C-H functionalization, is determined by the substituents on the amide nitrogen atom of the acrylamides instead of by the reaction conditions.

Abstract:Given the profound effect of fluorine‐containing groups in improving the chemical, physical, and biological properties of the parent compound, the fluorine‐containing groups were receiving increasing attention in the medicinal, pharmaceutical, agricultural, and material sciences. Unlike well‐developed trifluoromethylation and difluoromethylation reactions, the efficient and mild methods for rapid introduction of CF2Me are still underdeveloped, despite their important applications in pharmaceuticals and agrochemicals. This concept describes recent progress in difluoroethylation functionalization.

Abstract:Difluoroethyl products have a wide range of applications in the fields of biomedicine and materials, and it is important to develop new method to synthesize difluoroethyl compounds. Herein, a palladium-catalyzed chlorodifluoroethylation was disclosed via ortho-C-H activation of acetanilide, using chlorodifluoroethyl(mesityl) hypervalent iodine reagent as the source of CH2CF2Cl. Moreover, adding organic base 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) to the above system, the difluorovinylated acetanilide products were obtained via an elimination reaction.

Abstract:The first example of decarboxylative difluoroacetamidation of α,β-unsaturated carboxylic acids by using difluoromethyl-substituted carbonyl compounds was disclosed. The procedure, which was mediated by low-cost and benign CuSO4, furnished a broad range of difluorinated alkenes in remarkable yields with only the E configuration in most of the cases. Moreover, the product could be smoothly transformed to the corresponding difluorofunctionalized ester and alcohol in high yields.

Abstract:N -(Difluoropropenyl)amides/amines are an important class of fluorinated compounds. Here, we report an efficient method for synthesizing these compounds without the use of transition metals. Under simple base-promoted conditions, 3-bromo-3,3-difluoroprop-1-ene reacts with N -methylanilines or N -arylacrylamides, with the elimination of one molecule of HBr, to give the target compound. Another efficient method for synthesizing difluoroalkenes is the reaction of 2-bromo-3,3,3-trifluoroprop-1-ene with indole or its analogues. Abstract:Herein, we report a simple and efficient method for the direct installation of chlorodifluoroethyl group onto aromatic molecules of various aromatic amides with a new 2-chloro,2,2-difluoroethyl(mesityl)iodonium salt (CDFI). Moreover, the chlorodifluoroethyl compounds could be smoothly converted into difluorovinyl compounds in a one-pot or discrete procedure and regarded as a steady source of difluorovinyl compounds with "HCl-mask".

Abstract:Difluoroacrylamides/amines are an important class of fluorinated compounds. Here, we report an efficient synthesis method for difluoroacrylamides without the use of transition metals. Under simple base promoted conditions, 3-bromo-3,3-difluoroprop-1-ene can react with aniline or mono-substituted aniline, removing one molecule of HBr to obtain the target compound. The substituent can be methyl or acryloyl. Another efficient method for synthesizing difluoroalkenes is the reaction of 2-bromo-3,3,3-trifluoroprop-1-ene with indole, which can also yield difluoroalkenes under standard conditions.

Abstract:Deuterated mono-and difluoroalkyl groups have attracted increasing attention in medicinal and biological studies.Controllable snipping one or two fluorine atoms from widely accessible trifluoromethyl groups followed by deuteration is an attractive method to access these molecules, whereas this has been hindered by the challenges associated with the chemoselectivity control during the defluoriantion process. Herein, we report a strategy for chemoselective deuterodefluorination reaction of trifluoro-and difluoroalkylacetamides via spin-center shifts. The reaction starts with the attack of a deuterated 4-dimethylaminopyridine（DMAP）-boryl radical（DMAP-BD 2 ·） to the amide oxygen atom, followed by a spin-center shift process to trigger the C–F bond scission. The resulting α, α-difluorocarbonyl radicals undergo deuterium atom transfer to afford the CF 2 D-products in high yields and high levels of D-incorporation. Notably, increasing the amount of DMAP-BD 3 enables the selective cleavage of the two C-F bonds, leading to CFD 2 -products in good yields meanwhile maintaining good levels of D-incorporation.

Abstract:Acrylamides are a class of important organic compounds that play key roles in biomedical chemistry, materials science, life science, and other fields. Due to the strong electron‐withdrawing effect of trifluoromethyl, the synthesis of trifluoromethylated acrylamides, especially those containing double bonds, has long been a challenge. Here, we report an example of the formation of trifluoromethyl acrylamides through palladium catalyzedcarbonylation of fluorinated olefins. By optimizing the reaction conditions, we can obtain different amide compounds, including alkyl acrylamides. aromatic acrylamides and heterocyclic acrylamides. This method has good substrate adaptability and functional group tolerance,so it is expected to be used in the synthesis of key amides in the fields of medicine and other fields.

Abstract:A facile silver-catalyzed decarboxylative radical addition/cyclization reaction of alpha,alpha-difluoroarylacetic acids and acrylamides has been disclosed. The method provides a highly attractive approach to synthesize a series of difluorinated oxindoles that contain various functional groups in moderate to good yields under mild conditions. Moreover, experimental studies reveal that the CF2 group of the ,-difluoroarylacetic acids plays a vital role in the transformation.

Abstract:A new radical difluoromethylation was developed by using inexpensive and readily available difluoroacetic anhydride and N-phenyl-4-methylbenzenesulfonamide for the first time. The reaction of arylboronic acids with the new difluoromethylation reagent, N-phenyl-N-tosyldifluoroacetamide, proceeded smoothly in the presence of palladium catalyst to provide difluoromethylarenes in satisfactory to excellent yields. The electronic property (electron-donating or electron-withdrawing) of the substituent linked to the aromatic ring did not considerably influence the reactivity of arylboronic acid. Various groups, including the synthetically useful functional groups Cl, CN, and NO2, were tolerated well under the current reaction conditions.
