Abstract:Asprellols A-C (1-3, resp.), three new 24-nortriterpenoids, were isolated from the CHCl3-soluble fraction of 95% EtOH extract of the roots of Ilex asprella, together with a known nortriterpenoid. The structures of the new compounds were elucidated as 2,6,20-trihydroxy-3-oxo-11,12-epoxy-24-norursa-1,4-dien-28,13-olide (1), 2,6-dihydroxy-3-oxo-11,12-epoxy-24-norursa-1,4,20(30)-trien-28,13-olide (2), and 2,6-dihydroxy-3-oxo-11,12-epoxy-24-noroleana-1,4-dien-28,13-olide (3) on the basis of spectroscopic analyses.

Abstract:Two novel 18,19-seco-ursane triterpenoid saponins, ilexasprellanosides J-K (1-2, resp.), 3-O-α-l-Rhamnopyranosyl-(1 → 2)-β-d-xylopyrannosyl-19-O-β-d-glucopyranosyl-16-β-hydroxyl-18,19-seco-13(18)-urs-ene-21, 28-lactone (1), 3-O-β-d-Xylopyrannosyl-19-O-α-l-rhamnopyran osyl-(1 → 2)-α-l-arabinopyranoside-16, 21-epoxy-18, 19-seco-13(18)-urs-ene-28-oic acid (2), five known compounds (3-7) were isolated from the leaves of Ilex asprella (Hook. et Arn.) Champ. ex Benth. (Gangmeiye). The chemical structures of these compounds were elucidated through UV, IR, ESI-MS, 1H NMR and 13C NMR analyses. In MTT and SRB assays, compounds 1-4 presented cytotoxic activities against several human cancer cell lines, namely, the HL-60 human acute promyelocytic leukaemia, Bel 7402 liver cancer, BGC-823 gastric cancer and KB human nasopharyngeal carcinoma cell lines. Compound 1 exhibited weak cytotoxic activities against the human tumour cell lines HL-60, Bel 7402 and KB with inhibition rates of 27.97%, 21.00% and 25.60%, respectively. Compound 2 exhibited weak cytotoxic activities against the human tumour cell lines HL-60, Bel 7402 and BGC-823 with inhibition rates of 19.34%, 7.50% and 4.26%. Respectively, the compounds exerted no statistically different effects on mast cell degranulation in rats. This result indicates that the compounds do not affect mast cell degranulation.

Abstract:Six new triterpene saponins, ilexasprellanosides A-F (1-6, resp.), together with eleven known compounds were isolated from the roots of Ilex asprella. The new saponins were characterized as ursa-12,18-dien-28-oic acid 3-O-β-D-xylopyranoside (1), 19α-hydroxyursolic acid 3-O-β-D-(2'-O-acetylxylopyranoside) (2), 19α-hydroxyursolic acid 3-O-β-D-glucuronopyranoside (3), 3β,19α-dihydroxyolean-12-en-23,28-dioic acid 28-O-β-D-glucopyranoside (4), 19α-hydroxyoleanolic acid 3-O-β-D-(2'-O-acetylxylopyranoside) (5), 19α-hydroxyoleanolic acid 3-O-β-D-glucuronopyranoside (6). The structures of the new compounds were elucidated by analysis of their spectroscopic data and chemical degradation. Compounds 2, 4, oleanolic acid 3-O-β-D-glucuronopyranoside, 3-β-acetoxy-28-hydroxyurs-12-ene, and pomolic acid showed significant cytotoxic activities against human tumor cell line A549 (IC50 values of 1.87, 2.51, 1.41, 3.24, and 5.63 μM, resp.).

Abstract:Objective To study the chemical constituents in the leaves of Ilex cornuta.Methods The compounds were isolated and purified by silica gel,Sephadex LH-20 column chromatography,and semi-preparative HPLC,and their structures were identified by spectroscopic data and physicochemical properties.Results Twelve compounds were obtained from the leaves of I.cornuta and identified as(2R,3S,4S)-4-(4-hydroxy-3-methoxybenzyl)-2-(5-hydroxy-3-methoxyphenyl)-3-hydroxymethyl-tetrahydrofuran-3-ol(1),physcion(2),11-keto-α-amyrin palmitate(3),lupole(4),30-oxolupeol(5),uvaol(6),3β-hydroxy-urs-11-en-13β(28)-olide(7),ursolic acid(8),β-sitosterol(9),daucosterol(10),pomolic acid-28-O-β-D-glucopyranoside(11),and pedunculoside(12).Conclusion Compounds 1 and 2 are obtained from the plants in Ilex L.for the first time,and compounds 11 and 12 are obtained from this plant for the first time.

Abstract:Encouraged by the in vivo anti-inflammatory effect of aqueous extract of Ilex asprella stems, a further phyto-chemical investigation on I. asprella stems oriented by the in vitro NO production inhibition in RAW264.7 cells was conducted, which led to the isolation of eight new phenolic constituents, namely asprenols A-H (1-8), together with 12 known ones (9-20). The structures of the new compounds were established by extensive spectroscopic data analyses of HR-ESI-MS, IR, UV, and 1D and 2D NMR, and the absolute configurations were determined by comparison of experimental and calculated ECD analyses. All isolated were evaluated for the inhibition against NO production in RAW 264.7 cells, and several compounds showed moderate inhibitory effect.

Abstract:Five new triterpene saponins, Ilexpublesnins N-R (1-5), along with seven known analogs were isolated from the root of Ilex pubescens. Their structures were elucidated on the basis of extensive spectroscopic analysis, including 1D and 2D NMR experiments. Ilexpublesnin N (1) possessed a rare 20-hydroxyursolic acid scaffold from natural resource. These compounds were evaluated in vitro for their cytotoxic effects on human cancer cell lines HepG2, HLE, BEL7402, BEL7403, BEL7405, MCF-7, HeLa. Among them, only compounds 5 and 10 showed cytotoxic potentiality against BEl-7403 and HEL cell lines [inhibition (%): 35.38 and 45.12, respectively].

Abstract:Eleven new triterpene saponins, ilexpublesnins C-M (1-11), along with ten known analogues were isolated from the roots of Ilex pubescens. Their structures were elucidated on the basis of extensive spectroscopic analysis, including 1D and 2D NMR experiments. Compounds 1, 2, 10, and 11 contain a 24-aldehyde, which is rare for triterpene saponins from Ilex. These compounds were evaluated in vitro for their cytotoxic effects on human cancer cell lines HepG2, HLE, BEL7402, BEL7403, BEL7405, MCF-7, and HeLa. Among them, only compounds 6 and 19 showed cytotoxicity against the MCF-7 cell line [inhibition (%): 33.14 and 34.03, respectively].

Abstract:Two new guaiacene-type sesquiterpenes 13 alpha-dihydroixerin acid, ixerin acid and one new secoguaiacene-type sesquiterpene secoixerin Z, along with four known compounds, were separated from ethanol extract of Ixeris sonchifolia. The structures were determined based on the detailed spectroscopic and physicochemical methods. The cytotoxic activity of the isolates was tested against A549 cells. Among them, compound 3 exhibited potent cytotoxicity against A549 cells with the IC50 of 5.6 +/- 0.9 mu M. [GRAPHICS] .

Abstract:OBJECTIVE:To investigate the chemical constituents of Ilex cornuta.METHOD:The chemical constituents were isolated by column chromatographic methods. And the structures were identified by spectral data.RESULT:Seven compounds were isolated and identified as follows: 2alpha-hydroxy ursolic acid (1), arjunolic acid (2), 23-hydroxy ursolic acid (3), 27-0-p-(Z) -coumaroyl ursolic acid (4), 27-O-p-(E)-coumaroyl ursolic acid (5), asiatic acid (6).CONCLUSION:Compound 1, 2 were obtained from this genus for the first time and 3-6 from this plant for the first time.

Abstract:Twelve new nortriterpenoids, namely wuweizidilactones J-P (1-7), schindilactones I-K (8-10), preschisanartanin N (11), and schisdilactone J (12), as well as 23 known analogues, were isolated from the leaves and stems of Schisandra chinensis. The absolute configurations of 1 and 11 were established by calculated electronic circular dichroism (ECD) spectra and that of 7 was confirmed by single-crystal X-ray diffraction. The absolute configurations of 2-6, 8, and 9 were determined by an empirical comparison of their experimental ECD spectra with that of 1. It is the first time that the ECD spectra of 18(13 -> 14)-abeoschiartane-, 18-norschiartane-, and schisanartane-type nortriterpenoids possessing an alpha,beta-unsaturated-gamma-lactone moiety are presented, this method providing an alternative means of obtaining absolute configurational assignments for these types of nortriterpenoids. All isolates were evaluated for their anti-acetylcholinesterase and anti-butyrylcholinesterase activities, and compounds 8, 13, 23, and 31 show anti-acetylcholinesterase activity at concentrations of 50 mu M, with 12.7, 10.7, 16.6, and 32.1% inhibition, respectively. Abstract:Five new nortriterpenoids, schicagenins D-F (1-3, resp.) and negleschidilactones A and B (4 and 5, resp.), together with eleven known ones, were isolated from the stems of Schisandra neglecta. Their structures were established on the basis of extensive spectroscopic analyses. All the compounds were evaluated for their activities regarding insulin sensitivity in 3T3-L1 differentiated adipocytes. None of them showed a significant bioactivity at 10M concentration.

Abstract:Ilex asprella is one of representative medicinal plants in South of the Five Ridges of China.The roots and rhizomes of I.asprella have the effects of clearing heat and detoxifying,stimulating salvia,and reducing thirst,which has been used to treat windheat cold,acute and chronic pharyngitis,and sore throat.Contemporary studies showed that L asprella contains the major triterpenoids and glycosides,phenolic acids,and minor steroids.The extracts and compounds show activities of anti-inflammatory,antiviral,antitumor,and regulating lipid metabolism.The present paper summarizes a phytochemical and pharmacological advance on this species to provide reference for clarification of its pharmacologically active ingredients,quality evaluation,and further explorations.
