Abstract:Tanaconitine (1), a novel Aconitum alkaloid with a C(29) skeleton, together with six known diterpenoid alkaloids, was isolated from the roots of Aconitum tanguticum. Abstract:Through phytochemical research, the composition of diterpenoid alkaloids in Aconitum sessiliflorum was analyzed, and sixteen compounds were identified. Among them, there are seven napelline type C 20 -diterpenoid alkaloids ( 1 – 4 , 7 – 8 , 15 ), three denudatine type C 20 -diterpenoid alkaloids ( 5 – 6 , 16 ), and six aconitine type C 19 -diterpenoid alkaloids ( 9 – 14 ). On the basis of NMR spectroscopy, the structural analysis of the separated compounds was achieved by comparing with previous data. All sixteen compounds were isolated for the first time from the Aconitum sessiliflorum , of which five compounds ( 1 , 6 , 9 , 13 , 15 ) were isolated for the first time from series Brachypoda, where Aconitum sessiliflorum is classified. These diterpenoid alkaloids not only enrich the chemical diversity of Aconitum sessiliflorum , but also facilitate the analysis of their chemotaxonomic implications. Importantly, the diverse types and structural features of these diterpenoid alkaloids provide new insights into both the potential phylogenetic affiliations within the series Brachypoda and the varying degrees of evolutionary divergence among these related taxa.

Abstract:Two new lappaconitine‐type C18‐diterpenoid alkaloids, named as leucostosines C (1) and D (2), together with six known compounds (3‐8), were isolated from the roots of Aconitum leucostomum Worosch. Their structures were elucidated by various spectroscopic analyses, including IR, HR‐ESI‐MS, NMR spectra and X‐ray experiments. Leucostosine C is the first diterpenoid alkaloid bearing the 7‐amino group. The isolated compounds were tested for the acetylcholinesterase (AChE) inhibitory effect and neuroprotective activity, none of them showed significant activities.

Abstract:Three new C19-norditerpenoid alkaloids (1⁻3), along with two known C19-norditerpenoid alkaloids (4,5), have been isolated from Aconitum szechenyianum. Based on extensive spectroscopic techniques (1D, 2D-NMR, IR, and MS) and chemical methods, their structures were established as szechenyianine D (1), szechenyianine E (2), szechenyianine F (3), 8-O-methyl-14-benzoylaconine (4), and spicatine A (5). The immunosuppressive effects of compounds 1⁻5 were studied using a ConA-induced or LPS-induced splenocyte proliferation model. In vitro tests showed that Compounds 2, 4, and 5 suppressed ConA-induced or LPS-induced splenocyte proliferation in a concentration-dependent manner. The CC50/IC50 values of 2, 4, and 5 suggested that these compounds were potential immunosuppressive agents for the treatment of autoimmune diseases characterized by arthritis, such as rheumatoid arthritis.

Abstract:Seven new aconitine‐type C19‐diterpenoid alkaloids, apetaldines K‐Q (1‐7), were isolated from the roots of Aconitum apetalum (Huth) B. Fedtsch. Their structures were established on the basis of extensive spectroscopic analyses (HRESIMS, 1D and 2D NMR). Among them, compounds 1 and 2 possess a unique 2‐(E)‐(2‐methylbut‐2‐enamido)benzoate moiety at the C‐18 position. Furthermore, cytotoxic activities of these diterpenoid alkaloids were also evaluated.

Abstract:Phytochemical investigation on the roots of Aconitum polycarpum Chang ex W. T. Wang (Ranunculaceae) led to the isolation of twenty-six compounds for the first time. Their structures were determined by NMR analysis and compared with reported data. The chemotaxonomic value of these compounds was further discussed by network analysis. The structural features of the isolated diterpenoid alkaloids indicate that the relatively advanced evolutionary status of A . polycarpum within subgenus Aconitum . Some compounds, such as 12 , 18 , 20 , 21 , and 22 with a limited distribution in the genus Aconitum exhibited certain potential chemotaxonomically values.

Abstract:A new C20‐denudatine‐type diterpenoid alkaloid (DA) 11S‐aconicarnine D (1) and fifteen known DAs were isolated from the lateral roots of Aconitum brachypodum Diels. Their structures were identified on the basis of extensive spectroscopic analyses, NMR calculations and DP4+ analysis. The antimicrobial activities of compound 1 was evaluated, compound 1 exhibited potent antimicrobial activity against Alternaria panax and Fusarium solani with MICs of 2.00 and 16.00 μg/mL (Nystatin, 1.00 μg/mL), respectively.

Abstract:A phytochemical investigation on the roots of Aconitum austroyunnanense afforded three undescribed aconitine–type C 19 –diterpenoid alkaloids, austroyunnanines A–C (1–3). Structural elucidation of all the compounds were performed by spectral methods such as 1 D and 2 D ( 1 H– 1 H COSY, HMQC, and HMBC) NMR spectroscopy. The isolated alkaloids were tested in vivo for their antinociceptive properties. Consequently, austroyunnanine B (2) exhibited significant antinociceptive effect and its ID 50 value (48.0 μmol/kg) was 2-fold less than those of the positive control drugs aspirin and acetaminophen. Abstract:A phytochemical investigation on the roots of Aconitum austroyunnanense led to the isolation of two previously undescribed diterpenoid alkaloids, austroyunnanines D and E ( 1 and 2 ). Their structures were determined by spectral methods such as 1D and 2D ( 1 H– 1 H COSY, HMQC, NOESY, and HMBC) NMR spectroscopy, in addition to high-resolution mass spectrometry. The isolated compounds were tested in vitro for cytotoxic activity against three human gastric carcinoma cell lines. As a result, compound 1 exhibited some cytotoxicities against all the tested tumor cell lines with IC 50 values less than 20.0 μM.

Abstract:Six new C19-aconitine-type diterpenoid alkaloids, pendulumines A-F (1-6), together with six known ones (7-12), were isolated from the rhizomes of Aconitum pendulum. Their structures were elucidated using extensive spectroscopic data analysis, including 1D and 2D NMR, MS, and single-crystal X-ray diffraction analysis. The isolates were also tested for their analgesic activity based on the thermal avoidance response of the roundworm Pirstionchus pacificus, and 9 showed significant biological activity with an EC50 value of 0.08 ± 0.02 mg.mL-1.

Abstract:Two previously uncharacterized compounds, an aconitine‐type C19‐diterpenoid alkaloid (1) and a napelline‐type diterpenoid alkaloid C20‐diterpenoid alkaloid (2), as well as ten known compounds (3–12), were isolated from Aconitum pendulum. Their structures were elucidated based on spectroscopic data, including 1D and 2D NMR, IR, HR‐ESI‐MS, and single‐crystal X‐ray diffraction analysis. The anti‐insecticidal activities of these compounds were evaluated by contact toxicity tests against two‐spotted spider mites, and compounds 1, 2, and 9 showed moderate contact toxicity, with LC50 values of 0.86±0.09, 0.95±0.23, and 0.89±0.19 mg/mL, respectively. This study highlights the potential use of diterpenoid alkaloids as natural plant‐derived pesticides for the management of plant pests.

Abstract:Twenty-five diterpenoid alkaloids were isolated from the roots of two Aconitum species. The structures of seven new C19-diterpenoid alkaloids, apetaldines A-G (1-7), and 10 known alkaloids (8-17) from Aconitum apetalum and eight known alkaloids (18-25) from Aconitum franchetii var. villosulum were elucidated via HRESIMS, IR, and NMR data. Alkaloids 1-10, 15, 16, and 18-25 were screened for their antifeedant activity. Among the compounds tested, chasmanthinine (19) showed highly potent antifeedant activity with an effective concentration for 50% feeding reduction (EC50) at 0.07 mg/cm2. The antifeedant structure-activity relationship of the diterpenoid alkaloids is also discussed.

Abstract:Five new aconitine-type C19-diterpenoid alkaloids, apetalrines A–E (1–5), were isolated from Aconitum apetalum. Their structures were determined by analysis of 1D and 2D NMR, IR, and HRESIMS data. Semisynthesis of apetalrine B (2) from its parent compound aconorine was achieved to confirm the structure proposed. Twenty derivatives of 2 (11a–11l, 12a, 12b, 12d, 12e, 12j, 12k, 12m, 12n) were synthesized via a unified approach relying on simple coupling reactions. The evaluation of neuroprotective effects of compounds (1–5, 11b, 11c, 11f–11i, 12a, 12b, 12d, 12e, 12k, 12m, 12n) with low cytotoxicity revealed compound 2 to exhibit good neuroprotective effects in H2O2-treated SH-SY5Y cells at a concentration of 50 μM. Abstract:Former study suggests alkaloids from herbs of Aconitum genus plants possess excellent bioactivities, which exert great value for related deeper chemical constituent investigation. Herein, chemical isolation was performed and four alkaloids were isolated from Fuzi, of which two were new ones, and the other two were reported NMR data for the first time. Their chemical structures were identified by NMR data, high resolution MS, UV and IR analysis. Additionally, the MS fragmentation patterns were explored, formerly, that of hetisane alkaloid was rarely reported, and fragmentation mechanism of the diagnostic ion was proposed. Based on these fragment pathway, metabolites and metabolic pathways of four compounds were investigated in rat liver microsomes using UPLC-Q/TOF-MS, and dehydrogenation product was firstly found from metabolites of hetisane alkaloid.
