Abstract:The feasibility of employing Diels-Alder reaction for improving the grafting efficiency and controllability of poly (ethylene glycol) (PEG) towards chitosan (CS) was evaluated. Diene-capped PEG (PEG-diene) was conveniently obtained through thio-acrylate reaction between furfuryl mercaptan and poly( ethylene glycol) methyl ether methacrylate (PEGMEMA). By using sodium dodecyl sulphate-chitosan complex (SCC) as an organo-soluble precursor, the dienophile, (N-(2-carboxyethyl) maleimide), was regioselectively conjugated to the hydroxy groups of chitosan backbone through conventional coupling method. Sodium dodecyl sulphate (SDS) was then removed from the conjugate simply by precipitating the reaction solution into tris (hydroxymethyl) aminomethane (Tris) solution to give the chitosan intermediate carrying dienophile moieties (CS-dienophile). In situ H-1-NMR was further used for studying the Diels-Alder reactions between CS-dienophile and PEG-diene. It was found that PEG could be grafted onto chitosan backbone in aqueous media under mild conditions. In addition,the PEG grafts could be detached from the graft copolymers at high temperature through retro Diels-Alder reaction.

Abstract:Under mild conditions, a series of alpha,alpha-difluoro-beta-ketoamides were synthesized with Selectfluor (R) as the F+ source in the presence of K2CO3 in the mixed-solvent of water and PEG-400 (V-H2O : VPEG-400 = 3 : 1), without additional phase-transfer catalyst. Abstract:An efficient Co-catalyzed hydro-difluoromethylthiolation/hydro-trifluoromethylthiolation reaction of unactivated alkenes was described. Both reactions were conducted at room temperature and a variety of common functional groups such as halogen, ester, aldehyde, enolizable ketone or ester, nitro or cyano group, sulfonate and carbamate were compatible with the reaction conditions. Radical cyclization and radical inhibitor experiments suggested that the reaction proceeds through a free radical process.

Abstract:A novel FeCl3·6H2O-catalyzed tandem Friedel–Crafts alkylation–hydrolysis reaction between chain α-oxo ketene dithioacetals and alcohols to afford α-alkylated β-oxo thioesters has been successfully developed. The reaction is efficient in the presence of catalyst loading as low as 30 mol% in MeCN at room temperature, and a wide variety of α-alkylated β-oxo thioesters are efficiently synthesized in good yields.

Abstract:Alcohols can be efficiently converted into the useful thioethers by a transition metal- and base-free dehydrative S-alkylation reaction with thiols or disulfides by employing alkyl halides as the effective catalyst. This simple and efficient method is a green and practical way for the preparation of thioethers, as it tolerates a wide range of substrates such as aryl and alkyl thiols, as well as benzylic, allylic, secondary, tertiary, and even the less reactive aliphatic alcohols.

Abstract:An effective deprotective method of spiro 3-cyclic thiaza ketal of steroidal 1,4-dien-3-ones using alkyl vinyl ether in the presence of protic acid followed by the treatment of aqueous alkali was described. This novel protocol could be fulfilled under mild condition with high yield. The mechanism mediated by a carbonium ion formed in situ was clarified by the capture of the cleaved fragment.

Abstract:Oxalic acid monothioester (OAM), an easily accessible and storable reagent, was reported herein as a thioester synthetic equivalent for palladium-catalyzed decarboxylative thiocarbonylation of organohalides and hydrothiocarbonylation of unsaturated carbon-carbon bonds at room temperature with high chemo- and regioselectivity. The reaction is applicable to the synthesis of cysteine-derived thioesters, thus allowing chemical modification of cysteine-containing peptides. Decarboxylation of OAM proceeds through oxidative addition of Pd(0) to the acyl-S bond, which accounts for the very mild reaction conditions.

Abstract:A synthetic method of dialkylideneacetones has been developed. Compared with known protocols, the method employed catalytic Ca(OH)2 as the cheap, mild base catalyst and dilute aqueous EtOH (20%, v/v) as the green and safe solvent. The procedure was easily operated: In most cases, the product could be isolated by a simple filtration, and purified by washing with water. This paper provided experimental details of the reactions, which could be applied in gram-scale synthesis and should be a very reliable and practical protocol to prepare these useful compounds in laboratory and at the industrial level.

Abstract:The use of alkylthio electrophiles in synthesis has remained elusive because of the lack of a suitable reagent that is practical and of excellent enantioselectivity and appropriate reactivity. In this work we introduce a novel alkylthio reagent based on the 2-mercapto-5-methyl-1,3,4-thiadiazole (MMTD) fragment for direct alkylsulfenylation of ketones and aldehydes. It can be readily prepared by the oxidative coupling between thiadiazole and other alkylthio reagents and be combined with chiral primary aminocatalysis. This protocol provides facile access to diverse α-alkylthio quaternary carbon centers with good stereoselectivities.

Abstract:Treatment of lithio derivative of novel PEG-supported alpha-phenylselenopropionate with aldehydes, followed by oxidation-elimination with 30% hydrogen peroxide, formed Baylis-Hillman products, which were then reacted with sodium arylsulfinate. The resulting sulfonylated products were cleaved from the PEG efficiently affording methyl (2Z)-2-arylsulfonylmethyl-2-alkenoates in good yields and high purities.
