Abstract:Rh-catalyzed dynamic kinetic intramolecular [4+2] cycloaddition reaction of 1,3-disubstituted allene-1,3-dienes afforded cis -fused [4.3.0]bicyclic products with an excellent chemo-, diastereo-, and enantio-selectivity. Many synthetically useful functional groups are tolerated. The synthetic utility has been demonstrated. Based on the careful experimental studies, a mechanism involving the rapid racemization of the allene moiety in the starting materials has been proposed.

Abstract:<p>The first example of Rh‐catalyzed kinetic resolution of 1,3‐disubstituted allene‐1,3‐dienes involving intramolecular [4+2]‐cycloaddition has been developed. Followed by C=C isomerization, the reaction finally afforded aza‐[4.3.0]bicyclic compounds with a very high enantio‐ and E‐selectivity. The mechanism has been unveiled by careful control experiments. Abstract:The first rhodium-catalyzed intramolecular [5 + 2] cycloaddition of 3-acyloxy 1,4-enyne and alkene was developed. The cycloaddition is highly diastereoselective in most cases. Various cis-fused bicyclo[5.3.0]decadienes were prepared stereoselectively. The chirality in the propargylic ester starting materials could be transferred to the bicyclic products with high efficiency. Electron-deficient phosphine ligand greatly facilitated the cycloaddition. Up to three new stereogenic centers could be generated. The resulting diene in the products could be hydrolyzed to enones, which allowed the introduction of more functional groups to the seven-membered ring.

Abstract:An asymmetric intermolecular [4 + 2] cycloaddition of 1,3-dienes with dialkyl acetylenedicarboxylates, which was catalyzed by a rhodium(i)-chiral phosphoramidite complex, was developed. This protocol provided a highly enantioselective access to prepare carbonyl substituted cyclohexa-1,4-dienes with up to 96% yield and >99% ee. Notably, a cycloaddition on the 10 g scale gave the product in 92% yield and with 99% ee, which showed great potential for the scale-up synthesis of carbonyl substituted cyclohexa-1,4-dienes. In addition, oxidative aromatizations and hydrolysis of the products were also investigated.

Abstract:Because of the inherent difficulty in differentiating two olefins, the development of metal-catalyzed asymmetric cyclization of 1,6-dienes remains challenging. Herein, we describe the first rhodium(III)-catalyzed asymmetric borylative cyclization of cyclohexadienone-tethered mono-, 1,1di-, and (E)-1,2-disubstituted alkenes (1,6-dienes), affording optically pure cis-bicyclic skeletons bearing three or four contiguous stereocenters with high yields (25-93%), and excellent diastereoselectivities (>20:1 dr) and enantioselectivities (90-99% ee). This mild catalytic approach is generally compatible with a wide range of functional groups, which allows several facile conversions of the cyclization products. Furthermore, on the basis of our SAESI-MS experiment and computational study, a Rh(I)/(III) catalytic cycle is proposed in this tandem reaction, and the Rh(I) active species catalyzes the overall transformation via sequential oxidative addition of B(2)pin(2), olefin insertion, cyclizing conjugate addition, and reductive elimination. The irreversible conjugate addition determines the overall regioselectivity of borylative cyclization, and the ring strain favors the formation of 5,6-bicyclic structure. This highlights the control of ring strain in diene cyclizations, which provides a useful basis for future reaction designs.

Abstract:We recently developed a completely new type of Rh‐catalyzed [5+2] cycloaddition by using 3‐acyloxy‐1,4‐enyne (ACE) as the 5‐carbon building block. In this update, we show that ACE can undergo intramolecular [5+2] cycloaddition with either an alkene or an allene in the presence of an appropriate rhodium catalyst and ligands to afford bicyclic compounds with multiple stereogenic centers. In most cases, cis‐fused bicyclo[5.3.0]decadienes are prepared highly diastereoselectively.

Abstract:Allenes are important 2π building blocks in organic synthesis and engage as 2-carbon components in many metal-catalyzed reactions. Wender and co-workers discovered that methyl substituents on the terminal allene double bond counterintuitively change the reactivities of allenes in [Rh(CO)2Cl]2-catalyzed intermolecular (5 + 2) cycloadditions with vinylcyclopropanes (VCPs). More sterically encumbered allenes afford higher cycloadduct yields, and such effects are also observed in other Rh(I)-catalyzed intermolecular cycloadditions. Through density functional theory calculations (B3LYP and M06) and experiment, we explored this enigmatic reactivity and selectivity of allenes in [Rh(CO)2Cl]2-catalyzed intermolecular (5 + 2) cycloadditions with VCPs. The apparent low reactivity of terminally unsubstituted allenes is associated with a competing allene dimerization that irreversibly sequesters rhodium. With terminally substituted allenes, steric repulsion between the terminal substituents significantly increases the barrier of allene dimerization while the barrier of the (5 + 2) cycloaddition is not affected, and thus the cycloaddition prevails. Computation has also revealed the origin of chemoselectivity in (5 + 2) cycloadditions with allene-ynes. Although simple allene and acetylene have similar reaction barriers, intermolecular (5 + 2) cycloadditions of allene-ynes occur exclusively at the terminal allene double bond. The terminal double bond is more reactive due to the enhanced d-π* backdonation. At the same time, insertion of the internal double bond of an allene-yne has a higher barrier as it would break π conjugation. Substituted alkynes are more difficult to insert compared with acetylene, because of the steric repulsion from the additional substituents. This leads to the greater reactivity of the allene double bond relative to the alkynyl group in allene-ynes.

Abstract:A highly enantioselective Rh(I)-catalyzed intramolecular [3 + 2] cycloaddition of 1-yne-VCPs to bicyclo[3.3.0] compounds with an all-carbon chiral quaternary stereocenter at the bridgehead carbon was developed. DFT calculations of the energy surface of the catalytic cycle (complexation, cyclopropane cleavage, alkyne insertion, and reductive elimination) of the asymmetric [3 + 2] cycloaddition reaction indicated that the rate- and stereo-determining step is the alkyne-insertion step. Analysis of the alkyne-insertion transition states revealed that the serious steric repulsion between the substituents in the alkyne moiety of the substrates and the rigid H(8)-BINAP backbone is responsible for not generating the disfavored [3 + 2] cycloadducts.

Abstract:Reported herein is the first rhodium‐catalyzed [4+2+1] cycloaddition of in situ generated ene/yne‐ene‐allenes and CO to synthesize challenging seven‐membered carbocycles fused with five‐membered rings. This reaction is designed based on the 1,3‐acyloxy migration of ene/yne‐ene‐propargyl esters to ene/yne‐ene‐allenes, followed by oxidative cyclization, CO insertion, and reductive elimination to form the final [4+2+1] cycloadducts. The possible competing [4+1], [4+2], and [2+2+1] cycloadditions were disfavored, making the present reaction an efficient way to access functionalized 5/7 rings.

Abstract:A diastereoselective [4 + 2] cycloisomerization of asymmetric allenyl dienes is reported. The asymmetric dienyl allenes are synthesized using the method reported by Ma. These substrates readily undergo diastereoselective intramolecular rhodium catalyzed [4 + 2] cycloisomerization analogous to thermal intramolecular Diels–Alder reactions. Overall, 29 examples are presented with tethers possessing nitrogen, oxygen, and carbon. Abstract:Recently we reported the first conjugated diene. assisted, rhodium-catalyzed allylic C-H bond activation and addition to alkenes to synthesize multifunctional tetrahydropyrroles, tetrahydrofurans, and cyclopentanes from ene-2-diene substrates, with good to excellent diastereoselectivities. Here we report a DFT study of the mechanism of this reaction, aiming to obtain a detailed potential energy surface, to understand factors determining its stereochemistry, and to determine why conjugated diene is very critical for the success of this reaction. DFT calculations unveiled that the catalytic cycle of this reaction involves a sequence of substrate catalyst complex formation, allylic C-H activation, alkene insertion into the Rh-H bond, and di-pi-allyl-assisted C(sp(3))-Rh-C(sp(3)) reductive elimination, among which the C-H activation and alkene insertion steps are reversible. The main reason for the formation of the cis-divinyl product is that the irreversible reductive elimination transition state from the bis-allylic Rh complex favors a cis 5/5 bicyclic conformation to reduce the ring strain. Moreover, formation of the cis-divinyl product is also assisted by the alkene coordination to the Rh center in the reductive elimination transition state. DFT insights revealed that the conjugated diene, which is very critical for the target reaction, disfavors the double-bond isomerization and facilitates the reductive elimination for the bis-allylic Rh complex, causing the C-H activation and alkene insertion to occur. The computational results showed that the bridgehead double-bond distortion, as suggested by the Bredt's rule, is responsible for not generating the type II [4+2] cycloadducts from ene-2-dienes.
