Experimental study on combined culture in vitro of macromolecular scaffold and bone marrow-originated osteoblast

Abstract:【Objective】To observe the effect of macromolecular scaffold on the proliferation and ossification activity of bone marrow-originated osteoblast. 【Methods】Osteoblast of young New Zealand rabbit, which were originated from marrow stroma cell via isolation, cultured in vitro and derivated, were inoculated in the macromolecular scaffold prepared with lysine disocyanate-glycerol polymer (LDIG) for combinating culture with porous polyglycolic acid (PGA) scaffold as contrast. Subsequently, the proliferating osteoblast amount and ossification activity were detected by means of cell count,Ⅰ-type collagen and alkaline phosphatase activities (APA) test and scanning electron microscope (SEM). 【Results】Bone marrow-originated osteoblast prosperously proliferated in the macromolecular scaffold and the proliferating osteoblast amount and ossification activity was higher than that in the PGA scaffold. 【Conclusion】The tissue engineering scaffold prepared with macromolecular material is capable of constructing tissue engineering bone because of its ideal porous network structure and favourable osteoblastic compatibility.
