Abstract:A direct catalytic asymmetric aldol-type reaction of 3-substituted-2-oxindoles with glyoxal derivatives and ethyl trifluoropyruvate, catalyzed by a chiral N,N'-dioxide-Sc(OTf)(3) (Tf = trifluoromethanesulfonyl) complex, has been developed that tolerates a wide range of substrates. The reaction proceeds in good yields and excellent enantioselectivities (up to 93% yield, 99:1 diastereomeric ratio (dr), and >99% enantiomeric excess (ee)) under mild conditions, to deliver 3-(alpha-hydroxy-beta-carbonyl) oxindoles with vicinal quaternary-tertiary or quaternary-quaternary stereocenters. Even with 1 mol % catalyst loading or on scaleup (10 mmol of starting material), maintenance of ee was observed, which showed the potential value of the catalyst system. In studies probing the reaction mechanism, a positive nonlinear effect was observed and Sc(III)-based enolate intermediates were detected by using ESIMS. On the basis of the experimental results and previous reports, a possible catalytic cycle was assumed.

Abstract:A Cs2CO3-catalyzed alkylation reaction of indoles with trifluoromethyl ketones was presented. Both alicyclic and aromatic trifluoromethyl ketones as well as various substituted indoles are compatible with the methodology. Good to excellent yields of the corresponding trifluoromethyl substituted tertiary alcohols 2,2,2-tritrifluoro-1-(1H-indol-3-yl)-ethan-1-ols were acquired as the sole products.

Abstract:A highly enantioselective Mukaiyama aldol reaction of silyl enol ethers with isatins was developed. A series of chiral 3-substituted 3-hydroxy-2-oxindoles could be obtained exclusively with high yields (up to 95%) and excellent enantioselectivities (up to 99%). In particular, water was essential to improve the diastereoselectivity.

Abstract:Chiral N,N'-dioxide/Y(OTf)3 and Sc(OTf)3 complexes have been developed as efficient catalysts for the bisvinylogous Mukaiyama aldol reaction of silyl ketene acetal with α-ketoesters and aldehydes, respectively. The catalytic systems were highly ε-selective, and the substrate scope was wide. The corresponding ε-hydroxy-α,β,γ,δ-unsaturated esters were obtained in up to 95% yield and 98% ee.

Abstract:The In(OAc)3-catalyzed reaction of bromo- and iodoalkanes with PhSiH3 in THF at 70 degrees C gave dehalogenated alkanes in good to high yields. In the presence of Et3B and air, the reduction proceeded smoothly at 30 degrees C. When 2,6-lutidine and air were used as additives, the In(OAc)3-catalyzed system enabled an efficient reduction of simple and functionalized iodoalkanes in EtOH. Catalytic use of GaCl3 was found to be effective in the reduction of haloalkanes with poly(methylhydrosiloxane) (PMHS). These catalytic reductions probably involve a radical chain mechanism in which indium or gallium hydride species work as the actual reductants.

Abstract:Indium nanoparticles have been employed in the reactions of various carbonyl compounds with allyl (crotyl) chloride in water, affording the corresponding alcohols with high yields. The crotylation gave exclusive gamma-adducts with a dominant syn-isomer.

Abstract:Sulfonate derivatives of chiral 1,1′-binaphthol were used as chiral anionic surfactants in asymmetric aldol-type reaction in water to give aldol adducts with moderate to good diastereo- and enantioselectivities; Ga(OTf)3 and Cu(OTf)2 were better than Sc(OTf)3 as Lewis acid catalysts.

Abstract:A new efficient strategy for the synthesis of 3-trifluoromethyl-indoles was developed through tandem cyclization/trifluoromethylation of 2-alkynylanilines and Umemoto's reagent under effect of CuBr. The reaction features the use of cheap copper salt, mild reaction conditions and high yield of products. The preliminary mechanism study indicates the solvent dimethylacetamide acts as the role of dealkylation.

Abstract:An efficient method for the highly regio‐ and stereoselective synthesis of 1,2,3‐trisubstituted indanes from diarylmethanols and allylamides through iron(III) chloride hexahydrate‐catalyzed intermolecular [3+2] cycloaddition reaction is reported. A range of 1,2,3‐trisubstituted indanes were prepared in good to excellent yields with excellent stereoselectivities for various combinations of diarylmethanols and allylamides. The choice of solvent is critical to the catalysis. Mechanistic studies indicated that the generation of a benzylic cation is involved in this catalytic process, and the reaction proceeds via the initial formation of diarylmethyl ethers. This reaction features ready availability of the starting materials, high level of functional tolerance and simple experimental operation. magnified image

Abstract:N-Heterocyclic carbene (NHC)-catalyzed vinylogous Mukaiyama aldol reaction of ketones was developed. Under the catalysis of 5 mol% NHC, α-keto esters and α-trifluoromethyl ketones reacted with 2-(trimethysilyloxy)furan efficiently to produce γ-substituted butenolides containing adjacent quaternary and tertiary carbon centers in high yields with good diastereoselectivities.

Abstract:The combination of Ga(OTf)(3) with chiral semi-crown ligands (la-e) generates highly effective chiral gallium Lewis acid catalysts for aqueous asymmetric aldol reactions of aromatic silyl enol ethers with aldehydes. A ligand-acceleration effect was observed. Water is essential for obtaining high diastereo-selectivity and enantioselectivity. The p-phenyl substituent in aromatic silyl enol ether (2 h) plays an important role and increases the enantioselectivity up to 95% ee. Although aliphatic silyl enol ethers provided low enantioselectivities and silylketene acetal is easily hydrolyzed in aqueous alcohol, the aldol reactions of silylketene thioacetal (12) with aldehydes in the presence of gallium-Lewis acid catalysts give the beta-hydroxy thioester with reasonable yields and high diastereo- (up to 99: 1) and enantioselectivities (up to 96% ee).

Abstract:Herein, chiral Brønsted acid-catalyzed intramolecular asymmetric allylic alkylation of indoles with allylic primary alcohols is described. The allyl alcohols were directly employed as the allylic precursors in this metal-free protocol, without preactivation or any additional activating reagents. This method provides the convenient synthesis of a broad range of functionalized tetrahydrocarbazoles in excellent yields (≤97%) with good enantioselectivity (≤93% ee). The optimal conditions are compatible for gram-scale reaction.
