Abstract:To generate and characterize a monoclonal antibody (mAb) against humanin (HN), BALB/c mice were immunized with the purified pet-44a-HN in adjuvant and their splenic lymphocytes were fused with myeloma SP2/0 cells. The hybridoma cell lines were screened for HN-specific antibodies by indirect enzyme-linked immunosorbent assay (ELISA), and anti-HN mAb-producing hybridoma clones were obtained using a limiting dilution assay. The specificity and affinity of the antibodies were characterized by western blot assays and indirect ELISA. Following fusion, screening and cloning, four hybridoma clones were obtained, and the clone 5A8H3 was demonstrated to stably produce anti-HN IgG2a. Further characterization of 5A8H3 revealed that this mAb specifically recognized HN, the fusion proteins of pet44a-HN protein and pGEMEX-1-HN, but not control (Escherichia coli proteins). This mAb interacted with HN at an affinity constant (Ka) of 2.0 × 10 8 M –1 . The HN-specific IgG2a mAb was successfully generated. It interacted with HN specifically and sensitively, providing a valuable tool for further study of the biological functions of HN.
Production of Monoclonal Antibodies Against Phosphoprotein 65 from Human Cytomegalovirus.

Abstract:Human cytomegalovirus (HCMV) has a very high infection rate in the normal population. Early diagnosis is of great value, especially in immunosuppressed individuals. HCMV phosphoprotein 65 (pp65) is the most widely used antigen for diagnosis. In this report, we prepared a monoclonal antibody against pp65, which could be used for both immunohistochemistry and Western blot diagnosis of HCMV infection.
Relationship Between T-cell Receptor Beta Chain Sequences and Human Cytomegalovirus Infection in Allogeneic Hematopoietic Stem Cell Transplant Recipients.

Abstract:In the present study, clonal amplifications of T-cell receptor β variable (TCR BV) linked to human cytomegalovirus (HCMV) infection were detected in recipients of allogeneic hematopoietic stem cell transplants (HSCT), and certain relationships between them were identified. Furthermore, the relationship between TCR BV sequences and HCMV infections was investigated. The results indicated that the 3 recipients of HSCT had monoclonal expansion of specific TCR BV clones following HSCT. Among these recipients, 2 suffered from pp65 and immediate early (IE) antigenemia. These patients demonstrated preferential expansion of TCR BV9 (QVRGGTDTQ) and TCR BV11 (VATDFQ). The remaining recipient did not express TCR BV9 and TCR BV11, nor did this individual have pp65 and IE antigenemia. These results suggest that expression of TCR BV9 and TCR BV11 may be associated with HCMV antigenemia, and may be involved in the immune response. The amino acid sequences 'QVRGGTDTQ' and 'VATDFQ' may be involved in HCMV reactivation in patients who have undergone HSCT. Assessment of the TCR BV families may provide valuable insight into HCMV pathogenesis and may aid in the diagnosis and therapy for HSCT recipients infected with HCMV.
The clinical significance of CMV - pp65 antigen detcetion in hepatitis B virus (HBV) related hepatocellular carcinoma patients

Abstract:Objective: To investigate CMV-pp65 antigenemia in hepatitis B virus(HBV) related hepatocellular carcinoma patients and discuss the clinical significance of CMV-pp65 antigen detcetion.Methods: Immunohistochemistry two-step method and enzyme-linked immunosorbent assay(ELISA) were used to detect the CMV-pp65 antigen and CMV-specific IgM antibodies in 30 cases of HBV related hepatocellular carcinoma patients;Simultaneously the CMV-pp65 antigen of 18 healthy volunteers were detected as a control group.Chi-square test was used for statistical analysis.Results: The positive rate of CMV-pp65 antigen in HBV related hepatocellular carcinoma patients was 43.3%(13/30);while the positive rate of CMV-specific IgM antibodies was 6.7%(2/30).The difference had significance in statistics(χ2=10.756,P0.01).In Healthy control group,0 in 18 cases was CMV-pp65 antigen positive.The difference was of significance in statistics(χ2=10.697,P 0.01).Conclusion: Hepatitis B virus related hepatocellular carcinoma patients have high positive rate of CMV-pp65 antigenemia,suggesting active CMV infection;CMV-IgM antibody is not suitable for the early diagnosis of active CMV infection in patients with hepatocellular carcinoma.

Abstract:Human cytomegalovirus (HCMV) has evolved several immune evasion strategies. One strategy is controlling the movement of peripheral blood mononuclear cells (PBMCs) by encoding homologues of chemokines. Our aim was to determine whether HCMV open reading frame (ORF) UL128 could encode a protein that attracts PBMCs like a β-chemokine. The recombinant UL128 protein was synthesized by construction of a stably transfected CHO-UL128 cell line, and a chemotaxis assay showed that UL128 was able to attract PBMCs with a potency equal to that of MIP-1α in vitro. We hypothesize that UL128 protein may act as a β-chemokine homologue in viral pathogenesis.

Abstract:Human cytomegalovirus (CMV)-specific cytotoxic T lymphocyte (CTL) immune response must be reconstituted for long-term protection against CMV relapse and disease in hematopoietic stem cell transplantation (HSCT) recipients. We phenotypically quantitated absolute numbers of CMV-pp65 peptide-specific CTLs (CTL(CMV)) in 50 related donor unmanipulated allografts infused into HLA-matched or -mismatched recipients and examined the incidence of CMV reactivation. High CTL(CMV) with terminally differentiated effector CD45RO(-)CD62L(-) cell (T(EMRA)) phenotype in the allografts were associated with reduced risk of CMV reactivation, in the presence of sufficient CD45RO(+)CD62L(-) cell (T(EM)) infusion (>/=0.208 x 10(6)/kg). Early after transplantation, there was significant expansion of CTL(CMV) with the central memory CD45RO(+)CD62L(+) cell (T(CM)) phenotype when CMV was reactivated. The frequencies of CTL(CMV) T(Naive) (CD45RO(-)CD62L(+)), T(CM), and T(EM) at day 90 posttransplantation and of CTL(CMV) T(EMRA) at day 60 posttransplantation were greater in recipients with higher infusions of CTL(CMV) T(EMRA), suggesting protective immunity transferred by infusion of CTL(CMV) within allografts. Moreover, the majority of the CTL(CMV) identified in the recipients early after HSCT was of donor origin. Our findings support that measuring levels of CTL(CMV) and its subsets in the donor grafts and manipulating these cells early after transplantation may help control CMV reactivation, which is closely correlated with immune reconstitution and differentiation of CTL(CMV) subsets.
[Application of Flow Cytometry to Detect PP65 Antigenemia for Diagnosis and Monitoring of Human Cytomegalovirus Infection].

Abstract:OBJECTIVES:To evaluate the clinical significance of flow cytometry (FCM) to detect the cytomegalovirus (CMV) PP65 antigen in patients with CMV infection. METHODS:Samples from 35 patients without CMV infection were used as negative control. The definite diagnosis of CMV infection was based on the national criteria for CMV infection. All 136 patients with CMV infection were examined with the FCM to detect CMV PP65 antigen, real-time fluorescence quantitative-polymerase chain reaction assay (RFQ-PCR) to detect CMV-DNA and ELISA to measure the serum level of IgM antibody against CMV. The results of these 3 assays in 2 groups (isolated organ involvement and disseminated diseases) were compared and the significance of PP65 antigenemia was evaluated. A short-term follow-up was undertaken in 18 patients. RESULTS:The percentages of PP65 positivity in blood mononuclear cells (MNC) and polymorphic nuclear leukocyte (PMNL) from 35 negative control patients were 0.21% +/- 0.09% with a range of 0 - 0.41% and 0.24% +/- 0.10% with a range of 0.12% - 0.48%, respectively, which were not significantly different (t = 0.425, P > 0.05). The 95(th) percentiles (P(95)) of PP65 in MNC and PMNL were 0.39% and 0.45%, respectively, so a cutoff value of >/= 0.50% was set. Of the 136 patients with CMV infection, 118 samples from 118 patients were positive for PP65 antigenemia with a positive rate of 86.8%, which was not statistically different from that (90.4%, chi(2) = 0.91, P > 0.05) of CMV-DNA detected by RFQ-PCR assay but it was significantly higher than that (45.6%, chi(2) = 51.50, P < 0.005) of the detection by IgM measurement. PP65 detection was correlated with urine CMV DNA amplification (chi(2) = 63.78, P < 0.01) while the different detection rates between the two assays were not statistically significant (chi(m)(2) = 1.78,P > 0.05). PP65 detection was not correlated with serum IgM measurement while the detection rates between the two were significantly different (chi(m)(2) = 52.92,P < 0.01). No significant difference was found between the detection rates of CMV infection in MNC (45/53, 84.9%) and PMNL (43/53, 81.1%) (chi(m)(2) = 0.25, P > 0.05). Higher PP65 antigenemia level was correlated with systemic CMV infection, while lower level of PP65 was either in the patients with isolated organ involvement by CMV (chi(2) = 38.51, P < 0.005) or less severe in patient's situation. PP65 antigenemia of CMV infection returned to lower level or negative in recovery stage and increased when condition of patients deteriorated. CONCLUSIONS:PP65 antigenemia detection by FCM is effective in the diagnosis of the active CMV infection. Quantitative monitoring of PP65 antigenemia is useful in the evaluation of patients with CMV infection.
Human Cytomegalovirus Pp65 Protein Induces Specific Immune Response

Abstract:Objective To investigate if recombinant HCMVpp65 can induce specific immune response in T cells.Methods The stimulating effect of recombinant HCMV pp65 protein on PBMC were observed in vitro. Cytotoxicity of PBMC (stimulated by rhHCMVpp65 protein) was detected by using PBMCs incubated with K562 cells or HCMV-infected human embryo lung fibroblasts (HEL) respectively, as well as HEL cells infected at various time points.Results rhHCMV pp65 protein could activate PBMCs and cause PBMC proliferation in vitro. The proliferating PBMCs had specific cytotoxicity to HEL cells infected by HCMV, but not to K562 cells and uninfected HEL cells. CTL could kill the HEL cells 2 h after it was infected with HCMV, and its cytotoxicity persisted for 68 h.Conclusion Recombinant HCMV pp65 protein could induce specific immune response in T cells in vitro. CTL could kill the HCMV-infected HEL cells, and the cytotoxicity of CTL is independent of viral gene expression and requires no protein synthesis. It is important to the immunotherapy of HCMV infection.

Abstract:There is no vaccine available for human cytomegalovirus (HCMV) infection and the treatment is very limited; therefore, it is still an important cause of morbidity and occasional mortality in transplant recipients. An HCMV peptide was selected based on the GenBank sequence M60929. The peptide was conjugated with keyhole limpet haemocyanin and used to immunise New Zealand rabbits to prepare polyclonal antibody. After immunisation, the serum of the rabbits was obtained and purified. ELISA, immunoprecipitation and immunocytochemical staining were used to identify antibodies and antibody titre. The identification of the synthetic peptide antibody was confirmed by ELISA, immunoprecipitation and immunofluorescence against HCMV Towne and AD169 strains. The antibody can be used to detect the HCMV infection by immunocytochemical staining. The synthetic peptide showed favourable immunogenicity. The serum from immunised rabbits could be used to identify HCMV AD169 and Towne strains. Future research should be directed to epitope screening of synthetic HMCV peptides, which could help to understand HCMV infection and virus-neutralising antibodies more fully and to prepare HCMV vaccines and antiviral drugs.

Abstract:T-cell responses directed against human cytomegalovirus (HCMV) glycoprotein B (gB) contribute to protective immunity against HCMV infection in both animal models and humans. However, the gB-specific human CD8(+) T cell responses remain poorly understood. gB antigen-specific CD8(+) T cells were stained with seven major histocompatibility complex (MHC)-peptide pentamers in 16 human leukocyte antigen (HLA)-A*1101-positive, HCMV-seropositive patients following hematopoietic stem cell transplantation (HSCT). Of these seven pentamers, the most frequent CD8(+) T-cell responses were directed against the gB(332-340) peptide. These gB(332-340)-specific CD8(+) T cells were strongly associated with the presence of plasma HCMV immunoglobulin M in all HSCT recipients and exhibited a probable causal relationship with the level of pp65 antigenemia. Together, these data suggest a role for gB(332-340)-specific CD8(+) T cells in HCMV reactivation after HSCT. Furthermore, the pentamer assay may be valuable in detecting antigen-specific CD8(+) T cells. (C) 2014 Elsevier B.V. All rights reserved.
Development of Two Potential Diagnostic Monoclonal Antibodies Against Human Cytomegalovirus Glycoprotein B

Abstract:Human cytomegalovirus glycoprotein B (gB) represents a target for diagnosis and treatment in view of the role it plays in virus entry and spread. Nevertheless, to our knowledge, rare detection of a gB antigen has been reported in transplant patients and limited information is available about diagnostic gB monoclonal antibodies (mAbs). Our aim was to develop gB mAbs with diagnostic potential. Hydrophilic gB peptides (ST: amino acids 27-40, SH: amino acids 81-94) of favorable immunogenicity were synthesized and used to immunize BALB/c mice. Two mAbs, named ZJU-FH6 and ZJU-FE6, were generated by the hybridoma technique and limited serial dilution and then characterized by indirect ELISA, Western blotting, immunoprecipitation, and immunohistochemical staining. The mAbs displayed high titers of specific binding affinities for the ST and SH synthetic peptides at an mAb dilution of 1:60,000 and 1:240,000, respectively. Western blotting and immunoprecipitation indicated that these mAbs recognized both denatured and native gB of the Towne and AD169 strains. The mAbs, when used as the primary antibody, showed positive staining in cells infected with both Towne and AD169 strains. The mAbs were then tested on patients submitted to allogeneic hematopoietic stem cell transplantation. The gB antigen positivity rates of the patients tested using ZJU-FH6 and ZJU-FE6 were 62.0 and 63.0%, respectively. The gB antigen showed a significant correlation with the level of pp65 antigen in peripheral blood leukocytes. In conclusion, two potential diagnostic gB mAbs were developed and were shown to be capable of recognizing gB in peripheral blood leukocytes in a reliable manner.
Human cytomegalovirus pp65 lower matrix protein: a humoral immunogen for systemic lupus erythematosus patients and autoantibody accelerator for NZB/W F1 mice

Abstract:Both the infection of human cytomegalovirus (HCMV) and the immunization of its recombinant glycoprotein (gB) in mice have been known to induce autoimmunity, resulting in symptoms similar to those of human systemic lupus erythematosus (SLE). Research has also found that the murine cytomegalovirus (MCMV)-specific monoclonal antibody (mAb) is able to react with a human U1-70K-like autoantigen. To investigate HCMV involvement in autoimmunity, we analysed the humoral responses to HCMV by autoimmune patients and normal adults. Our studies show unambiguously that sera from SLE patients exhibited an elevated IgG titre to HCMV when compared with those observed in controls and other connective tissue disease (CTD) patients (P < 0.001). The IgM titres to HCMV and IgG to HBV were evaluated, and no significant differences were noted among all testing groups. In addition to initiating T cell activity, as reported by many investigators, we found that the HCMV pp65 antigen (also known as lower matrix protein) was able to induce humoral responses in SLE patients. Immunoblot assays showed that 82.56% of sera from SLE patients reacted with the HCMV pp65 antigen, but only 11.11%, 23.53% and 31.17% of patients from normal control, rheumatoid arthritis (RA) and CTD patients, respectively, reacted to it. Unlike HCMV pp65, HCMV pp150 induced B cell activity in most collected sera (92.22%-98.04%). Finally, female NZB/W F1 mice immunized with plasmids encoding HCMV pp65 open reading frame (pcDNApp65) developed an early onset of autoantibody activity and more severe glomerulonephritis. Thus, we conclude that the HCMV pp65 antigen triggers humoral immunity in SLE patients and autoimmune-prone mice and that it could very well exacerbate the autoimmune responses in susceptible animals.
Efficacy of pp65‐specific TCR‐T cell therapy in treating cytomegalovirus infection after hematopoietic stem cell transplantation

Abstract:Cytomegalovirus (CMV) infection remains a major cause of mortality after hematopoietic stem cell transplantation (HSCT). Current treatments, including anti‐viral drugs and adoptive cell therapy with CMV‐specific cytotoxic T lymphocytes (CTLs),only show limited benefits in patients. T‐cell receptor (TCR)‐T cell therapy offers a promising option to treat CMV infections. Here, using tetramer‐based screening and single‐cell TCR cloning technologies, we identified various CMV antigen‐specific TCRs from healthy donors, and generated TCR‐T cells targeting multiple pp65 epitopes corresponding to three major HLA‐A alleles. The TCR‐T cells showed efficient cytotoxicity towards epitope‐expressing target cells in vitro. After transfer into immune‐deficient mice bearing pp65+HLA+ tumor cells, TCR‐T cells induced dramatic tumor regression and exhibited long‐term persistence. In a phase I clinical trial (NCT04153279), CMV TCR‐T cells were applied to treat patients with CMV reactivation after HSCT. Except one patient who withdrew at early treatment stage, all other 6 patients were well‐tolerated and achieved complete response (CR), no more than grade 2 cytokine release syndrome (CRS) and other adverse events were observed. CMV TCR‐T cells persisted up to 3 months. Among them, 2 patients have survived for more than one year. This study demonstrates the great potential in the treatment and prevention of CMV infection following HSCT or other organ transplantation. This article is protected by copyright. Anti-HCMV effect of recombinant protein of HCMV pp65 and combination with Jin-Ye-Bai-Du granules

Abstract:Objective To compare the effect of recombinant HCMV pp65 protein against HCMV with Chinese medicine Jin-Ye-Bai-Du granules and to study the relation between them.Methods The development of cytopathic effect(CPE) of human embryo lung fibroblast cell(HEL) caused by HCMV were observed in vitro before and after the treatment of pp65-CTL,Jin-Ye-Bai-Du granules,pp65-CTL+Jin-Ye-Bai-Du granules and ganciclovir,respectively.At the same time,the expression of the HCMV mRNA in infected HEL cells were measured by quantitative RT-PCR.Results There are less CPE in HCMV-infected HEL cells after 72h which were treated with pp65-CTL.pp65-CTL+Jin-Ye-Bai-Du granules had strongest inhibiting effect both on HCMVmRNA and CPE among five groups.Conclusion These data suggested that CTL induced by HCMV pp65 had an obvious inhibition on HCMV,and could cooperated with Jin-Ye-Bai-Du granules on action against HCMV.pp65-CTL+Jin-Ye-Bai-Du granules used as a safe and valuable medicine for treatment of active HCMV infection.
[Development of HPV16 Positive Cervical Cancer Model in the Hu-Pbl-scid Mouse and Its Immunological Features].

Abstract:OBJECTIVE:To develop a HPV16 positive cervical cancer model in the hu-PBL-SCID mouse and investigate its immunological features.METHODS:Thirty-two CB17SCID mice were randomly divided into 4 groups: group A (5 mice) subcutaneously injected with phosphate-buffered saline, group B (5 mice) intraperitoneally injected with human peripheral blood lymphocyte (PBL) for immune reconstruction, group C (11 mice) subcutaneously injected with human cervical carcinoma cell line SiHa, and group D (11 mice) intraperitoneally injected with PBL and subcutaneously injected with SiHa cells after 24 hours of PBL transplantation. The tumor growth, behaviors and status of xenogeneic graft versus host disease (XGVHD) were observed. Human immunoglobulins G (IgG) in mouse serum, the percentage of human CD3(+), CD4(+) and CD8(+) T cells in peripheral blood and spleen, spleen weight, tumor infiltrating lymphocytes and human CD4(+) T cells, and cytotoxicity test of spleen cells were detected.RESULTS:The rate of successful tumor transplantation was 100%. XGVHD was not found. On the 5th day, human IgG level in the group B (0.98 microg/ml +/- 0.20 microg/ml) and group D (1.39 microg/ml +/- 0.25 microg/ml) was significantly higher than that in the group A (t = 7.655, 9.937, both P = 0.000). Human IgG level in group D was significantly higher than that in the group B (t = 3.200, P = 0.006). Only very low levels of human serumal IgG were detected in the group C and group A with no significantly difference. The level of human serumal IgG was gradually elevated in all the humanized SCID mice as the the time after PBL transplantation went on, and was significantly higher than that in non-humanized mice (P < 0.05). The percentage of human CD3(+), CD4(+) and CD8(+) T cells was significantly increased in the peripheral blood and spleen of immunoreconstituted SCID mice. The weight of spleen was markedly increased in the group D. TIL infiltrating in the tumor were remarkable and human CD4(+) T cells was detected by immunohistochemistry in the group D but not in the group C. The spleen cells in the group D displayed stronger cytotoxicity to the target cells (P < 0.05).CONCLUSION:Human immune function can be successfully reconstructed in SCID mouse via intraperitoneally injecting with human PBL, and induce anti-tumor immune response to the transplantated tumor of HPV16 positive cervical cancer.
B cell epitope of human cytomegalovirus phosphoprotein 65 (HCMV pp65) induced anti-dsDNA antibody in BALB/c mice

Abstract:Background: HCMV phosphoprotein 65 (HCMVpp65) is a putative immunogen that acts as an accelerator, inducing autoantibody and exacerbating autoimmune response in susceptible animals. The immunity to pp65336-439 instigates autoimmunity, suggesting that pp65336-439 contains crucial B cell epitope(s) for the development of nephritis. This study narrowed down the target epitope to pp65422-439 for immunization of BALB/c mice and mapping of B cell epitope. Methods: The target epitope pp65422-439 reactivity and B cell epitope mapping was examined in serum from pp65422-439-immunized mice and patients with systemic lupus erythematosus (SLE). Kidney tissue from immunized mice was examined for signs of immune complex nephritis. Results: Anti-pp65422-439 antibody in serum either from patients with SLE or from pp65422-439-immunized mice exhibited cross-reactivity to several nuclear components such as double-stranded DNA (dsDNA). Moreover, the pp65422-439-immunized mice developed initial signs of glomerulonephritis such as deposition of immunoglobulin G/M (IgG/IgM) and third complement component (C3). With B cell epitope mapping by pp65422-439-derived decapeptides, one dominant epitope, pp65428-437, was identified in serum from pp65422-439-immunized mice and patients with SLE with anti-pp65422-439 antibody. Epitope spreading from pp65428-437 to pp65430-439 was found in pp65422-439-immunized mice in which we generated monoclonal antibodies to pp65425-434 and pp65430-439. However, dsDNA positive reactivity was exclusively observed in Crithidia luciliae stains with pp65430-439-reactive monoclonal antibody. Additionally, we observed the amelioration of autoimmunity following the elevation of IgM targeting pp65428-437. CONCLUSIONS: Our data suggest that pp65428-437 may be an autoimmune or lupus-prone B cell epitope and may catalyze further epitope spreading for inducing autoantibodies in lupus-susceptible individuals.
The Study on High-Resolution HLA and Human Cytomegalovirus (HCMV) Viremia in Bone Marrow Transplantation Recipients

Abstract:OBJECTIVE:To study the correlation between high-resolution HLA-A * 1101, HLA-A * 0201, HLA-A * 2402, HLA-B * 4001, HLA-DRB1 * 0901 with HCMV pp65 antigenemia after bone marrow transplantation (BMT) in China.METHODS:48 recipients doing BMT during 2009. 2-2010. 10 were selected in my hospital; HCMV pp65 was detected by ELISA or immunohistochemical methods. The frequency of HLA-A * 1101, HLA-A * 0201, HLA-A * 2402, HLA-B * 4001, HLA-DRB1 * 0901 alleles were determined by Polymerase chain reaction-sequence based typing (PCR - SBT).RESULTS:(1) The BMT recipients were HCMV pp65 antigenic positive(100%); (2) The positive rate of HLA-A * 1101, HLA-A * 0201, HLA-A * 2402, HLA-B * 4001 showed no obvious difference between 12 lower antigenemia group and 36 higher antigenemia group, the positive rate: HLA-A * 1101 were 33.3% (8/24) and 20.8% (15/72), HLA-A * 0201 were 4.2% (1/24) and 13.9% (10/72), HLA-A * 2402 were 12.5% (3/24) and 19.4% (14/72), HLA-B* 4001 were 16.7% (4/24) and 12.5% (9/72); (3) HLA-DRB1 * 0901 positive rate in higher antigenemia group was higher than the lower (P = 0.048), the positive rate were 4.2% (1/24) and 19.4% (14/72); (4) HLA-DRB1 * 0901 recipients were higher pp65 antigenemia than HLA-A * 2402 recipients (P = 0.007) and HLA-A * 1101 recipients (P = 0.028), HLA-A * 0201 recipients were higher pp65 antigenemia than HLA-A * 2402 (P = 0.02), the pp65 antigenemia showed no obvious difference among the rest of high-resolution HLA groups (P > 0.05).CONCLUSION:HLA-DRB1 * 0901 alleles might be correlated with BMT recipients happened higher pp65 antigenemia, HLA-A * 2402 alleles might be correlated with BMT recipients happened lower pp65 antigenemia.

Abstract:Human cytomegalovirus (HCMV) is a β-herpesvirus that increases morbidity and mortality in immunocompromised individuals including transplant recipients and newborns. New anti-HCMV therapies are an urgent medical need for diverse patient populations. HCMV infection of a broad range of host tissues is dependent on the gH/gL/gO trimer and gH/gL/UL28/UL130/UL131A pentamer complexes on the viral envelope. We sought to develop safe and effective therapeutics against HCMV by generating broadly-neutralizing, human monoclonal antibodies (mAbs) from VelocImmune® mice immunized with gH/gL cDNA. Following high-throughput binding and neutralization screening assays, 11 neutralizing antibodies were identified with unique CDR3 regions and a high-affinity (KD 1.4-65 nM) to the pentamer complex. The antibodies bound to distinct regions within Domains 1 and 2 of gH and effectively neutralized diverse clinical strains in physiologically relevant cell types including epithelial cells, trophoblasts, and monocytes. Importantly, combined adminstration of mAbs with ganciclovir, an FDA approved antiviral, greatly limited virus dissemination. Our work identifies several anti-gH/gL mAbs and sheds light on gH neutralizing epitopes that can guide future vaccine strategies.
