Abstract:Reaction of (S)- or (R)-3-aminoquinuclidine with 2-chloropyrimidine or 2-bromopyrimidine led to an unexpected formation of both cis- and trans-octahydropyrrolo [2,3]pyridine derivatives. A single-step synthesis of two of the four stereoisomers of these octahydropyrrolo[2,3]pyridine derivatives provides a convenient way of generating stereochemically defined isomers. Optimization of reaction conditions was carried out by (1)H NMR monitoring. The relative and absolute stereochemistry of all four stereoisomers was determined by a combination of (1)H, (13)C, and (15)N NMR spectroscopy and vibrational circular dichroism spectroscopy.

Abstract:A three-step liquid-phase protocol for the synthesis of furanoquinolines and pyranoquinolines is described. The one-pot three-component aza-Diels-Alder reaction of PEG-supported benzaldehyde, anilines and 2,3-dihydrofuran or dihydropyran under TFA gave PEG-supported tetrahydroquinolines, which was assuredly oxidized by DDQ and then cleaved from the support to afford the corresponding furanoquinolines and pyranoquinolines with reasonable yields and purity.

Abstract:The Winterfeldt oxidation (NaOH, DMF, air, rt) of substituted 1,2,3,4-tetrahydro-gamma-carbolines has been developed, which provides a convenient and efficient method for the synthesis of the corresponding dihydropyrrolo[3,2-b]quinolones in moderate to excellent yields (38-94%). The generality and substrate scope of this reaction are explored and a possible mechanism is proposed. The results imply that electron-withdrawing groups on N-2 of tetrahydro-gamma-carbolines and N-5-H are necessary. The synthesis of 5 or 7-substituted pyrrolo[3,2-b]quinolones in near quantitative yields was also achieved through deprotection and aromatization of N-1-Boc-dihydropyrrolo[3,2-b]quinolones.

Abstract:A series of 1,3-cyclodiketone- and tetrahydroepoxyisoindole-fused β-enamino dicarbonyl heterocycles were synthesized via a 1,4-diazabicyclo[2.2.2]octane-catalyzed, CH3NO2-mediated three-component reaction of 1,3-cyclodiketone, furfural, and allylamine in toluene. The target compounds were generated via the formation of β-enamino diketone as a key intermediate, followed by intramolecular Diels-Alder cycloaddition. The prepared molecules bearing a quinoline-2,4-dione moiety could be further brominated with N-bromosuccinimide and diastereoselectively reduced by NaBH4 to afford pyrano[3,2-c]quinolin-5-one-derived heterocycles with six vicinal stereogenic centers.

Abstract:A facile one-pot, four-component reaction for the synthesis of pyrrolo[1,2-a]quinolines is described. Knoevenagel condensation reaction of malononitrile and aromatic aldehydes gave 2-arylmethylidenemalononitriles which on treatment with quinoline and cyclohexyl isocyanide under solvent-free conditions afforded 1-(cyclohexylamino)-2-arylpyrrolo[1,2-a]quinoline-3-carbonitriles in good yields.

Abstract:A strategy for the syntheses of benzofuranoquinolines and its analogues via the irradiation of o-phenylfuranyl/thienylpyridines/pyrimidines in DCM with UV light at rt under an argon atmosphere is described. The mechanism of this reaction through the process of 6π-electrocyclization, [1,5]-hydrogen shift, and 1,3-eneamine tautomerism leading to H2 evolution was elucidated. Notably, the syntheses of cis-8b-methyl-8b,13a-dihydrobenzo[f]benzofuro[3,2-h]quinolone via the photoinduced rearrangement of 2-(3-methylbenzofuran-2-yl)-3-phenylpyridine relevant to the mechanism of this reaction highlights the importance of the developed methodology.

Abstract:The synthesis of bis(pyrrolo[2,1- a ]isoquinolinyl)methanes was achieved through TfOH-catalyzed sequential Friedel–Crafts alkylation of pyrrolo[2,1- a ]isoquinoline and aldehyde. A series of highly functionalized bis(pyrrolo[2,1- a ]isoquinolinyl)methane derivatives can be obtained in acceptable to excellent yields (11 examples, up to 96% yield). Interestingly, deformylation was observed when treating pyrrolo[2,1- a ]isoquinoline-derived aldehyde and indole under the current reaction conditions. Furthermore, the replacement of aldehyde with isatin resulted in the formation of methylene-bridged dimeric pyrrolo[2,1- a ]isoquinoline. Abstract:A concise synthesis of 8, 9-dihydo-2H-pyrano-[5,6-g] quinolin-2-one derivatives was reported, using 3-amino-phenol as starting material. Treatment of 3-amino-phenol with ketones bearing alpha C - H at the presence of a catalyst PTS .H2O affords compound 2a similar to 2d, followed by treatment with ethyl acetoacetate, gives target compound 3a similar to 3d with regioselectivity.

Abstract:An efficient procedure for the synthesis of 7‐(aryl)‐8‐nitro‐2,3,6,7‐tetrahydroimidazo[1,2‐a]pyridinones, 8‐(aryl)‐9‐nitro‐3,4,7,8‐tetrahydropyridone[1,2‐a]pyrimidines and 9‐(aryl)‐10‐nitro‐2,3,4,5,8,9‐hexahydropyridone[1,2‐a]diazepine via one‐pot three component reaction of diamine, nitroketene dithioacetal (1,1‐bis(methylsulfanyl)‐2‐nitroethene), and coumarine‐3‐ carboxylic acid derivatives in EtOH under reflux conditions is reported. The advantages of this procedure are simplicity, easy purification, good yields, and catalyst‐free conditions. All products were confirmed by 1H‐ and 13C‐NMR, IR, MS, and X‐ray crystal structure analyses.
