Abstract:OBJECTIVE To investigate the chemical constituents of Ilex pernyi. METHOD The chemical constituents were isolated by various column chromatographic methods. The structures were identified by spectral data. RESULT Eight triterpenoid compounds were isolated and identified as ursolic acid (1), lupeol (2), alpha-amyrin (3), uvaol (4), 3beta-hydroxyurs-11-ene-13beta-olide (5), pomolic acid (6), lup-20 (29)-ene-3beta, 24-diol (7), 3beta, 23-dihydroxy-urs-12-en-28-oic acid (8). CONCLUSION The eight compounds were obtained from this plant for the first time.

Abstract:A new compound and five known compounds were isolated from the ethanolic extract of the leaves of Ilex pernyi Franch. Their structures were established on the basis of spectral analysis and identified as trans-isoeugenyl-alpha-L-arabinopynosyl (1 --> 6) -beta-D-glucopyranoside (1) , kaempferol-3-O-sambubioside (2), quercetin-3-O-sambubioside (3), isoquercitrin (4), (+) -syringaresinol-O-beta-D-glucopyranoside (5), amarantholidoside IV (6). Among them, compound 1 is a new phenolic glycoside, named as ilexperphenoside A, and compounds 2-6 were isolated from this plant for the first time.

Abstract:Five new triterpene saponins, Ilexpublesnins N-R (1-5), along with seven known analogs were isolated from the root of Ilex pubescens. Their structures were elucidated on the basis of extensive spectroscopic analysis, including 1D and 2D NMR experiments. Ilexpublesnin N (1) possessed a rare 20-hydroxyursolic acid scaffold from natural resource. These compounds were evaluated in vitro for their cytotoxic effects on human cancer cell lines HepG2, HLE, BEL7402, BEL7403, BEL7405, MCF-7, HeLa. Among them, only compounds 5 and 10 showed cytotoxic potentiality against BEl-7403 and HEL cell lines [inhibition (%): 35.38 and 45.12, respectively].

Abstract:Eleven new triterpene saponins, ilexpublesnins C-M (1-11), along with ten known analogues were isolated from the roots of Ilex pubescens. Their structures were elucidated on the basis of extensive spectroscopic analysis, including 1D and 2D NMR experiments. Compounds 1, 2, 10, and 11 contain a 24-aldehyde, which is rare for triterpene saponins from Ilex. These compounds were evaluated in vitro for their cytotoxic effects on human cancer cell lines HepG2, HLE, BEL7402, BEL7403, BEL7405, MCF-7, and HeLa. Among them, only compounds 6 and 19 showed cytotoxicity against the MCF-7 cell line [inhibition (%): 33.14 and 34.03, respectively].

Abstract:An abietane diterpenoid, triptobenzene Y and six sesquiterpene polyol esters, wilforsinines C–H, together with 14 known compounds, have been isolated from the roots of Tripterygium wilfordii. The structures of the compounds were elucidated on the basis of spectroscopic analyses. The quinone reductase (QR) induction assay indicated that two compounds showed moderate QR-inducing activities at concentrations of 25μM and 50μM, respectively.

Abstract:Ilex latifolia Thunb is a traditional Chinese tea and herbal medicine. In this study, one new triterpene saponin (1) and six known triterpenoids (2-7) were isolated from the methanol extract of I. latifolia using a PC12 cell bioassay system. The structures and stereochemistry of these compounds were elucidated using spectroscopic methods and chemical derivatization. This new triterpene saponin (1) was characterized as an ursolic type acid with a 19α-hydroxyl and a trisaccharide moiety at C-3. Compound 1 significantly promoted the neurite outgrowth in PC12 cells by 52% at 10 μM, whereas compounds 2-7 showed less neuritogenic activity. Structure activity relationship studies indicated that introducing a trisaccharide moiety at C-3 is important for the neuritogenic activity, but the sugar group at C-28 decreased this activity. In addition, compound 1 increased the neurite outgrowth length in primary cortical neuron cells of mice and also exhibited a neuronal protection effect on H2O2-damaged PC12 cells at optimum concentrations.

Abstract:Two new phenolic glycosides (1-2), along with six existing compounds (3-8), were isolated from the ethanolic extract of Ilex pubescens roots, a traditional folk medicine. These structures were determined using HR-ESI-MS, IR, UV, and NMR (including 1 D, 2 D-NMR). The anti-inflammatory activities of three phenolic glycosides (1-3) were evaluated in the human HepG2 cell lines. The results showed that compound 3 could induce P-gp and BCRP expression through the Nrf2-mediated pathway.

Abstract:Six new triterpene saponins, ilexasprellanosides A-F (1-6, resp.), together with eleven known compounds were isolated from the roots of Ilex asprella. The new saponins were characterized as ursa-12,18-dien-28-oic acid 3-O-β-D-xylopyranoside (1), 19α-hydroxyursolic acid 3-O-β-D-(2'-O-acetylxylopyranoside) (2), 19α-hydroxyursolic acid 3-O-β-D-glucuronopyranoside (3), 3β,19α-dihydroxyolean-12-en-23,28-dioic acid 28-O-β-D-glucopyranoside (4), 19α-hydroxyoleanolic acid 3-O-β-D-(2'-O-acetylxylopyranoside) (5), 19α-hydroxyoleanolic acid 3-O-β-D-glucuronopyranoside (6). The structures of the new compounds were elucidated by analysis of their spectroscopic data and chemical degradation. Compounds 2, 4, oleanolic acid 3-O-β-D-glucuronopyranoside, 3-β-acetoxy-28-hydroxyurs-12-ene, and pomolic acid showed significant cytotoxic activities against human tumor cell line A549 (IC50 values of 1.87, 2.51, 1.41, 3.24, and 5.63 μM, resp.).

Abstract:Two new triterpenoid saponins, ilemaminosides A and B (1 and 2) along with six known saponins (3-8) were isolated from 70% ethanolic extract of the leaves of Ilex mamillata C.Y. Wu ex C.J. Tseng. The new saponins were characterised as 3-O-alpha-L-arabinopyranosyl-ilexgenin B (1) and 3-O-beta-D-glucopyranosyl-(1 -> 3)-alpha-L-arabinopyranosyl-ilexgenin B (2). The structures of compounds 1 and 2 were elucidated on the basis of the chemical and spectroscopic methods, and the structures of known compounds were identified by comparison of their spectroscopic data with those reported in the literature. The compounds showed inhibitory activities in anti-inflammatory assay in vitro with IC50 values in the range 25.37-38.33 mu gmL(-1).

Abstract:OBJECTOVE To investigate the liposoluble components from the leaves of llex comuta Lindl. ex Paxt.. METHODS The compounds were isolated by repeated column chromatography on silica gel and Sephadex LH-20, and the structures were elucidated by spectral analysis. RESULTS Eleven compounds were obtained. Their structures were identified as lupeol(Ⅰ), 11-keto-α-amyrin pahlmitate(Ⅱ), α-amyrin palmitate(Ⅲ), 12-ursene-3,28-diol(Ⅳ), ursolic acid(Ⅴ), 3β-hydroxylup-20(29)-en-30-al(Ⅵ), 3β-hydroxy-20-oxo-30-norlupane(Ⅶ), tanacetene(ⅧA),β-sitosterol (Ⅸ), n-behenic acid(Ⅹ), and n-hexacosane (Ⅺ). CONCLUSION Compound Ⅳ, Ⅶ, Ⅷ, Ⅹ and Ⅸ are firstly isolated from the genus Ilex. In addition, compound Ⅱ, Ⅲ and Ⅸ are isolated from l. comuta for the first time.

Abstract:Seven triterpenoid saponins, named ilexsaponin I–O, along with twelve known ones, were isolated from the roots of Ilex pubescens. The structures of all compounds were elucidated by use of extensive spectroscopic methods (IR, HR-ESI-MS, and 1D and 2D NMR). Sugar residues obtained after acid hydrolysis were identified by TLC and HPLC. The in vitro anti-inflammatory effects of the triterpenoid saponins were also evaluated in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Among the isolated saponins, seven compounds were shown to inhibit LPS-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production by suppressing the expression of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2), respectively, in LPS-stimulated RAW 264.7 cells. Ilexsaponin I and β-d-glucopyranosyl 3-β-[β-d-xylopyranosyl-(1 → 2)-β-d-glucopyranosyloxy]-olea-12-en-28-oate exerted more potent anti-inflammatory effects than the other compounds tested.
