Abstract:Virus-free materials of Lilium spp.were obtained from meristem culture in vitro.A series of optimization experiments for cultural medium composition and concentration of phytohormones were conducted with a view to aceelerate the propagation of virus-free materlals.The best condition of chromosome determination was established.The obtained results indicated that the most suitable rapid propagation media of Lililum spp.was MS+ 6-BA 1.0mg/L+ IAA0.2mg/L+ KT1.0mg/L+ PP3334.0mg/L.The optimized medium for rooting was 1/2MS+IBA0.5mg/L.These results could meet the technical requirements of rapid-propagation in large scale production.Virus-free materials were observed repeatedly under electron microscope,10 virus-free samples were screened out which could be used to the fast propagation for production.Some physiological data were tested among the virus-free lines and control lines.There was difference between the lines.

Abstract:The segments from various part of \%Liriope platyphylla \%Wang et Tang var. \%variegata \%Hort. were cultured on MS medium supplemented with variable ratio of hormones. The results showed that the highest induction frequency (100%) of adventitious buds was obtained by culturing the buds of rootstocks on MS medium supplemented with BA 1.5 mg/L+NAA 0.5 mg/L, but the mosaic of the plant can't be kept by this way. In order to solve this problem, the liquid culture with MS medium supplemented with BA 3.0 mg/L+NAA 0.5 mg/L can avoid dedifferentiation and keep the mosaic of the proliferated buds. The adventitious buds could be transplanted once a month, while MS medium supplemented with IBA 0.25 mg/L is suitable for rooting. After a period of training, the rooted plants could be planted on soil.

Abstract:A regeneration protocol for cultured endosperm of Actinidia chinensis were set up via organogenesis from callus tissues derived from endosperm explants of Actinidia chinensis ＇ Jintao＇ in vitro. The effects of different media, plant growth regulators, sucrose concentrations and dark treatment on callus induction and shoot formation were studied. The addition of 2,4-D significantly increased percentage of calli formation form the endosperm explants. Addition of 20g/L of sucrose resulted in better organogenesis form, endosperm callus. Dark culture could promote the growth of endosperm callus and organ formation, and the best efficiency was obtained after 7d of culture in the dark.

Abstract:Rice cells were precultured for 10 d in medium containing 60 g/L sucrose and subsequently for 1 d in medium supplemented with 0. 4 M sorbitol. After loading with 25%PVS2 at 22°C for 10 min and dehydration in 100%PVS2 at 0°C for 7. 5 min,they were plunged into liquid nitrogen directly. Survival was 45. 0 ±5.1% (based on the reduction of triphenyl tetrazolium chloride)following warming and unloading. For regrowth, cells were plated on semi-solid medium replenished with 40%(w/v) starch for 2d prior to reculture. Cell suspensions were reestablished and plants were regenerated from recovered cells. Twenty eight plants set seeds in the greenhouse.

Abstract:This paper studied on the difference of a shallow liquid culture system and the solid culture system in the system of rapid propagation technology of virus-free ginger. The multiplication coefficient?the length of root and plant height in shallow liquid culture were prior to those of solid culture. As the regeneration frequency of root and the average root number of each explant were analyzed, there was no significant difference between the two systems. The survival rate of transplantation of seedlings from the shallow liquid system was much higher than that from solid culture system. Furthermore, the cost of chemicals of shallow liquid system was only less than 1/3 compared to that of solid system. Meanwhile, the load of manual work and time were reduced much more. So it was more economical and convenient. For all of the above, the shallow liquid culture system was proposed during rapid propagation process of virus-free ginger.

Abstract:The primarily technology of tissue culture and propagation of Siraitia grosvenorii was reported,contains the condition of tissue culture,confection of medium,selection and sterilization of explant,inoculation and culture,preservation of superior provenance,hardening-seedling and transplanting,plant pack and transportation. And 5 kinds of referenced medium were reported too. The medium of induced stem was MS+BA0.5～1.0 mg/L+IAA(NAA)0.05～0.1 mg/L+sugar 3%+agar 4.5 g/L,pH5.8. The medium of induced stem apex was MS+BA0.5～1.0 mg/L+NAA0.05～0.1 mg/L+coconut milk 100 mL+sugar 3%+agar 4.5 mg/L,pH5.8. The medium of subculture of fasciculate bud was MS+BA0.3～0.7 mg/L+NAA0.05/IAA0.1 mg/L+sugar 3%+agar 4.5 mg/L,pH5.8. The medium of subculture of cutting was MS+BA0.1 mg/L+IAA0.3 mg/L+active carbon 0.07 g/L+sugar 3%+agar 4.5 mg/L,pH5.8. The rooting medium was MS+BA0.07 mg/L+IBA0.15 mg/L+IAA0.1 mg/L active carbon 0.1 g/L+sugar 3%+agar 4.5 mg/L,pH5.8. On the other hand,the cause of ill phenomenon in the process of tissue culture was analyzed,preventive measure was put forward. And feasible condition of hardening-seedling and transplanting was clear,the relevant manage method was formed. The integrated technology procedure of tissue cultured plantlets of S.grosvenorii was established.

Abstract:The tissue culture technology of Dioscorea bulbifera L. is of great significance for the effective protection and utilization of its germplasm resources. This paper reviews the research progress of tissue culture technology of Dioscorea bulbifera L. with different explants. It introduces the historical development of tissue culture technology of Dioscorea bulbifera L., including early exploration and gradual improvement of modern technology. The paper elaborates various methods and strategies of tissue culture technology of Dioscorea bulbifera L. at present, including research progress in aspects such as explant selection, medium formulation, and application of growth regulators. It also looks forward to the future development direction and potential applications of tissue culture technology of Dioscorea bulbifera L., and discusses its application prospects in germplasm resource preservation, new variety breeding, and improvement of disease and pest resistance. This paper is of important theoretical and practical significance for promoting the research and application of tissue culture technology of Dioscorea bulbifera L.

Abstract:The plants of the Lycoris genus are precious flower bulb resources of China. However, they are restricted by the long life cycle and low proliferation, and cannot meet the needs of commercial production. For effective asexual propagation methods, scales cutting, spray cultivation and tissue culture were conducted in L. radiata. For the scales cutting in substrate, the highest budding rate and propagation coefficient reached 80.5% and 7.2, respectively. Spray cultivation of bulb pieces was proved to be an effective method for bulblet reproduction of Lycoris, which generated more buds and higher propagation coefficient as 9.8. 5 g/L activated carbon effectively prevented the browning of explants during tissue culture. Medium component with 10 mg/L BA and 0.5 mg/L NAA was proved best for buds inducing and proliferating. Furthermore, we carried out the enrichment analysis of pathways during bulb development, and got some genes related to starch and sucrose metabolism, which will beneficially support the further molecular breeding of Lycoris.

Abstract:The propagation in vitro system of fig variety 'Zhongguoziguo' was studied.The result showed that the most suitable medium for proliferation was MS+ 0.5mg/L 6-BA +0.05 mg/L NAA and that for shoot rooting was 1/2MS+0.2 mg/L IBA.Plants were obtained from rooted plantlets after acclimatization and transplanting into pots.Meanwhile,it also indicated that GA3 had significant effect on the shoots growth and 0.2% GA3 could accelerate the elongation of the shoots distinctly.Furthermore,the conservation method of this genetic resource by restricting the growth of the explants in vitro condition was also tested.It was found that the shoot had almost no growth in the medium 1/2MS with 1.5% sucrose,and after about seven months,it could resume the growth ability rapidly when transplanting on the proliferation medium with 0.2% GA3.So,this would be a better way for germplasm preservation in short time.

Abstract:Plant regeneration from protoplasts has proved a very useful technique for crop genetic improvement programs,genetic transformation,and somatic hybridization etc.Now reliable procedures are available for protoplast isolation and culture from a range of high plants including cotton.There were many successful reports on plant regeneration from protoplast.Great progresses was also made for cotton somatic hybridization via protoplast fusion,several inter-specific hybrids were created.As is known,there were 47 wild species,which have advantageous properties such as disease and herbicide resistance that the genetic improvements need.Somatic hybridization is a unique useful way to transfer both nuclear and cytoplasmic genes of different wild species simultaneously in comparison to chromosomal and gene engineering.The success of protoplast fusion in cotton was based on the effective system of cotton protoplast culture,and plant regeneration via somatic embryogenesis from protoplasts was a key step to somatic hybridization.So it is necessary to establish a highly reproducible system for efficient plant regeneration from protoplasts or fused protoplasts in cotton.Advances on fundamental techniques,research status,problems,and promising development of cotton protoplast culture,as well as the somatic hybridization were reviewed,in order to offer beneficial references for cotton gene and chromosome engineering.

Abstract:In vitro culture of Amorphophallus konjac C.Koch with Rhizomes as explants.The optimum media for callus induction was screened out by using orthogonal design.Different results can get when cultivate on different media.The results show that the media of NN+1.0mg/L BA+1.0mg/L NAA is the best for callus induction.Media of MS+2.0mg/L BA+1.0mg/L IAA,can get the highest ratio of adventitious buds differentiation,and IAA is better than NAA in this phase.MS+0.5mg/L IBA+0.5mg/L GA_3 can induce roots occurrence preferably,up to 100%.Antioxidants of 100mg/L PVP,100mg/L Vc and sorbent of 500mg/L AC can control browning efficiently.PVP and Vc restrain callus growth,whenas AC can stimulate it.

Abstract:Tissue culture and rapid propagation of a Yunyan 97 giant variation plant was carried out. Result indicated that the most suitable medium for callus induction and proliferation of bud clusters was MS+6-BA2mg?L-1+NAA0.2mg?L-1, while the suitable medium for buds amplification was 1/2MS+6-BA 0.10 mg?L-1+IBA 0.5mg?L-1. It took about 80 days from explants inoculated to small seedling. Attention was also paid to slicing time of explants with bud clusters and how phytohormone was added. Result suggested that, to slice explants 15 to 20 days after bud spots were formed could promote healthy bud clusters which were also easy to slice. Phytohormone added before and after medium sterilization resulted little influence to phytohormone activity.

Abstract:A new hormone was used for sugarcane tissue culture and rapid propagation.The stem apexes of ROC22 and its sub-cultured plantlets were used as the materials.MS basal medium supplied with different concentrations of LFS as the treatment,while 6-BA as the control in the differentiating and multiplication stage,and NAA as the control in the rooting stage,respectively.It was revealed that LFS could significantly retard the browning of the stem apexes and increase stem apexes' survival ratio.It also demonstrated that 2.5～3.0 mg/L LFS was suitable for differentiation,while 3.0～5.0 mg/L for multiplication,and 8.76 times of multiplication rate was got after three subcultures while 8.53 times for the control.Compared with NAA,LFS could induce the occurrence of adventitious roots and the appropriate concentrations were between 2.0～4.0 mg/L with the rooting rate more than 90%.LFS showed favorable application prospects in sugarcane tissue culture for rapid propagation.
A STUDY ON THE RAPID PROPAGATION OF ANOECTOCHILUS FORMOSANUS HAYATA VIATISSUE CULTURE IN VITRO

Abstract:This paper deals with the rapid propagation ofAnoectochilus formosanus Hayata via tissue culture in vitro. The stem segments of A.frormosanus Hayata were used as explants to establish the sterile materials through the surface sterile and initial culture. Four basic media,including MS,H,B_5 and N_6were tested, and of which H was the optimal medium for the multiplication culture of A. formosanus Hayata. The order of the four media for the multiplication culture of A.formosanus Hayata was:H≥N_6B_5≥MS.16 hormone treatments, involving 3 kinds of hormone (KT, BA and NAA), and their different concentrations and different matchings,were tested. The experimental results showed that "2.0mg/L BA+1.0mg/L NAA" was the optimal hormone matching. Under this hormone condition, the axillary buds were germinated from the 3～4 nodes of the basic part of almost 100% of the plantlets of A. formosanus Hayata and the clustered shoots were formed. The recommended optional medium prescription was "H+100mg/L inositol+0.5mg/L nicotinic acid+1.0mg/L thiamine· HCI+0.5mg/L pryidoxine· HCI+2.0mg/L glycine+2.0mg/L BA+1.0mg/L NAA+2.5% sucrose+0.7%argar". 7 kinds of cultivated materials for the transplantation of A. formosanus Hayata were tested, of which the mixed cultivated material, "River sand:Garden soil:Coir=1∶1∶1", was the optimal cultivated material for the cultivation of A. formosanus Hayata.

Abstract:In recent years, the optimum development of land resources has become an important task for ensuring the security of food production in China. Soil microorganisms have been considered to play an important role in conferring soil fertility and productivity. In order to obtain the plant-growth-promoting bacteria in newly reclaimed land, a total of 988 bacterial strains were isolated from nine soil samples collected from different sites in wastelands in Hangzhou (Zhejiang Province, China), a rural mountainous area. Among them, five strains exhibited substantial potential of phosphate solubilization, nitrogen fixation, siderophore production and indole acetic acid production at both pH 5.0 and pH 7.0, and also promoted eggplant growth in immature soil from newly reclaimed land. Furthermore, bacterial strains ZJ62 and ZJ3-12 were identified as Pantoeadispersa and Pantoea ananatis, respectively, while strains ZJ5, ZJ9 and ZJ174 were identified as Burkholderiaarboris, Burkholderia pyrrocinia and Burkholderia pyrrocinia, respectively, based on colony morphology observation and phylogenetic analysis of 16S rDNA and the housekeeping genes sequences. Overall, the result of this study showed that the 5 obtained bacterial strains have a great potential in promoting plant growth in immature soil from newly reclaimed land.

Abstract:Objective To optimize the tissue culture and rapid-proliferation techniques of Stephania kwangsiensis for producing large-scale seedlings. MethodsThe tender stems were used as explants and cultivated in different MS media. The optimum media for inducing buds, subproliferation, and rooting were selected by adjusting the various kinds and concentrations of plant hormones. ResultsThe medium MS+NAA 0.1 mg/L+BA 1.0 mg/L was suitable for buds inducing and could be used in the first generative cultivation. Subproliferation needed to be cultivated alternately in the following three media: MS+IBA 0.4 mg/L+NAA 0.2 mg/mL+GA 0.5-1.0 mg/L, MS+NAA 0.1 mg/L+GA 1.0 mg/L, and MS+IBA 0.2 mg/L+BA 0.4-0.8 mg/L+GA 0.5-1.0 mg/L; propagation coefficient was 6.0 times/50 d. The medium 1/2 MS+IBA 0.8 mg/L was apt to induce rooting and the rooting rate was 75%, so it can be used as rooting medium. Rooting plantlets were transplanted between spring and summer and its surviving rate was up to 90%. ConclusionThe rapid-proliferation technique of S. kwangsiensis can be used for producing large-scale seedlings.

Abstract:Solanum nigrum, which belongs to the Solanaceae family, is an essential plant for food and medicine. It has many important secondary compounds, including glycoproteins, glycoalkaloids, polyphenolics, and anthocyanin‐rich purple berries, as well as many ideal characteristics such as self‐fertilization, a short life cycle and a small genome size that make it a potential model plant for the study of secondary metabolism and fruit development. In this study, we report a highly efficient and convenient tissue culture, transformation and genome editing method for S. nigrum using leaf segments after 8 weeks of tissue culture, with a required period from transformation initiation to harvest of about 3.5 months. Our results also show multi‐shoot regeneration per leaf segment and a 100% shoot regeneration efficiency in a shoot regeneration medium. Moreover, over 82% of kanamycin‐resistant plants exhibited strong green fluorescence marker protein expression, with genetic integration confirmed by PCR results and green fluorescence protein expression in their T1 progeny. Furthermore, we successfully applied this transformation method to achieve an average of 83% genome editing efficiency of SnMYB1, a gene involved in regulating the anthocyanin biosynthetic pathway of S. nigrum in response to missing nutrients. Taken together, the combination of highly efficient tissue culture, transformation and genome editing systems can provide a powerful platform for supporting fundamental research on the molecular mechanisms of secondary metabolism, fruit development, and production of important compounds by biotechnology.
