Abstract:An unexpected process from tetrahydrodiazirino[3,1-a]isoquinolines and diversified sulfonyl chlorides RSO2Cl (R=aryl, alkyl) to deliver highly yields 4,5-dihydro-3H-benzo[d][1,2]diazepine skeleton compounds was disclosed via N-insertion ring-opening reaction. The structural framework of 2,3-benzodiazepines is unambiguously confirmed by single-crystal X-ray crystallographic analysis. The present protocol features mild reaction conditions, high yields and feasibility of large-scale synthesis and catalyst-free access to benzo[2,3]diazepines skeleton. In contrast, azomethine imines were produced by employing tetrahydrodiazirino[3,1-a]isoquinoline reacts with aroyl chlorides under the same conditions.

Abstract:A new copper-catalyzed oxidative cyclization protocol was developed for the synthesis of 5,6-dihydropyrrolo[2,1-a]isoquinolines via a three-component reaction of tetrahydroisoquinolines with bromoketones and electron-deficient alkenes with air as a terminal oxidant. A variety of functional groups survived under the reaction conditions and the target products were obtained in good yields. This reaction features such advantages as eco-friendly reaction conditions, a simplified operation process and a broad substrate scope.

Abstract:Here, a general copper-catalyzed radical cascade carbocyclization reaction with 2-arylbenzoimidazoles and a Togni reagent was realized. Structurally diverse CF3-containing tetracyclic core benzimidazo[2,1-a]isoquinoline-6(5H)-ones were obtained in moderate to good yields. The wide substrate scope, good functional group tolerance, and ease of scale-up of this method are expected to promote its potential applications in pharmacy and biotechnology.

Abstract:An efficient synthesis of 1,3-/1,1-dialkoxy 1,2-dihydroisoquinolines from o-ethynylbenzacetals and sulfonyl azides via a cascade process combining copper-catalyzed alkyne-azide cycloaddition (CuAAC), Dimroth rearrangement, 1,5-OR shift/1,5-H shift, and 6π-electrocyclic ring closure (6π-ERC) is described. Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:The 1-arylpyrazolo[5,1-b]quinazolin-9(1H)-one derivatives were obtained via intramolecular alkyne hydroamination reaction of 2-amino-N′-arylbenzohydrazide and alkynal diethyl acetal in the presence of CuBr and Cs2CO3. Abstract:A series of 15-arylisoquinolino[2′,1′:1,2]imidazo[4,5-f][1,10]phenanthrolines was synthesized by a copper(I)-catalyzed reaction of 2-(2-bromophenyl)-1H-imidazo[4,5-f][1,10] phenanthrolines and terminal alkyne in the presence of Cs2CO3. It was proved to be an efficient method for the synthesis of hexacyclic compounds bearing three kinds of heterocycles which were isoquinoline, imidazole and phenanthroline.

Abstract:We have developed a versatile and efficient method for copper-catalyzed synthesis of both 2-amino-4(3H)-quinazolinone and 2-aminoquinazoline derivatives. The protocol uses readily available substituted 2-halobenzoic acids, 2-bromobenzaldehyde, 2-bromophenyl ketones and guanidines as the starting materials, inexpensive copper(I) iodide as the catalyst, and the method has important application values for construction of N-heterocycles in organic chemistry and medicinal chemistry.

Abstract:A selective copper(I)-catalyzed benzylic C(sp3)–H geminal difunctionalization reaction of a benzylic-type sp3 carbon was developed. This novel strategy allowed simultaneous introduction of amide and hydroxyl group in a highly selective and efficient way via successive oxidative intramolecular amidation and hydroxylation. This method was also applied to the synthesis of 3-hydroxy-2,3-dihydro-1H-pyrrolo[3,4-b]quinoxalinones from readily available 3-methyl-N-substituted quinoxaline-2-carboxamides in moderate to good yields. The five-membered cyclic hemiaminal moiety of the pyrrolo[3,4-b]quinoxalinones can serve as an intermediates for subsequent transformations into other useful functional groups.
