Abstract:A new class of highly oxygenated Schisandra nortriterpenoids, lancolides A-D(1-4), from Schisandra lancifolia, represents the first example of natural products that possess a tricyclo[6.3.0.0(2.11)]undecane-bridged system. Their structures were elucidated by NMR spectra, X-ray diffraction, and quantum chemical calculations. Lancolides A (1) and D (4) had specific antiplatelet aggregation induced by platelet-activating factor (PAF).

Abstract:Three unique nortriterpenoids, schilancitrilactones A-C (1-3), were isolated from the stems of Schisandra lancifolia . Compound 1 possesses a 5/5/7/5/5/5-fused hexacyclic ring system with a C(29) backbone, while 2 and 3 feature a C(27) skeleton with a 5/7/5/5/5-fused pentacyclic ring system. Their absolute stereochemistries were established by CD and single-crystal X-ray diffraction experiments. Compound 3 showed anti-HIV-1 activity with an EC(50) value of 27.54 μg/mL, and 1 exhibited antifeedant activity at 15.73 μg/cm(2).

Abstract:Ten schinortriterpenoids with biogenetically related lancischiartane scaffolds, including the first 3-norlancischiartane (1) with unusual configuration inversions occurring at C-1 and C-10, were isolated from Schisandra lancifolia. Unusual ECD curve patterns observed in 6–8 were confirmed to be caused by nondegenerate exciton coupling, suggesting that ECD spectrum empirical comparison should be used with caution in configuration determination. Additionally, structure revision of 2, originally proposed as arisanlactone A, was completed using NMR computation and X-ray diffraction analysis.

Abstract:Schicagenins A-C (1-3), three unprecedented nortriterpenoids characterized with a tetracyclic oxa-cage motif and C(9) side chain, were discovered from the leaves and stems of Schisandra chinensis. Their structures were determined on the basis of extensive spectroscopic analysis, and the absolute stereochemistries were established by single-crystal X-ray diffraction and CD experiments. A plausible biosynthetic pathway of 1-3 was also discussed.

Abstract:Twelve new nortriterpenoids, namely wuweizidilactones J-P (1-7), schindilactones I-K (8-10), preschisanartanin N (11), and schisdilactone J (12), as well as 23 known analogues, were isolated from the leaves and stems of Schisandra chinensis. The absolute configurations of 1 and 11 were established by calculated electronic circular dichroism (ECD) spectra and that of 7 was confirmed by single-crystal X-ray diffraction. The absolute configurations of 2-6, 8, and 9 were determined by an empirical comparison of their experimental ECD spectra with that of 1. It is the first time that the ECD spectra of 18(13 -> 14)-abeoschiartane-, 18-norschiartane-, and schisanartane-type nortriterpenoids possessing an alpha,beta-unsaturated-gamma-lactone moiety are presented, this method providing an alternative means of obtaining absolute configurational assignments for these types of nortriterpenoids. All isolates were evaluated for their anti-acetylcholinesterase and anti-butyrylcholinesterase activities, and compounds 8, 13, 23, and 31 show anti-acetylcholinesterase activity at concentrations of 50 mu M, with 12.7, 10.7, 16.6, and 32.1% inhibition, respectively. Abstract:An ultra performance liquid chromatography (UPLC) coupled with quadrupole time-of-flight mass spectrometry (QTOF-MS) procedure is designed for the first simultaneous analysis of nortriterpenoids and lignans in Schisandra samples. The method consists of three individual mass spectrometric experiments, including the full scan MS, MS/MS experiment and in-source collision induced dissociation (CID) MS/MS, which enable the identification of diagnostic fragmentation pathways of nortriterpenoids and lignans. As such, a total of 6 nortriterpenoids and 10 lignans were unequivocally identified, and one nortriterpenoid and 20 lignans were tentatively identified from different Schisandra samples within 12.5min. In addition, 6 nortriterpenoids and 10 lignans were quantified in 48 samples of S. chinensis and S. sphenanthera using an extract ion chromatogram (XIC) of the full scan MS experiment. Dataset obtained from UPLC–MS was processed with principal component analysis (PCA) and orthogonal partial least squared discriminant analysis (OPLS-DA) to compare the difference between the two Schisandra species.

Abstract:Two distinctive rearranged 19-nor-7,8-seco-labdane diterpenoids (1 and 2) with a novel tetracyclo[5.2.1.02,5.04,10]decane skeleton, a derivative of the open tetrahydrofuran ring (7), three dimeric compounds (8−10), and four revised homologs (3−6) were obtained from Chinese liverwort Pallavicinia ambigua. Their structures were identified via combined analysis of their spectroscopic data, single-crystal X-ray diffraction patterns, and ECD calculations. The light-driven conversion of compound 5 to compounds 1−4 demonstrated that photochemically induced postmodification involved in biosynthesis is an important way to diversify natural structures. A preliminary cytotoxicity assay revealed that compound 5 showed significant inhibition in the human prostate cancer (PC-3) cell line via an apoptotic pathway.

Abstract:Two new triterpenoids, schisphendilactone A and B ( 1 and 2 ), together with three known triterpenoids, were isolated from the stems of Schisandra sphenanthera . Their structures were elucidated by spectroscopic methods, and the absolute configuration of 1 was determined by single-crystal X-ray diffraction. Compound 2 showed moderate inhibitory activity against SW480 cancer cell line, and compound 5 exhibited promising anti-HIV-1 activity with EC 50 value of 0.52 μg ml −1 and therapeutic index value of 117.12.

Abstract:Introduction - Dibenzocyclooctadiene lignans are bioactive constituents in Schisandra chinensis (Turcz.) Baill. They are often present as isomers and this makes structural discrimination difficult.Objective - To develop a rapid approach towards the characterisation and identification of isomeric dibenzocyclooctadiene lignans from S. chinensis using HPLC-PAD-ESI-MSn.Methodology - The fragmentation behaviours of seven dibenzocyclooctadiene lignans from S. chinensis were studied using ion trap mass spectrometry with positive electrospray ionisation and loop injection. The fragmentation patterns of angeloylgomisin H, tigloylgomisin H, benzoylgomisin H and gomisin D were supported by high-resolution mass spectrometric analysis of some characteristic ions using a time-of-flight mass spectrometer. Combined with high-performance liquid chromatography and photodiode array detection, the established approach to the structural identification of dibenzocyclooctadiene lignans by ion trap mass spectrometry was applied to the analysis of extracts of S. chinensis.Results - According to the HPLC retention behaviour, the diagnostic UV spectra and the molecular structural information provided by multistage mass spectrometry spectra and the literature, a total of 25 dibenzocyclooctadiene lignans, including seven groups of lignan isomers, were identified or tentatively characterised in S. chinensis rapidly. One of these was reported as a constituent of S. chinensis for the first time.Conclusion - This study has shown that HPLC-PAD-ESI-MSn may be used as an effective and rapid method for the characterisation and identification of isomeric dibenzocyclooctadiene lignans from S. chinensis. Abstract:An unusual pyridine-containing sesterterpenoid, leucosceptrodine (1), and five new nor-leucosceptrane sesterterpenoids including bisnor- (C23, 2), tetranor- (C21, 3) and pentanor- (C20, 4−6) skeletons, were isolated from the leaves of Tibetan Leucosceptrum canum. Their structures including the absolute configurations were determined by extensive spectroscopic analyses and quantum chemical calculations. Single crystal of one epimer (5) was crystallized from a pair of inseparable epimers, and its structure including absolute configuration was determined by X-ray crystallographic analysis. Immunosuppressive activity of compounds 1−4 with different potencies through inhibiting the secretion of cytokines TNF-α and IL-6 in LPS-induced macrophages RAW264.7 was observed.

The postulated biogenetic sequences of sixteen highly oxygenated nortriterpenoids and bisnortriterpenoids with new skeletons from three Schisandra species are discussed and have been compared from a chemotaxonomic standpoint.

Abstract:LC-MS has been a widely used analytical technique for identification of natural compounds. However, sophisticated and laborious data analysis is required to identify chemical components, especially new compounds, from a large LC-MS dataset. The aim of this study is to develop an integrated data-mining strategy that combines molecular networking (MN), in-house polygonal mass defect filtering (MDF), and diagnostic fragment ion filtering (DFIF) to identify phytochemicals in Stephania tetrandra based on LC-MS data. S. tetrandra samples were prepared by matrix solid-phase dispersion extraction methods and then raw MS spectra were acquired using LC-QTOF-MS/MS. MN and in-house polygonal MDF classified the compounds roughly. Modified DFIF were then used in succession to place each spectrum into a specific class. Finally, the exact structures were deduced by fragmentation pathways and related botanical biogenesis, with the help of the narrowed classification from MN and MDF. The total workflow was a combination of data filtering and identification methods for rapid characterization of known compounds (dereplication) and discovery of new compounds. Consequently, 144 compounds were identified or tentatively identified in the aerial parts and roots of S. tetrandra, including 11 potentially new compounds and 63 compounds first identified in this species. Among 144 compounds, 61 were from the aerial parts exclusively, 8 were from the roots exclusively, and 75 were found in both parts. Furthermore, two new biflavonoids were isolated with the guide of LC-MS analysis and structurally elucidated by spectroscopic methods. In conclusion, the proposed data-mining strategy based on LC-MS can be used to profile chemical constituents with high efficiency and guide the isolation of new compounds from medicinal plants. The comparison of the components of the aerial parts and roots of S. tetrandra would be helpful for the rational utilization of the medicinal plant.

Abstract:From the whole plant of Senecio tsoongianus, two pairs of epimers, tsoongianolides A (1) and B (2), and tsoongianolides C (3) and D (4), and two new sesquiterpene lactones, tsoongianolides E (5) and F (6), all of which possess the eremophilane skeleton, were isolated. Two known eremophilane dimers (7) and (8) were also obtained from this species. Their absolute structures were elucidated by 1D- and 2D-NMR techniques and by X-ray diffraction studies, along with chemical evidence.
