Abstract:Two previously uncharacterized compounds, an aconitine‐type C19‐diterpenoid alkaloid (1) and a napelline‐type diterpenoid alkaloid C20‐diterpenoid alkaloid (2), as well as ten known compounds (3–12), were isolated from Aconitum pendulum. Their structures were elucidated based on spectroscopic data, including 1D and 2D NMR, IR, HR‐ESI‐MS, and single‐crystal X‐ray diffraction analysis. The anti‐insecticidal activities of these compounds were evaluated by contact toxicity tests against two‐spotted spider mites, and compounds 1, 2, and 9 showed moderate contact toxicity, with LC50 values of 0.86±0.09, 0.95±0.23, and 0.89±0.19 mg/mL, respectively. This study highlights the potential use of diterpenoid alkaloids as natural plant‐derived pesticides for the management of plant pests.

Abstract:From the roots of Aconitum toxicum, acotoxicine (1), a new bisnorditerpene alkaloid, was isolated together with the structurally related delavaconitine, delavaconine, dolaconine, aconosine, and the norditerpene alkaloid neolinine. The structure of compound 1 was elucidated by spectroscopic data interpretation. Further, eight lipo-alkaloids (2-9) were detected in the plant material by LC-APCI-MSn analysis. This is the first report of a C18 bisnorditerpene alkaloid and lipo-alkaloids from A. toxicum.

Abstract:Two new lappaconitine‐type C18‐diterpenoid alkaloids, named as leucostosines C (1) and D (2), together with six known compounds (3‐8), were isolated from the roots of Aconitum leucostomum Worosch. Their structures were elucidated by various spectroscopic analyses, including IR, HR‐ESI‐MS, NMR spectra and X‐ray experiments. Leucostosine C is the first diterpenoid alkaloid bearing the 7‐amino group. The isolated compounds were tested for the acetylcholinesterase (AChE) inhibitory effect and neuroprotective activity, none of them showed significant activities.

Abstract:Three new C19-norditerpenoid alkaloids (1⁻3), along with two known C19-norditerpenoid alkaloids (4,5), have been isolated from Aconitum szechenyianum. Based on extensive spectroscopic techniques (1D, 2D-NMR, IR, and MS) and chemical methods, their structures were established as szechenyianine D (1), szechenyianine E (2), szechenyianine F (3), 8-O-methyl-14-benzoylaconine (4), and spicatine A (5). The immunosuppressive effects of compounds 1⁻5 were studied using a ConA-induced or LPS-induced splenocyte proliferation model. In vitro tests showed that Compounds 2, 4, and 5 suppressed ConA-induced or LPS-induced splenocyte proliferation in a concentration-dependent manner. The CC50/IC50 values of 2, 4, and 5 suggested that these compounds were potential immunosuppressive agents for the treatment of autoimmune diseases characterized by arthritis, such as rheumatoid arthritis.

Abstract:A phytochemical investigation on the roots of Aconitum austroyunnanense led to the isolation of two previously undescribed diterpenoid alkaloids, austroyunnanines D and E ( 1 and 2 ). Their structures were determined by spectral methods such as 1D and 2D ( 1 H– 1 H COSY, HMQC, NOESY, and HMBC) NMR spectroscopy, in addition to high-resolution mass spectrometry. The isolated compounds were tested in vitro for cytotoxic activity against three human gastric carcinoma cell lines. As a result, compound 1 exhibited some cytotoxicities against all the tested tumor cell lines with IC 50 values less than 20.0 μM.

Abstract:Seven previously undescribed diterpenoid alkaloids, including five C20-diterpenoid alkaloids, barpuberudine, barpubesines A-D, and two C18-diterpenoid alkaloids, barpubenines A-B, along with 11 known diterpenoid alkaloids were isolated from the whole plant of Aconitum barbatum var. puberulum Ledeb. (Ranunculaceae). Barpuberudine is an unprecedented carbon skeleton of C20-diterpenoid alkaloid, while barpubenines A-B are the first example of rearranged types in C18-diterpenoid alkaloids. Their structures were elucidated based on a comprehensive spectroscopic data analysis. The probable pathway of biogenesis of barpuberudine and barpubenines A-B were discussed. Additionally, the antiarrhythmic, cytotoxic and antimicrobial activities of isolates were also evaluated.

Their structures were established on the basis of extensive spectroscopic analyses (HRESIMS, 1D and 2D NMR). Among them, compounds 1 and 2 possess a unique 2‐(E)‐(2‐methylbut‐2‐enamido)benzoate moiety at the C‐18 position. Furthermore, cytotoxic activities of these diterpenoid alkaloids were also evaluated.

Abstract:A phytochemical investigation on the roots of Aconitum austroyunnanense afforded three undescribed aconitine–type C 19 –diterpenoid alkaloids, austroyunnanines A–C (1–3). Structural elucidation of all the compounds were performed by spectral methods such as 1 D and 2 D ( 1 H– 1 H COSY, HMQC, and HMBC) NMR spectroscopy. The isolated alkaloids were tested in vivo for their antinociceptive properties. Their structures were determined by spectral methods such as 1D and 2D ( 1 H– 1 H COSY, HMQC, NOESY, and HMBC) NMR spectroscopy, in addition to high-resolution mass spectrometry. The isolated alkaloids were tested in vivo for antinociceptive potential. The results show that pseudostapines A and B possess more potent antinociceptive effect than the positive control drugs aspirin and acetaminophen.

Abstract:Aconicumines A–D, an advanced class of norditerpenoid alkaloids, and seven known alkaloids, were isolated from Aconitum taipaicum Hand.-Mazz. (Ranunculaceae). The structures of the previously undescribed compounds, including their absolute configurations, were fully elucidated based on spectroscopic and single-crystal X-ray diffraction data analysis. Aconicumines A–D exhibit interesting cage-like structure, characterised by an unprecedented N,O- diacetal moiety (C6–O–C19–N–C17–O–C7) that has not been previously observed in diterpenoid alkaloids. Possible biosynthetic pathways for aconicumines A–D were proposed. Aconitine, hypaconitine, and aconicumine A showed significant inhibition of nitric oxide production in RAW 264.7 macrophages induced by lipopolysaccharide with IC 50 values ranging from 4.1 to 19.7 μM compared to positive control (dexamethasone, IC 50 = 12.5 μM). Furthermore, the primary structure–activity relationships for aconicumines A–D were also represented.

Abstract:Seven new diterpenoid alkaloids, acotarines A-G (1–7), along with four known diterpenoid alkaloids, were isolated from Aconitum taronense. Their structures were established by 1D, 2D NMR spectra and HRESIMS data. Acotarine A (1) represents a franchetine-type alkaloid characterized by a N-butanone moiety. All the compounds were tested for their activities to induce lysosomal biogenesis through LysoTracker Red staining, compounds acotarines B (2), E (5), F (6), indaconitine (8), 7,8-epoxy-franchetine (9), and 6-epiforesticine (10) showed inducing lysosomal biogenesis activity.

Abstract:Through phytochemical research, the composition of diterpenoid alkaloids in Aconitum sessiliflorum was analyzed, and sixteen compounds were identified. Among them, there are seven napelline type C 20 -diterpenoid alkaloids ( 1 – 4 , 7 – 8 , 15 ), three denudatine type C 20 -diterpenoid alkaloids ( 5 – 6 , 16 ), and six aconitine type C 19 -diterpenoid alkaloids ( 9 – 14 ). On the basis of NMR spectroscopy, the structural analysis of the separated compounds was achieved by comparing with previous data. All sixteen compounds were isolated for the first time from the Aconitum sessiliflorum , of which five compounds ( 1 , 6 , 9 , 13 , 15 ) were isolated for the first time from series Brachypoda, where Aconitum sessiliflorum is classified. These diterpenoid alkaloids not only enrich the chemical diversity of Aconitum sessiliflorum , but also facilitate the analysis of their chemotaxonomic implications. Importantly, the diverse types and structural features of these diterpenoid alkaloids provide new insights into both the potential phylogenetic affiliations within the series Brachypoda and the varying degrees of evolutionary divergence among these related taxa.

Abstract:Phytochemical investigation on the roots of Aconitum polycarpum Chang ex W. T. Wang (Ranunculaceae) led to the isolation of twenty-six compounds for the first time. Their structures were determined by NMR analysis and compared with reported data. The chemotaxonomic value of these compounds was further discussed by network analysis. The structural features of the isolated diterpenoid alkaloids indicate that the relatively advanced evolutionary status of A . polycarpum within subgenus Aconitum . Some compounds, such as 12 , 18 , 20 , 21 , and 22 with a limited distribution in the genus Aconitum exhibited certain potential chemotaxonomically values.

Abstract:A new C20‐denudatine‐type diterpenoid alkaloid (DA) 11S‐aconicarnine D (1) and fifteen known DAs were isolated from the lateral roots of Aconitum brachypodum Diels. Their structures were identified on the basis of extensive spectroscopic analyses, NMR calculations and DP4+ analysis. The antimicrobial activities of compound 1 was evaluated, compound 1 exhibited potent antimicrobial activity against Alternaria panax and Fusarium solani with MICs of 2.00 and 16.00 μg/mL (Nystatin, 1.00 μg/mL), respectively.
