Abstract:A protocol for the Knoevenagel condensation of aromatic aldehydes with methylene compounds catalyzed by task specifed ionic liquid 1,8-diazabicyclo[5.4.0]-undec-7-en-8-ium lactate under solvent-free conditions has been developed. The reactions proceed at ambient temperature and the work-up procedures are very simple. Good to excellent yields could be obtained. The ionic liuid could be simply prepared and recycled efficiently.

Abstract:Esterase BioH, which is obligatory for biotin synthesis in Escherichia coli, was found to exhibit a promiscuous ability to catalyse Aldol and Knoevenagel reactions with moderate to good yields. The reaction conditions including organic solvent, molar ratio of ketone to aldehyde, enzyme amount, and reaction time were investigated to evaluate the effect of different reaction conditions on yield. Target compounds were afforded in the best yield of 91.2% for Aldol reaction and 54.7% for Knoevenagel reaction. In addition, because the enzyme could be prepared with a low cost, this protocol could provide an economic route to conduct Aldol and Knoevenagel reactions, which expand the field of enzymatic promiscuity.

Abstract:The chemical synthesis of organic molecules involves, at its very essence, the creation of carbon–carbon bonds. In this context, the aldol reaction is among the most important synthetic methods, and a wide variety of catalytic and stereoselective versions have been reported. However, aldolizations yielding tertiary aldols, which result from the reaction of an enolate with a ketone, are challenging and only a few catalytic asymmetric Mukaiyama aldol reactions with ketones as electrophiles have been described. These methods typically require relatively high catalyst loadings, deliver substandard enantioselectivity or need special reagents or additives. We now report extremely potent catalysts that readily enable the reaction of silyl ketene acetals with a diverse set of ketones to furnish the corresponding tertiary aldol products in excellent yields and enantioselectivities. Parts per million (ppm) levels of catalyst loadings can be routinely used and provide fast and quantitative product formation in high enantiopurity. In situ spectroscopic studies and acidity measurements suggest a silylium ion based, asymmetric counteranion-directed Lewis acid catalysis mechanism.

Abstract:A series of poly(2-oxazoline) (POX) derivatives bearing prolinamide pendants were designed as organocatalysts and evaluated in the direct asymmetric aldol reaction between aromatic aldehydes and cyclic ketones. The structural variation of the alkyl spacer connecting the polymer backbone with the catalytic unit was applied so as to deduce structure–performance relationships combined with comparable experiments from model catalysts. Results showed that the POX-bound prolinamides can promote the aldol reaction more effectively as compared to their small-molecular and non-POX-bound analogs. The catalyst P3 containing the pyrrolidine moiety closer to the tertiary amide backbone exhibited the overall best catalytic efficiency, affording anti-products in 84% yield with 89% ee in the representative aldol addition of cyclohexanone to 4-nitrobenzaldehyde at a 10 mol.% catalyst loading. Furthermore, the influence of trifluoroacetic acid as an additive on the asymmetric transformation was investigated. Theoretical calculations revealed that the protonation of the aldehyde carbonyl group switched the activation mode of the aldol acceptor through hydrogen bond interactions, thereby changing the relative energy barrier of the enamine/aldehyde reaction transition states, which accounted well for the significant improvement in the enantioselectivity of the acidic additives observed experimentally.

Abstract:The direct asymmetric aldol reactions of equivalent molar amounts of aldehydes and ketones were carried out at -20 degrees C over alkaline Al(2)O(3) with 20 mol % of Pro-Trp as catalyst and 20 mol % of N-methylmorpholine or 1,4-diazabicyclo[2.2.2]octane as additive. After simple and environmentally friendly work-up, moderate to high isolated yields (up to 95%), good diastereoselectivities (>99:1), and enantioselectivities (up to 98% ee) have been achieved for the reactions of different kinds of ketones with various aldehydes. The catalytic system could be reused without decrease of activity by addition of 10 mol % catalyst and base in the catalytic system.

Abstract:The dispersion of active sites plays an imperative role in the promotion of catalytic performance on methyl acrylate synthesis via direct vaporous aldol condensation. Herein, a novel PEG-assisted impregnation method was proposed to synthesize Cs/Zr-SiO2 catalyst with high dispersion of acid and base sites. A series of characterization techniques including NMR, TEM, physical N-2 adsorption isotherms, XRD, XPS, NH3-/CO2-TPD, and Py-IR were used to investigate the physico-chemical properties of these prepared catalyst series. The introduction of PEG improved the specific surface area, dispersion of active sites, and density of acid and base sites, resulting in superior catalytic performance of Cs/PEG@Zr-SiO2 by comparison with that of Cs/SiO2 and Cs/Zr-SiO2. The kinetic experiments revealed that the activation energy of aldol condensation was 80.8 kJ/mol over the Cs/PEG@Zr-SiO2 catalyst. The deactivation behavior derived from carbon deposition was discovered, and the spent catalyst could be regenerated by simple thermal treatment.

Abstract:A novel efficient procedure has been developed for the conjugate addition of amines to electron deficient alkenes. A series of KF supported on different carriers have been synthesized for the conjugate addition amines to alkenes. After optimizing the reaction conditions, KF/MgO was chosen as the most efficient catalyst for the reactions. The results showed that the catalyst was very efficient for the conjugate addition of amines to electron deficient alkenes with excellent yields in several minutes. Operational simplicity, no need of any solvent, low cost of the catalyst used, high yields, reusability, excellent chemoselectivity and wide applicability are the key features of this methodology.

Abstract:The direct dehydrogenation of ketones has been widely utilized in both academic and industrial settings as the most efficient means of constructing unsaturated ketones and phenols. In contrast to the induction of dehydrogenation at the alpha-site of carbonyl by enol activation, direct and efficient activation of the carbonyl beta-C(sp(3))-H bond to form unsaturated ketones remains unexplored. Herein, we report a polarity-matching-based activation strategy for the carbonyl beta-C(sp(3))-H bond, enabling the photoinduced intramolecular dehydrogenation of ketones without prefunctionalization via the merger of photoredox and cobalt catalysis. This photoinduced mode employs a benign and mild approach to construct unsaturated ketone and phenol skeletons via acceptorless catalytic dehydrogenation, showcasing potential applications in the field of drug synthesis.

Abstract:A novel carbon-based strong acid catalyst, methylnaphthalenesulfonic acid - formaldehyde condensate, has been synthesized successfully. The catalytic activity for acetalization and ketalization at room temperature under solvent-free condition was investigated. The results showed that the novel catalyst was very efficient for the reaction. Also the novel heterogeneous catalyst owned the advantages of high activity, wide applicability, strikingly simple work-up procedure, non-pollution, stability and reusability, which made the catalyst hold great potential for the "green process".

Abstract:Aldol condensation cyclopentanone is an attractive reaction for bio‐jet fuel production. KOH is an efficient homogeneous catalyst for aldol condensation but suffering the problem of separation. In this paper, a heterogeneous catalyst, diatomite stabilized KOH (KOH/diatomite) was prepared in a simple way (dry‐mixed impregnation process), and exhibited good activity (overall yield: 85.0%) and reusability. To clarify the support effect of diatomite, KOH/SiO2 catalyst was also prepared. According to characterizations of these two catalysts, we suggest that the excellent catalytic activity of KOH/diatomite can be attributed to the formation of K‐O‐Si bond. In addition, the impurity elements existed in diatomite (e.g. CaO, Na2O, Al2O3 ) and higher BET surface areas may also benefit to this reaction. This study provides a new possibility for converting homogeneous catalyst (KOH) to heterogeneous catalyst (KOH/diatomite). In this way, KOH/diatomite performed excellent catalytic activity and exhibited stable reusability, which can avoid the problem of separation and pollution.
