Culture of Rabbit Bone Marrow Mesenchymal Stem Cells on Porous Silk Fibroin Scaffold in Vitro

Abstract:Objective To explore the potential significance of porous silk fibroin as an extracellular matrix material in bone tissue engineering. Methods Bone marrow were obtained from 4 week old New Zealand rabbit by aspiration under Nembutal anesthesia, and rabbit bone marrow mesenchymal stem cells (MSCs) were separated by gradient centrifugation by percoll, and incubated in DMEM containing 15% FBS. The rabbit MSCs were purified through passage cultures and inoculated into 24 well dishes and cultured with porous silk fibroin scaffold of three different pores size 58, 78, 111μm immersed with DMEM-L containing 15% FBS for 8 days. The porous silk fibroin scaffold growing MSCs were sampled at the culture day 1, 4, and 8, and the cell proliferation and growth characteristics were observed under phase microscope, scanning electron microscope and laser scan confocal miroscope. Results Rabbit bone marrow MSCs grew well in porous silk fibroin scaffold of three different pore size, especially in size 111μm silk fibroin materials, as demonstrated by the morphological investigations. Conclusions Under selection of suitable pore size, porous silk fibroin may serve as one prospective scaffold material with good biocompatibility, cellular adhesion and degradability in bone tissue engineering.
