Abstract:Four piperazine-appended [60] fullerenes (1a-1d) were synthesized and characterized. Their activities of scavenging radical were studied via UV-Vis and EPR. The results indicate that light irradiation is of advantage to increase the activity of scavenging radical.

Abstract:AbstractA novel fullerene‐maleic anhydride derivative was synthesized via radical polymerization. It is soluble in polar solvents such as water, dimethyl sulfoxide, and tetrahydrofuran etc. The product was characterized by FTIR, UV‐Vis, GPC, and Transmission electron microscope (TEM). TEM analysis shows that the average particle diameter is about 38 nm. The in vitro antitumor activity of the fullerene‐maleic anhydride derivative has been tested and the result shows that the derivative exhibits better antitumor activity against HeLa cells and bone tumor cells. To prove water‐soluble fullerene derivatives photodynamic effect on tumor cell, i.e., the photodynamic effect on mouse bone tumor in vivo we injected the fullerene ‐maleic anhydride derivatives into the mouse bone tumor body. We found that with I‐W lamp (500 W) illumination the mouse bone tumor body was strongly damaged. The scavenging of reactive oxygen species and the potential for cell protection by fullerenols with different isoelectric points

Abstract:Fullerenols were known as the major water-soluble derivatives of fullerene that possess particular significance biological effects such as anticancer active and free radical scavengers. We have reported a novel method for purifying nanoparticles of fullerenols, which based on different isoelectric points (pIs). Consistent with their cytoprotective abilities, these samples can scavenge the stable 2,2diphenyl-1-picryhydrazyl (DPPH) radical in vitro with the following relative potencies: pI 7.0 > pI 5.29> pI 4.40> pI 2.81. The observed differences in free radical-scavenging capabilities support the hypothesis that physicochemical properties, such as surface chemistry induced differences in electron affinity, and degree of aggregation, influence the biological and biomedical activities of fullerenols with different pIs. These purified fullerenols with pI 2.81, pI 4.40, pI 5.29 and pI 7.0 were performed to determine the effect of purified fullerenols on cytotoxicity and DNA stability in Raw 264.7 cells. There no significantly different by statistical product and service solutions (SPSS) analysis. But only the sample of fullerenols with pI 2.81 caused obvious DNA damage to RAW264.7 cells in the continuous buffer system.

Abstract:A water soluble -cyclodextrin (-CD) functionalized [60]fullerene (C-60-CD) was synthesized. The C-60-CD-induced generation of reactive oxygen species (ROS), and DNA-cleavage ability and cytotoxicity of C-60-CD were studied upon visible light irradiation, demonstrating that the compound is very promising in the applications of photodynamic therapy. The histological analyses demonstrate that C-60-CD has no acute or subacute toxicity to living body.

Abstract:Hydrophilic fullerene derivatives get notable performance in various biological applications, especially in cancer therapy and antioxidation. The biological behaviors of functional fullerenes are much dependent on their surface physicochemical properties. The excellent reactive oxygen species-scavenging capabilities of functional fullerenes promote their outstanding performances in inhibiting pathological symptoms associated with oxidative stress, including neurodegenerative diseases, cardiovascular diseases, acute and chronic kidney disease, and diabetes. Herein, fullerene derivatives with reversed surface charges in aqueous solutions are prepared: cationic C60-EDA and anionic C60-(EDA-EA). Under the driving force of membrane potential (negative inside) in the cell and mitochondria, C60-EDA is much rapidly taken in by cells and transported into mitochondria compared with C60-(EDA-EA) that is enriched in lysosomes. With high cellular uptake and mitochondrial enrichment, C60-EDA exhibits stronger antioxidation capabilities in vitro than C60-(EDA-EA), indicating its better performance in the therapy of oxidation-induced diseases. Abstract:Fullerene skeleton modification has been investigated through selective cleavage of the fullerene carbon-carbon bonds under mild conditions. Several cage-opened fullerene derivatives including three [59]fullerenones with an 18-membered-ring orifice and one [59]fullerenone with a 19-membered-ring orifice have been prepared starting from the fullerene mixed peroxide 1, C60(OOtBu)6. The prepositioned tert-butyl peroxy groups in 1 serve as excellent oxygen sources for formation of hydroxyl and carbonyl groups. The cage-opening reactions were initiated by photoinduced homolysis of the tBu-O bond, followed by sequential ring expansion steps. A key step of the ring expansion reactions is the oxidation of adjacent fullerene hydroxyl and amino groups by diacetoxyliodobenzene (DIB). Aminolysis of a cage-opened fullerene derivative containing an anhydride moiety resulted in multiple bond cleavage in one step. A domino mechanism was proposed for this reaction. Decarboxylation led to elimination of one carbon atom from the C60 cage and formation of [59]fullerenones. The cage-opened [59]fullerenones were found to encapsulate water under mild conditions. All compounds were characterized by spectroscopic data. Single-crystal structures were also obtained for five skeleton-modified derivatives including two water-encapsulated fulleroids.

Abstract:Fullerene is a cosmic material with a buckyball-like structure comprising 60 carbon atoms. It has attracted significant interest because of its outstanding antioxidant, antiviral, and antibacterial properties. Natural fullerene (NC60) in shungite meets the demand of biomedical fields to scavenge reactive oxygen species in many diseases. However, its hydrophobicity and poor solubility in water hinder its use as an antioxidant. In this study, highly water-dispersed and stable Pluronic-coated natural fullerene nanoaggregates (NC60/Plu) were prepared from various Pluronic polymers. The water dispersity and stability of NC60 were compared and optimized based on the characteristics of Pluronic polymers including F68, F127, L35, P123, and L81. In particular, NC60 coated with Pluronic F127 at a weight ratio of 1 to 5 showed excellent antioxidant effects both in situ and in vitro. This suggests that the high solubilization of NC60 in Pluronic polymers increases its chance of interacting with reactive oxygen radicals and improves radical scavenging activity. Thus, the optimized NC60/PF127 may be a novel biocompatible antioxidant for treating various diseases associated with oxidative stress.

Abstract:Three novel fullerene derivatives containing a C60-fluorene core and thiophenyl, terthiophenyl and tetrathiophenyl groups grafted at the 2-position of the fluorene ring, respectively, were synthesized and characterized. Their photophysical and electrochemical properties were investigated systematically. The results showed that their UV-vis absorption bands in the range of 350–500 nm were much wider and stronger than that of the traditional fullerene derivative PC61BM, and the LUMO energy levels of the synthesized fullerene derivatives were gradually up-shifted by increasing of the number of thiophene rings in the oligothiophene units.

Abstract:Aminopolycarboxylic esters react with C-60 under photolysis to produce fullerene multicarboxylates. Irradiation of tetramethyl ethylenediaminetetraacetate (EDTA) with C-60 yields the EDTA-containing fullerene monoadduct C-60(MeOOCCH)(2)NCH2CH2N(CH2COOMe)(2). In addition, several other C-60 monoadducts are also isolated and characterized, including compounds due to EDTA fragmentation. Similar results are observed with pentamethyldimethylenetriaminepentaaceta (DTPA). When partially methylated nitrilotriacetic acid is irradiated with C-60, decarboxylation occurs and organo dihydrofullerene derivatives such as C-60(H)(CH2N(CH2COOMe)(2)) are formed. Radical mechanisms are proposed for both types of photoreactions. The fullerene derivatives are characterized by their spectroscopic data. Photoreactions of C-60 with other analogous molecules also support the conclusions.

Abstract:A series of bromo-terminated four-armed homopolymers [polystyrene (PSt) and poly(methyl methacrylate) (PMMA)], prepared by atom transfer radical polymerization (ATRP), were subsequently functionalized with fullerene C-60 by atom transfer radical addition (ATRA) reaction using CuBr/2,2'-bipyridine (bipy) as the catalyst system. The C-60 end-capped four-armed polymer derivatives were then characterized by gel permeation chromatography (GPC), UV-vis, FT-IR, thermal gravimetric analyses (TGA) and differential scanning calorimeter (DSC). Another technique of synthesizing C-60-functionalized four-armed polymers from azide functional polymers was also applied for comparison. The optical limiting behavior of C-60 end-capped four-armed polymers prepared via ATRA reaction were measured in THF solution at 532 nm. Both groups of fullerene-functionalized polymers (C-60-PSts and C-60-PMMAs) are provided with optical limiting properties.

Abstract:tert-Butylperoxy radicals add to C(60) selectively to form multi-adducts C(60)(O)(m)(OO(t)Bu)(n) (m = 0, n = 2, 4, 6; m = 1, n = 0, 2, 4, 6) in moderate yields under various conditions. Visible light irradiation favors epoxide formation. High concentration of tert-butylperoxy radicals mainly produces the hexa-homoadduct C(60)(OO(t)Bu)(6) 6; low concentration and long reaction time favor the epoxy-containing C(60)(O)(OO(t)Bu)(4) 7. The reaction can be stopped at the bis-adducts with limited TBHP. A stepwise addition mechanism is discussed involving mono-, allyl-, and cyclopentadienyl C(60) radical intermediates. m-CPBA reacts with the 1,4-bis-adduct to form C(60)(O)(OO(t)Bu)(2) and C(60)(O)(3)(OO(t)Bu)(2). The C-O bond of the epoxy ring in 7 can be cleaved with HNO(3) and CF(3)COOH. Nucleophilic addition of NaOMe to 7 follows the S(N)1 and extended S(N)2' mechanism, from which four products are isolated with the general formula C(60)(O)(a)(OH)(b)(OMe)(c)(OO(t)Bu)(d). Visible light irradiation of the hexa-adduct 6 results in partial cleavage of both the C-O and O-O bonds of peroxide moieties and formation of the cage-opened compound C(60)(O)(O)(2)(OO(t)Bu)(4). All the fullerene derivatives are characterized by spectroscopic data. A single-crystal structure has been obtained for an isomer of C(60)(O)(OH)(2)(OMe)(4)(OO(t)Bu)(2).

Abstract:Novel water-soluble fullerene–glycine derivatives were synthesized by means of simple organic chemistry. They are completely soluble in water, yielding a clear brown solution. The products were characterized by fourier transform infrared (FTIR), ultraviolet–visible spectroscopy (UV–Vis), 1H NMR, 13C NMR, thermogravimetric analyses (TGA), and scanning electron microscopy (SEM). The assembly behavior of water-soluble fullerene–glycine derivatives was investigated by SEM. The results show that the fullerene–glycine derivatives create morphology that is sphere-like. The cytotoxicity to cancer cell lines of the fullerene–glycine derivatives was evaluated by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) and flow cytometry. The results show that fullerene–glycine derivatives exhibit mortality and apoptosis of the cells which increased with the increase of fullerene–glycine derivative concentration. The cytotoxicity mechanism of fullerene–glycine derivatives was investigated for the first time.

Abstract:A soluble fullerene, TAZC60, bearing an electron transport moiety (triazole) is synthesized and characterized. TAZC60 possesses a band gap of 2.2 eV, and HOMO and LUMO energy levels of 5.9 and 3.7 eV, respectively. Thin films of TAZC60 exhibit an electron mobility of 2.2 x 10(-8) m(2)/V s, as determined from the current-voltage characteristics that fit with a space-charge limited current (SCLC). These properties are similar to those of [6,6]-phenyl C61 butyric acid methyl ester, PCBM, a soluble fullerene widely used in organic photovoltaics. The strategy of modifying fullerenes with a solublizing electron transport moiety may open avenues for the design of more specific polymers for controlling the morphology and transport properties of polymer-fullerene blends for polymer solar cells.

Abstract:Addition of C60 moiety, a powerful electron acceptor to poly(N-vinylcarbazole) (PVK) by chemical reaction modifies considerably the physical and chemical properties of PVK. The characterization techniques employed are UV-visible, IR, DSC, TGA, ESR, 13C-NMR spectroscopy, scanning electron microscopy, XRD, and cyclic voltammetry. The fullerenated PVK, which has a visibly earthy yellow cast when compared with the unreacted polymer, has a new structure in the UV-vis absorption spectrum with the active range extending from about 280 to 870 nm, its apparent temperature sensitivity is intriguing, and an unusual temperature dependence for the ESR spectrum is observed. Considerable difference of electronic structure between pure PVK and C60-PVK copolymer is indicated. The thermal stability and oxidation-reduction activation of pure PVK are enhanced by C60-chemical modification. © 1996 John Wiley & Sons, Inc.

Abstract:1,2-(1-Acridin-10'-yl-2-aza-2-methylprop-1,3-ylene)fullerene was synthesized firstly and characterized by UV-Vis, H-1 NMR, C-13 NMR and electrospray ionization mass spectroscopy, which is capable of forming a stable complex with zinc tetraphenylporphyrin via the axial ligation. The steady state fluorescence studies show efficient quenching of the zinc tetraphenylporphyrin emission upon axial coordination of acridine attached to C-60.
Catechol amide derivatized polyhydroxylated fullerene as potential chelating agents of radionuclides: Synthesis, reactive oxygen species scavenging, and cytotoxic studies

Abstract:Radionuclide internal contamination can induce chemical and radioactive intoxication and produce harmful free radicals in vivo. At present, administration of chelating agents is the most effective treatment against nuclide contamination. However, traditional studies on chelating agents have ignored the damage caused by free radicals to the body. The present study aimed to develop a type of a bifunctional sequestering agent that can chelates nuclides and scavenges free radicals simultaneously. Therefore, a novel catechol amide-derivatized polyhydroxylated fullerene was designed and prepared. The poor water solubility of fullerene was ameliorated by chemically modifying hydrophilic catechol amide and multiple hydroxyl groups, and obtaining high water-soluble fullerene derivatives. The affinities of chelators were investigated via sulfochlorophenol competitive complexing method and antioxidant capacities were examined by electron paramagnetic resonance. The results revealed the good complexation of the designed and synthesized chelating agent with uranyl ions; and its efficiency in scavenging hydroxyl radicals. This chelating agent showed extremely low toxicity and notable protective effect against oxidative stress on A549 cells. Besides, in U(VI)-exposed A549 cells, immediate treatment with catechol amide-derivatized polyhydroxylated fullerene significantly decreased the lactate dehydrogenase (LDH) release by inhibiting the cellular U(VI) intake, promoting the intracellular U(VI) release and inhibiting the production of intracellular reactive oxygen species (ROS). These results suggest that this fullerene derivative may be a valuable in vivo antioxidant and radionuclide decorporation agent.
