Abstract:A HFIP-promoted highly selective hydroxyalkylation of aniline derivatives with arylglyoxal hydrates has been realized. The reaction produces various N,N-dialkylanilines and their derivatives with α-hydroxy carbonyl units in good to excellent yields under mild conditions. Furthermore, the synthetic potential of this method has been demonstrated by the facile synthesis of several structurally interesting molecules such as benzil, 1,2,4-triazine, quinoxaline, hydantoin, and 2-thiohydantoin with aromatic amine units.

Abstract:The rearrangement and demethylaromatization of cyclohexadienimines (namely cyclohexadienone imines) were detailedly investigated under metal-free conditions. Treating 4-aryl-4-methylcyclohexadienimines with acyl chloride at 100 °C in dichloromethane led to the smooth formation of m-arylaniline derivatives in good to excellent yields, in which [1,2]-migration of aryl group at C-4 occurred exclusively. The demethylaromatization of 4-aryl-4-methylcyclohexadienimines mediated by iodotriphenylphosphonium iodide (in-situ prepared via the reaction of triphenylphosphine with iodine) in toluene at 100 °Cproceeded well generating p-arylanilines in moderate to good yields. An efficient and alternate method for the synthesis of polysubstituted aryl amines especially m-arylaniline derivatives which are otherwise difficult to synthesize through traditional method was developed.

Abstract:A new pair of reaction partners has been established for the aromatic C-H functionalization of benzyl electrophiles with nucleophiles via palladium-catalyzed benzylic C-N cleavage. A range of N-(1-naphthylmethyl)sulfonimides, N-(2-thienylmethyl)sulfonimides, and N-(2-furanylmethyl)sulfonimides smoothly underwent palladium-catalyzed aromatic C-H allylation with allyl Grignard reagents at room temperature, delivering structurally diverse substituted 1-allylnaphthalenes and 2-allylheteroarenes in moderate to excellent yields with extremely high regioselectivities. Replacing the N-(arylmethyl)sulfonimide with an (arylmethyl)ammonium salt, an arylmethyl chloride, or an arylmethyl phosphate as the benzyl electrophile leads to a dramatic erosion of the regioselectivity.

Abstract:Nitrogen containing compounds, such as anilines, are some of the most widespread and useful chemical species, although their high and unselective reactivity has prevented their incorporation into many interesting transformations, such as the functionalization of alkenes. Herein we report a method that allows the trifluoromethylarylation of alkenes using anilines, for the first time, with no need for additives, transition metals, photocatalysts or an excess of reagents. An in-depth mechanistic study reveals the key role of hexafluoroisopropanol (HFIP) as a unique solvent, establishing a hydrogen bonding network with aniline and trifluoromethyl reagent, that is responsible for the altered reactivity and exquisite selectivity. This work uncovers a new mode of reactivity that involves the use of abundant anilines as a non-prefunctionalized aromatic source and the simultaneous activation of trifluoromethyl hypervalent iodine reagent.

Abstract:alpha,beta-Unsaturated nitriles were found to be suitable substrates for aminochlorination with N,N-dichloro-p-toluenesulfonamide (4-TsNCl2) in the presence of CuCl as the catalyst (10 mol-%) and 4 angstrom molecular sieves. The reaction is very convenient to carry out at room temperature without the protection of inert gases, and this method provides an easy route to vicinal haloamino nitriles with excellent regio- and stereoselectivities and in good chemical yields. Abstract:Anilines are fundamental motifs in various chemical contexts, and are widely used in the industrial production of fine chemicals, polymers, agrochemicals and pharmaceuticals. A recent development for the synthesis of anilines uses the primary amination of C-H bonds in electron-rich arenes. However, there are limitations to this strategy: the amination of electron-deficient arenes remains a challenging task and the amination of electron-rich arenes has a limited control over regioselectivity-the formation of meta-aminated products is especially difficult. Here we report a site-directed C-C bond primary amination of simple and readily available alkylarenes or benzyl alcohols for the direct and efficient preparation of anilines. This chemistry involves a novel C-C bond transformation and offers a versatile protocol for the synthesis of substituted anilines. The use of O-2 as an environmentally benign oxidant is demonstrated, and studies on model compounds suggest that this method may also be used for the depolymerization of lignin.

Abstract:Anilines generally act as N-nucleophiles in transition-metal-catalyzed allylic substitution reactions. In this paper, a highly enantioselective intramolecular Friedel-Crafts-type allylic alkylation of aniline derivatives was realized by using an iridium catalyst derived from [Ir(cod)Cl](2) and (R-a)-BHPphos. Various tetrahydroisoquinilin-5-amines were obtained in moderate to good yields, excellent enantioselectivity and regioselectivity under mild reaction conditions. BHPphos=N-benzhydryl-N-phenyldinaphthophosphoramidite.

Abstract:A combination of cupric bromide and Oxone® serves as a facile, mild and effective reagent for the bromination of aromatic amines. Primary, secondary and tertiary aromatic amines are all suitable substrates. The reaction possesses high regioselectivity and functional group tolerance, and mono- and multi-brominated products can be obtained controllably in moderate to excellent yields.

Abstract:In the presence of chloroformates, the in situ formed allylic stannanes trapped the N-alkoxycarbonyl salt of azaaromatics to give allylation products. Several kinds of azaaromatics were used as substrates, such as imidazole, benzothiazole, pyridine, pyrimidine, etc. The reaction of crotyl bromide showed high regioselectivity.

Abstract:An efficient and broadly applicable rhodium-catalyzed direct ortho-arylation of anilines with aryl iodides relying on readily available aminophosphines as traceless directing groups is reported. Its scope and functional group compatibility were both found to be quite broad as a large variety of both aminophosphines and (hetero)aryl iodides, including complex ones, could be utilized. The ortho-arylated anilines could be obtained in high average yields, without any competing diarylation and with full regioselectivity, which constitutes a major step forward compared to other processes. The reaction is moreover not limited to aryl iodides, as an aryl bromide and a triflate could be successfully used, and could be extended to diarylation. Mechanistic studies revealed the key and unique role of the aminophosphine, acting not only as a substrate but also as a ligand for the rhodium catalyst.

Abstract:The charge-accelerated aza-Claisen rearrangement of ammonium salts serves as a key step in the construction of complex nitrogen-containing molecules. However, much less attention has been paid to the aromatic aza-Claisen rearrangement than to the aliphatic one. Herein, we report an unprecedented aromatic aza-Claisen rearrangement of arylpropargylammonium salts, generated in situ from arynes and tertiary propargylamines, delivering structurally diverse 2-propargylanilines in moderate to good yields with high regioselectivity. This rearrangement proceeds in the absence of strong bases or transition metals, is compatible with moisture and air, tolerates a wide variety of functional groups, and is amenable to forming 11- to 13-membered heterocycles with a triple bond. The 2-propargylaniline products were treated with aluminum chloride in ethanol to afford multisubstituted indoles in moderate to excellent yields. Finally, a series of deuterium-labeling experiments was performed to elucidate the reaction mechanism.

Abstract:Six Cp*Ir complexes containing NN-bitentate chelate ligands [Cp*IrCl(C5H4CH2C5H3OH)][Cl] (1), [Cp*IrCl(C5H4CH2C5H3O)] (2), [Cp*IrCl(C5H4C5H3OH)] [Cl] (3), [Cp*IrCl(C5H4CH2C5H4)][Cl] (4), [Cp*IrCl(CH3OC5H3CH2C5H3OCH3)][Cl] (5), and [Cp*IrCl(CH3OC5H3CH2C5H3OH)][Cl] (6) were synthesized and characterized. Complex 1 could be transformed to 2 when reacted with (NaOBu)-Bu-t or NEt3 via -OH deprotonation. These six complexes were tested as catalysts for mono-N-alkylation of amines with trialkyl amines, and complex 1 exhibited highest activity. The coupling reactions proceed under air condition, with 1 mol % catalyst loading without extra base in methanol at 120 degrees C and can be further accelerated by adding NR3.HCl.
