Abstract:Organic compounds bearing both silyl and boryl groups are important building blocks in organic synthesis because of the adequate reactivity of the silyl and boryl groups and high stereospecificity in their derivatization reactions. The difference in reactivity between the silyl and boryl groups enables stepwise derivatization of these groups to afford complex molecules. Here, we report the copper(I)-catalyzed silaboration of terminal allenes to produce multisubstituted allylic boronates embedded with an alkenyl silane structure. The reaction can proceed with a variety of allenes and silylboranes. Furthermore, the silyl and boryl groups were successfully converted into other functional groups, while retaining the stereochemistry of the alkene moiety.

Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

Abstract:A highly regio- and stereoselective copper-catalyzed borylcupration of 1,2-allenylsilanes affords an unexpected regioreversed allylic boronate bearing an extra C-Si bond at the 3-position, with a thermodynamically disfavored Z geometry. Such stereodefined allylic boronates containing an extra alkenyl silane moiety are very useful organodimetallic reagents for organic synthesis.

Abstract:A copper-catalyzed three-component reaction of cyclobutanone oxime esters and 1,3-enynes in the presence of TMSCN or TMSCF3 has been developed. This mild protocol enjoys a broad substrate scope tolerating many functional groups, providing a facile access to 1,7-double-functionalized allenes, which are difficult to prepare. The allenyl nitrile products may be easily transformed into allenoic acid derivatives and stereodefined tetrasubstituted alkenes, demonstrating their potentials as platform molecules in synthesis. Abstract:A highly chemo- and regioselective copper-catalyzed borylcupration of 1,2-allenylsilanes affords 2-borylallylsilanes by applying the ligand effect. Many synthetically attractive functional groups are well tolerated. As demonstrated, such 2-borylallylsilanes are very useful dimetallic reagents in organic synthesis. A rationale for the regioselectivity switch is provided.

Abstract:A catalytic enantioselective conjugate addition with commercially available allenylboronic acid pinacol ester as nucleophile promoted by a chiral copper complex of N-heterocyclic carbene (NHC) is disclosed. This process constitutes an unprecedented instance of the conjugate addition that introduces an allenyl group into α,β-unsaturated carbonyl compounds, affording products that are otherwise difficult to access in up to 92% yield, >98% allenyl addition selectivity and 96:4 enantiomeric ratio. DFT calculations were performed to elucidate the origins of enantioselectivity.

Abstract:The Cu-catalyzed 1,4-protosilylation and protoborylation of trifluoromethyl-substituted conjugated enynes were developed to access functionalized homoallenylsilanes and homoallenylboronates. This protocol also provides a general method to synthesize optically active homoallenylsilanes and homoallenylboronates in moderate to excellent yields with high enantiomeric excess by using new designed chiral bisoxazoline ligands. Simultaneously, the transformations of homoallenylsilanes and homoallenylboronates were also explored to synthesize useful building blocks.

Abstract:A copper-catalyzed highly regio- and stereoselective silaboration of alkynes was developed. In this work, direct cis-difunctionalization of alkynes was realized with silaboronate reagent and copper catalyst in aprotic solvents. The regiodivergent silaborations were controlled by tuning the copper catalysts and phosphine ligands used in reactions. This protocol provides an efficient and practical method to synthesize the multisubstituted functionalized alkenes with specific stereoselectivity.

Abstract:Recent years have witnessed the growing interest in the remote functionalization of alkenes for it offers a strategy to activate the challenging C-H bonds distant from the initiation point via alkene isomerization/functionalization. However, the catalytic enantioselective isomerization/functionalization with one single transition metal catalyst remains rare. Here we report a highly regio- and enantioselective cobalt-catalyzed remote C-H bond borylation of internal alkenes via sequential alkene isomerization/hydroboration. A chiral ligand featured twisted pincer, anionic, and non-rigid characters is designed and used for this transformation. This methodology, which is operationally simple using low catalyst loading without additional activator, shows excellent enantioselectivity and can be used to convert various internal alkenes with regio- and stereoisomers to valuable chiral secondary organoboronates with good functional group tolerance.

Abstract:Allenenitriles bearing different synthetically versatile functional groups have been prepared smoothly from 5-alkynyl fluorosulfonamides in decent yields with an excellent chemo- and regio-selectivity under redox neutral conditions. The resulting allenenitriles can be readily converted to useful functionalized heterocycles. Based on mechanistic study, it is confirmed that this is the first example of radical-based non-activated propargylic C-H functionalization for allene syntheses.

Abstract:Ethyne is employed as coupling partner in copper-mediated cross-coupling reactions with N-tosylhydrazones and α-diazoacetate, leading to the development of a new synthetic method for terminal allenes. With this novel coupling method, the terminal allenes were obtained in good yields and with excellent functional group tolerance. Copper carbene migratory insertion is proposed as the key step in these transformations.

Abstract:Chiral allene and N -heteroaryl motifs are present in an ever-growing list of biologically active natural products and synthetic drugs. Although significant progress has been made in asymmetric syntheses of chiral allenes, general and practical protocols for enantioselective syntheses of chiral N -heteroaryl-substituted allenes from readily available starting materials still remain rare. Here we report a highly enantioselective synthesis of quinolinyl-substituted chiral allenes through a copper-catalyzed asymmetric allenylation of quinoline N -oxides with readily available 1,3-enynes. A variety of 1,3-enynes react with quinoline N -oxides, affording the corresponding quinolinyl-substituted allenes in high yields (up to 95%) and high enantioselectivities (up to 99% ee). This transformation tolerates a variety of functional groups, such as chloro, bromo, trifluoromethyl ether, tertiary amine, siloxy, carboxylic ester, imide, pyridine, and thiophene moieties. DFT calculations suggest a pathway involving an intramolecular nucleophilic addition of an allenyl copper intermediate with a coordinated quinoline N -oxide through a five-membered, rather than seven-membered, transition state.

Abstract:Borylation of 1,3-enynes with bis(boronate) compounds often ends up with the formation of hydroborylated products, leaving the diborylation of 1,3-enynes for the formation of 1,4-diborylated allenes to be challenging. Herein, a copper-catalyzed chemo-, regio-, and stereo-selective diborylation of 1,3-enynes for the efficient construction of 1,4-diborylated allenes under base-free conditions was reported. A wide range of 1,3-enynes bearing various functional groups can participant in the reaction and afforded the corresponding 1,4-diborylated allenes in good to excellent yields, which was enabled by the protocol of Bpin to BF 3 K conversion. the borylcopper species was supposed to selectively attack the C−C triple bond of the 1,3-enynes.

Abstract:The synthesis of furan-substituted allenes using conjugated ene-yne ketones as carbene source has been developed. For this reaction, bases play vital roles in controlling the reaction pathways, allowing for access to two types of allene products through trapping of different electrophiles. Mechanistically, the catalytic procedure generated a Cu(I) (2-furyl)carbene intermediate, which is subsequently followed by a migratory insertion process to afford nucleophilic organocopper species. The organocopper species thus generated can be trapped by proton or allyl halide, affording tri- or tetrasubstituted allenes, respectively. The reaction, which is characterized by its mild reaction conditions and the utilization of cheap copper(I) iodide as catalyst, allows for synthesis of a variety of furan-substituted allenes with a wide range of functional groups tolerance.

Abstract:Herein, we developed the first example of copper-catalyzed silicon radical-initiated 1,4-silylcyanation of unactivated 1,3-enynes, which provided an efficient method to access CN-bearing tri- and tetra-substituted homoallenylsilane derivatives in high yields with excellent regioselectivities. This protocol featured good functional group compatibility and broad substrate scopes, enabling the formation of C-Si bond under cheap copper catalyst with a low loading. Furthermore, this means showed potential application value in the late-stage functionalization of natural products.

Abstract:Ene-allenes serve as versatile building blocks in organic synthesis, and the development of their efficient preparation is valuable. Herein the synthesis of boryl-substituted ene-allenes utilizing a Cu/Pd-catalyzed borylallenylation of alkynes with propargylic carbonates and bis(pinacolato)diboron is reported. Densely (tetra-, penta-, and hexa-) substituted ene-allenes were synthesized in acceptable yield with high regio- and stereoselectivity. Abstract:A copper-catalyzed efficient regioselective silylation reaction of chloro-substituted allenyl-Bdan was developed. Under mild reaction conditions, allenyl and propargyl silane compounds can be selectively obtained in moderate to high yields by adjusting the bases and solvents used in the reactions.
