Abstract:OBJECTIVE:To study lysosomes involvement in the degradation of ricin A chain.METHODS:A lysosome-targeted singal KFERQ was added to the C terminus of rRTA by DNA recombinant technology. A pKK223.3 expression system in E. coli was used to produce recombinant ricine A chain (rRTA) and rRTA-KFERQ. Recombinant proteins were purified by affinity chromatography using Blue-Sepharose 6B. The cytotoxicity of recombinant proteins was measured by the MTT method.RESULTS:Recombinant RTA-KFERQ was 49.87%, 54.18% and 88.68% less cytotoxic than RTA itself on the three cell lines HEPG2, Hela and A549, respectively.CONCLUSION:Lysosomes can degrade, but not completely inactivate RTA in different cells, suggesting cells may have other degradation pathways for RTA.
A novel neutralizing monoclonal antibody against cell-binding polypeptide of ricin.

Abstract:One strain of neutralizing monoclonal antibody (MAb) against cell-binding polypeptide of ricin, named 3E1, was generated efficiently. The antibody recognized the linearity epitope of RTB located in a toxin structure domain characterized by Western blotting. The safe period of mice for intraperitoneal injection of 100 mu g of antibody was 20 min after intraperitoneal injection of 2 mu g of Ricin (10 times LD50). The neutralizing MAb we obtained could be developed into an immunotherapeutic agent to counteract the use of ricin as a terrorist or biological warfare weapon. It might be useful, as well, for antibody-based prophylaxis.

Abstract:Ricin enters the cells by receptor-mediated endocytosis, followed by translocation across the membranes of intracellular organelles. A trans-Golgi retention peptide signal YQRL was fused to the C-terminus of ricin A chain (RTA) by polymerase chain reaction. The recombinant RTA and RTA–YQRL were expressed in Escherichia coli using plasmid pKK223.3 under the control of a tac promoter. The recombinant proteins were purified by affinity chromatography on a Blue-Sepharose 6B column. The cytotoxicities of RTA and the fusion toxin RTA–YQRL were measured by the MTT assay in HeLa, SKOV-3, and WISH cells following fluid-phase endocytosis. The rRTA–YQRL was 2-, 10-, and 40-fold more cytotoxic than rRTA itself in the three cell lines, respectively. The results indicate that addition of a TGN retention signal YQRL to the C-terminus of RTA can markedly increase its cytotoxicity, suggesting TGN may play an important role in the intracellular routing and translocation of RTA.
Expression and purification of recombination ricin toxin A and its monoclonal antibody based sensitive ELISA

Abstract:Objective To construct a prokaryotic expression vector in which the maximum level of rRTA-Trx fusion protein was attained with homogeneous,soluble,and full bioactive,and to prepare anti-rRTA monoclonal antibody(mAb) and establish sandwich ELISA for ricin detection.Methods A pair of RTA special primer was designed based on computer-aided design,RNA secondary structure,and also Escherichia coli partialism codon.RTA gene was cloned from PUC19/RA to pET32a(+) by PCR.Prokaryotic expression vector pET32a(+)/rRA/JM109 was eatablished and identified by double enzyme digestion and DNA sequencing.After induction with IPTG,the rRTA protein was obtained,and then purified by Ni2+-NTA resin column and evaluated by ELISA and Western blot assays.An anti-RTA mAb was obtained by using hybridoma technique and identified by Western blot assay.A sandwich ELISA for ricin detection was also established.Results a prokaryotic expression vector pET32a(+)/rRA/BL21 was constructed.Recombinant RTA proteins with high biological activity were successfully produced in the pET32a vector expression system.After purification with Ni2+-NTA resin column,the extraction showed a yield of approximately 10-20 mg/L.An anti-RTA mAb was obtained with hybridoma technique and specific ELISA screening.Western blotting showed that the mAb had specific binding ability to RTA in reduced SDS-PAGE.A sandwich ELISA for detects ricin was established with a ricin detection limit of 3.125 ng/mL.Conclusion rRTA,anti-RTA mAb,and a sandwich ELISA for ricin detection have practical use in biotherapy against carcinoma and in prophylaxis,diagnosis and therapeutics against bioterror attack of in the future.
Conformation-Dependent High-Affinity Potent Ricin-Neutralizing Monoclonal Antibodies

Abstract:Ricin is a potential biothreat agent with no approved antidote available for ricin poisoning. The aim of this study was to develop potent antibody-based antiricin antidotes. Four strong ricin resistant hybridoma clones secreting antiricin monoclonal antibodies (mAbs) were developed. All four mAbs are bound to conformational epitopes of ricin toxin B (RTB) with high affinity (KD values from 2.55 to 36.27 nM). RTB not only triggers cellular uptake of ricin, but also facilitates transport of the ricin toxin A (RTA) from the endoplasmic reticulum to the cytosol, where RTA exerts its toxic activity. The four mAbs were found to have potent ricin-neutralizing capacities and synergistic effects among them as determined by an in vitro neutralization assay. In vivo protection assay demonstrated that all four mAbs had strong efficacy against ricin challenges. D9 was found to be exceptionally effective. Intraperitoneal (i.p.) administration of D9, at a dose of 5 μ g, 6 weeks before or 6 hours after an i.p. challenge with 5 × LD50 of ricin was able to protect or rescue 100% of the mice, indicating that mAb D9 is an excellent candidate to be developed as a potent antidote against ricin poisoning for both prophylactic and therapeutic purposes.
Monoclonal Antibody, Mab 4c13, an Effective Detoxicant Antibody Against Ricin Poisoning

Abstract:Ricin is a glycoprotein produced in castor seeds and consists of two polypeptide chains named Ricin Toxin A Chain (RTA) and Ricin Toxin B Chain (RTB), linked via a disulfide bridge. Due to its high toxicity, ricin is regarded as a high terrorist risk for the public. However, antibodies can play a pivotal role in neutralizing the toxin. In this research, the anti-toxicant effect of mAb 4C13, a monoclonal antibody (mAb) established using detoxicated ricin as the immunized antigen, was evaluated. Compared with mAb 4F2 and mAb 5G6, the effective mechanism of mAb 4C13 was analyzed by experiments relating to its cytotoxicity, epitope on ricin, binding kinetics with the toxin, its blockage on the protein synthesis inhibition induced by ricin and the intracelluar tracing of its complex with ricin. Our result indicated that mAb 4C13 could recognize and bind to RTA, RTB and exert its high affinity to the holotoxin. Both cytotoxicity and animal toxicity of ricin were well blocked by pre-incubating the toxin with mAb 4C13. By intravenous injection, mAb 4C13 could rescue the mouse intraperitoneally (ip) injected with a lethal dose of ricin (20μg/kg) even at 6h after the intoxication and its efficacy was dependent on its dosage. This research indicated that mAb 4C13 could be an excellent candidate for therapeutic antibodies. Its potent antitoxic efficiency was related to its recognition on the specific epitope with very high affinity and its blockage of protein synthesis inhibition in cytoplasm followed by cellular internalization with ricin.

Abstract:BACKGROUND AND AIM:Overexpression of the human epidermal growth factor receptor 2 (HER-2) protein has been detected in gastric cancer and has been associated with an unfavorable prognosis. We investigated the anti-cancer effects of anti-p185(HER-2) ricin A chain (RTA) immunotoxin, alone or in combination with 5-flurouracil on SGC7901-HER-2+ cells.METHODS:SGC7901-HER-2+ cells were obtained by transfecting SGC7901 cells with HER-2-pcDNA3.1. Anti-p185(HER-2)-RTA was prepared by chemical conjugation of anti-HER-2 monoclonal antibody (mAb) and RTA. The SGC7901-HER-2+ cells were incubated with RTA, anti-p185(HER-2)-RTA, and/or 5-flurouracil. The effects of drugs on cells were evaluated by MTT assay and Annexin V-fluorescein isothiocyanate and propidium iodide double staining flow cytometry. The expression of caspase-3, caspase-9, cyclooxygenase-2, and nuclear factor-kappaB/p65 were assayed by western blot. SGC7901-HER-2+ cells were transplanted into BALB/c nude mice to produce solid tumors in an attempt to study the immunotoxin activity in vivo.RESULTS:In vitro, anti-p185(HER-2)-RTA inhibited cell growth and induced apoptosis in SGC7901-HER-2+ cells. Anti-p185(HER-2)-RTA enhanced caspase-3 and caspase-9 activity, while downregulating the expression of cyclooxygenase-2 and nuclear factor-kappaB/p65. Its combination with 5-flurouracil further inhibited the growth of SGC7901-HER-2+ cells. In vivo, our data showed that anti-p185(HER-2)-RTA significantly inhibited the growth of SGC7901-HER-2+ cells-transplanted tumors.CONCLUSIONS:Anti-p185(HER-2)-RTA inhibits the growth of SGC7901-HER-2+ cells. The effect may be related to the activation of caspase-3 and caspase-9 and inhibition of cyclooxygenase-2 and nuclear factor-kappaB/p65. Anti-p185(HER-2)-RTA plus 5-FU enhance anti-cancer activity, suggesting useful clues for further study for the treatment of HER-2 positive gastric cancers.
A Novel Neutralizing Monoclonal Antibody Against Both Ricin Toxin A and Ricin Toxin B, and Application of A Rapid Sandwich Enzyme-Linked Immunosorbent Assay

Abstract:Two strains of neutralizing monoclonal antibodies (MAbs) anti-ricin, both B chain and A chain, named 3D74 and 4C13, were generated efficiently. The two antibodies recognized different epitopes located in a separated toxin structure domain characterized by enzyme-linked immunosorbent assay ( ELISA) and Western blotting. 3D74 recognized space conformation epitope, whereas 4C13 recognized linearity epitope of ricin. 4C13 possessed a novel neutralizing ability; the safe period for intraperitoneal injection of 100 mu g of antibody was 30 min after intraperitoneal injection of 2 mu g of ricin (10 times LD50). Using 3D74 and 4C13 as pair-matching antibodies, we established sandwich ELISA and rapid ELISA for detection of ricin. The detection sensitivities were 31.3 and 156 mu g/ml, respectively. Rapid sandwich ELISA was sufficiently sensitive and was performed easily for ricin detection without any special instruments.
Rational design, expression and biological activity assessment of a novel peptide based on ricin toxin antibody

Abstract:Objective To design and express a novel peptide based on ricin toxin antibody in E. coli, and to evaluate its biological activity. Methods Based on the crystal structure of ricin toxin A chain (RTA) and the RTA-rRNA interact in the complex model, the steric conformation of RTA was theoretical modeled and its functional domain was preliminarily determined. The humanized single-domain RTA antibody was designed rationally by computer-guidod molecular design method. Its coding sequence was ob- tained by overlapping extension PCR, and cloned into the pET-32a vector. The fusion protein was then ex-pressed in E. coli BL21 (DE3), identified by Western blot, and purified with Ni-NTA agarose. The binding and neutralizing activity of this novel peptide for riein was evaluated by competitive ELlSA assay and MTT assay. Results A recombinant human single-domain antibody expressing a polypeptide against RTA in the CDR3 loop was designed. The fusion protein was successfully expressed in E. coll. The purified protein can bind to ricin, and neutralize its activity in SP2/0 viability assay. Conclusion The success of the novel pep-tide based on riein toxin antibody provides a novel method to develop new generation of ricin antagonists.
A Novel Designed Single Domain Antibody on 3-D Structure of Ricin A Chain Remarkably Blocked Ricin-Induced Cytotoxicity

Abstract:Ricin A chain (RA), an N-glycosidase, is able to fatally disrupt protein synthesis by attacking the Achilles heel of the ribosome RNA (rRNA). As specific immunotoxins, emergence of inhibitors for RA may obtain access to antagonistics against ricin intoxication and contribute to ameliorate the concomitant side effects. Many experimental results showed that the engineered VHs, which possessed solubility, stability, small size and consequently easier to express, purify and manipulate in vitro, were self and long-lived molecules compared to synthetic peptides. In this study, based on the crystal structure of RA, a novel recombinant human single-domain antibody expressing a polypeptide against RA in the CDR3 loop (named rVHPT) was obtained using computer-guided molecular design method. Theoretically, rVHPT could penetrate deeply into the active cleft of RA and act as a potent antagonist analogue to block the RA–rRNA interaction. Followed results showed that the recombinant VHPT was easily expressed of high-yield production and in a partially soluble fusion form in Escherichia coli. In vitro cytotoxicity experiments demonstrated that it possessed remarkable ability to block ricin-induced cytotoxicity. This study highlights the potential of human VH to display biostructure and biofunction of peptides designed on RA functional domain and could be useful in developing new antidotes with potential therapeutic uses to neutralize unintended exposure to ricin.

Abstract:So far, no specific therapeutic agent is available for the treatment of ricin intoxication. Here, V(H) and V(L) genes were cloned from a hybridoma cell line secreting anti-ricin mAb 4C13, which could neutralize the toxicity of ricin. A chimeric antibody, c4C13, containing 4C13 mAb variable region genes fused to human constant region genes (gamma 1, kappa), was constructed. C4C13 retained the binding activity and recognized the same, or a closely related, epitope as the original mouse antibody. Furthermore, c4C13 blocked ricin-induced cytotoxicity to SP2/0 cells. Compared with its parental mouse antibody, c4C13 will be safer when used in human body to reverse clinical ricin intoxication.
BCMab1-Ra, a Novel Immunotoxin That BCMab1 Antibody Coupled to Ricin A Chain, Can Eliminate Bladder Tumor.

Abstract:Bladder cancer is one of the most common malignancies. However, there is no ideal therapy to cure bladder cancer so far, especially invasive carcinoma. Here, we developed a new antibody-based drug BCMab1-Ra, which was generated by conjugation of BCMab1 (a new monoclonal antibody that specifically recognized the aberrantly glycosylated Integrin a3b1 in bladder cancer) with the ricin A chain (Ra). A patient with multiple bladder cancer received intravescical administration of BCMab1-Ra treatment as a volunteer. After 30 weeks of treatment, no tumor was observed by cystoscope examination. We did not observe any local or systemic side effects. Human anti-mouse antibody (HAMA) was not detectable in the circulation. Results follow-up showed no tumor had been found in every half year review in 3 years.
Anti-Ricin Protective Monoclonal Antibodies

Abstract:Development of anti-ricin protective monoclonal antibodies (mAbs) started in the early 1980s. Much progress has been made since then. Antibodies possess great potential for development as antidotes against toxins. These can be used either prophylactically to prevent, or therapeutically to treat, toxin-mediated intoxications in an emergency situation. Unlike many other therapeutic products, antibodies offer unique and high target specificity, and long half-life in serum. There are several mAbs which are currently in the discovery stage for medical countermeasures against ricin intoxication. This review summarizes these mAbs, including their anti-ricin mechanism, generation, and efficacies in vivo.

Abstract:Ricin and its A chains can be used to conjugate with monoclonal antibodies to prepare immunotoxins. Ricin A chain (RTA) and its modification RTA-KDEL (ER-retrieval signal) were expressed with the pKK223.3 system in Escherichia coli under control of a tac promoter. The recombinant proteins can be purified by one-step affinity chromatography on a column of Blue-Sepharose 6B. The toxicities of RTA and its mutant RTA-KDEL were evaluated by the MTT assay in HeLa, MCF, and ECV-304 cells following fluid-phase endocytosis. RTA-KDEL was somewhat more cytotoxic than RTA itself in the different cell lines. The results suggest that rRTA-KDEL may be useful for the synthesis of more potent immunotoxins.
Activity of Recombinant Toxin Ricin A Chain and Several Wild Type Single Chain Ribosome Inactivating Proteins

Abstract:Recombinant ricin toxin A chain(rRTA)is one of the important elements used for constructing immunotoxin,which may be useful as a treatment of many diseases.To obtain rRTA,ricin toxin A chain gene was cloned into pExSecⅠ vector and expressed in E.coli BL21(DE3),the proportion of expressed rRTA in total bacteria protein was about 20%.The expression products obtained a purity of 80% or so by a CM Sepharose column.Recombinant ricin toxin A chain,and several wild type single chain ribosome inactivating proteins,were analyzed by cleavage assay of supercoiled DNA and 3 H Leu labeled protein synthesis inhibition assay.The results suggest that rRTA had the similar activity as the others while there had no linear correlation between the two methods.
Structure-Based Affinity Maturation of A Chimeric Anti-Ricin Antibody C4c13

Abstract:Ricin is a highly lethal toxin. Anti-ricin chimeric monoclonal antibody (mAb) C4C13 was prepared in our lab; however, its binding affinity was much weaker than that of the parent antibody 4C13. In this study, based on the computer-guided homology modeling and conformational optimization methods, the 3-D structure of C4C13 variable regions Fv was constructed and optimized. Using molecular docking and dynamics simulation methods, the 3-D complex structure of ricin and C4C13 Fv was obtained. Considering the orientation property, surface electrostatic distribution, residues chemical andphysical character and intermolecular hydrogen bond, the binding mode and key residues were predicted. According to C4C13 Fv fragment and ricin complementary binding surface, electrostatic attraction periphery and van der Waals interaction interface, three mutants (i.e., M1 (NH102F, WH103Y); M2 (WH103Y) and M3 (RL90G)) were designed, in which M1 and M2 were predicted to possess higher antigen-binding activity than C4C13, while M3 was weaker. The relative affinity assays by ELISA showed that M1 and M2 mutations had higher affinity (9.6 and 18.3 nmol/L) than C4C13 (130 nmol/L) and M3 had weaker affinity (234.5 nmol/L) than C4C13. The results showed that the modeling complex structure of the antigen (ricin) and antibody (C4C13) is reasonable. Our work offered affinity maturated antibodies by site mutations, which were beneficial for valuable anti-ricin antibody design and preparation in future.
Strong protection against ricin challenge induced by a novel modified ricin A-chain protein in mouse model

Abstract:Ricin toxin (RT) is an extremely potent toxin derived from the castor bean plant. As a possible bioterrorist weapon, it was categorized as a level B agent in international society. With the growing awareness and concerns of the "white powder incident" in recent years, it is indispensable to develop an effective countermeasure against RT intoxication. In this study we used site-directed mutagenesis and polymerase chain reaction (PCR) techniques to modify the gene of ricin A-chain (RTA). As a result, we have generated a mutated and truncated ricin A-chain (mtRTA) vaccine antigen by E.coli strain. The cytotoxicity assay was used to evaluate the safety of the as-prepared mtRTA antigen, and the results showed that there was no residual toxicity observed when compared to the recombinant RTA (rRTA) or native RT. Furthermore, BALB/c mice were subcutaneously (s.c.) vaccinated with mtRTA 3 times at an interval of 2 weeks, and then the survivals were evaluated after intraperitoneal (i.p.) or intratracheal challenge of RT. The vaccinated mice developed a strong protective immune response that was wholly protective against 40 × LD50 of RT i.p. injection or 20 × LD50 of RT intratracheal spraying. The mtRTA antigen has great potential to be a vaccine candidate for future application in humans.

A Biparatopic Intrabody Renders Vero Cells Impervious to Ricin Intoxication

Abstract:Expression of camelid-derived, single-domain antibodies (VHHs) within the cytoplasm of mammalian cells as intrabodies has opened-up novel avenues for medical countermeasures against fast-acting biothreat agents. In this report, we describe a heterodimeric intrabody that renders Vero cells virtually impervious to ricin toxin (RT), a potent Category B ribosome-inactivating protein (RIP). The intrabody consists of two structurally defined VHHs that target distinct epitopes on RTs enzymatic subunit (RTA): V9E1 targets RTAs P-stalk recruitment site, and V2A11 targets RTAs active site. Resistance to RT conferred by the biparatopic VHH construct far exceeded that of either of the VHHs alone and effectively inhibited all measurable RT-induced cytotoxicty in vitro. We propose that targeted delivery of bispecific intrabodies to lung tissues may represent a novel means to shield the airways from the effects of inhalational RT exposure.

Abstract:3048 Background: The receptor tyrosine kinase RON is critical in tumorigenesis and a validated drug target for cancer therapy. Here we report the development and therapeutic efficacy of anti-RON mAb Zt/g4-maytansinoid conjugates (Zt/g4-DM1) with enhanced cytotoxicity against colorectal cancer cells (CRC) in mouse xenograft tumor models. Methods: Zt/g4 (IgG1a/κ) was selected as the drug carrier for its ability to induce RON endocytosis in CRC cells. Zt/g4 was conjugated to DM1 via the thioether link to form antibody-drug conjugate Zt/g4-DM1 with an antibody-drug ratio of 1:4 molecules. CRC HCT116, HT29, and SW620 cells were used as the model for in vitro and in vivo studies. Results: Zt/g4-DM1 retains the intrinsic activity that causes rapid RON internalization. Targeted payload delivery induced cell cycle arrest at G2/M phase, reduced cell viability with IC50 values at ~1.2 μg/ml, which results in massive cell death within a period of 72h. Abstract:Ricin toxin's enzymatic subunit (RTA) has been subjected to intensive B cell epitope mapping studies using a combination of competition ELISAs, hydrogen exchange-mass spectrometry and X-ray crystallography. Those studies identified four spatially distinct clusters (I-IV) of toxin-neutralizing epitopes on the surface of RTA. Here we describe A9, a new single domain camelid antibody (VHH) that was proposed to recognize a novel epitope on RTA that straddles clusters I and III. The X-ray crystal structure of A9 bound to RTA (2.6 Å resolution) revealed extensive antibody contact with RTA's β-strand h (732 Å2 buried surface area; BSA), along with limited engagement with α-helix D (90 Å2) and α-helix C (138 Å2). Collectively, these contacts explain the overlap between epitope clusters I and III, as identified by competition ELISA. However, considerable binding affinity, and, consequently, toxin-neutralizing activity of A9 is mediated by an unusual CDR2 containing five consecutive Gly residues that interact with α-helix B (82 Å2), a known neutralizing hotspot on RTA. Removal of a single Gly residue from the penta-glycine stretch in CDR2 reduced A9's binding affinity by 10-fold and eliminated toxin-neutralizing activity. Computational modeling indicates that removal of a Gly from CDR2 does not perturb contact with RTA per se, but results in the loss of an intramolecular hydrogen bond network involved in stabilizing CDR2 in the unbound state. These results reveal a novel configuration of a CDR2 element involved in neutralizing ricin toxin.
