Abstract:We have developed a versatile and efficient method for copper-catalyzed synthesis of both 2-amino-4(3H)-quinazolinone and 2-aminoquinazoline derivatives. The protocol uses readily available substituted 2-halobenzoic acids, 2-bromobenzaldehyde, 2-bromophenyl ketones and guanidines as the starting materials, inexpensive copper(I) iodide as the catalyst, and the method has important application values for construction of N-heterocycles in organic chemistry and medicinal chemistry.

Abstract:An efficient and practical copper-catalyzed one-pot method for synthesis of tetrahydroisoquinolino[2,1-a]quinazolinone derivatives has been developed, and the corresponding target products were prepared in moderate to good yields. The one-pot approach underwent sequential copper-catalyzed N-arylation, intramolecular aerobic oxidative cyclization, and the newly synthesized products provided diverse structures for screening of biological molecules.

Abstract:A highly efficient method for the facile access of isoquinolines and isoquinoline N -oxides via a Cu( I )-catalyzed intramolecular cyclization of ( E )-2-alkynylaryl oxime derivatives in water has been developed. This protocol was performed under simple and mild conditions without organic solvent, additives or ligands. By switching on/off a hydroxyl protecting group of oximes, the selective N–O/O–H cleavage could be triggered, delivering a series of isoquinolines and isoquinoline N -oxides, respectively, in moderate to high yields with good functional group tolerance and high atom economy. Moreover, the practicality of this method was further demonstrated by the total synthesis of moxaverine in five steps.

Abstract:A convenient and efficient copper-catalyzed cascade method has been developed for synthesis of benzimidazoisoquinoline derivatives via reactions of readily available substituted 2-(2-halophenyObenzoimidazoles with alkyl cyanoacetates under mild condition, and the corresponding alkyl 6-aminobenzimidazo[2,1-a]isoquinoline-5-carboxylates were obtained in good to excellent yields. The novel method provides diverse and useful poly N-heterocyclic compounds for combinatorial chemistry and medicinal chemistry.

Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:A new copper-catalyzed oxidative cyclization protocol was developed for the synthesis of 5,6-dihydropyrrolo[2,1-a]isoquinolines via a three-component reaction of tetrahydroisoquinolines with bromoketones and electron-deficient alkenes with air as a terminal oxidant. A variety of functional groups survived under the reaction conditions and the target products were obtained in good yields. This reaction features such advantages as eco-friendly reaction conditions, a simplified operation process and a broad substrate scope.
