Abstract:The power output and stability of enzyme-based biofuel cells (BFCs) is greatly dependent on the properties of both the biocathode and bioanode, which may be adapted for portable power production. In this paper, a novel highly uniform three-dimensional (3D) macroporous gold (MP-Au) film was prepared by heating the gold "supraspheres", which were synthesized by a bottom-up protein templating approach, and followed by modification of laccase on the MP-Au film by covalent immobilization. The as-prepared laccase/MP-Au biocathode exihibited an onset potential of 0.62 V versus saturated calomel electrode (SCE, or 0.86 V vs NHE, normal hydrogen electrode) toward O2 reduction and a high catalytic current of 0.61 mAcm(-2). On the other hand, mutated glucose dehydrogenase (GDH) surface displayed bacteria (GDH-bacteria) were used to improve the stability of the glucose oxidation at the bioanode. The as-assembled membraneless glucose/O2 fuel cell showed a high power output of 55.8 ± 2.0 μW cm(-2) and open circuit potential of 0.80 V, contributing to the improved electrocatalysis toward O2 reduction at the laccase/MP-Au biocathode. Moreover, the BFC retained 84% of its maximal power density even after continuous operation for 55 h because of the high stability of the bacterial surface displayed GDH mutant toward glucose oxidation. Our findings may be promising for the development of more efficient glucose BFC for portable battery or self-powered device applications.
A Membraneless Starch/O2 Biofuel Cell Based on Bacterial Surface Regulable Displayed Sequential Enzymes of Glucoamylase and Glucose Dehydrogenase

Abstract:Enzymatic biofuel cells (EBFCs) provide a new strategy to enable direct biomass-to-electricity conversion, posing considerable demand on sequential enzymes. However, artificial blend of multi-enzyme systems often suffer biocatalytic inefficiency due to the rambling mixture of catalytic units. In an attempt to construct a highperformance starch/O-2 EBFC, herein we prepared a starch-oxidizing bioanode based on displaying a sequential enzyme system of glucoamylase (GA) and glucose dehydrogenase (GDH) on E.coli cell surfaces in a precise way using cohesin-dockerin interactions. The enzyme stoichiometry was optimized, with GA & GDH (3:1)-E.coli exhibiting the highest catalytic reaction rate. The bioanode employed polymerized methylene blue (polyMB) to collect electrons from the oxidation of NADH into NAD(+), which jointly oxidized starch together with co displayed GA and GDH. The bioanode was oxygen-insensitive, which can be combined with a laccase based biocathode, resulting in a membranless starch/O-2 EBFC in a non-compartmentalized configuration. The optimal EBFC exhibited an open-circuit voltage (OCV) of 0.74 V, a maximum power density of 30.1 +/- 2.8 mu W cm(-2), and good operational stability.

Abstract:This review discusses different types of nano-gold hybrid materials used in electrodes for fuel cells and their application for the electrochemical reduction of oxygen, with the majority operating in alkaline media. The various synthesis methods for these materials are also described. Successful applications of these emerging hybrid materials with respect to parameters such as size and crystallographic orientations of nano-gold materials in electrodes, nano-gold composites on different substrates and electrode materials, nano-gold as binary catalytic materials, as well as dealloyed nanoporous gold electrocatalysts are highlighted. Gold nanoparticles (Au NPs) are also discussed in the context of bioengineering topics such as bio-sensors, bio-fuel cells, and doping of biological molecules like DNA with NPs. Furthermore, limitations of these applications in addition to future applications that could be explored in the field of nano-gold hybrid materials are mentioned. (C) 2016 Elsevier Ltd. Abstract:Enzymatic biofuel cells (EBFCs) are attractive power sources capable of utilizing biomolecules to generate electrical power. However, their practical utility is limited because of their relatively low power output and short lifetime, which arise from the low enzyme loading capacity on the support materials, activity loss during enzyme immobilization, intrinsically poor enzyme stability, and limited substrate and electron transport. Nanobiocatalytic approaches, in which enzymes are incorporated with nanostructured supports, have been recognized as a potent solution to resolve these limitations. They construct high-performance enzyme electrodes utilizing the affirmative characteristics of the materials coupled with appropriate immobilization strategies. In this review, the roles of nanostructured supports and catalytic mechanisms for generating biofuel-driven electrical power, the recent research progress on EBFCs based on widely investigated nanocarbons, nanoparticles, and polymeric nanofibers, and newly emerged metal-organic frameworks, nanoflowers, and other nanohybrids are discussed. The current challenges and prospects for utilizing nanostructured supports in EBFCs are also discussed.
A Hybrid Biofuel Cell with High Power and Operational Stability Using Electron Transfer‐Intensified Mediators and Multi‐Interaction Assembly

Abstract:Here, an electron transfer enhanced redox mediator is presented that significantly enhances electron transfer between the host electrode and enzymes, as well as between adjacent enzymes, promoting seamless interfaces through multiple interactions. Biofuel cells (BFCs) offer an eco‐friendly route to convert biochemical energy into electricity. However, their performance is hindered by insufficient enzyme immobilization as well as limited electron transfer within the enzymatic electrode. While the incorporation of redox mediators (RMs) into enzyme layers has been shown to improve BFC performance through enhanced electron transfer, progress has plateaued in the last decade. Herein, a major breakthrough is presented realized by a novel strategy that exploits electron transfer‐intensified RM layers. Metal nanoparticles covalently bridged between neighboring RMs facilitate electron transfer ubiquitously. Electron transfer characteristics are enhanced not only within the RM layers themselves, but also at the glucose oxidase (GOx)/host electrode and GOx/GOx interfaces. This leads to a remarkable performance boost in the enzymatic anode. A hybrid BFC constructed with innovative anode and Pt‐based cathode exhibits a striking combination of high power output (2.3 and 8.5 mW cm−2 at 10 and 300 mmol L−1 glucose, respectively) and exceptional operational stability (≈80% and 47% power retention after 10 days and 1 month, respectively), outperforming all previously reported BFCs by a significant margin.

Abstract:The electrode material is a pivotal component in achieving efficient electron transfer in an enzymatic biofuel cell (EBFC). Herein, novel hydrophilic carbon fibers (HCFs) with a helical structure are synthesized through chemical vapor deposition and are used as an electrode material for the immobilization of biocatalysts to fabricate bioelectrodes, where glucose dehydrogenase, serving as the anodic enzyme, is co‐immobilized with 1,4‐naphthoquinone on the HCFs for the catalysis of the glucose oxidation reaction (GOR), and bilirubin oxidase, as the cathodic enzyme, is co‐immobilized with protoporphyrin IX for the catalysis of the oxygen reduction reaction (ORR). It is found that thermal annealing the HCFs improves the bioelectrocatalytic performances of the as‐fabricated bioanode for the GOR and biocathode for the ORR, owing to the enhanced electron transfer kinetics. Subsequently, a membraneless glucose/O2 EBFC assembled by using the as‐synthesized bioelectrodes is able to generate a high open circuit voltage of 0.8 V and a maximum power density of 0.7 mW cm−2 at 0.44 V with good operational stability. The practicability in fabricating membraneless EBFCs makes the synthesized HCFs promising in miniature bioenergy devices. Trying to cell it: Hydrophilic carbon fibers with a helical architecture are synthesized to immobilize redox enzymes in a membraneless glucose/O2 biofuel cell. The enhanced electron transfer kinetics and the hydrophilic property show affinity toward glucose oxidation and oxygen reduction reactions, resulting in superior cell performance.

Abstract:Abstract Presently, one of the most important aspects for the development of enzymatic biofuel cells (EBFCs) is to synthesize the novel electrode materials that possess high current density, low open-circuit voltage (OCV) and long-term stability. To achieve the above attributes, lots of new strategies are being used by the researchers for the development of advanced materials. Nowadays, nanomaterials and nanocomposites are the promising material that has been utilized as effective electrode material in solar cells, supercapacitors and biofuel cells application. Herein, we account for a novel electrocatalyst as electrode material that comprised ZnO nanoparticles decorated on the surface of polyindole (PIn)-multi-walled carbon nanotube (MWCNT), for the immobilization of glucose oxidase (GOx) enzyme and mediator (Ferritin). The PIn-MWCNT scaffold is prepared via in situ chemical oxidative polymerization of indole on the surface of MWCNT and assessed by myriad techniques. The micrograph of scanning electron microscopy (SEM) designated the interconnected morphology of MWCNTs in the polymer matrix. X-ray diffraction spectroscopy (XRD) and Fourier transform infrared spectroscopy (FTIR), confirm the crystallinity and different functional groups available in the synthesized material, respectively. The electrochemical assessment demonstrates that the ZnO/PIn-MWCNT/Frt/GOx nanobiocatalyst exhibits much higher electrocatalytic activity towards the oxidation of glucose with a maximum current density of 4.9 mA cm −2 by consuming 50 mM glucose concentration in phosphate buffer saline (PBS) (pH 7.4) as the testing solution by applying 100 mVs −1 scan rates. The outcomes reflect that the as-prepared ZnO/PIn-MWCNTs/Frt/GOx biocomposite is a promising bioanode for the development of EBFCs.

Hierarchical Porous Carbon Fibers for Enhanced Interfacial Electron Transfer of Electroactive Biofilm Electrode

Abstract:The nanoporous carbon fiber materials derived from electrospun polyacrylonitrile (PAN) fibers doped with zeolitic imidazolate framework are developed here and applied in the microbe fuel cell anode for enhanced interfacial electron transfer. Zeolitic imidazolate fram-8 (ZIF-8) could introduce a large number of mesopores into fibers, which significantly promote indirect electron transfer mediated by flavins (IET). Moreover, it is noted that thinner fibers are more suitable for cytochromes-based direct electron transfer (DET). Furthermore, the enlarged fiber interspace strengthens the amount of biofilm loading but a larger interspace between thick fibers would hinder the formation of continuous biofilm. Consequently, the nanoporous carbon fiber derived from PAN/ZIF-8 composite with a 1:1 wt ratio shows the best performance according to its suitable mesoporous structure and optimal fiber diameter, which delivers a 10-fold higher maximum power density in microbial fuel cells compared to carbon fabric. In this work, we reveal that the proportion of IET and DET in the interfacial electron transfer process varies with different porous structures and fiber diameters, which may provide some insights for designing porous fiber electrodes for microbial fuel cells and also other devices of bioelectrochemical systems.

Abstract:Enzymatic biofuel cells (EBFCs) utilize enzymes to convert chemical energy present in renewable biofuels into electrical energy and have shown much promise in the continuous powering of implantable devices. Currently, however, EBFCs are greatly limited in terms of power and operational stability with a majority of reported improvements requiring the inclusion of potentially toxic and unstable electron transfer mediators or multicompartment systems separated by a semipermeable membrane resulting in complicated setups. We report on the development of a simple, membrane/mediator-free EBFC utilizing novel electrodes of graphene and single-wall carbon nanotube cogel. These cogel electrodes had large surface area (∼ 800 m(2) g(-1)) that enabled high enzyme loading, large porosity for unhindered glucose transport and moderate electrical conductivity (∼ 0.2 S cm(-1)) for efficient charge collection. Glucose oxidase and bilirubin oxidase were physically adsorbed onto these electrodes to form anodes and cathodes, respectively, and the EBFC produced power densities up to 0.19 mW cm(-2) that correlated to 0.65 mW mL(-1) or 140 mW g(-1) of GOX with an open circuit voltage of 0.61 V. Further, the electrodes were rejuvenated by a simple wash and reloading procedure. We postulate these porous and ultrahigh surface area electrodes will be useful for biosensing applications, and will allow reuse of EBFCs.
Iron cobalt-doped carbon nanofibers anode to simultaneously boost bioelectrocatalysis and direct electron transfer in microbial fuel cells: Characterization, performance, and mechanism

Abstract:A self-supporting electrode (FeCo-MOF/CNFs) combining iron cobalt bimetallic metal-organic frameworks (FeCo-MOFs) with carbon nanofibers (CNFs) was applied as the anode of a microbial fuel cell (MFC). The introduction of FeCo-MOFs enhanced graphitization degree and electrical conductivity, which endowed FeCo-MOF/CNFs with excellent electrocatalytic performance and good biocompatibility. The hierarchical porous structure of FeCo-MOF/CNFs provided abundant attachment sites for electroactive bacteria (EAB) and facilitated rapid electron transfer. The MFC equipped with FeCo-MOF/CNFs anode (FeCo/CNFs-MFC) exhibited considerable power generation output (maximum power density: 5.3 ± 0.2 W/m2, coulombic efficiency: 54 ± 4 %). In addition, FeCo/CNFs-MFC achieved a direct electron transfer (DET) catalytic current density of 0.63 A/m2. FeCo-MOF/CNFs could simultaneously enhance the bioelectrocatalysis activity and promote the DET process of EAB, which provided an effective way to improve the sluggish extracellular electron transport process of the MFC anode.

Abstract:The enzymatic biofuel cell (EBFC) has been considered as a promising implantable energy generator because it can extract energy from a living body without any harm to the host. However, an unprotected enzyme will be destabilized and even eventually be deactivated in human blood. Thus, the performance of implantable EBFC has received barely any improvement. It is therefore a breakthrough in realizing a superior efficient EBFC that can work stably in human blood which relies in protecting the enzyme to defend it from the attack of biological molecules in human blood. Herein, we innovatively created a single-walled carbon nanotube (SWCNT) and cascaded enzyme-glucose oxidase (GOx)/horseradish peroxidase (HRP) coembedded hydrophilic MAF-7 biocatalyst (SWCNT-MAF-7-GOx/HRP). The SWCNT-MAF-7-GOx/HRP is highly stable in electrocatalytic activity even when it is exposed to high temperature and some molecular inhibitors. In addition, we were pleasantly surprised to find that the electrocatalytic activity of GOx/HRP in hydrophilic SWCNT-MAF-7 far surpasses that of the GOx/HRP in hydrophobic SWCNT-ZIF-8. In human whole blood, the SWCNT-MAF-7-GOx/HRP catalytic EBFC exhibits an eightfold increase in power density (119 mu W cm(-2) vs 14 mu W cm(-2)) and 13-fold increase in stability in comparison with the EBFC based on an unprotected enzyme. In this study, the application of metal-organic framework-based encapsulation techniques in the field of biofuel cells is successfully realized, breaking a new path for creating implantable bioelectrical-generating devices.

Abstract:A hybrid system of carbon nanotubes (CNTs) coated with poly (amidoamine) (PAMAM) dendrimer-encapsulated platinum nanoparticles (Pt-DENs) and glucose oxidase (GOx) was prepared through the layer-by-layer (LbL) self-assembly approach and then used as anode in enzyme-based biofuel cells (BFCs). The assembly process was monitored by ζ-potential measurement, and the as-resulted Pt-DENs/CNTs nanocomposites were characterized by transmission electron microscopy (TEM). The performance of electrodes modified by Pt-DENs/CNTs was also investigated by electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). We found that the Pt-DENs/CNTs could enhance the electron transfer between the redox centers in enzyme and the electrode surfaces. Furthermore, by employing the Pt-DENs/CNTs modified electrodes as anode, the enzyme-based BFCs operated in a solution containing glucose generated an open-circuit voltage of approximately 640.0mV and a maximum current density of about 90.0μA/cm2, suggesting that Pt-DENs/CNTs may serve as an alternative anode to previously used noble metals in BFC applications.

Abstract:Nanoporous gold (NPG) membranes made by dealloying consist of a bicontinuous network of Au ligaments and open pore channels, which have gained considerable attention as a platform for the design of carbon-free electrodes for proton exchange membrane fuel cells (PEMFCs). Benefiting from a unique combination of high electronic conductivity, high surface area, and modifiable surface chemistry, these self-supporting membrane type electrodes allow integration of various structural functions required for specific electrode reactions and simultaneously facilitate the entire fuel cell kinetic process. In this review, we summarize the major research progresses in this area, with an emphasis on how to customize the surface structures of these three-dimensional electrocatalysts for desired fuel molecule oxidation and oxidant reduction. We will also discuss these designed structural characteristics that can be readily accommodated in membrane electrode assemblies (MEA), thus effectively bridging the technological gap between electrocatalysts’ intrinsic activities and their actual performances in fuel cells.

Abstract:Microbial fuel cells (MFCs) are innovative devices to extract renewable energy using exoelectrogens from wastewater. The performance of MFCs mainly depends on the electron transfer efficiency between the exoelectrogens and the anode materials. In this work, iron carbide (Fe3C) nanoparticles encapsulated with graphitic carbon layers embedded into biochar (nano-Fe3C@C) were prepared as a binder-free anode material for MFCs. The encapsulated carbon layers can avoid the direct contact between Fe3C nanoparticles and the electrolyte, thereby effectively inhibiting the dissolution and over-aggregation of nanoparticles, and over 96% of the in 10-70 nm. Moreover, this configuration enhanced the interfacial contact between the Fe3C nanoparticles and the biochar matrix, resulting in a significantly lower charge transfer resistance (Rct) of 39.30 omega compared to carbon cloth (CC) and sugarcane carbon (SC) anodes. This hybrid structure also promoted biocompatibility and extracellular electron transfer (EET) of exoelectrogens with nano-Fe3C anode, to obtain a fast start-up time of 67 h. Modified anode material achieved a maximum load voltage of 0.62 V along with significant enrichment of the Geobacter genus. Consequently, the nano-Fe3C anode exhibited an exceptional power density of 2316 mW m- 2 in the acetate-fed MFCs, which was higher than the reported studies involving Fe3C-based non-graphene anode materials. The nano-Fe3C@C material is a promising and sustainable anode for MFCs in wastewater treatment and renewable energy generation. Current findings have opened a new gateway for the preparation of other dispersive, durable, and high-performance metal-based materials for MFCs and bioelectrochemical sensors.
Self-encapsulated Enzyme Through In-Situ Growth of Polypyrrole for High-Performance Enzymatic Biofuel Cell

Abstract:Lacking of electron flow pathway within enzymes, enzymatic biofuel cells (EBFCs) always suffer from low power and poor stability, which limits their further application. Aiming to improve enzyme's conductivity and stability, we constructed a conductive enzyme nanocapsule n(GOx-PPy). A self-encapsulation was triggered by adding glucose to initiate an in-situ growth of PPy network within and around the glucose oxidase (GOx). Conductive pathways, as well as protective shell, were formed between single GOx active site and PPy network, allowing enhanced electron transfer and enzymatic stability. The n(GOx-PPy)-based EBFC demonstrated 245-fold increased power output, providing a promising strategy for constructing high-performance EBFCs.
Enhanced Oxygen Reduction upon Ag-Fe-doped Polyacrylonitrile@UiO-66-NH 2 Nanofibers to Improve Power-Generation Performance of Microbial Fuel Cells

Abstract:Microbial fuel cells (MFCs) have great potential as a new energy technology that utilizes microorganisms to produce electrical energy by decomposing organic matter. A cathode catalyst is key to achieving an accelerated cathodic oxygen reduction reaction (ORR) in MFCs. We prepared a Zr-based metal organic-framework-derived silver-iron co-doped bimetallic material based on electrospun nanofibers by promoting the in situ growth of UiO-66-NH 2 on polyacrylonitrile (PAN) nanofibers and named it as CNFs-Ag/Fe-m:n doped catalyst (m:n were 0, 1:1, 1:2, 1:3, and 2:1, respectively). Experimental results combined with density functional theory (DFT) calculations reveal that a moderate amount of Fe doped in CNFs-Ag-1:1 reduces the Gibbs free energy in the last step of the ORR. This indicates that Fe doping improves the performance of the catalytic ORR, and MFCs equipped with CNFs-Ag/Fe-1:1 exhibit a maximum power density of 737. 45 mW m −2 , significantly higher than that obtained for MFCs using commercial Pt/C (457.99 mW m −2 ).

Abstract:Flexible and wearable energy devices have become an important support and indispensable product of the era of artificial intelligence and Internet of Things. Enzyme biofuel cells (EBFCs) have received much attention for their ability to convert the body's abundant biochemical energy into electricity. It is still a challenge to realize the flexibility of EBFC system and improve its stability under mechanical deformation. Therefore, we establish a laminated stack of EBFC that is fabricated by flexible enzymatic bioelectrodes and stretchable hydrogel. The bioelectrodes were synthesized through enzyme encapsulation and in-situ growth strategies based on polyurethane/regenerated cellulose (PU/RC) nanofiber, since it has been proved that mental organic frameworks (MOFs) could effectively maintaining biological activity and nanomaterials could promote electron transport efficiency. The results show that a piece of as-fabricated stretchable EBFC (5 × 1 cm) demonstrates considerable energy harvesting while produces a EOCV of 0.35 V. More importantly, the stretchable EBFC exhibits stable energy output performance even under repeated stretching, bending and twisting cycles. The assembled EBFC with stable output performance under multiple mechanical deformation states in this work shows important practical application value in the field of wearable energy devices.
