Abstract:Penaeus monodon is a crucial economic crustacean globally and a focal point of aquatic research. Germplasm resources provide a diverse food source for humans, meeting various nutritional needs; they also maintain plant and animal gene banks, enhancing resilience, adaptability, and promoting ecological balance in terms of genetic diversity. To accurately assess the germplasm resources of P. monodon, we conducted a comprehensive investigation. We analyzed muscle characteristics, nutrient content, amino acids, fatty acids, and genetic diversity using whole-genome sequencing in six P. monodon groups. The results showed that the crude protein content of the six groups of P. monodon was 21.17–24.60 g/100 g, crude fat content was 0.60–0.80 g/100 g, essential amino acid content was 5.44–6.41 g/100 g, total amino acid content was 15.68–18.63 g/100 g, and total fatty acid content was 354.78–587.43 mg/100 g. Genetic differentiation index (Fst) values ranged from 0.029 to 0.604 for six groups. PmSZ has a good genetic condition. There was no significant difference in crude fat in conventional nutrients, and the difference in biochemical indexes T-AOC, CAT, and SOD was not significant (P > 0.05). The noteworthy point is that the analysis of genetic diversity reveals a moderate level of genetic diversity. PmDF and PmZH exhibit the highest degree of differentiation, while PmRP and PmZH show the lowest degree of differentiation. Our survey results establish a critical foundation for comprehending and harnessing the germplasm resources essential for selectively breeding robust P. monodon populations.

Abstract:To improve the quality of cultured large yellow croaker (Larimichthys crocea), this study was performed to study the impacts of glycerol monolaurate (GML) on the nutritional value, growth performance, muscle texture, and taste intensity of L. crocea. The results showed that GML as a feed additive significantly increased the crude lipid content and reduced the diameters of muscle fibers, which in turn markedly altered the flesh texture in terms of cohesiveness. Moreover, the taste indicators (umami and richness) and flavor-related amino acid (glutamic acid, glycine, and proline) contents of L. crocea muscle were significantly higher in the GML group. Metabolomic and gene expression analyses showed that GML supplementation could significantly improve amino acid biosynthesis and metabolism, promote protein and lipid synthesis, and activate myogenic-related signaling pathways of L. crocea. Consequently, adding an appropriate amount of GML to fish feed would be conducive to providing healthy, nutrient-rich and acceptably flavored aquatic-products.

Abstract:Background At present, fish meal (FM) resources are in short supply, and competition for food between humans and animals is becoming increasingly critical. Finding non-grain protein sources that can replace FM is the key to solving the rapid development of aquaculture. Methods Seven trial diets were prepared with 0 g/kg (EP0), 30 g/kg (EP3), 60 g/kg (EP6), 90 g/kg (EP9), 120 g/kg (EP12), 150 g/kg (EP15), and 180 g/kg (EP18) of enzyme-digested poultry by-product meal (EPBM) by replacing of FM. A total of 630 hybrid groupers ( Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂) were equally portioned into 21 tanks. At 8:00 and 16:00 each day, groupers were fed until they were full for a cumulative period of 8 weeks. Results The results showed that 30 g/kg of EPBM significantly increased the rates of weight gain and special growth ( P < 0.05). Significantly higher activities of serum glutamic pyruvic transaminase, glutamic oxaloacetic transaminase, catalase, and superoxide dismutase were observed in the EP3 group ( P < 0.05). The categories and numbers of the top 10 dominant bacteria in the phylum and genus levels were not significantly influenced by feed ( P > 0.05). In the proximal intestine and distal intestine, there were significantly higher expressions of SNAT3, LAAT1, CAT2 , and CAT1 in the EP3 group compared with the EP0 group ( P < 0.05). In the EP3 group, the expressions of PepT1, LAAT1, B 0, + AT , and CAT2 were significantly increased in MI than those in all other groups (except the EP0 group, P < 0.05). Conclusion When FM was replaced by 30 g/kg of EPBM, growth performance, antioxidant capacity, and the ability to transport amino acids and peptides of hybrid grouper were significantly improved.

Abstract:Abstract The feeding regime is a critical aspect of white shrimp ( Litopenaeus vannamei ) aquaculture, but research on intensive shrimp culture using automatic feeders is scarce. Our study investigated different feeding strategies for adult white shrimp in intensive aquaculture tanks, evaluating growth, survivability, feed utilization, shrimp digestive and antioxidant capacity, and conducting an economic analysis. Shrimp (8.0 ± 0.5 g) were cultured for 35 days with feeding frequencies of 6, 8, 10, and 12 times/day (A6, A8, A10, and A12 groups, respectively) using automatic feeders, and one group was fed manually 6 times/day (M6). The results show that the automatic feeding groups had a higher final body weight and specific growth rate, and a lower feed conversion ratio compared to the manual group. Among the automatic feeding groups, A6 and A8 exhibited the best performance, with a quadratic regression suggesting 7.83 times/day as an optimal feeding frequency. Digestive enzyme activity (TP and α-AMS) was significantly higher in the A6 and A8 groups ( p < 0.05). However, the A6 and A8 groups showed significantly higher levels of antioxidant enzymes (SOD and GPx) and lower levels of lipid oxidant enzyme (MDA), indicating better digestive and antioxidant abilities in shrimp with higher growth rates. Moreover, the antioxidant genes expression of MnSOD and GPx testified the enzyme changing. Pearson's correlation confirmed the positive correlation between SOD and GPx. Economically, the A8 group showed the highest profitability among all groups. Overall, the study recommends a feeding frequency of 6–8 times/day using automatic feeders as a suitable and feasible approach for white shrimp culture in intensive settings.

Abstract:Groupers, as a popular economic fish species, are now more frequently cultured in land-based facilities, however, traditional pond-farming methods are plagued by frequent disease outbreaks, unstable economic benefits, and environmental pollution. To explore a new farming mode for a grouper, an 80-day cultivation experiment was conducted using both a simulated pond system (SPS) and a recirculating aquaculture system (RAS). The research aims to evaluate the growth performance, health (pathogenic bacteria, intestinal microbiota), off-flavor compounds (geosmin, 2-methylisoborneol), and nutritional component (amino acids, fatty acid composition and content) of a hybrid grouper ( Pinephelus fuscoguttatus ♀ × Epinephelus lanceolatus ♂ ) under different aquaculture systems. The results showed that the hybrid grouper in a RAS exhibited better growth performance; the concentration of Vibrio in the fish tissue in the RAS was significantly lower than that in the SPS. Moreover, the content of fresh amino acids in the RAS was significantly higher, and the levels of saturated fatty acids (SFAs) and ω-6 polyunsaturated fatty acids (ω-6PUFAs) were significantly higher in the RAS. This finding indicates the superior flavor and nutritional value of the grouper. These results prove that the RAS is suitable for the widespread cultivation of grouper.

Abstract:The present study was conducted to investigate the effects of dietary phospholipids on growth performance, fatty acid composition, and lipid metabolism in juvenile hybrid grouper (Epinephelus fuscoguttatus ♀ × E. lancolatus ♂). Five isonitrogenous and isolipidic diets with graded levels of phospholipids (14.10, 32.10, 52.50, 70.20, and 91.30 g/kg) were fed to triplicate groups of juvenile grouper (initial body weight 13.00 ± 0.02 g) for 61 days. Results showed that dietary phospholipids produced no statistical influence on growth performance of grouper. The fatty acids profiles of experimental fish are correlated well with dietary fatty acids composition. The increase of dietary phospholipids significantly elevated the crude lipid content in whole fish body, liver and muscle. Meanwhile, the increased dietary phospholipids significantly promoted hepatic triglyceride (TG) accumulation. The results of gene expression analysis revealed that dietary phospholipids did not significantly affect the expression of lipogenesis related genes, FAS, ACC and DGAT2. In addition, high dietary phospholipids inclusion significantly decreased the expression of fatty acids oxidation related genes, PPARɑ and CPT1. In summary, juvenile grouper don’t have a specific requirement for dietary phospholipids which inclusion aggravated lipid deposition through the regulation of fatty acids oxidation at the transcriptional level.

Abstract:Dietary phospholipids supplementation is essential for normal development of fish larvae due to their limited phospholipid biosynthetic ability. The present study was conducted to investigate the effects of dietary phospholipids on growth performance, fatty acid composition, antioxidant capacity and lipid metabolism of larval hybrid grouper. The larvae (40 day after hatching, 0.37 +/- 0.01 g) were fed the diet with graded level of phospholipids (1.87, 3.61, 5.53, 7.25 and 9.69%) four times daily for 23 days. Results showed that the specific growth rate of grouper larvae was significantly improved with the increase of dietary phospholipids, and optimal dietary phospholipids was estimated to be 9.17%. Fatty acids composition of whole fish body and liver was highly reflective of dietary fatty acid profile. In addition, the increase of dietary phospholipids resulted in a significantly elevated hepatic crude lipid and triglyceride content in a dose-dependent manner. The analysis of gene expression revealed that increased dietary phospholipids did not produce any statistical effect on the expression of lipogenesis genes including LXRalpha, FAS and ACC, but significantly facilitated the expression of DGAT2, a rate-limiting enzyme involved in triglyceride synthesis. The expression of genes, CPT1, involved in I3-oxidation was not significantly influenced as dietary phospholipids increased. However, phospholipids inclusion significantly reduced the expression of ATGL, an rate-limiting lipolytic enzyme. Additionally, the supplementation of dietary phospholipids significantly elevated the activity of antioxidant relate enzymes, CAT, SOD and T-AOC, but significantly decreased the MDA content, an end product of fatty acid peroxidation. In summary, dietary phospholipids supplementation was beneficial in improving growth performance and antioxidant capacity of grouper larvae, and meanwhile, promoted hepatic lipid deposition through regulating the synthesis and hydrolysis of triglyceride.

Abstract:Abstract Background: Nano-Se-enriched Gracilaria lemaneiformis ( G. lemaneiformis ) as feed ingredient on aquatic animals remained largely unknown. Feeding experiment was conducted for a total of 45 days to research the biological impacts of nano-selenium (Nano-Se) (group N), G. lemaneiformis (group G) and Nano-Se-enriched G. lemaneiformis (group NG) on growth, selenium concentration, oxidation, immunity and nutrition of Epinephelus coioides . Results: At each tested time point, the weight gain rate, specific growth rate and feed conversion rate of group N, group G and group NG were significantly increased in contrast to those of the control group (group C). A remarkable increase of the hepatosomatic index (HSI) and viscerosomatic index (VSI) was observed in group N and group NG. The activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) of fish in group N, group G and group NG were significantly enhanced relative to those in group C. The immune system was also activated by showing an elevated expression level of various immunological genes including Immunoglobulin M, tumor necrosis factor-α and Interleukin -8. The muscle Se content in group N and group NG was 31- and 40.5-fold that of the control group (group C), respectively. The liver Se value of group N and group NG was 1.06 and 2.50 times higher than that of group C, respectively. The lipid and moisture values of group N and group NG are increased and reduced, respectively, in comparison to that of group C. The diet supplementation of Nano-Se-enriched G. lemaneiformis led to significant increase of the protein content, delicious amino acids along with total amino acids content in grouper. Conclusions: Our results demonstrate a great application potential of Nano-Se-enriched G. lemaneiformis as dietary additive in grouper.
Effects of High-Lipid Dietary Protein Ratio on Growth, Antioxidant Parameters, Histological Structure, and Expression of Antioxidant- and Immune-Related Genes of Hybrid Grouper

Abstract:The hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus) is a new species of grouper crossed from giant grouper (E. lanceolatus) as the male parent and brown-marbled grouper (E. fuscoguttatus) as the female parent. We hypothesized that optimal levels of dietary protein may benefit liver function. High-lipid diets are energetic feeds that conserve protein and reduce costs, and are a hot topic in aquaculture today. Therefore, the objective of the research is to investigated the effects of dietary protein level in high-lipid diets on serum and liver biochemistry, liver histology, and liver immune and antioxidant indexes and gene mRNA expression of the juvenile hybrid grouper (♀ Epinephelus fuscoguttatus × ♂ E. lanceolatus). Six iso-lipidic (161 g/kg) diets were formulated containing graded levels of protein (510 as control, 480,450, 420, 390 and 360 g/kg). Each treatment consisted of three replicates and 30 fish (6.70 ± 0.02 g) in one replicate. After an 8-week feeding experiment, the results indicated the following: (1) With the decreasing of dietary protein level, the specific growth rate (SGR) of groupers increased gradually and then decreased; SGRs of the 390 and 360 g/kg groups were significantly lower than other groups (p < 0.05). (2) In terms of serum and liver, the activity of antioxidant enzymes such as catalase (CAT) and superoxide dismutase (SOD), and the total antioxidant capacity (T-AOC) content, and the activity of immune enzymes such as lysozyme (LYS) and immunoglobulin (IgM) was significantly increased under the appropriate protein level. (3) Based on liver histology, we know that high or low dietary protein levels cause liver damage. (4) Dietary protein levels can significantly affect the mRNA expression levels of an anti-inflammatory factor gene (tgfβ), pro-inflammatory factor genes (il6, il8), heat shock proteins, and antioxidant and immune genes (hsp70 and hsp90, gpx, nrf2, keap1). It is concluded that the appropriate protein level can promote the growth performance of groupers, improve antioxidant activity and immune enzyme activity in serum and liver, and enhance the expression of immune genes.

Abstract:This experiment was conducted to determine the optimum amount of peanut cake (PNK) in the diet of hybrid grouper by studying the effect of PNK on the growth, antioxidant capacity, liver fat metabolism, and intestinal microbiota composition by replacing fish meal (FM) protein in the diet of the juvenile hybrid grouper. A total of 840 juvenile hybrid grouper with uniform body weight (13.20 ± 0.05 g) were selected from 7 treatments and 4 replicates (30 fish each) farmed for eight weeks. Results showed that the weight gain rate (WGR) of the low-substitution group was significantly higher than that of the high-substitution group (P < 0.05) and that the feed conversion and specific growth rates of the 44%, 55%, and 66% groups were significantly higher than those of the other groups (P < 0.05). The crude lipid content of the 44% group was significantly higher than those of other groups (P < 0.05), and the crude protein content of the 66% group was lowest and significantly lower than that of the control group (P < 0.05). The total protein content showed a decreasing trend and then an increasing trend. The total cholesterol content showed a gradually decreasing trend and was significantly higher in the control and 11% groups than those in the other groups (P < 0.05). The glutamic oxalacetic transaminase content in the 66% group was significantly higher than those in other groups (P < 0.05). The total antioxidant capacity and the malondialdehyde content of the 66% group were significantly higher (P < 0.05) and significantly lower (P < 0.05), respectively, than those of other groups. The oil red-stained sections of the liver showed that the deposited fat gradually increased with increasing levels of PNK replacement in the diet. A replacement level higher than 22% caused a decrease in WGR. When the replacement level was higher than 44%, a significant increase (P < 0.05) in harmful bacteria in their intestines was observed, and when the replacement level was higher than 55%, a significant increase (P < 0.05) in liver fat accumulation was observed. Based on the PNK replacement level corresponding to WGR, the appropriate PNK replacement level in the diet was 15%.

Abstract:The effect of hydrolyzed fish protein powder (HFP) on the growth, intestinal development, gene mRNA expression, and enzyme activity in the intestine and liver of juvenile hybrid grouper ( Epinephelus fuscoguttatus ♀ and Epinephelus lanceolatus ♂) was assessed after an 8-week feeding trial. Seven isonitrogenous (50%) and isolipidic (9%) diets were fed to hybrid grouper with 0% (CT), 1% (H1), 1.5% (H2), 2% (H3), 2.5% (H4), 3% (H5), and 4% (H6) HFP. No significant difference ( p > 0.05) in weight gain rate (WGR), specific growth rate (SGR), feed conversion ratio (FCR), and survival rate (SR) was observed in all the groups. The crude protein content in the H6 group was significantly higher than in the other groups ( p < 0.05). Intestinal lipase and trypsin activity were significantly higher in H3 and H5 groups ( p < 0.05). In the serum, superoxide dismutase (SOD) activity was significantly higher in H5 and H6 groups, while malondialdehyde (MDA) activity was lower ( p < 0.05) compared to other treatments. Insulin-like growth factor ( IGF-I ) and target of rapamycin ( TOR ) mRNA expression levels in the intestine and muscle were significantly higher in the H2 group and H1 group ( p < 0.05), respectively. The most abundant intestinal bacteria found at the genus level are Acinetobacter , Vibrio , and Flavobacteriaceae. The villus was significantly longer in hybrid grouper fed with different levels of HFP compared to the control, and fish in the H2 group had thicker intestinal muscle compared to the other groups ( p < 0.05). In conclusion, the addition of HFP to the low fishmeal (FM) diets of juvenile grouper improved the intestinal development and increased the levels of intestinal digestive enzymes.

Abstract:In a context where the search for plant-derived additives is a hot topic, glycerol monolaurate (GML) was chosen as our subject to study its effect on grouper (Epinephelus fuscoguttatus♀ × Epinephelus lanceolatus♂). Seven gradient levels of GML (0, 600, 1200, 1800, 2400, 3000, and 3600 mg/kg) were used for the experiment. Based on our experiments, 1800 mg/kg GML significantly increased the final body weight (FBW) and weight gain rate (WGR). GML increased the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) and decreased malondialdehyde (MDA). Adding 1800 mg/kg GML also significantly increased the levels of lauric acid (C12:0) (LA), n-3 polyunsaturated fatty acids (PFA), and the n-6 PFA-to-n-3/n-6 ratio, while significantly decreasing the levels of saturated fatty acids (SFA). Dietary supplementation with GML significantly inhibited the expression of pro-inflammatory factors and reduced the occurrence of inflammation. GML improved intestinal flora and the abundance of beneficial bacteria (Bacillus, Psychrobacter, Acinetobacter, Acinetobacter, Stenotrophomonas, and Glutamicibacter). It provides a theoretical basis for the application of GML in aquafeed and greatly enhances the possibility of using GML in aquafeed. Based on the above experimental results, the optimum level of GML in grouper feed is 1800 mg/kg.

Screening of host gut-derived probiotics and effects of feeding probiotics on growth, immunity, and antioxidant enzyme activity of hybrid grouper (Epinephelus fuscoguttatus♀ × E. lanceolatus♂)

Abstract:In recent years, the widespread use of antibiotics in intensive grouper mariculture has resulted in the ineffectiveness of antibiotic treatment, leading to an increasing incidence of diseases caused by bacteria, viruses, and parasites, causing serious economic losses. Hence, it is crucial to develop alternative strategies to antibiotics for healthy and sustainable development of the mariculture industry. Here, we aimed to screen host gut-derived probiotics and evaluate its effects on growth and immunity of grouper. In this study, 43 bacterial strains were isolated from the intestine of the hybrid grouper (Epinephelus fuscoguttatus♀ × E. lanceolatus♂), and a potential probiotic strain G1-26, which can efficiently secrete amylase, protease, and lipase, was obtained using different screening mediums. Based on 16S rDNA sequencing, the potential probiotic strain G1-26 was identified as Vibrio fluvialis. The results of a biological characteristic evaluation showed that V. fluvialis G1-26 could grow at 25-45 °C, pH 5.5-7.5, salinity 10-40, and bile salt concentration 0-0.030%, and produce amylase, lipase, and protease under different culture conditions. Additionally, V. fluvialis G1-26 is sensitive to many antibiotics and does not exhibit aquatic biotoxicity. Subsequently, hybrid groupers were fed diets containing V. fluvialis G1-26 at different concentrations (0, 106, 108, and 1010 CFU/g) for 60 d. The results showed that V. fluvialis G1-26 at 108 CFU/g did not significantly affect the growth performance of the hybrid grouper (P > 0.05). V. fluvialis G1-26 supplementation at 108 and 1010 CFU/g significantly promoted the relative expression of immune-related genes in hybrid groupers (TLR3, TLR5, IL-1β, IL-8, IL-10, CTL, LysC, TNF-2, and MHC-2) and improved the activities of alkaline phosphatase, acid phosphatase, total superoxide dismutase, and total protein in the liver. In conclusion, V. fluvialis G1-26, a potential probiotic strain isolated from the intestine of the hybrid grouper, can be used as an effective immunopotentiator at an optimal dose of 108 CFU/g diet. Our results provide a scientific basis for the development and utilization of probiotics in the grouper mariculture industry.

Abstract:Seaweed draws a lot of attention for its vital role in aquaculture as it contains beneficial biological compounds that undoubtedly might help in the development of this field. The current study sheds light on the potential efficiency of dietary supplements of Grateloupia acuminata and G. doryphore (Halymeniaceae) nanoparticles (GNS) at different levels with bionanocomposite cellulose acetate membranes (CA/bio-AgNps) on improved growth performance, digestive enzyme activity, immunity, antioxidative, resistance against infectious pathogens, and characterization of water quality treated with CA/bio-AgNps that is used in rearing Nile tilapia ( Oreochromis niloticus ). Four concentrations (0.1, 0.25, 0.5, and 1.0 ml/L) of GNS extract were tested as potential anti-bacterial and for the efficacy of being parasitic. Fish with an average weight (24.46 ± 0. 50 g) were apportioned into six experimental groups (T0, T1, T2, T3, T4, and T5) represented as 0.0%, 0.0%, 0.1%, 0.25%, 0.5%, and 1.0% GNS in diets with CA/bio-AgNps, respectively. Injection of fish with Aeromonas hydrophila was performed at the end of the trial. Chemical and bacteriological water indices significantly showed improvement after being treated with CA/bio-AgNps than the control group. Growth, carcass composition, digestive enzyme, and hematological and biochemical indices were significantly noticed positive ( p< 0.05), especially T4 and T5, than the control group. In parallel, a significant improvement was noticed in serum lysozyme, total immunoglobulin, complement C3, antioxidative enzyme, and the relative expression of hepatic and inflammatory genes with an increased level of GNS ( p< 0.05) are upregulated than the control group. Remarkably, GNS-supplemented diets and extracts provided positive efficacy against A. hydrophila with a decreased percentage of fish mortality, besides efficacy on antibacterial strains and Cichlidogyrus tilapiae , respectively. To sum up, the seaweed extract with CA/bio-AgNps resulted in better growth performance of fish, antipathogenic effect, and health status. Furthermore, CA/bio-AgNps were vital in improving water characteristics. They should be studied and applied more in the future.

Abstract:Abstract The purpose of subject was to explore the optimum protein requirement of juvenile grouper ( Epinephelus coioides ). In the test, 450 juveniles with an average weight (10.02 ± 0.22) g were randomly divided into six groups with triplicate, and were fed with 350, 400, 450, 500, 550 and 600 g/kg iso-lipid test diet twice 1 day for 8 weeks, respectively. The results showed that: (1) With the increase of protein level, the body weight gain rate and specific growth rate first increased and then reduced, while the feed coefficient rate first decreased and then increased, while the protein efficiency significantly decreased ( P < 0.05). (2) With the increase of protein level, the condition factor, hepaticsomatic index and visceralsomatic index significantly reduced ( P < 0.05). (3) With the increase of protein level, the crude protein content of whole fish and muscle gradually increased, while the crude lipid content gradually decreased. (4) High-protein diet (550–600 g/kg) significantly increased the plasma total protein content and decreased the triglyceride content of orange-spotted grouper ( P < 0.05). (5) Compared with the 350 g/kg group, 500, 550, 600 g/kg groups significantly increased the activities of glutamic-pyruvic transaminase and glutamic oxaloacetic transaminase in liver ( P < 0.05). (6) With the increase of protein level, the protease activity of intestine first increased and then decreased, and reached the maximum at the protein level of 500 g/kg, while lipase and amylase decreased significantly ( P < 0.05). (7) The activities of acid phosphatase, superoxide dismutase and lysozyme in liver increased first and then decreased with the increase of protein level, and reached the maximum in the 400 g/kg protein group. According to the analysis specific growth rate, the optimum protein level of juvenile orange-spotted grouper is 521.84 g/kg.
