Abstract:In this paper,the effects of 6-BA and NAA combinations of different organs and different concentrations on the rapid culture of Caryopteris clandonensis are lucubrated.The results show that the germination and rootability of the middle section of newly-growing twigs are the highest;the inducing ratio is higher than 85%;MS + 6-BA 0.5 mg/L + NAA 0.5 mg/L is the optimal successive transfer culture medium,and the germination and rootability of the twigs of Caryopteris clandonensis are high.The rootability of stem apex seedlings is high during rooting,and the rooting ratio of stem apex seedlings can be up to 85% in short time.Short but strong seedlings can be cultured in PP333,thus,the survival ratio of the seedlings of Caryopteris clandonensis can be increased.

Abstract:The segments from various part of \%Liriope platyphylla \%Wang et Tang var. \%variegata \%Hort. were cultured on MS medium supplemented with variable ratio of hormones. The results showed that the highest induction frequency (100%) of adventitious buds was obtained by culturing the buds of rootstocks on MS medium supplemented with BA 1.5 mg/L+NAA 0.5 mg/L, but the mosaic of the plant can't be kept by this way. In order to solve this problem, the liquid culture with MS medium supplemented with BA 3.0 mg/L+NAA 0.5 mg/L can avoid dedifferentiation and keep the mosaic of the proliferated buds. The adventitious buds could be transplanted once a month, while MS medium supplemented with IBA 0.25 mg/L is suitable for rooting. After a period of training, the rooted plants could be planted on soil.

Abstract:Virus-free materials of Lilium spp.were obtained from meristem culture in vitro.A series of optimization experiments for cultural medium composition and concentration of phytohormones were conducted with a view to aceelerate the propagation of virus-free materlals.The best condition of chromosome determination was established.The obtained results indicated that the most suitable rapid propagation media of Lililum spp.was MS+ 6-BA 1.0mg/L+ IAA0.2mg/L+ KT1.0mg/L+ PP3334.0mg/L.The optimized medium for rooting was 1/2MS+IBA0.5mg/L.These results could meet the technical requirements of rapid-propagation in large scale production.Virus-free materials were observed repeatedly under electron microscope,10 virus-free samples were screened out which could be used to the fast propagation for production.Some physiological data were tested among the virus-free lines and control lines.There was difference between the lines.

Abstract:The tissue culture and rapid proliferation techniques of Morinda officinalis How were studied by using apical buds and tender stems as explants.The explants were cultivated in different MS medium with different types and concentrations of plant hormones.The results showed that the most suitable procedure for sterilization was to soak explants into 0.1% HgCl2 solution for 10 minutes after pretreatment with 70% alcohol for 60 seconds.The tender stems were superior to apical buds in vitro culture procedure.The medium MS+IBA 0.05 mg/L+BA 1.0 mg/L was effective to induce shooting and suitable for the first generation cultivation;MS+IBA 0.2 mg/L+BA 1.0 mg/L could effectively induce fasciculate buds and be suitable for subculture and proliferation,the propagation coefficient was 6.0 every 50 days;the most optimum rooting medium was 1/2MS+IBA 0.4～0.8 mg/L,and the rooting rate was 100%.The rooting seedlings were transplanted in sand or burnt soil with plant ash.The highest survival rate was 90%.

Abstract:The tissue culture technology of Dioscorea bulbifera L. is of great significance for the effective protection and utilization of its germplasm resources. This paper reviews the research progress of tissue culture technology of Dioscorea bulbifera L. with different explants. It introduces the historical development of tissue culture technology of Dioscorea bulbifera L., including early exploration and gradual improvement of modern technology. The paper elaborates various methods and strategies of tissue culture technology of Dioscorea bulbifera L. at present, including research progress in aspects such as explant selection, medium formulation, and application of growth regulators. It also looks forward to the future development direction and potential applications of tissue culture technology of Dioscorea bulbifera L., and discusses its application prospects in germplasm resource preservation, new variety breeding, and improvement of disease and pest resistance. Abstract:The primarily technology of tissue culture and propagation of Siraitia grosvenorii was reported,contains the condition of tissue culture,confection of medium,selection and sterilization of explant,inoculation and culture,preservation of superior provenance,hardening-seedling and transplanting,plant pack and transportation. The medium of induced stem apex was MS+BA0.5～1.0 mg/L+NAA0.05～0.1 mg/L+coconut milk 100 mL+sugar 3%+agar 4.5 mg/L,pH5.8. The medium of subculture of fasciculate bud was MS+BA0.3～0.7 mg/L+NAA0.05/IAA0.1 mg/L+sugar 3%+agar 4.5 mg/L,pH5.8. The medium of subculture of cutting was MS+BA0.1 mg/L+IAA0.3 mg/L+active carbon 0.07 g/L+sugar 3%+agar 4.5 mg/L,pH5.8. The rooting medium was MS+BA0.07 mg/L+IBA0.15 mg/L+IAA0.1 mg/L active carbon 0.1 g/L+sugar 3%+agar 4.5 mg/L,pH5.8. On the other hand,the cause of ill phenomenon in the process of tissue culture was analyzed,preventive measure was put forward. And feasible condition of hardening-seedling and transplanting was clear,the relevant manage method was formed. The integrated technology procedure of tissue cultured plantlets of S.grosvenorii was established.

Abstract:Axillary node segment from test-tube plant provided by CIP was used as explants to establish the regeneration system of Ullucus tuberousus.Propagation was selected according to experimental design using orthogonal table using Mean multiple per generation and Mean length of shoot as index. Result showed that MS medium supplemented with 0.08 mg·L-1BA+0.03 mg·L-1NAA+0.5 mg·L-1GA3 was the most suitable one for proliferation of axillary node segment taken from growing shoot,and axillary bud could proliferation above 20 times per 25 days in 5 clones provided by CIP,except clone U2,mean length of total axillary node increasing above 7cm.In addition,1/2 primary medium supplement with 0.03 mg/L NAA was efficient for rooting of node,rhizogenesis start after 7 to 10 d,15 d later 3 to 6 fibres root could extend 1.0~2.0cm long,radication rate was 100%.

Abstract:The propagation in vitro system of fig variety 'Zhongguoziguo' was studied.The result showed that the most suitable medium for proliferation was MS+ 0.5mg/L 6-BA +0.05 mg/L NAA and that for shoot rooting was 1/2MS+0.2 mg/L IBA.Plants were obtained from rooted plantlets after acclimatization and transplanting into pots.Meanwhile,it also indicated that GA3 had significant effect on the shoots growth and 0.2% GA3 could accelerate the elongation of the shoots distinctly.Furthermore,the conservation method of this genetic resource by restricting the growth of the explants in vitro condition was also tested.It was found that the shoot had almost no growth in the medium 1/2MS with 1.5% sucrose,and after about seven months,it could resume the growth ability rapidly when transplanting on the proliferation medium with 0.2% GA3.So,this would be a better way for germplasm preservation in short time.

Abstract:The plants of the Lycoris genus are precious flower bulb resources of China. However, they are restricted by the long life cycle and low proliferation, and cannot meet the needs of commercial production. For effective asexual propagation methods, scales cutting, spray cultivation and tissue culture were conducted in L. radiata. For the scales cutting in substrate, the highest budding rate and propagation coefficient reached 80.5% and 7.2, respectively. Spray cultivation of bulb pieces was proved to be an effective method for bulblet reproduction of Lycoris, which generated more buds and higher propagation coefficient as 9.8. 5 g/L activated carbon effectively prevented the browning of explants during tissue culture. Medium component with 10 mg/L BA and 0.5 mg/L NAA was proved best for buds inducing and proliferating. Furthermore, we carried out the enrichment analysis of pathways during bulb development, and got some genes related to starch and sucrose metabolism, which will beneficially support the further molecular breeding of Lycoris.
A STUDY ON THE RAPID PROPAGATION OF ANOECTOCHILUS FORMOSANUS HAYATA VIATISSUE CULTURE IN VITRO

Abstract:This paper deals with the rapid propagation ofAnoectochilus formosanus Hayata via tissue culture in vitro. The stem segments of A.frormosanus Hayata were used as explants to establish the sterile materials through the surface sterile and initial culture. Four basic media,including MS,H,B_5 and N_6were tested, and of which H was the optimal medium for the multiplication culture of A. formosanus Hayata. The order of the four media for the multiplication culture of A.formosanus Hayata was:H≥N_6B_5≥MS.16 hormone treatments, involving 3 kinds of hormone (KT, BA and NAA), and their different concentrations and different matchings,were tested. The experimental results showed that "2.0mg/L BA+1.0mg/L NAA" was the optimal hormone matching. Under this hormone condition, the axillary buds were germinated from the 3～4 nodes of the basic part of almost 100% of the plantlets of A. formosanus Hayata and the clustered shoots were formed. The recommended optional medium prescription was "H+100mg/L inositol+0.5mg/L nicotinic acid+1.0mg/L thiamine· HCI+0.5mg/L pryidoxine· HCI+2.0mg/L glycine+2.0mg/L BA+1.0mg/L NAA+2.5% sucrose+0.7%argar". 7 kinds of cultivated materials for the transplantation of A. formosanus Hayata were tested, of which the mixed cultivated material, "River sand:Garden soil:Coir=1∶1∶1", was the optimal cultivated material for the cultivation of A. formosanus Hayata.

Abstract:This paper studied on the difference of a shallow liquid culture system and the solid culture system in the system of rapid propagation technology of virus-free ginger. The multiplication coefficient?the length of root and plant height in shallow liquid culture were prior to those of solid culture. As the regeneration frequency of root and the average root number of each explant were analyzed, there was no significant difference between the two systems. The survival rate of transplantation of seedlings from the shallow liquid system was much higher than that from solid culture system. Furthermore, the cost of chemicals of shallow liquid system was only less than 1/3 compared to that of solid system. Meanwhile, the load of manual work and time were reduced much more. So it was more economical and convenient. For all of the above, the shallow liquid culture system was proposed during rapid propagation process of virus-free ginger.

Abstract:Ficus tikoua Bureau is an ecologically functional perennial vine that belongs to the Moraceae family. Tissue culture is a common method for the rapid propagation of plants and is suitable for the rapid propagation of F. tikoua . The objective of the current study was to evaluate the process of F. tikoua organogenesis in vitro utilizing stem explants. An efficient tissue culture propagation method was developed by manipulating several plant growth regulators (PGRs) and their proportions, as well as evaluating alternative medium options for specific phases of differentiation. Explants were cultured on MS medium, supplemented with various thidiazuron (TDZ) concentrations with 6-benzylaminopurine (BA) and naphthaleneacetic acid (NAA), to investigate their efficacy during in vitro shoot organogenesis. The results demonstrated that the combination of 0.5 mg L −1 BA, 1.0 mg L −1 NAA, and 1.0 mg L −1 TDZ was the most effective for callus formation (91.1%) and that the combination of 2.0 mg L −1 BA and 0.5 mg L −1 TDZ was the most suited for adventitious shoot induction (61.11%). The medium supplemented with 1.0 mg L −1 BA, 1.0 mg L −1 NAA, and 0.5 mg L −1 TDZ yielded the highest bud multiplication number with a 6.05 proliferation factor. The cultured shoots rooted easily on half-strength MS medium containing 1.0 mg L −1 IBA and 0.1 mg L −1 NAA (68.52%). The subsequent regenerated plants were grown in a mixture of garden soil, perlite, and vermiculite in a ratio of 2:1:1, resulting in a survival rate of 86.17%.

Abstract:In order to breed virus-free plantlets of the kiwifruit cultivar “Guichang,” which belongs to Actinidia deliciosa, in this study, stem segments with buds were used as explants, the establishment of a tissue culture rapid propagation system was carried out, and then the virus status of tissue culture plantlets was detected via the real-time reverse transcription-polymerase chain reaction (RT-qPCR) method. The tissue culture rapid propagation system proved that the contamination and browning rates could be controlled below 20% and the survival rate could be exceeded by 70% when the single bud stem segment of “Guichang” kiwifruit was sterilized with 70% alcohol for 30–60 s and 15% NaClO for 15 min, respectively. Meanwhile, we screened the hormone concentration to get better results, and the appropriate medium for adventitious bud induction was MS + 6-BA (1.0 mg/L) + IBA (0.2 mg/L); for proliferation, it was MS + 6-BA (1.0 mg/L) + IBA (0.1 mg/L); and for rooting, it was 1/2 MS + IBA (0.3 mg/L), and the efficiency of induction, proliferation, and rooting could reach 74.07%, 79.63%, and 85.18%, respectively. In addition, the RT-qPCR results demonstrated that the infection rate of 9 viruses: apple stem grooving virus (ASGV), cucumber mosaic virus (CMV), Actinidia virus X (AVX), cucumber necrosis virus (CNV), ribgrass mosaic virus (RMV), citrus leaf blotch virus (CLBV), Actinidia virus B (AcVB), Pelargonium zonate spot virus (PZSV), and cherry leaf roll virus (CLRV) in the “Guichang” kiwifruit tissue culture plantlets was 0. This study could lay a foundation for the production of “Guichang” kiwifruit tissue culture seedlings, and the medium formula provided in this study was useful for the industrial rapid propagation of “Guichang” plantlets.

Abstract:Abstract As a traditional Chinese medicinal material, Cynanchum atratum Bunge has been widely used in traditional Chinese medicine for its treatment of abscesses, acute urinary infection and hectic fever.Thus, wild resources of it have been endangered by overharvesting. Plant tissue culture technology is an important measure to protect wild resources of medicinal plants, including C. atratum . Therefore, a fast and efficient propagation system of C. atratum through axillary bud proliferation pathwayhas been established. Through axillary bud proliferation, the medium [MS+sucrose 30 g/L+Agar 7 g/L+NAA 0.2 mg/l+IBA 1.5mg/l+KT 0.5 mg/l] can effectively proliferate adventitious buds, and the induction rate was 100 %, proliferation coefficient could reach 8.56. MS medium was used to induce adventitious bud rooting, with rooting rate of 98% and no callus. The highest survival rate was 90% when the ratio of grass mud pond and orchard red soil was 1:1. To our knowledge this is the first report of rapid propagation system in C. atratum , it achieve rapid reproduction of C. atratum .

Abstract:This study successfully established an efficient in vitro propagation system for Paphiopedilum SCBG Huihuang90 via protocorm-like body (PLB) formation from seed-derived calluses, PLB proliferation and differentiation, root induction and greenhouse acclimatization. Furthermore, 1/2 Murashige and Skoog (MS) + 0.025 mg/L 2,4-Dichlorophenoxyacetic acid (2,4-D) was suitable for the proliferation of PLBs, and 1/2MS + 10% coconut water (CW, v/v) + 0.5 g/L activated carbon (AC) was suitable for PLB differentiation. PLBs at different developmental stages required different kinds of sugars. This study provided a reference for research on the propagation techniques of other Paphiopedilum.

Abstract:Gentiana rhodantha is a perennial herb of the genus Gentiana (Tourn.) L. This study was novel in establishing a regeneration system of G. rhodantha using young leaves as explants on the Murashige and Skoog (MS) medium supplemented with different plant growth regulators (PGRs). The roots, stems, and leaves of G. rhodantha were used as explants. The effects of the optimal explant disinfection method, type of explant used, concentrations of PGRs added to the culture media on tissue culture, and rapid propagation of G. rhodantha were studied. The results showed that the optimal disinfection method for stems and roots consisted of disinfection using 75% ethanol for 50 s, followed by 4% sodium hypochlorite (NaClO) for 10 min. The optimal disinfection technique for leaves consisted of disinfection using 75% ethanol for 50 s, followed by 4% NaClO for 8 min. Root explant was the most suitable for inducing the callus of G. rhodantha on the MS medium supplemented with different PGRs. The optimal conditions for callus induction included 1.0 mg/L 6-benzylaminopurine (6-BA) and 0.5 mg/L α-naphthalene acetic acid (NAA). The callus induction rate using the root explant reached 94.28%. MS supplemented with 2.0 mg/L 6-BA and 0.1 mg/L NAA was the optimal medium for inducing adventitious shoots from the callus of G. rhodantha . The best medium for propagation and plantlets strengthening was MS supplemented with 0.8 mg/L 6-BA and 0.3 mg/L NAA, and the propagation index was 8.62. MS supplemented with 0.3 mg/L 3-indolebutyric acid was the best culture medium for inducing the rooting of adventitious buds, with the maximum rooting rate reaching up to 100%.

Abstract:A new hormone was used for sugarcane tissue culture and rapid propagation.The stem apexes of ROC22 and its sub-cultured plantlets were used as the materials.MS basal medium supplied with different concentrations of LFS as the treatment,while 6-BA as the control in the differentiating and multiplication stage,and NAA as the control in the rooting stage,respectively.It was revealed that LFS could significantly retard the browning of the stem apexes and increase stem apexes' survival ratio.It also demonstrated that 2.5～3.0 mg/L LFS was suitable for differentiation,while 3.0～5.0 mg/L for multiplication,and 8.76 times of multiplication rate was got after three subcultures while 8.53 times for the control.Compared with NAA,LFS could induce the occurrence of adventitious roots and the appropriate concentrations were between 2.0～4.0 mg/L with the rooting rate more than 90%.LFS showed favorable application prospects in sugarcane tissue culture for rapid propagation.
