Abstract:To generate and characterize a monoclonal antibody (mAb) against humanin (HN), BALB/c mice were immunized with the purified pet-44a-HN in adjuvant and their splenic lymphocytes were fused with myeloma SP2/0 cells. The hybridoma cell lines were screened for HN-specific antibodies by indirect enzyme-linked immunosorbent assay (ELISA), and anti-HN mAb-producing hybridoma clones were obtained using a limiting dilution assay. The specificity and affinity of the antibodies were characterized by western blot assays and indirect ELISA. Following fusion, screening and cloning, four hybridoma clones were obtained, and the clone 5A8H3 was demonstrated to stably produce anti-HN IgG2a. Further characterization of 5A8H3 revealed that this mAb specifically recognized HN, the fusion proteins of pet44a-HN protein and pGEMEX-1-HN, but not control (Escherichia coli proteins). This mAb interacted with HN at an affinity constant (Ka) of 2.0 × 10 8 M –1 . The HN-specific IgG2a mAb was successfully generated. It interacted with HN specifically and sensitively, providing a valuable tool for further study of the biological functions of HN.

Abstract:Chemokine-like factor1 (CKLF1), and its three isoforms (CKLF2, 3 and 4), are recently identified human cytokines. CKLF1 is a potent chemoattractant for human leukocytes and can stimulate inflammation and the regeneration of murine skeletal muscle. CKLF2 can promote proliferation and differentiation of C2C12 muscle cells directly by inducing expression of myogenin and activating transcription factors. In the present study, we cloned CKLF murine homologues, and based on their biological and structural features, named them murine chemokine-like factor 2, 4, 5 and 6 (mCKLF2, 4, 5 and 6). mCKLF2, 4, 5 and 6 encode 152, 120, 122 and 86 amino-acid proteins, respectively. mCKLFs map to mouse chromosome 8 and have high sequence similarity to human CKLFs. Compared to human CKLFs, which have a CC motif in the C-terminal region, mCKLF2 and 4 contain a CX3C motif. Using a PCR-based approach, it appeared mCKLF2 and 5 mRNA were highly expressed in adult testis, while mCKLF4 mRNA was detected only in differentiated C2C12 cells, a pattern different from human CKLFs. Conditioned media from COS-7 cells transfected with mCKLF2 and 4 was chemotactic for mouse neutrophils, macrophages and lymphocytes. Our results show that mCKLF2, 4, 5 and 6 are four splicing variants which are homologues of human CKLFs and murine CKLFs possess distinct features compared to their human counterparts.
Preparation and Characterization of A Monoclonal Antibody Against Cklf1 Using Dna Immunization with in Vivo Electroporation

Abstract:Chemokine-like factor 1 (CKLF1) is a newly cloned human cytokine from PHA-stimulated U937 cells in our laboratory, which belongs to a novel gene family and has at least three alternative RNA splicing forms: CKLF2, CKLF3, and CKLF4. Former studies demonstrated that CKLF1 has a chemotaxis effect on different leukocytes both in vitro and in vivo. It can also stimulate the proliferation of mouse skeletal muscle cells and bone marrow cells. This study was designed to generate CKLF1-specific monoclonal antibody (MAb) for further exploration of its structure and function. Mice were immunized intramuscularly with naked plasmid DNA encoding CKLF1 gene with in vivo electroporation. Hybridomas were generated by the fusion of the spleenocytes to Sp2/0 myeloma cells. One hybridoma cell line designated as M4, which is stable in secreting anti-CKLF1 MAb, was generated. It belongs to the IgG1 isotype and is specific for the unique C-terminal domain of CKLFs protein. The specificity of this antibody has been assessed by enzyme-linked immunosorbent assay (ELISA) and Western blot. Our results demonstrated that intramuscular injection of naked DNA encoding CKLF1 gene combined with in vivo electroporation is an effective and simple method to raise MAbs that can be used for basic and clinical research of CKLF1 protein.

Abstract:Cytokines are small proteins that have an essential role in the immune and inflammatory responses. The repertoire of cytokines is becoming diverse and expanding. Here we report the identification and characterization of a novel cytokine designated as chemokine-like factor 1 (CKLF1). The full-length cDNA of CKLF1 is 530 bp long and a single open reading frame encoding 99 amino acid residues. CKLF1 bears no significant similarity to any other known cytokine in its amino acid sequence. Expression of CKLF1 can be partly inhibited by interleukin 10 in PHA-stimulated U937 cells. Recombinant CKLF1 is a potent chemoattractant for neutrophils, monocytes and lymphocytes; moreover, it can stimulate the proliferation of murine skeletal muscle cells. These results suggest that CKLF1 might have important roles in inflammation and in the regeneration of skeletal muscle.

Abstract:Chemokine-like factor 1 (CKLF1) exhibits chemotactic effects on leukocytes. Its amino acid sequence shares similarity with those of TARC/CCL17 and MDC/CCL22, the cognate ligands for CCR4. The chemotactic effects of CKLF1 for CCR4-transfected cells could be desensitized by TARC/CCL17 and markedly inhibited by PTX. CKLF1 induced a calcium flux in CCR4-transfected cells and fully desensitized a subsequent response to TARC/CCL17, and TARC/CCL17 could partly desensitize the response to CKLF1. CKLF1 caused significant receptor internalization in pCCR4-EGFP transfected cells. Taken together, CKLF1 is a novel functional ligand for CCR4.

Abstract:Objective:To amplify and sequence of the variable region genes of anti CD3 McAb.Methods:The V H?V L genes were amplified by RT PCR from total RNA that were extracted from WuT3 hybridoma.Recombinant cloning vector was constructed and sequenced after the enzyme digestion.Results:It showed that V H gene consisted of 363 bp encoding amine acid residues,belongs to mouse heavy chain subgroup IIB;V L gene consisted of 330 bp encoding amine acid residues,belongs to mouse κ light chain subgroup III.Comparing with Kabat database,the V H?V L genes were in agreement with the characterization of DNA sequences present in the mouse Ig V H?V L regions respectively.Conclusion:The success of cloning of the V H?V L genes of WuT3 McAb lay a good foundation for the construction and expression of chimeric antibody.

Abstract:OBJECTIVE:To prepare the polyclonal anti-peptide antibody against chemokine-like factor1 (CKLF1) and apply it to the expression and functional studies of CKLF1.METHODS:CKLF1 was analyzed with bioinformatics methods. The 16 amino acids sequence peptide was selected from CKLF1 C terminal end. Antibody was raised by immunizing rabbits with the peptide conjugated to keyhole limpet hemocyanin (KLH).RESULTS:A high titer polycolonal antibody was obtained from the rabbit against the peptide. ELISA analysis proved that the titer of rabbit serum against anti-peptide of CKLF1 was up to 10(-4). Western blot analysis revealed that it could react not only with recombinant CKLF1 expressed in a cell-Free Protein Biosynthesis System and Drosophila S2 cells, but also recognize the endogenous CKLFs in the tissue array. Positive staining was detected in the normal bronchial cartilage, gastric mucosa, and gastric smooth muscle tissues. Normal rectum and well-differentiated rectal carcinoma showed strong positive staining, but the poor-differentiated rectal carcinoma samples revealed negative staining.CONCLUSION:The anti-peptide antibody can specifically recognize CKLFs and may be a useful reagent for the detection of CKLF1.

Abstract:Leukocyte migration is an essential function of innate and adaptive immune responses. Chemokines and their receptors control the migration system. The abundance of chemokines is controlled by atypical chemokine receptors (ACKRs), chemokine receptor-like molecules that do not couple to the G protein signaling pathways. Among them, ACKR4 regulates dendritic cell migration by controlling the ligands and is involved in tumor development in mouse models. Because no anti-mouse ACKR4 (mACKR4) monoclonal antibody (mAb) for flow cytometry has been reported, this study aimed to develop a novel mAb for mACKR4. Among the established anti-mACKR4 mAbs, A4Mab-1 (rat IgG2b, kappa), A4Mab-2 (rat IgG2b, kappa), and A4Mab-3 (rat IgG2b, kappa) recognized mACKR4-overexpressed Chinese hamster ovary-K1 (CHO/mACKR4) by flow cytometry. The dissociation constant (K D) values of A4Mab-1, A4Mab-2, and A4Mab-3 for CHO/mACKR4 were determined as 6.0 × 10-9 M, 1.3 × 10-8 M, and 1.7 × 10-9 M, respectively. Furthermore, A4Mab-1 and A4Mab-2 could detect mACKR4 by western blotting. These results indicated that A4Mab-1, A4Mab-2, and A4Mab-3 help to detect mACKR4 by flow cytometry and western blotting and obtain the proof of concept in preclinical models.

Abstract:Chemokine-like factor 1(CKLF1) is a newly cloned cytokine with three RNA splicing isoforms. It has chemotactic activities on leukocytes and plays an important role in skeletal muscle regeneration. Here we have isolated two rat homologues of human chemokine-like factors by expressed sequence tag assembly, which are designated as rat chemokine-like factor 1 and 2 (rat CKLF1, CKLF2). The full-length cDNAs of rat CKLF1 and -2 contain 523 and 682 nucleotides and the open reading frames encoding 98 and 151 amino acids, respectively. Rat CKLF1 and -2 share about 54.1 and 59.6% homologies with human CKLF1 and -2 at the amino acid level; both rat CKLF1 and -2 contain a CX3C motif at their C-terminal regions while human CKLFs have a CC motif at the same regions. Rat CKLFs are highly expressed in testis, while human CKLFs have a broad expression spectrum across multiple tissues. Recombinant rat CKLF1 can be secreted into the cell culture supernatants and has chemotactic effects on neutrophils, macrophages and lymphocytes, which is similar to human CKLF1, while recombinant rat CKLF2 has weaker chemotactic effects on these cells. These findings show that rat CKLFs have similar bioactivity with human CKLFs, although they are different in tissue distribution and contain different characteristic motifs.

Abstract:TM4SF11 is only 102 kb from the chemokine gene cluster composed of SCYA22, SCYD1, and SCYA17 on chromosome 16q13. CKLF maps on chromosome 16q22. CKLFs have some characteristics associated with the CCL22/MDC, CX3CL1/fractalkine, CCL17/TARC, and TM4SF proteins. Bioinformatics based on CKLF2 cDNA and protein sequences in combination with experimental validation identified eight novel genes designated chemokine-like factor superfamily members 1–8 (CKLFSF1–8). CKLFSF1–8 form gene clusters; the sequence identities between CKLF2 and CKLFSF1–8 are from 12.5 to 39.7%. Most of the CKLFSFs have alternative RNA splicing forms. CKLFSF1 has a CC motif and higher sequence similarity with chemokines than with any of the other CKLFSFs. CKLFSF8 shares 39.3% amino acid identity with TM4SF11. CKLFSF1 links the CKLFSF family with chemokines, and CKLFSF8 links it with TM4SF. The characteristics of CKLFSF2–7 are intermediate between CKLFSF1 and CKLFSF8. This indicates that CKLFSF represents a novel gene family between the SCY and the TM4SF gene families.
Two C-terminal Peptides of Human CKLF1 Interact with the Chemokine Receptor CCR4.

Abstract:Human chemokine-like factor 1 (CKLF1) exhibits chemotactic effects on leukocytes. A previous study demonstrated that CKLF1 is a functional ligand for human CC chemokine receptor 4 (CCR4). In this study, N-terminal amino acid sequencing of secreted CKLF1 protein showed that it contains at least two peptides, C27 and C19. To examine whether C27 or C19 play a role via CCR4, C27 and C19 were chemically synthesized and analyzed by chemotaxis, calcium mobilization, and receptor internalization assays in CCR4-tranfected HEK293 cells or Hut78 cells. The chemotaxis assay showed that C27 could induce chemotaxis to CCR4-transfected HEK293 cells or Hut78 cells while C19 had weaker chemotactic activity, especially in Hut78 cells. C27- or C19-induced chemotaxis was abolished by pertussis toxin, suggesting the involvement of a Gi/o pathway. C27- or C19-induced chemotaxis was also inhibited by an antagonist of CCR4 that show good binding potency, excellent chemotaxis inhibitory activity and selectivity toward CCR4, suggesting that their chemotactic activity specifically involved CCR4. The chemotactic response of CCR4-tranfected HEK293 cells to C27 or C19 was markedly inhibited by preincubation with TARC/CCL17. TARC/CCL17 effectively desensitized the calcium mobilization induced by C27 or C19. Similarly, both of C27 or C19 also desensitized the calcium mobilization and chemotaxis of CCR4-tranfected HEK293 cells in response to TARC/CCL17, suggesting that they might interact with a common receptor. Both C27- and C19-induced clear internalization of CCR4-EGFP. These results confirm that the secreted peptides of CKLF1, C27 and C19, have functional activation via CCR4.

Abstract:OBJECTIVE:Acute monocytic leukemia (AML)-M5 is the common type of acute myeloid leukemias in children. Studies have shown that there are abundant lipopolysaccharide (LPS) receptor (designated as CD14) molecules on the cell membrane of M5 cells and they play an important role in the diagnosis of M5, since they can be recognized and bound by mouse-anti-human CD14 monoclonal antibody (McAb). However, mouse-originated antibodies are largely not suitable for clinical application due to the severe side effects, thus "humanized antibody" is desired. As the first step for developing humanized antibody, the construction and expression of single chain antibody (scFv) with functional protein are necessary. The present study aimed to express ZCH-7-2F9 ScFv (scFv(2F9)) in eukaryotic cells and obtain the scFv(2F9) protein with a high biological activity.METHODS:Four primers were synthesized to construct the eukaryotic expressional vector, which included SfiI and EcoRI enzyme cleaving site, 6 x His and stop code TGA sequences. scFv(2F9) gene was amplified through splicing by overlap extension (SOE) using the high fidelity Taq polymerase. Positive recombinants (pSectag2A/scFv(2F9)) were identified through enzyme cleaving and sequenced before expression and were transformed into Chinese hamster ovary (CHO) cells for expression. Western-Blot and flow cytometry (FCM) were carried out to determine the relative molecular mass (Mr) and binding activity of scFv(2F9).RESULTS:The cloned scFv(2F9) gene was identified to be functional by sequencing and expressing. The interested protein could be detected in the culture supernatant of transformed CHO cells with an Mr of 31 000. The blocking test showed that the positive cell percentages, the mean fluorescence intensity (MFI) and the peak of channel (peak Ch) were reduced by 90.02%, 63.30% and 63.38%, respectively after blocking with scFv(2F9), while those were 4.55%, 10.09% and 5.02% after blockage using the supernatant from the CHO cells transfected with blanked vector pSectag2A.CONCLUSIONS:The scFv(2F9) against human CD14 antigen was successfully expressed in eukaryotic cells and showed a high biological activity, which may be useful for the further studies on its humanized antibodies.
3A4, a New Potential Target for B and Myeloid Lineage Leukemias

Abstract:Antibody-targeting therapy has drawn great interests to the hematologists and oncologists. Many antibodies have been studied for their potential targeting for hematopoietic malignancies. A few have been proved to be very effective for patients with these diseases. However, more antibodies are needed for clinical use. CD45 and its isoforms may convey clinical potential in terms of targeting therapy. Zhejiang Children's Hospital (ZCH)-6-3A4 (3A4), a novel antibody that can recognize an isoform of CD45 has been found to react with restricted cell components in hematopoietic system, which may have the potential for targeting therapy. Herein, we conducted an in vitro study of our newly prepared antibody 3A4 using various cellular and immunocytological methods. The results showed that the antibody 3A4 (murine IgG1 kappa) was a new clone of anti-CD45RA. It could block the binding to an epitope of CD45RA recognized by a standard anti-CD45RA antibody (Clone name L48). The reactivity of the 3A4 to both fresh leukemia cells from patients and well-defined leukemia cell lines was largely similar to those of L48, but the former recognized more leukemia cells than the latter. Cytometric analysis after papain treatment showed that the internalization rate of the 3A4 antibody to the target cells was as high as 71.3% after incubation at 37 degrees C for 4 h, which was significantly higher than that of L48 (20.4%). The norcantharidin (NCTD)-conjugated immunotoxin (NCTD-3A4) was generated using an active ester method. The targeting inhibition rate on KG1a was as high as 61.10% after 96 h incubation in a dose-dependent manner, which was significantly higher than that (3.56%, P < 0.01) with 3A4-negative Nalm-6 cells. In conclusion, our new anti-CD45RA antibody 3A4 is probably a new target molecule of leukemia cells and holds a targeting therapeutic potential for hematopoietic malignancies, which warrants further development of this agent.
Functional Identification of the Stable Transfection C5ar Cell Line Molt-4

Abstract:The complement C5 anaphylatoxin receptor is a member of the seven transmembrane-spanning G protein-coupled receptor superfamily that signals through G alpha i and G alpha 16. C5aR is mostly expressed on neutrophils, macrophages and endothelial cells. C5a and C5aR interaction plays an important role in numerous biological effects such as in vivo cytokine storm which results in inflammatory damage. Considering the limitation of collection of human peripheral blood neutrophils and their short half life, the stably transfected cell line for studying the biological effects of C5aR is needed. In this study, we transfected C5aR gene into Molt-4 cell line and examined the function of ectopic C5aR. Our results showed stable expression of the C5aR in Molt-4 cell line and their interaction with human C5a induced ERKI/2 phosphorylation, Ca++ influx. This stable transfected cell line may provide a useful tool for studying signal pathways related to C5a and C5aR interplay and antibody development specific for C5aR.

Abstract:Antibody-targeting therapy has drawn great interests to the hematologists and oncologists. 3A4, a novel antibody recognizing human CD45RA antigen, is a new target molecule for leukemias and holds a therapeutic potential for myeloid lineage leukemias. However, murine antibodies cannot be safely used in patients because of their strong immune reaction, humanization of the antibodies interested will be an important development step for therapeutic purpose. The aim of this study was to engineer the mouse 3A4 and to investigate the biological activity of its chimeric form. The humanized antibody composed of the 3A4 single-chain fragment of variable region and the human IgG1 Fc region, which was named human-mouse chimeric antibody 3A4 (Hm3A4). The function and biological activities of Hm3A4 were characterized using a variety of biological approaches. The results showed that Hm3A4 retained a strong binding activity to its antigen and could significantly block the binding of parental 3A4 to the antigen. In vitro experiments revealed that Hm3A4 could kill the target cells through complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity function. In vivo, Hm3A4 showed efficient antileukemia activity outperforming the nontreated mice. In conclusion, the chimeric antibody has an excellent biological activity after humanization and holds targeting therapeutic potential for myeloid leukemia, which warrants further development of this agent.

Abstract:Objective B7-H4, a member of the B7 family of immunoregulatory receptors, may participate in the negative regulation of cell-mediated immunity. Aberrant B7-H4 expression is detected in some tumors and it plays a role in the occurrence and development of tumors. The aim of this study was to elucidate the functional and structural properties of B7-H4. Methods We developed a monoclonal antibody (mAb) against the extracellular domain of B7-H4 through immunization of Balb/c mice with 3T3-mB7-H4 cells which expressed extrinsic B7-H4. A stable hybridoma cell line was established. Then, we analysised the characterization of the mAb through Enzyme linked immunosorbent assay (ELISA), Immunoprecipitation (IP), western blotting, Immunohistochemical (IHC), and tested the biological activity of the mAb. Results ELISA, IP, and western blotting analyses indicated that the mAb specifically recognized B7-H4. In addition, flow cytometry demonstrated that the mAb exhibits excellent reactivity when applied to leukemic cells. IHC staining revealed that the mAb stained in a predominantly diffuse plasmalemmal or cytoplasmic pattern when applied to certain tumor tissues. The preliminary results of the mAb's biological activity showed that the mAb could effectively inhibit the function of B7-H4 in the inhibition of T cell, while promotingg the growth of T cells and the secretion of Interleukin-2 (lL-2), Interleukin-4 (IL-4), Interleukin10 (IL-10) and Interferon-γ (IFN-γ). Conclusion This mAb will be a valuable tool for the further investigation of B7-H4 function.

Abstract:AIM: A single chain antibody fragment, 9E4scFv, against human midkine(MK) was constructed and expressed in E. coli, and its affinity and character were studied in mice bearing tumor.METHODS: VH and VL genes were amplified from hyrbridoma cell 9E4, secreting monoclonal antibody, by RT-PCR, and connected by linker (Gly4Ser) 3 to form scFv gene, which was cloned into expression vector pET 30a (+) and finally expressed in E. coli. The expressed product 9E4scFv was purified by metal affinity chromatography, its purity and biological activity were determined using SDS-PAGE, ELISA and western blotting.RESULTS: SDS-PAGE analysis showed that the relative molecular weight of recombinant protein was 30kDa. ELISA assay revealed that 9E4scFv retained the immunoactivity of parent mAb, being capable of binding specifically to human gastric carcinoma cell line (BGC823) expressing associated antigen.CONCLUSION: 9E4scFv was successfully developed and might be used for humane tumor detection. The scFv may be a good carrier for tumor-imaging and tumour drugs.
Interactions of Peptides from Secreted Human CKLF1 and the N-Terminal Extracellular Tail of CCR4 Analyzed by CZE

Abstract:Human chemokine-like factor 1 (CKLF1) exhibits chemotactic effects on leukocytes. A previous study demonstrated that CKLF1 is a functional ligand for human CC chemokine receptor 4 (CCR4). The N-terminal amino acid sequencing of secreted CKLF1 protein showed that it contains at least two peptides, CKLF1-C27 and CKLF1-C19, which have functional activation via CCR4. To quantitatively evaluate the interaction of CKLF1-C27 or CKLF1-C19 with CCR4, the N-terminal extracellular tail of CCR4 (ML40), CKLF1-C27 and CKLF1-C19 were chemically synthesized and analyzed by capillary zone electrophoresis. Both qualitative and quantitative characterizations of the peptide-peptide binding were determined. We used the macrophage-derived chemokine (MDC) as the positive control and its binding constant was (4.99 ± 0.86) × 104 M−1. The binding constant of the interactions between CKLF1-C27/CKLF1-C19 and ML40 was calculated as (2.96 ± 0.59) × 104 M−1 and (1.39 ± 0.38) × 104 M−1 by the Scatchard analysis. This result proved that CKLF1-C27 had a greater potent affinity with ML40 than CKLF1-C19 because of the excess eight amino acids. To understand the molecular basis for the interaction, a mutagenesis study of CKLF1-C19 was undertaken. CKLF1-C19pm and CKLF1-C19km were synthesized and their interactions with ML40 were analyzed by CZE. We found that the substitution of Lys or Pro to Ala within the residues of CKLF1-C19 strongly abolished or decreased its interaction with ML40, suggesting that the Lys residues of CKLF1-C19 play the important role for the interaction and the Pro residues of CKLF1-C19 affect its affinity. All the experimental results show that the reported method by CZE for the determination of the interactions of CKLF1 peptides and the N-terminal extracellular tail of CCR4 is powerful.
Effects of Novel Human Chemokine-Like Factor 1 (CKLF1) on Bone Marrow Hematopoietic Stem Cell/progenitor Cell in Vitro.

Abstract:OBJECTIVE To explore the effects of a novel human chemokine-like factor 1 (CKLF1) on stem cell/progenitor cells. METHODS Human bone marrow mononuclear cells were separated by Ficoll (1.077 g/ml), and cultured in suspension and semisolid colony culture. The effects of CKLF1 on CFU-GM and CFU-Mix were observed. RESULTS The number of CFU-GM was significantly increased in 10 groups by addition of CKLF1 plasmid supernatant. The mean value was 234.81 +/- 98.71/1 x 10(5) cells, 2.42 fold of control group (P < 0.05). The mean value of CFU-Mix in groups of negative control, CKLF1, PHA, GM-CSF and G-CSF were 82.00 +/- 20.25, 105.76 +/- 36.70, 146.63 +/- 27.09, 143.33 +/- 60.23 per 1 x 10(5) cells, respectively, no statistical differences were seen between control and CKLF1 groups. The CD(34)(+) cells were detected by FACS. The average percentage in the groups of normal control, CKLF1, PHA and GM-CSF were (0.75 +/- 0.62)%, (1.32 +/- 0.87)%, (0.15 +/- 0.02)%, and (0.29 +/- 0.23)%, respectively. Compared with control, no significant differences were seen between each group (P > 0.05). CONCLUSION Novel chemokine-like factor 1 can increase the proliferation of CFU-GM, which indicated that CKLF1 could increase the proliferation of human bone marrow hematopoietic progenitor cells and colony formation.
