Abstract:Transition metal-catalyzed asymmetric nitrene transfer is a powerful method for the generation of enantioenriched amines found in natural products and bioactive molecules. We report herein a highly chemo- and enantioselective intramolecular silver-catalysed aziridination that employs 2,2,2-trichloroethoxysulfonyl (Tces)-protected carbamimidates as an underexplored class of nitrene precursors. In contrast to previous reports, the use of carbamimidates allows for tuning of the sterics in the nitrene precursor that, when combined with silver counteranion and ligand effects, show promise for expanding the scope of asymmetric aziridinations. Several homoallylic carbamimidates containing di- and trisubstituted olefins were converted into [4.1.0]-bicyclic aziridines in good-to-excellent yields (up to 98%) and enantioselectivities (up to 99% ee) and promising results noted to furnish enantioenriched 1,2-aminoalcohols. Post-synthetic modifications afford difunctionalized amines bearing two contiguous, enantioenriched chiral centres. Computational studies highlight reasons why carbamimidates are superior to sulfamates and carbamates, but more importantly, provide the key insights needed to expand the utility of this chemistry to other intra- and intermolecular asymmetric aziridinations.
