Abstract:A new indole alkaloidal glucoside together with three known compounds aurantiamide acetate (2), eleutheroside E (3) and 1-O-caffeoyl-β-D-glucopyranoside (4) has been isolated from ethanol extract of the aerial parts of Clematis terniflora DC. On the basis of their spectroscopic and chemical evidence, the new compound was elucidated as (6-O-β-D-glucopyranosyl-1H-indol-3-yl) carboxylic acid methyl ester (1). Compounds 1 and 3 showed significant cytotoxicity against human ECA-109.
A Bioactive Alkaloid from the Flowers of Trollius Chinensis.

Abstract:A new compound, named as trolline was isolated from the ethanol extract of the flowers of Trollius chinensis Bunge. The chemical structure of this compound was elucidated by NMR, MS, IR, UV spectra and single-crystal X-Ray analysis. The in vitro antibacterial and antiviral experiments revealed that trolline exhibited appreciable antibacterial activity against respiratory bacteria such as Staphylococcus aureus, Streptococcus pneumoniae and Klebsiella pneumoniae. Trolline was also demonstrated to have moderate antiviral activity against influenza virus A and B.
Analysis of bioactive Amaryllidaceae alkaloid profiles in Lycoris species by GC-MS

Abstract:The genus Lycoris, a group of Amaryllidaceae plants distributed in temperate regions of Eastern Asia, is already known for containing representative alkaloids typical of this botanical family with a wide range of biological activities (for example, lycorine and galanthamine). In the present work, the alkaloid profiles of nine species, L. albiflora, L. aurea, L. chinensis, L. haywardii, L. incarnata, L. longituba, L. radiata, L. sprengeri, and L. squamigera, and one variety (L. radiata var. pumila) have been evaluated by GC-MS. Structures belonging to the lycorine-, homolycorine-, haemanthamine-, narciclasine-, tazettine-, montanine- and galanthamine-series were identified and quantified, with galanthamine- and lycorine-type alkaloids predominating and usually showing a high relative abundance in comparison with other alkaloids of the extracts. Interestingly, L. longituba revealed itself to be a potential commercial source of bioactive alkaloids. In general terms, our results are consistent with the alkaloid profiles reported in the literature for previously studied species.

Abstract:Three new tetramic acid derivatives, tolypocladenols A1, A2, and B (1-3), a new pyridone alkaloid, tolypyridone A (4), and a new coumarin derivative, 3,8-dihydroxy-4-(4-hydroxyphenyl)-6-methylcoumarin (5), together with four known compounds (6-9) were isolated from the endolichenic fungus Tolypocladium cylindrosporum, which inhabits the lichen Lethariella zahlbruckneri. Structures of these compounds were determined by comprehensive analysis of spectroscopic data and single-crystal X-ray diffraction determination. Bioassay of the isolated compounds found that pyridoxatin (7) was cytotoxic to human cancer cells by induction of G0/G1 cell cycle arrest and apoptosis.

Abstract:Eight alkaloids were isolated from the bulb of Fritillaria Wabuensis S.Y. Tang et S.C. Yueh the best "Chuan Bei" cultivated in Sichuan province, which is efficacious against cough. Five of them are known, identified as imperialine (I), peimisine (II), ebeinone (III), isoverticine (IV), and imperialine-beta-N-oxide (V). All the signals for both 1H and 12C-NMR spectra of base (V) were assigned by 2D NMR experiments, and an X-ray structural data of alkaloid (V) was obtained for the first time.

Abstract:This study is to investigate the chemical constituents from the whole plant Corydalis edulis. The chemical constituents were separated and purified by macroporous resin D101, silica gel, Sephadex LH-20, ODS, and semi-preparative HPLC. Their structures were determined by physicochemical properties and spectroscopic data. Four compounds were isolated from the dichloromethane and water extracts of the whole plant C. edulis, and identified as 6'-β-D-xylosylicariside B2(1),(3S,5R,6S,7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one(2), loliolide(3), and 5,5'-dimethoxybiphenyl-2,2'-diol(4), respectively. Compound 1 is a new compound, of which the absolute configuration was established by electronic circular dichroism(ECD) calculations. Compound 4 is obtained from the plants of Papaveraceae family for the first time. Compounds 2 and 3 are firstly isolated from the Corydalis genus.

Abstract:Chemical investigation of the EtOAc extract of an insect-associated fungus Letendraea sp. 5XNZ4-2 cultured in Potato Dextrose Broth (1/2 PDB) medium lead to the isolation of three new polyketides, named letendronol D (1), phomopsiketones H-I (2-3). The structures of new compounds were elucidated by the analysis of HRESIMS and NMR spectroscopic data, and the absolute configurations were determined by modified Mosher’s method, ECD calculation and single-crystal X-ray diffraction. Cytotoxicity and antibacterial activities of 1 were assayed and regrettably 1 didn’t display any cytotoxicity and antibacterial activity. 3 was the first phomopsiketone derivative obtaining the lactone.

Abstract:Lycoris aurea, celebrated for its visually striking flowers and significant medicinal value due to the presence of alkaloids such as lycorine and galanthamine, has intricate yet poorly understood regulatory mechanisms. This study provides a detailed examination of the transcriptomic, metabolomic and ecological dynamics of L. aurea, aiming to elucidate the underlying molecular mechanisms of alkaloid biosynthesis. Our comparative analysis across different ecological settings highlighted key genes involved in alkaloid biosynthesis, such as genes encoding aldehyde dehydrogenase and norbelladine 4′‐O‐methyltransferase, which were distinctively increased in the high alkaloids‐producing group. We identified a total of 6871 differentially expressed genes and 915 metabolites involved in pathways like terpenoid backbone biosynthesis, phenylalanine, tyrosine and tryptophan biosynthesis. Protein interaction network analysis revealed significant upregulation of photosynthesis, photosystem and photosynthetic membrane pathways in the alkaloids‐producing region. Furthermore, our research delineated the interactions among soil microbial communities, genes and plant and soil biochemical properties, noting that bacterial populations correlate with soil properties that favour the activation of metabolic pathways essential for alkaloid production. Collectively, this study advances our understanding of the genetic and metabolic alkaloid biosynthesis pathways in L. aurea, shedding light on the complex interactions that govern alkaloid production.

Abstract:Former study suggests alkaloids from herbs of Aconitum genus plants possess excellent bioactivities, which exert great value for related deeper chemical constituent investigation. Herein, chemical isolation was performed and four alkaloids were isolated from Fuzi, of which two were new ones, and the other two were reported NMR data for the first time. Their chemical structures were identified by NMR data, high resolution MS, UV and IR analysis. Additionally, the MS fragmentation patterns were explored, formerly, that of hetisane alkaloid was rarely reported, and fragmentation mechanism of the diagnostic ion was proposed. Based on these fragment pathway, metabolites and metabolic pathways of four compounds were investigated in rat liver microsomes using UPLC-Q/TOF-MS, and dehydrogenation product was firstly found from metabolites of hetisane alkaloid.

Abstract:A new alkaloid, named 2-methoxy-4-(2-(2-pyridine)-ethyl) phenol (1), together with two known compounds, was isolated from Zingiberis rhizoma. Their structures were elucidated on the basis of 1D and 2D NMR spectra and MS spectra. Compound 1 exhibited substantial bioactivity against Canidia albicans ATCC 10231 with a minimum inhibitory concentration of 1.0 mg/mL.

Abstract:Liriogerphines A–D (1–4, respectively), an unprecedented class of hybrids of germacranolide-type sesquiterpenoids and aporphine-type alkaloids, were isolated from the rare medicinal plant Liriodendron chinense. Their structures were elucidated by comprehensive spectroscopic analyses combined with electronic circular dichroism calculations and X-ray crystallographic data. Biosynthetically, an aza-Michael addition reaction is proposed to be involved in the assemblies of this class of hybrids. Compound 4 exhibited cytotoxicity against leukemia cells via inducing apoptosis and inhibiting Bcl-2 expression.

Abstract:Eight undescribed steroidal alkaloid derivatives, including three cevanine-type isosteroidal alkaloids (two N -oxide glycosides and one D-ring aromatization) ( 1 – 3 ), one verazine-type steroidal alkaloid derivative ( 4 ), three solanidine-type steroidal alkaloid glycosides ( 5 – 7 ), and one veratramine-type analogue ( 8 ), along with three known compounds ( 9 – 11 ) were isolated from the bulbs of Fritillaria sinica . Their structures were elucidated by comprehensive analysis of spectroscopic data, acidic hydrolysis, and X-ray crystal diffractions. In the in vitro bioassay, the anti-cancer effect, anti-oxidation and anti-inflammatory activities for the isolates were evaluated at a concentration of 10 μM.

Abstract:A undescribed phenolic glycoside, trochinenol A (1), was isolated from the flowers of Trollius chinensis Bunge and the structure was identified by spectroscopic methods. Its anti-inflammatory and antibacterial effects were investigated by broth microdilution and NF-kappa B reporter gene assays. Consequently, compound 1 exhibited an appreciable effect against Staphylococcus aureus with the MIC value of 6.25 mu g/mL. Besides, it showed moderate effect against TNF alpha-induced activation of NF-kappa B pathway.
