Abstract:A new class of highly oxygenated Schisandra nortriterpenoids, lancolides A-D(1-4), from Schisandra lancifolia, represents the first example of natural products that possess a tricyclo[6.3.0.0(2.11)]undecane-bridged system. Their structures were elucidated by NMR spectra, X-ray diffraction, and quantum chemical calculations. Lancolides A (1) and D (4) had specific antiplatelet aggregation induced by platelet-activating factor (PAF).

Abstract:Three unique nortriterpenoids, schilancitrilactones A-C (1-3), were isolated from the stems of Schisandra lancifolia . Compound 1 possesses a 5/5/7/5/5/5-fused hexacyclic ring system with a C(29) backbone, while 2 and 3 feature a C(27) skeleton with a 5/7/5/5/5-fused pentacyclic ring system. Their absolute stereochemistries were established by CD and single-crystal X-ray diffraction experiments. Compound 3 showed anti-HIV-1 activity with an EC(50) value of 27.54 μg/mL, and 1 exhibited antifeedant activity at 15.73 μg/cm(2).

Abstract:Ligustrazine and ferulic acid, which are useful compounds of Chinese traditional medicine, were used as leading compounds. Six ligustrazine-aromatic acid derivatives were designed and synthesized based on the twin drug principle. Their structures were characterized by IR, H-1 NMR, C-13 NMR and mass spectra. In the in vivo experiment, some compounds have significant inhibitory effect on adenosine diphosphate (ADP) induced platelet aggregation. Among them, the inhibitory effect of compound 1a was 5.7 times than ozagrel.

Abstract:Five new dibenzocyclooctadiene lignans, schisphenlignans A-E (1-5), together with eight known ones, were isolated from the stems of Schisandra sphenanthera. The structures of 1-5 were elucidated based on the analysis of their NMR, MS and circular dichroism (CD) spectra. Some isolates were tested for their acute activities on insulin sensitivity in 3T3-L1 differentiated adipocytes, but none of them showed significant bioactivity with 10 mu M administration of the tested compounds.

Abstract:Twelve new nortriterpenoids, namely wuweizidilactones J-P (1-7), schindilactones I-K (8-10), preschisanartanin N (11), and schisdilactone J (12), as well as 23 known analogues, were isolated from the leaves and stems of Schisandra chinensis. The absolute configurations of 1 and 11 were established by calculated electronic circular dichroism (ECD) spectra and that of 7 was confirmed by single-crystal X-ray diffraction. The absolute configurations of 2-6, 8, and 9 were determined by an empirical comparison of their experimental ECD spectra with that of 1. It is the first time that the ECD spectra of 18(13 -> 14)-abeoschiartane-, 18-norschiartane-, and schisanartane-type nortriterpenoids possessing an alpha,beta-unsaturated-gamma-lactone moiety are presented, this method providing an alternative means of obtaining absolute configurational assignments for these types of nortriterpenoids. All isolates were evaluated for their anti-acetylcholinesterase and anti-butyrylcholinesterase activities, and compounds 8, 13, 23, and 31 show anti-acetylcholinesterase activity at concentrations of 50 mu M, with 12.7, 10.7, 16.6, and 32.1% inhibition, respectively. Abstract:Schicagenins A-C (1-3), three unprecedented nortriterpenoids characterized with a tetracyclic oxa-cage motif and C(9) side chain, were discovered from the leaves and stems of Schisandra chinensis. Their structures were determined on the basis of extensive spectroscopic analysis, and the absolute stereochemistries were established by single-crystal X-ray diffraction and CD experiments. A plausible biosynthetic pathway of 1-3 was also discussed.

Compound 1 was the first example of naturally occurring N-containing lignans featuring a nicotinoyl group. All the new compounds were evaluated for their anti-HIV-1 activities and showed EC50 values in the range 17.89-138.23 mu g/mL.

Abstract:Four new highly oxygenated nortriterpenoids, lancifodilactones O-R (1-4), together with six known ones, i.e., 5-10, were isolated from the leaves and stems of Schisandra lancifolia. Their structures were elucidated by spectroscopic analyses, including 1D- and 2D-NMR experiments and mass spectrometry. Compounds 1-3 were evaluated for their cytotoxicity against NB4, A549, SHSY5Y, PC-3, and MCF-7 cell lines. No compounds exhibited significant cytotoxicity, the I(C5)0 values being above 50 mu M.

Abstract:Purpose To investigate the effects of three lignans:heteroclitin D (HD),schisanhenol (SAL) and (+)-anwulignan(AN)with different skeletons from Schisandraceae medicinal plants on platelet aggregation. Methods At concentrations 0.05～25 mg/L of the three lignans, rabbit platelet aggregation induced by adenosine diphosphate (ADP) and platelet activating factor(PAF) were studied by Born's turbidimetry,the maximum time of platelet aggregation and platelet aggregation at 1,3,5 min were also observed. Results ① HD and SAL inhibited the ADP- and PAF-induced platelet aggregation in a concentration-dependant manner.The inhibitory effect of AN was weak.② ADP-induced platelet aggregation at 1,3,5 min were inhibited by HD and SAL in a similar manner.At 5 mg/L,AN inhibited platelet aggregation at 3 and 5 min significantly;The PAF-induced platelet aggregation at 1,3,5 min,the inhibition of HD at 1,3 min and of SAL at 3,5 min were more potent.③ HD,SAL and AN had no effects on maximum time of ADP- and PAF-induced platelet aggregation. Conclusions HD,SAL and AN have inhibitory effects on platelets aggregation induced by ADP and PAF in vitro.The inhibitory effect of HD is the most potent and could be one of the important active components from Kadsura medicinal plants.

Abstract:A previously undescribed sesquiterpene lactone, named eupalinolide N (1), was isolated from traditional Chinese medicine "Ye-Ma-Zhui" (Eupatorium lindleyanum) by ethanol reflux extraction, reduced pressure concentration, macroporous resin column chromatography, and C-18 reversed-phase chromatography. Its structure was elucidated by a comprehensive interpretation of spectroscopy evidence as well as ECD calculations. 1 showed anti-inflammatory activity by inhibiting the gene expressions of pro-inflammatory factors including IL-1 beta, TNFa COX-2, and iNOS at the concentration of 7.5 mu M, as well as attenuating the excretion of NO, IL-6, and TNF-alpha in Raw 264.7 macrophages at the concentration of 15 mu M.

Abstract:Linderaggrenolides A–N (1–14), 14 new lindenane sesquiterpenoid dimers with oxygen bridges were isolated from the roots of Lindera aggregata. Their structures were elucidated on the basis of comprehensive spectroscopic data analysis, with the absolute configurations established by empirical approaches, electronic circular dichroism calculations, and X-ray crystallography. Abstract:Four undescribed ginkgolides, including two rare sesquiterpene ginkgolides (compounds 1 and 2 ) and two diterpenoid ginkgolides (compounds 3 and 4) , were isolated from Ginkgo biloba L. The structures of these four ginkgolides were identified based on extensive spectroscopic analysis, DP4+ probability analysis and X-ray diffraction. Compounds 1 and 2 exhibited excellent antiplatelet aggregation activities with IC 50 values of 1.20 ± 0.25 and 4.11 ± 0.34 μM, respectively.

Abstract:Twenty new polyoxygenated labdane diterpenoids (1-20) were isolated from the aerial parts of Leonurus macranthus. Their structures were elucidated on the basis of spectroscopic and spectrometric data (1D and 2D NMR, IR, and HRESIMS). The absolute configurations of macranthin A (1) and 6-O-deacetylmacranthin A (2) were determined by single-crystal X-ray crystallographic analysis and a modified Mosher's method, respectively. Compounds 1-9, 12, 14, and 19 showed inhibition of nitric oxide production in lipopolysaccharide-activated BV-2 microglial cells with IC50 values of 10.0-63.7 μM.

Abstract:Phytochemical investigation of the root tubers of Lindera aggregata resulted in the isolation of five new sesquiterpene lactones, linderagalactones A-E (1-5), along with eight known sesquiterpenoids, 3-eudesmene-1beta,11-diol, hydroxylindestenolide, strychnistenolide, hydroxyisogermafurenolide, atractylenolide III, linderane, neolinderalactone, and linderalactone. The structures and relative configurations of 1-5 were determined by spectroscopic methods, especially HRESIMS and 2D NMR techniques. The absolute configurations of 1-4 were defined by comparison of quantum chemical TDDFT calculated and experimental ECD spectra. Linderagalactone A (1) is a halogenated sesquiterpene lactone possessing a unique rearranged carbon skeleton. Linderagalactone E (5), linderane, hydroxylindestenolide, and linderalactone showed hepatoprotective activity against H2O2-induced oxidative damages on HepG2 cells with EC(50) values of 67.5, 167.0, 42.4, and 98.0 microM, respectively.
