Abstract:The venerable Friedel-Crafts reaction appends alkyl or acyl groups to aromatic rings through alkyl or acyl cation equivalents typically generated by Lewis acids. We show that phenyl cation equivalents, generated from otherwise unreactive aryl fluorides, allow extension of the Friedel-Crafts reaction to intramolecular aryl couplings. The enabling feature of this reaction is the exchange of carbon-fluorine for silicon-fluorine bond enthalpies; the reaction is activated by an intermediate silyl cation. Catalytic quantities of protons or silyl cations paired with weakly coordinating carborane counterions initiate the reactions, after which proton transfer in the final aromatization step regenerates the active silyl cation species by protodesilylation of a quaternary silane. The methodology allows the high-yield formation of a range of tailored polycyclic aromatic hydrocarbons and graphene fragments.

A series of carbonates and polyfluorobenzenes have been examined and found to give moderate to good yields. Abstract:Direct CAr-F arylation is effective and sustainable for synthesis of polyfluorobiaryls with different degrees of fluorination, which are important motifs in medical and material chemistry. However, with no aid of transition metals, the engagement of CAr-F bond activation has proved difficult. Herein, an unprecedented transition-metal-free strategy is reported for site-selective CAr-F arylation of polyfluoroarenes with simple (het)arenes. By merging N,N-bis(2,6-diisopropylphenyl)perylene-3,4,9,10-bis(dicarboximide)-catalyzed electrophotocatalytic reduction and anodic nitroxyl radical oxidation in an electrophotocatalytic cell, various polyfluoroaromatics (2F-6F and 8F), especially inactive partially fluorinated aromatics, undergo sacrificial-reagents-free C-F bond arylation with high regioselectivity, and the yields are comparable to those for reported transition-metal catalysis. This atom- and step-economic protocol features a paired electrocatalysis with organic mediators in both cathodic and anodic processes. The broad substrate scope and good functional-group compatibility highlight the merits of this operationally simple strategy. Moreover, the easy gram-scale synthesis and late-stage functionalization collectively advocate for the practical value, which would promote the vigorous development of fluorine chemistry.

The reaction employs aryltrifluoroborates as the arylation reagent, leading to the corresponding products in moderate to good yield with broad substrate scope. Abstract:Difluoroenoxysilane, a commonly used difluoroallylating reagent, has attracted considerable attention in recent years. However, its application in the annulation reaction for the construction of fluorinated heterocyclic compounds remains relatively limited. Presented here is the Bronsted acid-catalyzed efficient formal [4 + 2] annulation of difluoroenoxysilanes with alpha-cyano chalcones. The developed protocol demonstrates tolerance to various substituents under mild reaction conditions, providing a reliable approach to construct gem-difluoro-3,4-dihydro-2H-pyrans in good to excellent yields with high diastereoselectivities.

Abstract:A cooperative Pd/Cu-catalyzed dual C-H bond activation of electron-deficient polyfluoroarenes with aryl halides is developed. This protocol provides a unique and strategically distinct access to a variety of polyfluoroarene-substituted benzofuran derivatives. Abstract:The transition-metal-catalyzed α-arylation of carbonyl compounds is a widely practiced method for C-C bond formation. Several enantioselective versions of this process have been reported, but intermolecular, enantioselective coupling reactions of aryl electrophiles with α-fluoro carbonyl compounds have yet to be disclosed. We report enantioselective coupling of aryl and heteroaryl bromides and triflates with α-fluoroindanones catalyzed by palladium complexes of a BINOL-derived monophosphine and Segphos, respectively. The enolates were generated directly from α-fluoroindanones in the presence of potassium phosphate base during the reactions. We also report that reactions of α-fluorotetralones occur in high yields and enantioselectivities when conducted with enolates generated by elimination of trifluoroacetate from trifluoromethyl β-diketone hydrates. These reactions were catalyzed by palladium complexes of the commercially available bisphosphine Difluorphos. Thus, the formation of enantioenriched α-aryl-α-fluoroketones can be readily achieved by C-C bond formation when the appropriate palladium catalyst and α-fluoro enolate precursor were used.

Abstract:A nickel-catalyzed reductive cross-coupling reactions between polyfluoroarenes and alkyl electrophiles is reported to access substituted fluoroarenes through chelation-assisted C–F activation. Diverse primary and secondary alkyl（pseudo）halides can be employed to couple with polyfluoroarenes, showing excellent regioselectivity. Furthermore, the nickel-catalyzed asymmetric cross-coupling of polyfluoroarenes with racemic alkyl halides is preliminarily explored. In addition, the practicability of the title transformation is also demonstrated by total synthesis of losmapimod and an analog as key steps. The developed method exhibits many advantages, including economic catalytic systems, commercially available alkyl electrophiles, and lack of sensitive organometallic reagents.

Abstract:A palladium-catalyzed cross-coupling of gem-difluorinated cyclopropanes with boronic acids, providing the corresponding arylated/alkenylated/alkylated 2-fluoroallylic scaffolds, is generated. This new approach has good functional group compatibility for both gem-difluorinated cyclopropanes and boronic acids; thus, an array of synthetic building blocks of monofluoroalkene scaffolds including conjugated fluorodiene and skipped fluorodiene gave good yields with high Z-selectivity. Moreover, proficient application was described for monofluoroalkene, whereas the corresponding alkyl fluoride was constructed through hydrogenation.

Abstract:Palladium-catalyzed CF bond formation via directed CH activation has recently emerged as a significant method to introduce fluorine atoms into organic molecules in an atom- and step-economical path. At the interface between fluorine chemistry and synthetic organic chemistry, the development of direct fluorinations of the CH bond remains sluggish, due to the major challenge of reductive elimination to afford the CF bond. One strategy is to oxidize palladium(II) species to more electron-deficient palladium(IV or III) fluorides to promote the CF bond forming reductive elimination, which opens up palladium-catalyzed CH fluorination as a powerful transformation. This focused review gives a brief summary of the mere handful of examples, including C(sp(2))F and C(sp(3))F bond formations, in this field so far.

Abstract:Efficient palladium-catalyzed regioselective vinylic C–H polyfluoroarylation of gem-disubstituted ethylenes with polyfluoroarenes was realized to access a variety of polyfluorinated triarylethenes. An aryl to vinyl 1,4-palladium migration is proposed to achieve the high regio- and stereoselectivities for the target products. This strategy features broad substrate scopes and good functional group tolerance. Mechanistic studies have suggested that a protonation-deprotonation process reversibly occurs between a five-membered palladacycle and the vinyl-palladium intermediates, and aromatic C–H cleavage of polyfluoroarenes is contributed to the rate-limiting step in the overall catalytic cycle.

Abstract:Herein we report the first versatile and expeditious method for the site-selective C-H fluoromethylation of aryl iodides via Pd/norbornene cooperative catalysis, which could work as a robust toolbox for the diversity-oriented synthesis (DOS) of fluoromethylated arenes. This methodology features the use of the low-cost industrial raw material CH2IF as the fluoromethyl source, an excellent functional group tolerance, and a broad ipso termination scope and can be expanded to the late-stage modification of biorelevant molecules.

This reaction uses readily available starting materials and is operationally simple, thus representing a practical method for the construction of C(sp(2))C(sp(3)) bonds with polyfluoroarenes through direct CH bond functionalization. Abstract:A palladium-catalyzed fluorosulfonylvinylation reaction of organic iodides is described. Catalytic Pd(OAc)2 with a stoichiometric amount of silver(I) trifluoroacetate enables the coupling process between either an (hetero)aryl or alkenyl iodide with ethenesulfonyl fluoride (ESF). The method is demonstrated in the successful syntheses of eighty-eight otherwise difficult to access compounds, in up to 99 % yields, including the unprecedented 2-heteroarylethenesulfonyl fluorides and 1,3-dienylsulfonyl fluorides.
