Abstract:Catalytic amination of aryl bromides with in situ generated dimethylamines from N, N-dimethylacetamide (DMA) has been Successfully carried Out using Ni(Phen)Cl-2 as catalyst. Both electron-rich and electron-poor aromatic system reacted smoothly under the conditions to give N,N-dimethylarylamines in good yields.

Abstract:The incorporation of the privileged amino functionality is of paramount importance in organic synthesis. In contrast to the well-developed amination methods for alkenes, the dearomative amination of arenes is largely underexplored due to the inherently inert reactivity of arene π-bonds and selectivity challenges. Herein, we report an intermolecular dearomative aminofunctionalization via direct nucleophilic addition of simple amines to chromium-bound arenes. This multicomponent 1,2-amination/carbonylation reaction enables rapid access to complicated alicyclic compounds containing amino and amide functionalities from benzene derivatives under CO-gas-free conditions, which also represents the first application of nitrogen-based nucleophiles in η6 -coordination-induced arene dearomatizations.

Abstract:N-Heterocyclic carbene (NHC) catalyzed direct carbonylation of dimethylamine leading to the formation of DMF was successfully accomplished under metal-free conditions. The catalytic efficiency was investigated and the turnover numbers can reach as high as >300. The possible mechanism was also proposed.

Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:Aryl amines are highly useful organic chemicals, but large-scale, transition-metal-free syntheses of aryl amines are surprisingly underdeveloped. A mild and scalable (up to 500 mmol) aryl amine synthesis from benzyne chemistry was invented using easily accessible aryl chlorides as precursors, NaH as a stoichiometric base, and a new type of sodium alkoxide cluster, X@RONa, as a catalyst. The cluster catalyst X@RONa featured an externally hydrophobic dodecameric sodium alkoxide shell housing an encapsulated center anion. The cluster made from methoxy-tert-butanol was found to be the most effective. The intramolecular version of this reaction allowed the synthesis of indolines and indoles. Experimental and computational mechanistic studies revealed that the rate-determining step was likely the transport of solid NaH into the X@RONa cluster in the organic phase.

Abstract:With the readily available Gorlos-Phos as a ligand, halogenated diarylamines were prepared efficiently from Pd-catalyzed monoamination of bromophenyl chlorides and primary arylamines, tolerating functional groups such as aldehyde, ketone, nitrile, fluorine, sulphonyl and trifluoromethyl groups. The formation of the amination of the C-CI bond and diamination products were not observed.

Abstract:Reaction of electron-poor amines with N-(3-bromopropyl)phthalimide in equivalent Et3N under solvent-free condition at 120 ℃ gave 2-[3-(arylamino)-propyl]-isoindole-1,3-dione in moderate yield.These intermediates were easily converted into N1-(electron-poor aryl)-propane-1,3-diamines by hydrazinolysis with hydrazine hydrate in high yield.The structures of said compounds were identified by IR,1HNMR,13CNMR and HRMS spectra.This method had some advantages such as simple operation,good yield and readily available starting materials.

Abstract:Herein, we disclose the metal-free synthesis of arylamines via the direct amination of phenols using aminating reagents. This reaction procedure uses easy accessible aminating reagents and provides a versatile synthetic route to a broad range of arylamines with various functionalities in good to excellent yield. By using a two-step route of amination and oxidative coupling reaction, we synthesized three naturally occurring carbazole alkaloids: murrayafoline A, mukonine, and clausenine from two commercially available phenols.

Abstract:The cross-coupling of bromomagnesium diarylamides, generated in situ from diarylamines, with aryl bromides or iodides can be effected with a simple NiCl2(PPh3)(2)-PPh3 catalyst system under relatively mild conditions. This coupling reaction is an inexpensive, convenient, and practical method, functioning as an alternative to the corresponding Pd-catalyzed or Cu-mediated process for the synthesis of triarylamines.

Abstract:Regio- and enantioselective hydroarylamination, hydroalkylamination and hydroamidation of styrenes have been developed by NiH catalysis with a simple bioxazoline ligand under mild conditions. A wide range of enantioenriched benzylic arylamines, alkylamines and amides can be easily accessed by nitroarenes, hydroxylamines and dioxazolones, respectively as amination reagents. The chiral induction in these reactions is proposed to proceed through an enantiodifferentiating syn-hydronickellation step.

Abstract:A new efficient regioselective boron-assisted mono bromination methodology for arylamines is described. Our one-pot, three-stage approach (lithiation, boron amide formation and bromination) proved to be highly useful in mildly brominating a variety of arylamines in up to 94% yields.

Abstract:A general and efficient synthesis of α‐amino‐α‐aryl carbonitriles through the three‐component reaction of arynes, aroyl cyanides, and DMF in a single step is described. DMF was not only used as the solvent, but it also participated in the reaction as a component. Used as the cyanide sources, the aroyl cyanides solved the toxicity and solubility problems associated with the use of HCN or metal cyanides in the Strecker synthesis. This procedure furnished α‐amino‐α‐aryl carbonitriles in moderate to good yields with broad substrate scope. Moreover, this reaction could be performed under mild conditions with high atom economy.

Abstract:A simple, efficient, and broad-scope rhodium(I)-catalyzed direct N-arylation of arylazocarboxylates with arylboronic acids has been developed, allowing the access to a diverse range of unsymmetrical N,N-diarylhydrazides from readily available starting materials in high yields under extraordinary mild conditions. The reaction features easy operation, ambient temperature and low catalyst loading (as low as 0.1 mol%), as well as broad functional-group compatibility. The resulting N,N-diarylhydrazides could serve as versatile building blocks to furnish other structurally important or pharmaceutically interesting molecules. This protocol provides an attractive and complementary approach for the assembly of vaulable N,N-diarylamine scaffolds.

Abstract:A reliable method is introduced for the synthesis of branched α‐arylated amines from simple linear N‐sulfonyl amines and aryl boroxines utilizing dual palladium catalysis involving amine dehydrogenation. Bromobenzene serves as a hydride acceptor, and the ensuing imine intermediate undergoes a novel umpolung arylation. Given the wide availability of primary amines, this method can be expected to be useful for the synthesis of structurally complex amines with varied functionality. Functionalization of C(sp3)−H bonds adjacent to a nitrogen atom is an efficient means of preparing structurally complex amines and has long been of interest to synthetic chemists. Herein, dual palladium catalysis is used to achieve α‐C−H arylation of linear N‐sulfonyl amines with aryl boroxines, rather than N‐arylation (C−N bond formation). This method has a broad scope with respect to both the amine and the boroxine, providing convenient access to α‐branched amines. During the dehydrogenation step of the catalytic cycle, bromobenzene accepts a hydride from the α‐carbon on the amine, producing an imine that subsequently undergoes Pd‐catalyzed C‐arylation with the boroxine. Mechanistic studies revealed that imine arylation proceeds through an unusual Pd0/PdII catalytic cycle, in which oxidative cyclization of a palladium η2‐imine complex generates a three‐membered palladacycle that subsequently undergoes transmetallation reaction with the boroxine.

Abstract:A copper-catalyzed 1,3-aminoalkynylation or 1,3-aminobromination of arylcyclopropane with N -fluorobenzenesulfonimide (NFSI) and trimethoxy[( trimethylsilyl ) ethynyl ]silane or LiBr have been successfully developed. Through this strategy, a wide range of γ-alkylnyl-/γ-bromino-γ-branched amine derivatives were efficiently synthesized in good yields under mild conditions.

Abstract:A new and practical α-monomethylation strategy using an amine-borane/N,N-dimethylformamide (R3 N-BH3 /DMF) system as the methyl source was developed. This protocol has been found to be effective in the α-monomethylation of arylacetonitriles and arylacetamides. Mechanistic studies revealed that the formyl group of DMF delivered the carbon and one hydrogen atoms of the methyl group, and R3 N-BH3 donated the remaining two hydrogen atoms. Such a unique reaction pathway enabled controllable assemblies of CDH2 -, CD2 H-, and CD3 - units using Me2 NH-BH3 /d7 -DMF, Me3 N-BD3 /DMF and Me3 N-BD3 /d7 -DMF systems, respectively. Further application of this method to the facile synthesis of anti-inflammatory flurbiprofen and its varied deuterium-labeled derivatives was demonstrated.

Abstract:The systematic synthesis of a series of N-acyl chloroformamidines is achieved by reaction of carbodiimides with acid chlorides; the structure of an N-acyl chloroformamidine was confirmed by X-ray diffraction. The reactivity of N-acyl chloroformamidines towards diethyl malonate, phenylethynyllithium, pentane-2,4-dione, methyl acetoacetate, water, 1,3-dimethylurea, 1,3-dimethylthiourea, and sodium hydrosulfide, resulted in unique protocols for the synthesis of N-acyl-substituted 1,1-diaminoethylenes, amidines, ureas, and thioureas, respectively. These results show that N-acyl chloroformamidines have a good ability to accept nucleophilic attack for the construction of useful N-containing compounds.

Abstract:The rearrangement and demethylaromatization of cyclohexadienimines (namely cyclohexadienone imines) were detailedly investigated under metal-free conditions. Treating 4-aryl-4-methylcyclohexadienimines with acyl chloride at 100 °C in dichloromethane led to the smooth formation of m-arylaniline derivatives in good to excellent yields, in which [1,2]-migration of aryl group at C-4 occurred exclusively. The demethylaromatization of 4-aryl-4-methylcyclohexadienimines mediated by iodotriphenylphosphonium iodide (in-situ prepared via the reaction of triphenylphosphine with iodine) in toluene at 100 °Cproceeded well generating p-arylanilines in moderate to good yields. An efficient and alternate method for the synthesis of polysubstituted aryl amines especially m-arylaniline derivatives which are otherwise difficult to synthesize through traditional method was developed.

Abstract:De novo catalytic syntheses of diarylamines from a palladium‐catalyzed reductive Buchwald‐Hartwig amination of nitroarenes with aryl (pseudo)halides is described. The exquisite use of upstream nitroarenes as arylamine surrogates, the judicious selection of bis(pinacolato)diboron (B2pin2) as a stoichiometric reducing agent, and wide substrate scope including (hetero)aryl halides (Cl, Br and I) and aryl triflates, constitute the striking features of the current protocol. Moreover, application of this technique to the syntheses of advanced intermediates and active pharmaceutical ingredients also proved successful, thus providing an alternative step‐economical approach to the syntheses of diarylamine‐incorporated molecules. Preliminary mechanistic investigation demonstrates that an amine and a nitrosoarene intermediates might be involved in this reductive event.
