Abstract:As a new gene, the monoclonal antibody against URG11 protein is not currently available. In this study, one monoclonal antibody (MAb) against URG11 was obtained with standard cell fusion technique and enzyme-linked immunosorbent assay (ELISA) screening. The peptide of URG11 used in making the MAb in this study was synthesized as described. One of the newly developed MAbs is named MAb 3D2, the isotype of which is IgG2a. Immunohistochemistry and Western blot showed that MAb 3D2 could recognize URG11 protein in both native and denatured forms. MAb 3D2 will be a useful tool for the functional research of URG11 in future studies.

Abstract:The full-length hNdrg2 cDNA-coded 357 amino acids was cloned and expressed in Escherichia coli strain DH5α as a 6× His-tagged protein. The purified 6× His-fusion protein was used to immunize mice for preparing monoclonal antibodies (mAb) against N-myc downstream-regulated gene 2 (Ndrg2). A hybridoma secreting a monoclonal antibody against Ndrg2 was obtained and named FMU-Ndrg2.3. Western blot analysis confirmed that this mAb is specific only to Ndrg2 but not to Ndrg1, Ndrg3, and Ndrg4-B. Some tissue distribution features of Ndrg2 proteins, such as thyroid, kidney, testis, prostate, and pancreas islets, were present by immunohistochemistry.

Abstract:OBJECTIVE:To construct a prokaryotic plasmid to express the testis development related gene 1 (TDRG1) recombinant protein and obtain anti-TDRG1 mAb by immunizing mice, and to identify the biological properties of the mAb.METHODS:The coding sequence of TDRG1 was amplified by RT-PCR from normal human testis tissue and cloned into the vector pET21, and then was expressed in the E. coli BL 21(DE3) to get TDRG1 recombinant protein. The purified TDRG1 recombinant protein was injected to immunize the BALB/C mice to develop anti-TDRG1 mAb. Splenocytes of the immunized mice were collected and fused with the mouse myeloma cell line Sp2/0 cells. The hybridoma cells that secreted anti-TDRG1 mAb were subcloned with limited dilution. Enzyme-linked immunosorbent assay (ELISA) was used to evaluate titers and subtypes of mAb. Western blot and immunohistochemistry were used to detect specificity of mAb.RESULTS:The prokaryotic plasmid expressing the recombinant protein was constructed, and the TDRG1 recombinant protein was expressed and purified. Two hybridoma cell lines that secreted anti-TDRG1 mAb were obtained. The titer of the mAb in ascites was 1 : 1.6 x 10(6), and the subtype of the mAb was IgG(1). Westem blot and immunohistochemistry analysis indicated the mAb showed specific combination with TDRG1 protein in human testes.CONCLUSION:The TDRG1 recombinant protein is highly purified and has strong antigenicity. The anti-TDRG1 mAb is produced successfully.

Abstract:Objective:To amplify and sequence of the variable region genes of anti CD3 McAb.Methods:The V H?V L genes were amplified by RT PCR from total RNA that were extracted from WuT3 hybridoma.Recombinant cloning vector was constructed and sequenced after the enzyme digestion.Results:It showed that V H gene consisted of 363 bp encoding amine acid residues,belongs to mouse heavy chain subgroup IIB;V L gene consisted of 330 bp encoding amine acid residues,belongs to mouse κ light chain subgroup III.Comparing with Kabat database,the V H?V L genes were in agreement with the characterization of DNA sequences present in the mouse Ig V H?V L regions respectively.Conclusion:The success of cloning of the V H?V L genes of WuT3 McAb lay a good foundation for the construction and expression of chimeric antibody.
Generation and characterization of mAb 61H9 against junctional adhesion molecule-a with potent antitumor activity

Abstract:Junctional adhesion molecule-A (JAM-A) is an adhesion molecule that exists on the surface of certain types of cells, including white blood cells, endothelial cells, and dendritic cells. In this study, the cDNA sequences of JAM-A-Fc were chemically synthesized with optimization for mammalian expression. Afterward, we analyzed JAM-A protein expression through transient transfection in HEK293 cell lines. Mice were immunized with JAM-A-Fc protein, and hybridoma was prepared by fusing myeloma cells and mouse spleen cells. Antibodies were purified from the hybridoma supernatant and four monoclonal strains were obtained and numbered 61H9, 70E5, 71A8, and 74H3 via enzyme-linked immunosorbent assay screening. Immunofluorescence staining assay showed 61H9 was the most suitable cell line for mAb production due to its fluorescence signal being the strongest. Flow cytometric analysis proved that 61H9 possessed high affinity. Moreover, antagonism of JAM-A mAb could attenuate the proliferative, migrative, and invasive abilities of ESCC cells and significantly inhibit tumor growth in mice. By examining hematoxylin-eosin staining mice tumor tissues, we found inflammatory cells infiltrated lightly in the anti-JAM-A group. The expression of BCL-2 and IκBα in the anti-JAM-A group were decreased in mice tumor tissues compared to the control group. Ultimately, a method for preparing high-yield JAM-A-Fc protein was created and a high affinity mAb against JAM-A with an antitumor effect was prepared.

Abstract:Hepatocellular carcinoma (HCC) is the second most common malignancy in Asia, with a 5-year survival rate of less than 5% due to high recurrence after surgery and resistance to chemotherapy. A variety of therapeutic interventions to treat HCC, particularly gene therapy, have recently been investigated in tumor model systems to provide a more complete understanding of hepatocarcinogenesis and effectively design therapeutic strategies to treat this disease. In our study, we constructed an adenoviral vector expressing small interfering RNA (siRNA) targeting a newly discovered gene named upregulated gene 11 (URG11). We introduced this vector into HCC cells to investigate the role of URG11 in HCC carcinogenesis. We observed that upon URG11 knockdown, HCC cell proliferation was inhibited through downregulation of several G1-S phase related molecules including cyclin D1 and apoptosis was induced as a result of Bcl-2 downregulation. Besides decreased expression of cyclin D1, CDK4, pRb and Bcl-2, URG11 also suppressed several other proteins including CAPN9, which was identified by cDNA microarray and 2D gel electrophoresis. Moreover, Ad-URG11-siRNA significantly suppressed HCC tumor growth in nude mice. In conclusion, Ad-URG11-siRNA can significantly suppress HCC tumor growth in vitro and in vivo by silencing the URG11 gene, and the use of this vector for gene therapy may represent a novel strategy to treat human HCC.

Abstract:Objective B7-H4, a member of the B7 family of immunoregulatory receptors, may participate in the negative regulation of cell-mediated immunity. Aberrant B7-H4 expression is detected in some tumors and it plays a role in the occurrence and development of tumors. The aim of this study was to elucidate the functional and structural properties of B7-H4. Methods We developed a monoclonal antibody (mAb) against the extracellular domain of B7-H4 through immunization of Balb/c mice with 3T3-mB7-H4 cells which expressed extrinsic B7-H4. A stable hybridoma cell line was established. Then, we analysised the characterization of the mAb through Enzyme linked immunosorbent assay (ELISA), Immunoprecipitation (IP), western blotting, Immunohistochemical (IHC), and tested the biological activity of the mAb. Results ELISA, IP, and western blotting analyses indicated that the mAb specifically recognized B7-H4. In addition, flow cytometry demonstrated that the mAb exhibits excellent reactivity when applied to leukemic cells. IHC staining revealed that the mAb stained in a predominantly diffuse plasmalemmal or cytoplasmic pattern when applied to certain tumor tissues. The preliminary results of the mAb's biological activity showed that the mAb could effectively inhibit the function of B7-H4 in the inhibition of T cell, while promotingg the growth of T cells and the secretion of Interleukin-2 (lL-2), Interleukin-4 (IL-4), Interleukin10 (IL-10) and Interferon-γ (IFN-γ). Conclusion This mAb will be a valuable tool for the further investigation of B7-H4 function.

Abstract:AIM:To clone mouse anti-human gastric cancer mAb(3H11) variable genes and to construct 3H11 human-mouse chimeric antibody.METHODS: The entire VH and VL genes of anti-gastric cancer mAb 3H11 were cloned by RT-PCR method from 3H11 hybridoma cells, using 5' primers for leader sequences. The 3H11 VL gene was then inserted into human-mouse chimeric light chain expression vector and transfected into murine Sp2/0 myeloma cells.RESULTS: DNA sequence analysis indicated that the cloned genes included the whole leader sequences and the mature Ig variable region encoding sequences. After gene transfection, transient expression of chimeric light chain protein was detected.CONCLUSION: DNA sequences and transient expression indicated that the cloned gene was functional. This work laid basis for constructing 3H11 human mouse chimeric antibody in the future.
Molecular Cloning of a Tumor-Associated Antigen Recognized by Monoclonal Antibody 3H11

Abstract:Monoclonal antibody (MAb) 3H11 can bind specifically to different cancer cells from different tissues. MAb 3H11 labeled with radioactive isotopes has been used clinically to detect primary cancer and metastatic cancer. Molecular cloning of the antigen recognized by MAb 3H11 is important in studying tumor occurrence and in developing new biotherapy for cancer. Using MAb 3H11, we screened cDNA library made from the human gastric cancer cell line MGC 803, which reacts with MAb 3H11, and isolated one positive clone specifically recognized by the antibody. The insert cDNA fragment was 0.5 kb. After recombining with glutathione-S-transferase expression vector pGEX-4T, the cDNA fragment could be expressed into a fusion protein that specifically reacted with MAb 3H11. Moreover, the fusion protein could competitively inhibit MAb 3H11 binding to MGC 803 cells. Based on the nucleotide sequence of the cDNA fragment, the full length of the cDNA (2156 bp) was obtained by Rapid-Amplification-cDNA-End (RACE) and nested PCR. Its reading frame was 1767 bp encoding a protein of 589 amino acids. Sequence analysis indicated that there is no highly homologous gene in the GenBank. Northern blot and RT-PCR showed that the mRNA of MAb 3H11 antigen was extensively distributed in embryonic tissue and in different cancerous tissues, but not in corresponding normal tissues. Moreover, in producing antibodies to the antigen expressed prokaryotically, we found that the immunogenicity of the antigen was low in mammalian. Thus we believe that this novel antigen acts as an expression regulator in embryo cells and regains expression in tumor cells. In addition, this antigen is characterized by low differentiation and high proliferation. Molecular function of the antigen needs to be investigated.
Engineering and Characterization of Hu3A4: A Novel Humanized Antibody with Potential As a Therapeutic Agent Against Myeloid Lineage Leukemias

Abstract:Leukemia stem cells (LSCs) play a critical role in the initiation, recurrence, and resistance to treatment of leukemia. Eradicating LSCs is crucial for the complete elimination of the disease. CD45RA is identified as an important marker for LSC subsets in acute myeloid leukemia (AML), providing a strategic target for therapy. In this report, we introduce Hu3A4, an innovative humanized CD45RA antibody devised to target LSCs expressing this antigen. Hu3A4 retains the antigen-recognition ability of its parental antibody while removing sequences from the variable region that could elicit human anti-mouse immune reactions. The modified variable regions of the heavy and light chains were intricately fused with the constant regions of human IgG1 heavy and light chains, respectively, producing a humanized antibody that emulates the structure of natural IgG. Hu3A4 was produced through recombinant expression in Chinese Hamster Ovary (CHO) cells, which ensured stable gene integration. In vitro tests revealed that Hu3A4 could effectively target and lyse the cells. Further, in vivo studies highlighted Hu3A4's substantial anti-leukemic activity, significantly prolonging survival times in treated animal models compared to controls (P < 0.01). To summarize, Hu3A4 exhibits remarkable bioactivity and offers a promising therapeutic potential for the treatment of leukemia patients. Progressing Hu3A4 through additional preclinical and clinical studies is crucial to validate its efficacy as a therapeutic agent for leukemia.

Abstract:OBJECTIVE:Acute monocytic leukemia (AML)-M5 is the common type of acute myeloid leukemias in children. Studies have shown that there are abundant lipopolysaccharide (LPS) receptor (designated as CD14) molecules on the cell membrane of M5 cells and they play an important role in the diagnosis of M5, since they can be recognized and bound by mouse-anti-human CD14 monoclonal antibody (McAb). However, mouse-originated antibodies are largely not suitable for clinical application due to the severe side effects, thus "humanized antibody" is desired. As the first step for developing humanized antibody, the construction and expression of single chain antibody (scFv) with functional protein are necessary. The present study aimed to express ZCH-7-2F9 ScFv (scFv(2F9)) in eukaryotic cells and obtain the scFv(2F9) protein with a high biological activity.METHODS:Four primers were synthesized to construct the eukaryotic expressional vector, which included SfiI and EcoRI enzyme cleaving site, 6 x His and stop code TGA sequences. scFv(2F9) gene was amplified through splicing by overlap extension (SOE) using the high fidelity Taq polymerase. Positive recombinants (pSectag2A/scFv(2F9)) were identified through enzyme cleaving and sequenced before expression and were transformed into Chinese hamster ovary (CHO) cells for expression. Western-Blot and flow cytometry (FCM) were carried out to determine the relative molecular mass (Mr) and binding activity of scFv(2F9).RESULTS:The cloned scFv(2F9) gene was identified to be functional by sequencing and expressing. The interested protein could be detected in the culture supernatant of transformed CHO cells with an Mr of 31 000. The blocking test showed that the positive cell percentages, the mean fluorescence intensity (MFI) and the peak of channel (peak Ch) were reduced by 90.02%, 63.30% and 63.38%, respectively after blocking with scFv(2F9), while those were 4.55%, 10.09% and 5.02% after blockage using the supernatant from the CHO cells transfected with blanked vector pSectag2A.CONCLUSIONS:The scFv(2F9) against human CD14 antigen was successfully expressed in eukaryotic cells and showed a high biological activity, which may be useful for the further studies on its humanized antibodies.

Cloning of the Cdna Fragment of a Tumor Associated Antigen Recognized by Monoclonal Antibody 3H11

Abstract:Monoclonal antibody (McAb) 3H11 can specifically bind to different tumor cells.Consequently cloning of the corresponding antigen is significant.The exprssion library,which was constructed with human gastric cancer cell line MGC 803,was screened with the McAb 3H11.One positive clone specifically recognized by McAb 3H11 was isolated.The cDNA fragment in the clone is 554 bp and there is no homology in GenBank.After recombination of this cDNA with Glutathione S transferase (GST) expression vector pGEX 4T,revealed by Western blot the GST fusion protein could react with McAb 3H11,but not with other antibodies.Moreover,the protein derived from the cDNA could competitively inhibit the binding between McAb 3H11 and MGC 803 cells.This work will be helpful to further research on the 3H11 tumor associated antigen.

Abstract:Aberrant mucin O-glycosylation is a pathological alteration that is widespread in cancer. The UDP-N-acetyl-D-galactosamine polypeptide N-acetylgalactosaminyltransferases (ppGalNAc-T) family of enzymes regulates the initial key steps of mucin O-glycosylation. ppGalNAc-T2, as a member of the ppGalNAc-T family, was recently described as an altered expression in oral squamous cell carcinoma and colorectal and breast carcinoma. In order to gain further insight into the role of ppGalNAc-T2, we produced the anti-human ppGalNAc-T2 monoclonal antibody (MAb) 5F3. The IgM κ isotype of this MAb was further characterized using ELISA, Western blot analysis, flow cytometry, and immunofluorescent staining. In the present study, MAb 5F3 can specifically recognize human ppGalNAc-T2 protein in various formats by Western blot analysis, flow cytometry, and immunofluorescent staining. For the first time, it was shown that both Jurkat and HepG2 cell lines clearly express ppGalNAc-T2.

Abstract:A monoclonal antibody, McAb9E (IgG3), was generated against a metastatic HCC cell line, MHCC-1. The antigen was characterized as human Caveolin-1 (Cav-1, 21kDa), with pI of 5.65. The Cav-1 antigen was found significantly over expressed in metastatic HCC cell lines as well as in tumor specimens. The Cav-1 specific McAb may be a useful molecular agent for metastatic HCC.

Abstract:Human adenovirus type 7 (HAdV7) is commonly associated with febrile acute respiratory disease (ARD) outbreaks. We have reported that 10G12, a mouse monoclonal antibody (mAb) specifically recognizing and neutralizing HAdV7, is a promising candidate for humanization. In this study, we engineered the six variants of 10G12 with increased degree of humanization and investigated their biological activity. The humanized monoclonal antibody (mAb) 10G12-M2 was shown to retain the parental antibody's high binding affinity, specificity and potent efficacy of viral suppression. The mAb 10G12-M2 recognized a conformational neutralization epitope of the hexon protein. Complex structure-based molecular docking simulation showed that the hexon protein formed several interactions with 10G12-M2, including hydrogen bonds and salt bridges interaction. Physicochemical properties analysis of 10G12-M2 demonstrated that it is stable and desirable lead candidate. In general, 10G12-M2 had excellent biological activity after humanization combined with the potential for use in prophylactic or therapeutic applications against HAdV7.

Abstract:Objective To design small hairpin RNA(shRNA) targeting Annexin A11(ANXA11),to transfect mouse hepatoma Hca-P with Annexin A11(ANXA11),to obtain the Hca-P cell line with Anxa11 stable knock-down and to establish material basis for future study.Methods Two Anxa11 shRNAs,shRNA-1 and shRNA-2,were synthesized,ligated to pGPU6/GFP/Neo expression plasmid and transfected into Hca-P cell.The monoclonal pGPU6/GFP/Neo-ANXA11-Hca-P cell with stable knockdown of ANXA11 was obtained by G418 screening and limited dilution.ANXA11 mRNA and protein expression levels were measured by RT-PCR and Western blot,and compared with empty vector transfected Hca-P and Hca-P control.Results The recombinant expression vector pGPU6/GFP/Neo-shRNA-ANXA11 was successfully constructed.The monoclonal pGPU6/GFP/Neo-ANXA11-Hca-P cell lines were obtained.Compared to normal Hca-P cell,the ANXA11 mRNA and protein level in Hca-P cells transfected with shRNA-1 wese down-regulated by about 80.2% and 77.7% with statistic significance(P0.01).Conclusion Mouse monoclonal hepatoma Hca-P cell with stable knock-down of ANXA11 was successfully constructed by RNAi and gene transfection,which provides a solid base for studying the effect and mechanism of ANXA11 in hepatoma lymphatic metastasis.

Abstract:AACR Centennial Conference: Translational Cancer Medicine-- Nov 4-8, 2007; Singapore B47 1. Cloning and functional study of novel genes Chronic hepatitis B, which is closely related to the development of cirrhosis and HCC, is a major public health problem worldwide ( Jie Liu, et al. The Lancet 2007 ). By cDNA subtraction and RACE-PCR full length cloning, totally 4 novel genes which are related to HBV have been successfully cloned by our group. (1) Novel gene URG7 : 745bp in full length, encoding a protein of 99 amino acids(published in Hepatology 2001 ). Functional study showed that this novel gene could stimulate cell growth, and also against Fas-mediated TNF killing in NF-κB dependant way. (2) Novel gene URG4 : 3607bp in length, encoding a 104kD protein (published in Neoplasia 2002 ). Homology analysis showed that this gene is on Chromosome 7,which has 34% homologous with caspase recruitment domain family 6. Initial study showed that URG4 ,as a novel oncogene, could stimulate cell growth by through cyclin D (published in Neoplasia 2006 ). (3) Novel gene URG11 : 3074 bp in length, encoding a 673 amino acids protein (published in Neoplasia 2003 ). Homology analysis demonstrated that URG11 is on Chromosome 11, without any homology with known genes. Functional study reveled that this gene could stimulate cell growth by up-regulating β-catenin expression (published in Hepatology 2006 ) with newly generated monoclonal antibody against URG11 (published in Hybridoma 2006 ). (4) HuSui1: 1370 bp in length, encoding 113 amino acids, is on human Chromosome17 (published in Oncogene 1999 ). The finding that HBx downregulating HuSui1 could induce malignant transformation of host cells. (5) S15a: 535 bp in length, encoding a 130 amino acids protein (published in Molecular Carcinogenesis 2004 ). Initial study showed that this gene functions as a oncogene which could stimulate cell growth. (6) VEGFR-3(S): the short form splice variant of VEGFR-3 (published in Hepatology 2007 ). The function study showed that HBV may short circuit VEGFR-3(S) signaling in liver cancer, suggesting that blocking VEGFR-3(S) signaling may be effective in preventing tumor development. 2. Predicting value of novel genes for HCC A 15-year retrospective study in patients with liver cirrhosis showed that elevated expression of URG11, URG7, URG4, S15a, VEGFR-3(S)and HuSui1 in the canceration group compared with non-canceration group. Combinational serum screening or even single protein along can be served as early predictors for HCC, with prediction of HCC 2-3 years earlier (published in Cancer Research 2004 ). These genes can be also the prognostic markers in patients with HCC (published in Cancer Research 2004 ). 3. Novel gene URG11-β-catenin pathway in HBV related HCC It was found that HBxAg could downregulate E-cadherin, the partner of β-catenin, both In Vivo and In Vitro by promoter methylation of E-cadherin (published in Oncogene 2006 ), and that HBx could stimulate Wnt signaling through URG11 and β-catenin, which contribiutes to the development of hepatocellular carcinoma (published in Hepatology 2006 ). These results were cited by Nat Rev Cancer 2007 as the mechanism whereby DNA virus contributes to the development of cancer.
