Abstract:We describe herein an intriguing method for the synthesis of biaryl aldehydes bearing both axial and central chirality through a desymmetric [3 + 2] cycloaddition reaction of activated isocyanides with prochiral biaryl dialdehydes under silver catalysis. This protocol features excellent enantioselectivity, 100% atom economy, good functional group compatibility, and operational simplicity.

Abstract:A series of planar chiral imidazolium salts derived from [2.2]paracyclophane have been synthesized and characterized. By using these imidazolium salts as carbene precursors, the Rh-catalyzed 1,2-addition of arylboronic acids to aldehydes proceeded readily with low catalyst loadings (0.03–0.3 mol %) and gave a variety of chiral diarylmethanols in excellent yields and moderate enantioselectivities.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:The asymmetric B & uuml;chner reaction and related arene cyclopropanations represent one type of the powerful methods for enantioselective dearomatization. However, examples of asymmetric B & uuml;chner reactions via a non-diazo approach are quite scarce, and the related arene cyclopropanation based on alkynes has not been reported. Herein, we disclose an asymmetric B & uuml;chner reaction and the related arene cyclopropanation by copper-catalyzed controllable cyclization of N-propargyl ynamides via vinyl cation intermediates, leading to chiral tricycle-fused cycloheptatrienes and benzonorcaradienes in high yields and enantioselectivities. Importantly, this protocol represents an asymmetric arene cyclopropanation reaction of alkynes and an asymmetric B & uuml;chner reaction based on vinyl cations. Asymmetric B & uuml;chner reactions are an effective methodology to bring about synthetic complexity from the common benzene motif, but effective, general catalytic methods are scarce. Here, the authors present divergent asymmetric arene dearomatization from ynamides using copper catalysis, wherein the catalytic cycle proceeds via a putative vinyl cation intermediate.

Abstract:An asymmetric intermolecular, three-component radical-initiated dicarbofunctionalization of 1,1-diarylalkenes with diverse carbon-centered radical precursors and electron-rich heteroaromatics by a copper(I) and chiral phosphoric acid cooperative catalysis strategy has been developed, providing straightforward access to chiral triarylmethanes bearing quaternary all-carbon stereocenters with high efficiency as well as excellent chemo- and enantioselectivity. The key to success is not only the introduction of a sterically demanding chiral phosphoric acid to favor radical difunctionalization over the otherwise remarkable side reactions but also the in situ generation of carbocation intermediates from benzylic radical to realize asymmetric induction with the aid of a removable hydroxy directing group via cooperative interactions with chiral phosphate. Density functional theory calculations elucidated the critical chiral environment created by the hydrogen-bonding and ion-pair interactions between the chiral phosphoric acid catalyst and substrates, which leads to the enantioselective C-C bond formation.

Abstract:A hitherto unknown class of C4-symmetric Caryl−Cβ (C3, C8, C13, C18) axially chiral porphyrins has been synthesized and the application of their iridium (Ir) complexes in catalytic asymmetric C(sp3)−H functionalization is documented. Cyclotetramerization of enantioenriched axially chiral 2-hydroxymethyl-3-naphthyl pyrroles under mild acidic conditions affords, after oxidation with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ), the C4-symmetric α,α,α,α-atropenantiomer as an only isolable diastereomer. Both regioisomeric Ir(Por*)(CO)(Cl) complexes catalyze the carbene C−H insertion reaction affording the same enantiomer, albeit with slight difference in enantioselectivity. With the optimum Ir-complex 3e, the 2-substituted arylacetic acid derivatives were generated from diazo compounds and cyclohexadiene in excellent yields and enantioselectivities.

Abstract:The efficient asymmetric preparation of the C-2-symmetric chiral 1,4-diamine, (1R,2S,4R,5S)-1,4-diamino-2,5-dimethylcyclohexane (5), and its salen ligands is described. With the Mitsunobu reaction as the key step, the overall yield of the four-step synthesis of 5 is 45%. The enantioselective alkynylation of meso-epoxides catalyzed by chiral gallium complexes is also achieved.

Abstract:Chiral C-2-symmetric diamines have emerged as versatile auxiliaries or ligands in numerous asymmetric transformations. Chiral 2,2'-bispyrrolidine-based salan ligands were prepared and applied to the asymmetric aryl transfer to aldehydes with arylboronic acids as the source of transferable aryl groups. The corresponding diarylmethanols were obtained in high yields with moderate to good enantioselectivitives of up to 83% ee.

Abstract:4-Aryl-3,4-dihydrocoumarins are one of the most important structural motifs. Herein, we disclose an enantioselective N -Heterocyclic Carbene catalyzed β-arylation/cyclization of α-bromoenals with 3-aminophenols under mild conditions. The protocol allows for the rapid preparation of 4-aryl-3,4-dihydrocoumarins in acceptable yields with good enantioselectivities. The products of this reaction could be converted into chiral diarylpropanoic acid derivatives without erosion of the enantioselectivity.

Abstract:The Cu-catalyzed 1,4-protosilylation and protoborylation of trifluoromethyl-substituted conjugated enynes were developed to access functionalized homoallenylsilanes and homoallenylboronates. This protocol also provides a general method to synthesize optically active homoallenylsilanes and homoallenylboronates in moderate to excellent yields with high enantiomeric excess by using new designed chiral bisoxazoline ligands. Simultaneously, the transformations of homoallenylsilanes and homoallenylboronates were also explored to synthesize useful building blocks.

Abstract:Herein, we report the rhodium-catalyzed domino arylation/cyclization reaction of unactivated internal alkynes to afford the optically active indenols in excellent yields (up to 92%) and extremely high enantioselectivities (up to 99% ee) in the presence of new amide substituted chiral diene ligands. Notably, a total of 21 new chiral diene ligands are synthesized, which are potential choices for other asymmetric reactions.

Abstract:CuBr and ZnI2 have been developed as catalysts or subcatalysts for the efficient asymmetric synthesis of axially chiral allenols with up to 97% ee from readily available propargylic alcohols, aliphatic or aromatic aldehyde, pyrrolidine, and commerically available ligands. The alcohol unit in the terminal alkynes plays a very important role for ensuring high enantioselectivity via coordination.

Abstract:A new C 2 symmetric chiral Schiff base compound A with relatively flexible chiral environment was synthesized from L (+) tartaric acid. Its structure was determined by NMR, IR, MS etc, and its preponderant conformation was modeled by CS Chem 3D Program. The results showed that its structure is not in a plane and has a big chiral cavity. It was showed that the complex B has different catalytic performance in asymmetric cyclopropanation of styrene and DMHD (2,5 dimethyl 2,4 hexadiene) with diazoacetate. The catalytic mechanism was also discussed.

Abstract:A highly efficient asymmetric arylation of cyclic N-sulfonyl aldimines with arylboronic acids catalyzed by a rhodium/sulfur-olefin ligand complex under mild conditions is described. A wide range 4-aryl-3,4-dihydro-1,2,3-benzoxathiazine 2,2-dioxides were obtained in extremely high yields (93-99%) and high enantiomeric purities (97-99% ee).

Abstract:A dirhodium(II)/phosphine catalyst with a chiral environment at the bridging site was developed for the asymmetric arylation of phenanthrene-9,10-diones with arylboronic acids. In contrast to the classic chiral bridging carboxylic acid (or derivatives) ligand strategy of bimetallic dirhodium(II) catalysis, in this reaction, tuning both axial and bridging ligands realized the first Rh2(OAc)4/phosphine-catalyzed highly enantioselective carbonyl addition reaction. The kinetic analysis reveals that dirhodium(II) and arylboronic acid follow the first-order kinetics, while phenanthrene-9,10-dione is zeroth-order. These data supported the proposed catalytic cycle, where the key intermediate in the rate-determining step involved the dirhodium(II) complex and arylboronic acid. Finally, axially chiral biaryls were prepared based on a newly developed oxidative ring-opening reaction of α-hydroxyl ketones with a base and molecular oxygen, which featured a central-to-axial chirality transfer radical β-scission step.

Abstract:Asymmetric diastereodivergent catalysis is a powerful strategy for accessing different stereoisomers of a molecule, which are important in the discovery of biologically and pharmacologically active candidates. While the asymmetric diastereodivergent synthesis of acyclic molecules has seen recent rapid development, the corresponding asymmetric diastereodivergent cycloaddition reactions have received comparatively less attention. Herein, we report the asymmetric diastereodivergent synthesis of chromanes with C2,C3-contiguous tetrasubstituted stereocenters via chiral phosphoric acid (CPA)-catalyzed [4 + 2] cycloadditions of ortho-quinone methides with γ-silyl-substituted allenones. A wide range of C3-epimeric chromanes were generated with high to excellent diastereoselectivities and enantioselectivities, starting from the same substrates but with modifications of the CPA catalysts. The diverse derivatizations of these chiral chromane stereoisomers into complex scaffolds illustrate the utility and value of this method.

Abstract:A palladium-catalyzed enantioselective decarboxylative allylic alkylation of α-benzyl cyanoacetates with methylene cyclic carbamates has been successfully developed. An array of enantioenriched products bearing nitrile-containing acyclic quaternary carbon stereocenters were obtained in high yields with good enantioselectivities (up to 99% yield and 93% ee). This method provides a new tactic for the construction of chiral nitrile-containing acyclic quaternary carbon stereocenters.

Abstract:AbstractDespite much progress has been made in the asymmetric α‐arylation reactions of cyclic ketones, lactones and lactams, the enantioselective α‐arylation of acyclic carbonyl compounds lagged much behind due to the in situ generated Z/E‐enolate intermediates leading to opposite enantiomers. Especially, the asymmetric α‐arylation of acyclic aldehydes is a long‐standing challenge, because of the highest activity among carbonyl compounds leading to various competitive side reactions. Herein we reported an efficient Pd‐catalyzed asymmetric intermolecular α‐arylation reaction of α‐alkyl‐α‐aryl disubstituted aldehydes with aryl bromides, which provides a rapid access to chiral aldehydes bearing an α‐all‐carbon quaternary stereocenter in moderate to good yields with good er in most cases. In addition, a pair of enantiomers could be easily prepared with the use of the same ligand by exchanging the aryl groups of aldehyde and aryl bromide.
