Abstract:A novel protocol for the facile synthesis of chiral 3-substituted 1,3,4,5-tetrahydrobenzo[e] [1,4]diazepin-2-ones via a domino reaction has been developed, which involves a cascade of S(N)2 nucleophilic substitution and copper-catalyzed C-N coupling reactions of a-amino acid amides and orthohalobenzyl halides. This protocol has some merits, such as one-pot process, easy operation, a wide scope of substrates. Furthermore, no heavy racemization occurs during the reaction.

Abstract:We describe herein an intriguing method for the synthesis of biaryl aldehydes bearing both axial and central chirality through a desymmetric [3 + 2] cycloaddition reaction of activated isocyanides with prochiral biaryl dialdehydes under silver catalysis. This protocol features excellent enantioselectivity, 100% atom economy, good functional group compatibility, and operational simplicity.

Abstract:The sodium salts of(S)-2-(1-imidazolyl)alkanoic acids(2) were prepared by the cyclization of natural amino acids with formaldehyde aqueous solution and glyoxal aqueous solution under basic condition.The sodium salts(2) were esterified to form(S)-2-(1-imidazolyl)alkanoates(3a~3g).The(S)-2-(1-imidazolyl)alkanols(4a~4e) were obtaind by reduction of 3a~3e via NaBH_(4) with satisfactory yields.The structures of the compounds(3a~3g,4a~4e) were confirmed by IR,~(1)H NMR,MS,and elemental analysis.

Abstract:2-Trifluoromethyl and 2-difluoromethyl substituted benzimidazole, benzoxazole and benzothiazole derivatives were efficiently prepared through a one-pot reaction of trifluoroacetic acid and difluoroacetic acid, respectively, with commercially available o-phenylenediamines, 2-aminophenols, and 2-aminobenzenethiols in good to excellent yields. Subsequential bromination of 2-difluoromethyl groups by photolysis with NBS led to the formation of bromodifluoromethyl benzo-1,3-diazoles which may be utilized to prepare the new generation of gem-difluoromethylene linked identical or non-identical twin molecules for drug synthesis.
Regioselective Synthesis of Acyclic Cis-Enediynes Via an Acid-Catalyzed Rearrangement of 1,2-Dialkynylallyl Alcohols. Syntheses, Computational Calculations, and Mechanism

Abstract:A novel synthesis of acyclic cis-enediynes 2 has been established by an acid-catalyzed rearrangement of 1,2-diyn-2-propen-1-ols 1 possessing a C-3-aryl group in the presence of water, alcohols, or thiols. Reactivity of allyl alcohols and regio- and cis/trans diastereoselectivity of the allylic migration were examined. In the presence of(+/-)-10-camphorsulfonic acid (CSA), the parent allyl alcohol 5 and the C-3-methyl-substituted 9 failed to give enediynes, whereas the C-3-aryl-substituted 12 and 29 underwent the allylic rearrangement to provide predominantly cis-enediynes 16 and 31 at room temperature or below. Under similar acidic conditions, enediyne alcohol 13 produced 16b and 16d with the same regio and cis/trans diastereoselectivity observed for 12. Allyl alcohol 30, an isomer of 29, also provided enediynes 31c and 32c after a prolonged reaction (90 h) at room temperature in the presence of CSA and EI;OH. These results suggested that the same allylic cations were obtained from allyl alcohols 12 and 13 or 29 and 30 even though the ease of ionization differed for each substrate. Involvement of allylic cations in the product-forming step was confirmed by the finding that chiral allyl alcohols (-)-12 and (-)-18c furnished racemic products. In general, the p-MeOPh-substituted allyl alcohol 29 gave a better regioselectivity than the Ph-substituted 12. In the reactions with alcohols, the regioisomeric ratios were 100:0 (31:33) for 29 and ca. 96:4 (16:17) for 12; the ratios decreased to ca. 90:10 (31:33) for 29 and ca. 10:30 (16:17) for 12 when thiols were used. The cis/trans diastereoselectivity is higher for allyl alcohol 12 (100% for 16 at 20 degrees C) compared to that for 29 (31:32 = 80:20-94:6 at 0 degrees C). Computational calculations at the RHF/3-21G level, carried out on the model compounds and allylic cations, indicated that nucleophilic trapping takes place preferentially at the Cg carbon to form the thermodynamically much more stable enediynes. Under the best reaction conditions (1 equiv of CSA and 2 equiv of EtOH in CH2Cl2, 20 degrees C), a number of acyclic cis-enediynes can be synthesized in three steps from the commercially available alpha-bromocinnamaldehyde (10).

Abstract:A new procedure for the highly diastereoseleetive synthesis of (1S)-trans-cyelopropane derivatives using a fructose derivative as the chiral auxiliary was developed. Chloroacetylated 1,2;4,5-di-O-isopropylidene-D-fructopyranose was reacted with a tertiary amine to form the ammonium salt, which was treated with cesium carbonate to give the cyclopropane derivative through ylide intermediate. The described procedure provides an efficient method to synthesize optically pure di- or poly-substituded cyclopropanes.

Abstract:We report, herein, aminocatalytic coupling of α-substituted acrylaldehydes with α-diazoesters, leading to chemoselective C–H insertion or cyclopropanation depending on α-substituents of diazoesters. A chiral primary–secondary diamine catalyst derived from <span class="smallcaps smallerCapital">l</span>-<i>tert</i>-leucine enabled the efficient promotion of both C–H insertion and cyclopropanation pathways in good yields and high enantioselectivities at room temperature without any metal or a Lewis acid cocatalyst. Abstract:The systematic synthesis of a series of N-acyl chloroformamidines is achieved by reaction of carbodiimides with acid chlorides; the structure of an N-acyl chloroformamidine was confirmed by X-ray diffraction. The reactivity of N-acyl chloroformamidines towards diethyl malonate, phenylethynyllithium, pentane-2,4-dione, methyl acetoacetate, water, 1,3-dimethylurea, 1,3-dimethylthiourea, and sodium hydrosulfide, resulted in unique protocols for the synthesis of N-acyl-substituted 1,1-diaminoethylenes, amidines, ureas, and thioureas, respectively. These results show that N-acyl chloroformamidines have a good ability to accept nucleophilic attack for the construction of useful N-containing compounds.

Abstract:A novel chiral diallylic diamine has been synthesized through double addition of allyl zinc bromide to chiral imines in this contribution. Firstly, (S)-N-Boc-2-amino-3-phenylpropanaI was prepared through N-Boc protection and Dess-Martin oxidation. Then, it was condensed with (S)-valine methyl ester to form a chiral imine, which was reacted with allyl zinc bromide in the presence of CeC1(3)center dot 7H(2)O to afford the corresponding chiral homoallylic amine with high optical activity. After removing Boc group, the newly formed chiral homoallylic amine was condensed with 2-thiophenecarboxaldehyde to provide a novel chiral imine with high optical purity. The following asymmetric addition of allyl zinc bromide to the novel chiral imine provided the target compound. The structure and the purity of desired compound and the key intermediates were demonstrated by H-1 NMR, C-13 NMR and MS spectra.

Abstract:An efficient approach to access functionalized 1,4- and 1,5-amino alcohols has been developed through the nucleophilic addition of semicyclic N,O-acetal with organozinc reagents. A number of substituted benzyl zinc reagents (including nitrile and ester substituted) could react with semicyclic N,O-acetals 1 and 2, affording the desired products 3a-3p and 4a-4o in good to excellent yields.

Abstract:Pseudo-C2-symmetric dodecaheterocyclic structures, which possess two acyl/aroyl groups disposed on either a cis- or trans-relative configuration, were prepared from the naturally occurring (-)-(1R)-myrtenal. Addition of Grignard reagents (RMgX) to the diastereoisomeric mixture of these compounds unexpectedly showed that nucleophilic additions to the two prochiral carbonyl centers gave the same stereochemical result in both cis/trans diastereoisomers, making unnecessary the separation of this mixture. Noticeably, both carbonyl groups showed different reactivity because one of them is attached to an acetalic carbon and the other to a thioacetalic carbon. Furthermore, addition of RMgX to the carbonyl attached to the former carbon takes place through the re face, while addition to the second one proceeds through the si face, thus affording the corresponding carbinols in a highly diastereoselective process. This structural feature allowed the sequential hydrolysis of both carbinols, yielding separately (R)- and (S)-1,2-diols after reduction with NaBH4. The mechanism of the asymmetric Grignard addition was explained by density functional theory calculations. This approach contributes to the development of the divergent synthesis of structurally and/or configurationally different chiral molecules.
