Abstract:An efficient and practical method for the synthesis of medicinally important acridines from readily available o-aminoaryl ketones and arylboronic acids was developed using copper(II)-mediated relay reactions that involve intermolecular Chan-Lam cross-coupling and subsequent intramolecular Friedel-Crafts-type reactions. A sole promoter, i.e., Cu(OTf)2, was used; therefore, strongly acidic and basic conditions, nonreadily available or expensive substrates, additives, and noble-metal catalysts were not needed.

Abstract:The reaction of o-aminoacetophenones and arylboronic acids catalyzed by copper(II) salts in the presence of pyridine under an O2 atmosphere provides a general and efficient one-pot preparation of biologically interesting acridones. This relay reaction comprises an intermolecular Suzuki cross-coupling, intramolecular oxidative C(sp3)-H amination, and C(sp2)-H activation with simultaneous rearrangement of the generated isatin intermediates. This strategy tolerates both electron-donating and -withdrawing functionalities to afford various acridones in good to excellent yields.

Abstract:A sequential multi-component reaction has been achieved via aerobic copper-catalyzed intermolecular C-N/C-O bonds cascade formation process. This mild, operationally simple method enables the transformation of commercially available aldehydes, along with aryl hydrazines, into the corresponding N',N'-diaryl acylhydrazines with high regioselectivity in moderate to good yields. This transformation employs aryl hydrazine as a dual synthon, a hydrazine and an aryl synthon. The value of this efficient synthetic protocol has been demonstrated through gram-scale experiment, late-stage modification of bioactive natural products and drug molecules. Mechanistic investigations support the generation of an acyl diazene intermediate and an aryl radical species, along with the dual role of air as the sole oxidant and source of oxygen functionality in this aerobic C-N bond coupling reaction.

Abstract:In this paper, aryldiazo sulfones are prepared from tandem sulfonylation/dehydrogenation reactions of arylhydrazines and sulfonyl chlorides. The transformations proceed well in the presence of catalytic CuSO4 center dot 5H(2)O, leading to aryldiazo sulfones in good to excellent yields. It is believed that this protocol represents a safe and convenient model for the synthesis of these versatile aryldiazo sulfones under mild conditions.

Abstract:An efficient heterogeneous copper(I)-catalyzed cross-coupling of aryl iodides and pinacolborane has been developed. The reaction proceeds smoothly in THF at room temperature by using 10 mol% of MCM-41-anchored NHC-copper(I) complex [MCM-41-NHC-CuI] as catalyst and sodium hydride as base and provides a general and practical route for the synthesis of a wide variety of arylboronates in good to high yields. This heterogenized copper(I) catalyst can be facilely prepared via a simple two-step procedure starting from easily accessible and inexpensive reagents, and exhibits a higher catalytic activity than CuI and can be recycled more than ten times without any significant loss of its catalytic activity.

Abstract:The reaction of alkenyl boronates and bromo-difluoroacetates in the presence of copper powder and TMEDA as the catalytic system was developed. The corresponding C–C coupling products were obtained in 35–93% yield. The method tolerated a variety of functional groups, namely, phenol, (thio)ether, protected amino-, hydroxy-, ketone, and carboxylic acid moieties, as well as various saturated carbo- and heterocycles, and was compatible with multigram scale up (to 76 g). The diastereoselectivity of the process was typically low, so that ca. 1:1 E/Z isomeric mixtures were formed from pure E or Z alkenyl boronates. Further functional group transformations demonstrated the utility of the prepared compounds as valuable building blocks for synthetic and medicinal chemistry. Based on the obtained results and the literature data, a plausible reaction mechanism was proposed involving the formation of radical and organocopper intermediates.

Abstract:A facile and efficient copper(I)-catalyzed protocol for the synthesis of 4-boraneyl-1,4-dihydroisoquinolin-3-ones via B–H insertion of copper carbenes derived from 4-diazoisoquinolin-3-ones into amine-borane Lewis adduct is reported. The reactions feature a good scope for the 4-diazoisoquinolin-3-ones, mild reaction conditions and high yields. The products are isolated as bench-stable Lewis adducts with Me3N.

Abstract:Pyranoquinolinones synthesized from citral were used for Cu-catalyzed N-arylation with a wide range of arylboric acids. The reaction proceeded well with a broad substrate scope, providing a direct way to access highly functional pyranoquinolinone core structure derivatives in yields of up to 80%. Compared to citral, the compounds we obtained have a much better inhibitory effect on HeLa cervical cancer cells, and compound 3p has an IC50 value of 4.6 μM, lower than cisplatin's 5.9 μM.

Abstract:Herein, we report a copper-catalyzed C-O cross-coupling of N-methoxy amides and arylboronic acids for the synthesis of aryl-N-methoxy arylimides. The fully selective O-arylation of the N-methoxy amides is found to be greatly prompted by the inexpensive and commercially available CuI. The reaction conditions tolerate a variety of functional groups and promote different reactivities depending on the electronic and steric properties of the distorted substrates.

Abstract:A Cu(I)-catalyzed aminoborylation reaction of styrenes is reported. In this transformation, diazo compounds are used as the electrophilic amination agent. The in situ generated benzylcopper species is trapped by the electrophilic nitrogen terminus of the diazo substrate to afford borylated hydrazones in a regioselective manner.

Abstract:A simple, efficient, and broad-scope rhodium(I)-catalyzed direct N-arylation of arylazocarboxylates with arylboronic acids has been developed, allowing the access to a diverse range of unsymmetrical N,N-diarylhydrazides from readily available starting materials in high yields under extraordinary mild conditions. The reaction features easy operation, ambient temperature and low catalyst loading (as low as 0.1 mol%), as well as broad functional-group compatibility. The resulting N,N-diarylhydrazides could serve as versatile building blocks to furnish other structurally important or pharmaceutically interesting molecules. This protocol provides an attractive and complementary approach for the assembly of vaulable N,N-diarylamine scaffolds.
