Abstract:A series of alpha,beta-divinyl tetrahydropyrroles, synthesized by asymmetric allylic C-H bond activation/conjugated diene addition reaction of ene-2-dienes, were found to be very efficient chiral chain diene ligands in the rhodium-catalyzed conjugated addition of organoboronic acids to various alpha,beta-unsaturated compounds, achieving the desired chiral adducts with good to excellent yields and ee values.
Density Functional Theory Study of the Mechanism of the Rhodium(I)-Catalyzed Conjugated Diene Assisted Allylic C–H Bond Activation and Addition to Alkenes Using Ene-2-dienes As Substrates

Abstract:Recently we reported the first conjugated diene. assisted, rhodium-catalyzed allylic C-H bond activation and addition to alkenes to synthesize multifunctional tetrahydropyrroles, tetrahydrofurans, and cyclopentanes from ene-2-diene substrates, with good to excellent diastereoselectivities. Here we report a DFT study of the mechanism of this reaction, aiming to obtain a detailed potential energy surface, to understand factors determining its stereochemistry, and to determine why conjugated diene is very critical for the success of this reaction. DFT calculations unveiled that the catalytic cycle of this reaction involves a sequence of substrate catalyst complex formation, allylic C-H activation, alkene insertion into the Rh-H bond, and di-pi-allyl-assisted C(sp(3))-Rh-C(sp(3)) reductive elimination, among which the C-H activation and alkene insertion steps are reversible. The main reason for the formation of the cis-divinyl product is that the irreversible reductive elimination transition state from the bis-allylic Rh complex favors a cis 5/5 bicyclic conformation to reduce the ring strain. Moreover, formation of the cis-divinyl product is also assisted by the alkene coordination to the Rh center in the reductive elimination transition state. DFT insights revealed that the conjugated diene, which is very critical for the target reaction, disfavors the double-bond isomerization and facilitates the reductive elimination for the bis-allylic Rh complex, causing the C-H activation and alkene insertion to occur. The computational results showed that the bridgehead double-bond distortion, as suggested by the Bredt's rule, is responsible for not generating the type II [4+2] cycloadducts from ene-2-dienes.

Abstract:An asymmetric intermolecular [4 + 2] cycloaddition of 1,3-dienes with dialkyl acetylenedicarboxylates, which was catalyzed by a rhodium(i)-chiral phosphoramidite complex, was developed. This protocol provided a highly enantioselective access to prepare carbonyl substituted cyclohexa-1,4-dienes with up to 96% yield and >99% ee. Notably, a cycloaddition on the 10 g scale gave the product in 92% yield and with 99% ee, which showed great potential for the scale-up synthesis of carbonyl substituted cyclohexa-1,4-dienes. In addition, oxidative aromatizations and hydrolysis of the products were also investigated.

Abstract:Because of the inherent difficulty in differentiating two olefins, the development of metal-catalyzed asymmetric cyclization of 1,6-dienes remains challenging. Herein, we describe the first rhodium(III)-catalyzed asymmetric borylative cyclization of cyclohexadienone-tethered mono-, 1,1di-, and (E)-1,2-disubstituted alkenes (1,6-dienes), affording optically pure cis-bicyclic skeletons bearing three or four contiguous stereocenters with high yields (25-93%), and excellent diastereoselectivities (>20:1 dr) and enantioselectivities (90-99% ee). This mild catalytic approach is generally compatible with a wide range of functional groups, which allows several facile conversions of the cyclization products. Furthermore, on the basis of our SAESI-MS experiment and computational study, a Rh(I)/(III) catalytic cycle is proposed in this tandem reaction, and the Rh(I) active species catalyzes the overall transformation via sequential oxidative addition of B(2)pin(2), olefin insertion, cyclizing conjugate addition, and reductive elimination. The irreversible conjugate addition determines the overall regioselectivity of borylative cyclization, and the ring strain favors the formation of 5,6-bicyclic structure. This highlights the control of ring strain in diene cyclizations, which provides a useful basis for future reaction designs.

Here we report the Rh-catalyzed d-arylation of aryldienes that gives nearly exclusive Z-1,4-addition products (generally with >95:5 positional and geometrical selectivity). This remote functionalization provides access to chiral diarylated butenes from readily available precursors poised for further functionalization, including in the synthesis of bioactive molecules. Mechanistic studies suggest that protonolysis of a Rh-allyl intermediate generated by diene insertion into a Rh-aryl is the rate determining step and occurs by an inner-sphere proton transfer pathway.

Abstract:A Cp*Rh-III-catalyzed annulative carbooxygenation of challenging 1,1-disubstituted alkenes triggered by C-H activation of N-aryloxyacetamides has been established, which affords 2,3-dihydrobenzofuran derivatives with a quaternary carbon center in good to excellent yields under mild redox-neutral conditions. An amide group on the alkenes is essential for the process, and may inhibit the beta-H elimination from C(sp3)-Rh species by saturating the rhodium center through coordination. Furthermore, mechanistic insights obtained from control experiments suggest a mechanism involving a Rh-III-Rh-V-Rh-III catalytic cycle.

Abstract:A variety of novel C-1-symmetric chiral diene ligands based on the dicyclopentadiene (DCP) skeleton were easily prepared from commercially available DCP. The application of these diene ligands in the rhodium-catalyzed asymmetric 1,4-addition of arylboronic acids to alpha,beta-unsaturated carbonyl compounds has been examined and excellent enantioselectivities (up to 97% ee) as well as good yields were achieved under mild reaction conditions. Abstract:The first rhodium-catalyzed intramolecular [5 + 2] cycloaddition of 3-acyloxy 1,4-enyne and alkene was developed. The cycloaddition is highly diastereoselective in most cases. Various cis-fused bicyclo[5.3.0]decadienes were prepared stereoselectively. The chirality in the propargylic ester starting materials could be transferred to the bicyclic products with high efficiency. Electron-deficient phosphine ligand greatly facilitated the cycloaddition. Up to three new stereogenic centers could be generated. The resulting diene in the products could be hydrolyzed to enones, which allowed the introduction of more functional groups to the seven-membered ring.

Abstract:A highly efficient P-stereogenic diphosphine-rhodium complex was applied to the chemo- and enantioselective hydrogenation of allylic hydrazones for the synthesis of chiral allylic hydrazines in 89-96 % yields and with 82-99 % ee values. This methodology was successfully applied to the preparation of versatile chiral allylic amine derivatives.

Abstract:We report the first Rh2(II)-catalyzed asymmetric cycloisomerization of activated enynes to provide cyclopropane-fused tetra-hydropyridines in good yields and excellent enantioselectivities under mild conditions. The activated group, CHZ (Z is electron-withdrawing group (EWG)), in the enyne substrates exerts two synergetic roles, one is to activate alkyne for the cyclopropanation reaction; the other is to introduce the C-H···O interaction between substrate and catalyst (reducing the energy barrier of the reaction). This double-mode activation was supported by both density functional theory (DFT) calculations and experimental tests. This strategy was also extended to other CH2Z (Z can be OH, OMe, F) as activating groups that made the CH2 more acidic so that the substrates could also form increased C-H···O interaction with the catalyst.

Abstract:Rh-catalyzed dynamic kinetic intramolecular [4+2] cycloaddition reaction of 1,3-disubstituted allene-1,3-dienes afforded cis -fused [4.3.0]bicyclic products with an excellent chemo-, diastereo-, and enantio-selectivity. Many synthetically useful functional groups are tolerated. The synthetic utility has been demonstrated. Based on the careful experimental studies, a mechanism involving the rapid racemization of the allene moiety in the starting materials has been proposed.

Abstract:Herein, we report a rhodium catalyzed directing-group free regioselective C-H allylation of simple arenes. Readily available gem-difluorinated cyclopropanes can be employed as highly reactive allyl surrogates via a sequence of C-C and C-F bond activation, providing allyl arene derivatives in good yields with high regioselectivity under mild conditions. The robust methodology enables facile late-stage functionalization of complex bioactive molecules. The high efficiency of this reaction is also demonstrated by the high turnover number (TON, up to 1700) of the rhodium catalyst on gram-scale experiments. Preliminary success on kinetic resolution of this transformation is achieved, providing a promising access to enantio-enriched gem-difluorinated cyclopropanes.

Abstract:An oxyacetamide-directed Rh(III)-catalyzed Z-type alkenyl C–H functionalization through a rare exo-rhodacyle intermediate is described, forming multisubstituted dienes and allenes. A variety of alkenes and propargylic carbonate coupling partners are suitable for this transformation with high regio- and stereoselectivity.
