Abstract:The power output and stability of enzyme-based biofuel cells (BFCs) is greatly dependent on the properties of both the biocathode and bioanode, which may be adapted for portable power production. In this paper, a novel highly uniform three-dimensional (3D) macroporous gold (MP-Au) film was prepared by heating the gold "supraspheres", which were synthesized by a bottom-up protein templating approach, and followed by modification of laccase on the MP-Au film by covalent immobilization. The as-prepared laccase/MP-Au biocathode exihibited an onset potential of 0.62 V versus saturated calomel electrode (SCE, or 0.86 V vs NHE, normal hydrogen electrode) toward O2 reduction and a high catalytic current of 0.61 mAcm(-2). On the other hand, mutated glucose dehydrogenase (GDH) surface displayed bacteria (GDH-bacteria) were used to improve the stability of the glucose oxidation at the bioanode. The as-assembled membraneless glucose/O2 fuel cell showed a high power output of 55.8 ± 2.0 μW cm(-2) and open circuit potential of 0.80 V, contributing to the improved electrocatalysis toward O2 reduction at the laccase/MP-Au biocathode. Moreover, the BFC retained 84% of its maximal power density even after continuous operation for 55 h because of the high stability of the bacterial surface displayed GDH mutant toward glucose oxidation. Our findings may be promising for the development of more efficient glucose BFC for portable battery or self-powered device applications.

Abstract:Enzymatic biofuel cells (EBFCs) provide a new strategy to enable direct biomass-to-electricity conversion, posing considerable demand on sequential enzymes. However, artificial blend of multi-enzyme systems often suffer biocatalytic inefficiency due to the rambling mixture of catalytic units. In an attempt to construct a highperformance starch/O-2 EBFC, herein we prepared a starch-oxidizing bioanode based on displaying a sequential enzyme system of glucoamylase (GA) and glucose dehydrogenase (GDH) on E.coli cell surfaces in a precise way using cohesin-dockerin interactions. The enzyme stoichiometry was optimized, with GA & GDH (3:1)-E.coli exhibiting the highest catalytic reaction rate. The bioanode employed polymerized methylene blue (polyMB) to collect electrons from the oxidation of NADH into NAD(+), which jointly oxidized starch together with co displayed GA and GDH. The bioanode was oxygen-insensitive, which can be combined with a laccase based biocathode, resulting in a membranless starch/O-2 EBFC in a non-compartmentalized configuration. The optimal EBFC exhibited an open-circuit voltage (OCV) of 0.74 V, a maximum power density of 30.1 +/- 2.8 mu W cm(-2), and good operational stability.
Controllable Display of Sequential Enzymes on Yeast Surface with Enhanced Biocatalytic Activity Toward Efficient Enzymatic Biofuel Cells.

Abstract:A precisely localized enzyme cascade was constructed by integrating two sequential enzymes, glucoamylase (GA) and glucose oxidase (GOx), on a yeast cell surface through an a-agglutinin receptor as the anchoring motif with cohesin-dockerin interaction. The overall catalytic activities of the combinant strains were significantly dependent on the assembly method, enzyme molecular size, enzyme order, and enzyme stoichiometry. The combinant strain with GA-DocC initially bound scaffoldin prior to GOx-DocT exhibited a higher overall reaction rate. The highest overall reaction rate (29.28 +/- 1.15 nmol H2O2 min(-1) mL(-1)) was achieved when GA/ GOx ratio was 2:1 with enzyme order: yeast-GA-GOx-GA, 4-fold enhancement compared to free enzyme mixture. Further, the first example of starch/O-2 enzymatic biofuel cells (EBFCs) using codisplayed GA/GOx based bioanodes were assembled, demonstrating excellent direct biomass-to-electricity conversion. The optimized EBFC registered an open-circuit voltage of 0.78 V and maximum power density (P-max) of 36.1 +/- 2.5 mu W cm(-2), significantly higher than the P-max for other starch/O-2 EBFCs reported so far. Therefore, this work highlights rational organization of sequential enzymes for enhanced biocatalytic activity and stability, which would find applications in biocatalysis, enzymatic biofuel cells, biosensing, and bioelectro-synthesis.

Abstract:In this work, a bacterial surface displaying enzyme based two-compartment biofuel cell for the direct electrical energy conversion from degradation products of lignocellulosic biomass is reported. Considering that the main degradation products of the lignocellulose are glucose and xylose, xylose dehydrogenase (XDH) displayed bacteria (XDH-bacteria) and glucose dehydrogenase (GDH) displayed bacteria (GDH-bacteria) were used as anode catalysts in anode chamber with methylene blue as electron transfer mediator. While the cathode chamber was constructed with laccase/multi-walled-carbon nanotube/glas sy-carbon-electrode. XDH-bacteria exhibited 1.75 times higher catalytic efficiency than GDH-bacteria. This assembled enzymatic fuel cell exhibited a high open-circuit potential of 0.80 V, acceptable stability and energy conversion efficiency. Moreover, the maximum power density of the cell could reach 53 mu Wcm(-2) when fueled with degradation products of corn stalk. Thus, this finding holds great potential to directly convert degradation products of biomass into electrical energy. (C) 2015 Elsevier Ltd. All rights reserved.

Abstract:Abstract Presently, one of the most important aspects for the development of enzymatic biofuel cells (EBFCs) is to synthesize the novel electrode materials that possess high current density, low open-circuit voltage (OCV) and long-term stability. To achieve the above attributes, lots of new strategies are being used by the researchers for the development of advanced materials. Nowadays, nanomaterials and nanocomposites are the promising material that has been utilized as effective electrode material in solar cells, supercapacitors and biofuel cells application. Herein, we account for a novel electrocatalyst as electrode material that comprised ZnO nanoparticles decorated on the surface of polyindole (PIn)-multi-walled carbon nanotube (MWCNT), for the immobilization of glucose oxidase (GOx) enzyme and mediator (Ferritin). The PIn-MWCNT scaffold is prepared via in situ chemical oxidative polymerization of indole on the surface of MWCNT and assessed by myriad techniques. The micrograph of scanning electron microscopy (SEM) designated the interconnected morphology of MWCNTs in the polymer matrix. X-ray diffraction spectroscopy (XRD) and Fourier transform infrared spectroscopy (FTIR), confirm the crystallinity and different functional groups available in the synthesized material, respectively. The electrochemical assessment demonstrates that the ZnO/PIn-MWCNT/Frt/GOx nanobiocatalyst exhibits much higher electrocatalytic activity towards the oxidation of glucose with a maximum current density of 4.9 mA cm −2 by consuming 50 mM glucose concentration in phosphate buffer saline (PBS) (pH 7.4) as the testing solution by applying 100 mVs −1 scan rates. The outcomes reflect that the as-prepared ZnO/PIn-MWCNTs/Frt/GOx biocomposite is a promising bioanode for the development of EBFCs.

Abstract:Enzymatic biofuel cells (EBFCs) utilize enzymes to convert chemical energy present in renewable biofuels into electrical energy and have shown much promise in the continuous powering of implantable devices. Currently, however, EBFCs are greatly limited in terms of power and operational stability with a majority of reported improvements requiring the inclusion of potentially toxic and unstable electron transfer mediators or multicompartment systems separated by a semipermeable membrane resulting in complicated setups. We report on the development of a simple, membrane/mediator-free EBFC utilizing novel electrodes of graphene and single-wall carbon nanotube cogel. These cogel electrodes had large surface area (∼ 800 m(2) g(-1)) that enabled high enzyme loading, large porosity for unhindered glucose transport and moderate electrical conductivity (∼ 0.2 S cm(-1)) for efficient charge collection. Glucose oxidase and bilirubin oxidase were physically adsorbed onto these electrodes to form anodes and cathodes, respectively, and the EBFC produced power densities up to 0.19 mW cm(-2) that correlated to 0.65 mW mL(-1) or 140 mW g(-1) of GOX with an open circuit voltage of 0.61 V. Further, the electrodes were rejuvenated by a simple wash and reloading procedure. We postulate these porous and ultrahigh surface area electrodes will be useful for biosensing applications, and will allow reuse of EBFCs.
An "On-Off" Switchable Power Output of Enzymatic Biofuel Cell Controlled by Thermal-Sensitive Polymer

Abstract:A novel "ON-OFF" switchable enzymatic biofuel cell (EBFC), controlled by in situ thermal-stimuli signal, has been consciously designed. Poly (N-isopropylacrylamide) (PNIPAm) chains were used to act as "ON" and "OFF" channels. Consecutively switching of temperature below and above the lower critical solution temperature (LCST), the reversible conformation changing of the PNIPAm chains between superhydrophilicity and superhydrophobicity was achieved, which constructed the "ON" and "OFF" channel for the transfer of the electrochemical probe to the underlying electrode correspondingly. Gold nanoparticles (AuNPs) protected glucose oxidase and laccase were successfully entrapped into the intelligent thermal-sensitive PNIPAm chains, and performed as the catalysts for the oxidation of glucose and the reduction of O2, respectively. Below the LCST, the fuels and the mediator could access to the catalytic centers of enzymes (set as "ON" state); while above the LCST, the reaction was impeded because the process of reactant transmission was blocked (set as "OFF" state). By switching the "valve" of mass transfer, the fabricated EBFC displayed the obvious "ON-OFF" controllable behavior. At the "ON" state, the open circuit voltage (Ecell(ocv)) and maximal power output density (Pmax) could reach to 0.70 V and 20.52 μW cm(-2), respectively; while at the "OFF" state, the Ecell(ocv) and Pmax were only 0.30 V and 3.28 μW cm(-2) correspondingly. The switchable process was repeatable, and the response time was only several minutes.

Abstract:Lacking of electron flow pathway within enzymes, enzymatic biofuel cells (EBFCs) always suffer from low power and poor stability, which limits their further application. Aiming to improve enzyme's conductivity and stability, we constructed a conductive enzyme nanocapsule n(GOx-PPy). A self-encapsulation was triggered by adding glucose to initiate an in-situ growth of PPy network within and around the glucose oxidase (GOx). Conductive pathways, as well as protective shell, were formed between single GOx active site and PPy network, allowing enhanced electron transfer and enzymatic stability. The n(GOx-PPy)-based EBFC demonstrated 245-fold increased power output, providing a promising strategy for constructing high-performance EBFCs.
Layer-by-Layer Assembled Enzyme Cascade Bioanode for Catalyzing Oxidation of Sucrose

Abstract:We report here a novel layer-by-layer (LbL) assembled enzyme cascade bioanode incorporated with CNT-invertase and CNT-glucose dehydrogenase for sucrose oxidation in a biofuel cell. Methylene green (MG) was used as redox mediator for electrocatalytic oxidation of NADH at reduced overpotentials on screen printed electrode (SPE). The LbL architecture showed advantages for sequential enzymatic reaction that favored the efficient penetration of substrate and products in a cascade system. The maximum current density and power density achieved were 412 ± 36 μA/cm2 and 85±6μW/cm2.The high current density/power density obtained showed LbL assembly is of great advantage and can be employed as a simple and efficient way to construct enzyme cascade bioanode for biofuel cell applications.

Abstract:Graphene oxide (GO) was prepared according to a modified Hummer's method,and graphene oxide aqueous solution,obtained by ultrasonic exfoliation,was then reduced by hydrothermal method to produce three-dimensional graphene (GN).The results showed that the as-prepared three-dimensional graphene material possessed a porous structure and some oxygen-containing groups disappeared after reduction.A glycerol enzymatic biofuel cell (EBFC) was prepared using three-dimensional GN as the carrier,and Meldola's blue as the mediator;Nafion (NF) was used to immobilize glycerol kinase (GK) and glycerol-3-phosphate oxidase (GPO) to prepare the anode catalyst,and platinum on carbon was used as the cathode catalyst.The results showed that the glycerol EBFC based on the NF/GN/MB enzyme-modified carbon paper electrode had good discharge performance.The open circuit potential was 0.77 V and the maximum power density was 42.05 tW/cm2 at 0.42 V.The assembly of the glycerol EBFC and the working conditions were optimized.The performance was also evaluated using polarization curve and alternating current (AC) impedance methods.This EBFC will provide a clean and reliable energy reuse approach for the treatment of glycerol,a by-product ofbiodiesel in the future.
Pseudocapacitance behavior enables efficient and stable electrochemical energy conversion and storage in glucose/air enzymatic biofuel cells

Abstract:Constructing an integrated system with high-efficiency and stable energy conversion and storage (ECS) is of great significance but remains challenges. Herein, we report a biodevice consisting of a bioanode of tetrathiafulvalene/glucose oxidase (TTF/GOx) immobilized on asphalt-derived porous carbon (APC) for catalyzing glucose oxidation and a biocathode of 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate)/bilirubin oxidase (ABTS/BOD) immobilized on APC for catalyzing oxygen reduction , to achieve ECS with high-efficiency and excellent stability. The redox mediators , TTF and ABTS, used at the bioanode and biocathode, respectively, simultaneously mediate electron transfer to release electric current on discharging and serve as pseudocapacitors to achieve charge storage at open-circuit conditions, enabling high power density and self-charge capability. The biodevice of the above configuration delivers a peak power output of 5.01 mW cm −2 in the pulse discharge mode with cell voltage remaining 93.7 % of its initial value after 100 pulse discharge/self-charge cycles. This work provides a highly efficient solution to address the low power density and poor stability of the conventional enzymatic biofuel cells, showing great implications for small, implantable and wearable electronics.

Abstract:Here, an electron transfer enhanced redox mediator is presented that significantly enhances electron transfer between the host electrode and enzymes, as well as between adjacent enzymes, promoting seamless interfaces through multiple interactions. Biofuel cells (BFCs) offer an eco‐friendly route to convert biochemical energy into electricity. However, their performance is hindered by insufficient enzyme immobilization as well as limited electron transfer within the enzymatic electrode. While the incorporation of redox mediators (RMs) into enzyme layers has been shown to improve BFC performance through enhanced electron transfer, progress has plateaued in the last decade. Herein, a major breakthrough is presented realized by a novel strategy that exploits electron transfer‐intensified RM layers. Metal nanoparticles covalently bridged between neighboring RMs facilitate electron transfer ubiquitously. Electron transfer characteristics are enhanced not only within the RM layers themselves, but also at the glucose oxidase (GOx)/host electrode and GOx/GOx interfaces. This leads to a remarkable performance boost in the enzymatic anode. A hybrid BFC constructed with innovative anode and Pt‐based cathode exhibits a striking combination of high power output (2.3 and 8.5 mW cm−2 at 10 and 300 mmol L−1 glucose, respectively) and exceptional operational stability (≈80% and 47% power retention after 10 days and 1 month, respectively), outperforming all previously reported BFCs by a significant margin.

Abstract:Designing an artificial enzyme, from the perspective of bionics, to mimic the catalytic activity of natural enzymes is highly desirable but remains challenges. In response to the simulation of biological structure, we developed cytochrome c oxidase (CcO)-like single-atom nanozymes with FeN5 active centers (FeN5 SAs) in this work. Similar to the spatial structure of heme a3 in natural CcO, the active center of FeN5 SAs is axial N-coordinated heme-like structure and can be served as oxygen-binding site to complete oxygen reduction reaction (ORR) in respiratory electron transport chain by catalyzing the oxidation of cytochrome c (Cyt c). Depending on this bionic structure, furthermore, FeN5 SAs exhibited competitive electrocatalytic performance towards ORR with a half-wave potential of 0.67 V (vs. RHE) at neutral condition. Coupling a glucose dehydrogenase (GDH) bioanode, the FeN5 SAs-based glucose/O2 enzymatic biofuel cell (EBFC) obtained a maximum power density of 149.2 ± 4.0 μW cm-2 with an open circuit potential of 0.40 ± 0.01 V. In this study, inspired by the native structure of enzymes, we develop CcO-like FeN5 SAs and expand its application in EBFCs, which may provide an rational research approach to advance the development of nanozymes.

Abstract:Biofuel cells that can convert the chemical energy of biomass such as glucose into electricity are expected to continuously obtain energy from living organisms and solve bottlenecks of energy supply for implanted electronics. However, the use of biofuel cells is limited mainly by the sharp drop in performance after implanting in living organisms caused by biofouling and electrode surface inactivation. Herein, a simple and effective strategy to overcome these problems by designing a porous antifouling interface on biofuel cells, is demonstrated. It resists the biofouling from body fluid while sustaining reactant penetration, and also enhances immobilization of enzymes. As a result, the biofuel cell can maintain almost 100% performance after implanting in living organisms, and a maximal output power of 76.6 mW cm-3 is achieved in vivo, which is ≈96 times of the highest performance reported to date. This strategy is universal and can be extended to the other electronic devices such as electrochemical sensors, which presents a new avenue for developing high-performance implanted electronics.

Abstract:Herein, we successfully construct the 3D biocompatible graphene through crosslinking 2D graphene nanosheet onto carbon fiber paper with poly(diallyldimethylammonium chloride) (PDDA) as anode of the alcohol biofuel cell. Compared with the bioanode without 3D graphene, the current density and output power of PDDA-graphene-ADH bioanode is increased by 23 % and 41 % at a high concentration of ethanol at pH 8.9, suggesting the stabilization role of graphene in enzyme loading. The study provides us a deep analysis on structures and performances of the bioanode incl. electrochemistry, X-ray photoelectron spectra, and atomic force microscopy images, which is significant to develop the new methods to construct 3D porous electrodes in energy conversion device.

Abstract:Enzyme immobilization is one of the key factors in constructing high-performance enzyme biosensors and biofuel cells (BFCs). Herein, we propose a new protocol for efficient immobilization of a glycoprotein enzyme based on the interaction of the 1, 2- or 1, 3-diols in the glycoprotein with a boronic acid functionalized monomer. Briefly, casting a mixture of glucose oxidase (GOx) and anilineboronic acid (ABA) followed by a NaAuCl4 solution to an Au-plated Au electrode surface yielded a GOx-poly(ABA) (PABA)-gold nanoparticle (Au-nano) bionanocomposite, and chitosan (CS) was then cast and air-dried. In the present protocol, the small-sized Au-nano or Au subnanostructures can form near/on the enzyme molecule, which greatly promotes the electron transfer of enzymatic reaction and enhances the amperometric responses. The thus-prepared CS/GOx-PABA-Au-nano/Au-plated Au electrode worked well in the first-/second generation biosensing modes and as a bioanode in a monopolar biofuel cell, with analytical or cell-power performance superior to those of most analogues hitherto reported. Abstract:This study demonstrated a novel nanographene platelets (NGPs)-based glucose/O2 biofuel cell (BFC) with the glucose oxidase (GOD) as the anodic biocatalysts and the laccase as the cathodic biocatalysts. The GOD/NGPs-modified electrode exhibited good catalytic activity towards glucose oxidation and the laccase/NGPs-modified electrode exhibited good catalytic activity towards O2 electroreduction. The maximum power density was ca. 57.8μWcm−2 for the assembled glucose/O2 NGPs-based BFC. These results indicated that the NGPs were very useful for the future development of novel carbon-based nanomaterials BFC device.

Abstract:In this paper, a method of carbon nanotube coated with the dendrimer encapsulated platinum nanoparticles (Pt-DENs/CNTs) was introduced, and the properties of enzyme electrode modified by the Pt-DENs/CNTs were investigated. The formation of the self-assembled (GOx/Pt-DENs)(n)/CNTs construction was expiored by high resolution transmission electron microscopy (HRTEM). And the results indicated that the uniform growth of the layer-by-layer nanostructures onto carboxyl-functionalized CNTs. The electrochemical properties of the enzyme anode modified by (GOx/Pt-DENs)(n)/CNTs, which act as biofuel cell anode, was studied by electrochemica measurements. Owing to its nanostructure, the (GOx/Pt-DENs)(n)/CNTs heterostructures have a large specific area, a high surface active center, good conductivity and macro tunnel effect, therefore the electrode modified by (GOx/Pt-DENs)(n)/CNTs has series of attractive electrochemical characteristics so as to enhance biofuel cell's function.
