Abstract:The segments from various part of \%Liriope platyphylla \%Wang et Tang var. \%variegata \%Hort. were cultured on MS medium supplemented with variable ratio of hormones. The results showed that the highest induction frequency (100%) of adventitious buds was obtained by culturing the buds of rootstocks on MS medium supplemented with BA 1.5 mg/L+NAA 0.5 mg/L, but the mosaic of the plant can't be kept by this way. In order to solve this problem, the liquid culture with MS medium supplemented with BA 3.0 mg/L+NAA 0.5 mg/L can avoid dedifferentiation and keep the mosaic of the proliferated buds. The adventitious buds could be transplanted once a month, while MS medium supplemented with IBA 0.25 mg/L is suitable for rooting. After a period of training, the rooted plants could be planted on soil.

Abstract:Based on in vitro culture system of high capacity and fast regeneration of pepper ( Capsicum annuum L.)cotyledon. Pepper cultivars were transformed with double Cecropin B and D genes constructed in plasmid pCDB Ⅱ which was transferred into Agrobaterium tumefaciens . With the regenerated plants resistance to Kanamycin, the rate of transgenic pepper were surveyed and analysed on four pepper cultivars and three different differentiation groups of the same explant. The results showed that there was remarkable difference among the different pepper cultivars and there was no remarkable difference among the different differentiation groups on the detection of PCR level . No remarkable difference was found between the different pepper cultivars and between the different differentiation groups on the detection of Southern blotting level.

Abstract:Anthracnose of capsicum casued by Colletotrichum spp. (C. spp. ) has been a great problems for capsicum production in Korea and China. Based on morphological characters, the capsicum anthracnose pathogen isolates K1 and C1, K2 and C2, K3 and C3 were identified as Colletotrichurn gloeosporioides G type, C. gloeosporioides R type and C. coccodes, respectively. Pathogenicity of Korea isolate and China isolate in capsicum lines of Korea and China did not show a difference. Results of RAPD-PCR analyses indicate that the similarities between K1 and C1, K2 and C2, and K3 and C3 were 85.71％, 71.43％ and 50.0％, respectively.

Abstract:Microspores of six hybrids between non-headed Chinese cabbage(Brassica campestris ssp. chinensis) and turnip (B. campestris ssp. rapifera Matzg) were cultured . The embryoid yield varied with the different materials used, and the two genotypes with the highest embryoid yield, B. campestris ssp. chinensis cv. aijiaohuang and shuzhouqing crossed with B. campestris ssp. rapifera Matzg cv. qisihai produced 21.2 and 17.2 embryoids per flower bud. The optimum period of inducing embryo was uninucleate period. Preheat of 33 ℃ for 24 h before the microspores were transferred to the culture condition of 25 ℃ could obviously enhance the yields, and the active carbon could improve embryo quality.

Abstract:To study the genetic transformation of Chinese cabbage-pe-tsai(Brassica compestris L.ssp.pekinensis Olsson‘Jinyu60’),its tissue culture system was established.Cotyledons with petiole were used as explants,and the culture conditions were optimized by an orthogonal design.The results showed that,before and after genetic transformation,the optimum organization culture medium combination for‘Jinyu60’was:MS+3 mg/L 6-BA+0.3 mg/L NAA+4 mg/L AgNO3+2%sucrose+0.8%agar and MS+3 mg/L 6-BA+0.01 mg/LNAA+4 mg/L AgNO3+2%sucrose+0.8%agar,respectively.The concentration rate between 6-BA and NAA was 3:0.01.

Abstract:Fruit quality has long been well known as an important prerequisite for evaluating pepper resources. In the study, 75 cultivars belonging to four Capsicum species were investigated on the bases of fruit morphology and concentrations of fruit secondary metabolites. The results showed that C. annuum had a greater variation than C. chinense and C. frutescens in terms of fruit weight, size and contents of carotenoids, anthocyanidins, and capsaicinoids. Moreover, there were significant differences in the concentrations of total phenol, total soluble sugar, total soluble solids, titratable acid, and total soluble protein of all samples. Capsaicin and dihydrocapsaicin were the most important capsaicinoids components in pepper fruits, of which C. chinense both had the highest levels, whereas some accessions of C. annuum were not detected. Eighteen fatty acids were detected in pepper fruits, and the dominant member was linoleic acid (a polyunsaturated fatty acid) therein. We integrated a set of methods for metabolites, and the results indicated that there was a positive correlation between concentrations of capsaicinoids and fatty acids. Four accessions had the highest contents of quality-related compounds, two of which belonged to C. chinense (S23 and S24) and possessed high levels of capsaicin. However, S67 had a high level of β-carotene and S68 contained higher contents of total fatty acid and ascorbic acid, and both of them belonged to C. annuum. This work could provide a valid experimental basis for the potential application value of the four accessions mentioned above.

Abstract:The tissue culture is the technical system of plant fast reproduction.In this article the present research progress in Chinese peony tissue culture was summarized,including seed culture,stem-tip culture,bud culture,young leaf culture,petiole culture,root culture and so on.The results indicated: the technique of peony tissue culture already gradually not only tended to the consummation,but also had the difficulty in the large scale production,fewer species cultured,the culturing material brown and vitrification,root growth and suitable medium,appropriate explants and so on.The technical objective,future direction and industrialization of Peony breeding were put forward.

Abstract:This paper compared the methods of anther and microspore-isolated cultures in Chinese cabbage.The results showed that the difference of genotype between varieties was the key factor affecting production of the pollen embryo.The embryo inducement efficiency of microspore-isolated culture was 10 times greater than that of pollen culture.But the regeneration rate of pollen plants of microspore-isolated culture was much lower than that of pollen culture.Many key techniques of microspore-isolated culture should be improved in order to widely apply to breeding work.

Abstract:The binary plasmid vector pBI35S-AMF was transferred into cv.Shanghaiqing of Chinese cabbage pak-choi(Brassica campestris ssp. Mokino chinensis var.communis Tsen et Lee) and cv.Youqing of flowering Chinese cabbage(B.campestris ssp. chinensis Makino var.parachinensis(Bailey) Tsen et Lee) by Agrobacterium-mediated method.The pivotal factors,which influenced the transformation frequency,were compared,and the parameters of transformation system were optimized.It was showed that selective medium containing 300 mg/L ampicillin,which inhibited growth of Agrobacterium,led to production of more Kan~R shoots;the time required for preculture and co-culture was three days and two days respectively;and the efficiency of inoculation with LBA4404/pBI35S-AMF was significantly higher than with EHA105/pBI35S-AMF.Molecular analysis to Kan~R seedlings showed that the frequency of Southern blotting was 82.2% and most of transformants were multi-copies integration.Moreover,the results indicated that the efficiency of transformation system was stable.

Abstract:The bulbs of narrow-leaf,broad-leaf and multi-seed cultivars of Fritillaria thunbergii Miq were used to investigate the effect of different sterilized methods and culture media with different hormone combinations on the callus induction,differentiation and plantlets regeneration.The results showed that the optimum sterilized method was treated with 0.1% mercuric chloride for 20 min;the callus induction rate was the highest on the medium with MS+NAA 1.5 mg ·L-1+KT 0.5 mg ·L-1,and the induction rate was 36.7%;the callus differentiation rate was the highest on the medium with MS+NAA 4.0 mg ·L-1+KT 1.0 mg ·L-1,and the differentiation rate was 80.7%.The results also demonstrated that the genotype had significant effects on callus induction,differentiation and plant regeneration of different cultivars of Fritillaria thunbergii.

Abstract:The leaf of Lilium tsingtauense Gilg. were used as experimental materials. And the effect of different sterilized time, kinds of hormone and its combination on advenitious buds proliferation, plant regeneration and root regeneration were studied the in this paper. The results showed that explant sterilized in 0.1% aqueous mercuric chloride for 5 min had better effect, and the contamination rate was 31%, the germinating rate was 79%, inducement rate was 65.2%. MS+6-BA 2.0 mg/L+NAA 0.2 mg/L was suitable for advenitious buds proliferation from the leaf. The optimum medium was MS+6-BA 1.0mg/L+NAA 0.2mg/L. The best medium for rooting was 1/2MS+NAA 0.2mg/L. The comparatively perfect tissue culture regeneration system was formed.

Abstract:The plants of the Lycoris genus are precious flower bulb resources of China. However, they are restricted by the long life cycle and low proliferation, and cannot meet the needs of commercial production. For effective asexual propagation methods, scales cutting, spray cultivation and tissue culture were conducted in L. radiata. For the scales cutting in substrate, the highest budding rate and propagation coefficient reached 80.5% and 7.2, respectively. Spray cultivation of bulb pieces was proved to be an effective method for bulblet reproduction of Lycoris, which generated more buds and higher propagation coefficient as 9.8. 5 g/L activated carbon effectively prevented the browning of explants during tissue culture. Medium component with 10 mg/L BA and 0.5 mg/L NAA was proved best for buds inducing and proliferating. Furthermore, we carried out the enrichment analysis of pathways during bulb development, and got some genes related to starch and sucrose metabolism, which will beneficially support the further molecular breeding of Lycoris.

Abstract:Pepper is an economically important vegetable worldwide, containing various specialized metabolites crucial for its development and flavor. Capsaicinoids, especially, are genus-specialized metabolites that confer a spicy flavor to Capsicum fruits. In this work, two pepper cultivars, YB (Capsicum frutescens L.) and JC (Capsicum baccatum L.) pepper, showed distinct differences in the accumulation of capsaicin and flavonoid. However, the molecular mechanism underlying them was still unclear. Metabolome analysis showed that the JC pepper induced a more abundant accumulation of metabolites associated with alkaloids, flavonoids, and capsaicinoids in the red ripening stages, leading to a spicier flavor in the JC pepper. Transcriptome analysis confirmed that the increased expression of transcripts associated with phenylpropanoid and flavonoid metabolic pathways occurred in the JC pepper. Integrative analysis of metabolome and transcriptome suggested that four structural genes, 4CL7, 4CL6, CHS, and COMT, were responsible for the higher accumulation of metabolites relevant to capsaicin and flavonoids. Through weighted gene co-expression network analyses, modules related to flavonoid biosynthesis and potential regulators for candidate genes were identified. The promoter analysis of four candidate genes showed they contained several cis-elements that were bonded to MYB, bZIP, and WRKY transcription factors. Further RT-qPCR examination verified three transcription factors, MYB, bZIP53, and WRKY25, that exhibited increased expression in the red ripening stage of the JC pepper compared to YB, which potentially regulated their expression. Altogether, our findings provide comprehensive understanding and valuable information for pepper breeding programs in the future.

Abstract:The purpose of this study was to find out a pepper which contains maximum capsaicin and dihydrocapsaicin, so that both can be extracted from it to be used as food ingredients or as powder after isolation. Several pepper germplasm in China were studied by HPLC (high performance liquid chromatography). Shuanla (Capsicum frutescens L. cv. Shuanlaense) was found in Capsaicin, Dihydrocapsaicin, Pungency (SHU) to be significantly higher than the others. We have identified that Shuanla is the hottest pepper variety in China.

Abstract:In order to improve the tissue culture technology of Siraitia grosvenorii (Swingle), stem cutting with single nod was used as explants, MS as basal medium and supplied with LFS or CPPU respectively. Results:(1)On the primary culture LFS 0.2(mg/L,as same as fallows) could induce the axillary bud of explants germinating and growing quickly, most of them could rooting, meanwhile the callus formation was controlled obviously during 25d cultured. But CPPU 0.02 could only induce a lout of callus formation, no bud germinated. (2)On the secondary culture for proliferation the bioactive of LFS to induce axillary bud germinating and plantlet rooting was enhanced by cooperation with NAA 0.1. At the same time on the help of NAA, CPPU also induced the axillary bud germinating and improved leaves growing bigger,however stem elongation and adventitious roots are strongly inhibited at the same time large numbers of callus are formde.

Abstract:The primarily technology of tissue culture and propagation of Siraitia grosvenorii was reported,contains the condition of tissue culture,confection of medium,selection and sterilization of explant,inoculation and culture,preservation of superior provenance,hardening-seedling and transplanting,plant pack and transportation. And 5 kinds of referenced medium were reported too. The medium of induced stem was MS+BA0.5～1.0 mg/L+IAA(NAA)0.05～0.1 mg/L+sugar 3%+agar 4.5 g/L,pH5.8. The medium of induced stem apex was MS+BA0.5～1.0 mg/L+NAA0.05～0.1 mg/L+coconut milk 100 mL+sugar 3%+agar 4.5 mg/L,pH5.8. The medium of subculture of fasciculate bud was MS+BA0.3～0.7 mg/L+NAA0.05/IAA0.1 mg/L+sugar 3%+agar 4.5 mg/L,pH5.8. The medium of subculture of cutting was MS+BA0.1 mg/L+IAA0.3 mg/L+active carbon 0.07 g/L+sugar 3%+agar 4.5 mg/L,pH5.8. The rooting medium was MS+BA0.07 mg/L+IBA0.15 mg/L+IAA0.1 mg/L active carbon 0.1 g/L+sugar 3%+agar 4.5 mg/L,pH5.8. On the other hand,the cause of ill phenomenon in the process of tissue culture was analyzed,preventive measure was put forward. And feasible condition of hardening-seedling and transplanting was clear,the relevant manage method was formed. The integrated technology procedure of tissue cultured plantlets of S.grosvenorii was established.
