Abstract:Asymmetric $\rm \beta$-sheet protein structures in spider silk should induce nonlinear optical interaction such as second harmonic generation (SHG) which is experimentally observed for a radial line and dragline spider silk by using an imaging femtosecond laser SHG microscope. By comparing different spider silks, we found that the SHG signal correlates with the existence of the protein $\rm \beta$-sheets. Measurements of the polarization dependence of SHG from the dragline indicated that the $\rm \beta$-sheet has a nonlinear response depending on the direction of the incident electric field. We propose a model of what orientation the $\rm \beta$-sheet takes in spider silk.

Abstract:Silica nanowires provide strong mode confinement in a cylindrical silica-core/air-cladding geometry and serve a model system for studying nonlinear propagation of short optical pulses inside fibers. We report on the fiber diameter dependence of the supercontinuum generated by femtosecond laser pulses in silica fiber tapers with average diameters in the range of 200 nm to 1200 nm. We observe a strong diameter-dependence of the spectral broadening, which can be attributed to the fiber's diameter-dependent dispersion and nonlinearity. The short interaction length (less than 20 mm) and the low energy threshold for supercontinuum generation (about 1 nJ) make tapered fibers with diameters between 400 nm and 800 nm an ideal source of coherent white-light source in nanophotonics.

Abstract:Measurements of the intensity transfer function of a silica microfiber resonator are shown to follow a wide variety of hysteresis cycles, depending on the cavity detuning and the scanning frequency of the range of input powers. We attribute these observations to a nonlinear phase shift of thermal origin and provide a simple model that reproduces well our measurements. The response time is found to be around 0.6 ms.

Abstract:A water soluble amorphous form of silk was made by ultra-short laser pulse irradiation and detected by nanoscale IR mapping. An optical absorption-induced nanoscale surface expansion was probed to yield the spectral response of silk at IR molecular fingerprinting wavelengths with a high ~ nm spatial resolution defined by the tip of the probe. Silk microtomed sections of 1–5 μm in thickness were prepared for nanoscale spectroscopy and a laser was used to induce amorphisation. Comparison of silk absorbance measurements carried out by table-top and synchrotron Fourier transform IR spectroscopy proved that chemical imaging obtained at high spatial resolution and specificity (able to discriminate between amorphous and crystalline silk) is reliably achieved by nanoscale IR. Differences in absorbance and spectral line-shapes of the bands are related to the different sensitivity of the applied methods to real and imaginary parts of permittivity. A nanoscale material characterization by combining synchrotron IR radiation and nano-IR is discussed.

Abstract:Synchrotron FTIR (S-FTIR) microspectroscopy was used to monitor the silk protein conformation in a range of single natural silk fibers (domestic and wild silkworm and spider dragline silk). With the selection of suitable aperture size, we obtained high-resolution S-FTIR spectra capable of semiquantitative analysis of protein secondary structures. For the first time, we have determined from S-FTIR the β-sheet content in a range of natural single silk fibers, 28 ± 4, 23 ± 2, and 17 ± 4% in Bombyx mori, Antheraea pernyi, and Nephila edulis silks, respectively. The trend of β-sheet content in different silk fibers from the current study accords quite well with published data determined by XRD, Raman, and (13)C NMR. Our results indicate that the S-FTIR microspectroscopy method has considerable potential for the study of single natural silk fibers.

Abstract:The nanoscale composition of silk defining its unique properties via a hierarchial structural anisotropy needs to be analysed at the highest spatial resolution of tens of nanometers corresponding to the size of fibrils made of β-sheets, which are the crystalline building blocks of silk. Nanoscale optical and structural properties of silk have been measured from 100 nm thick longitudinal slices of silk fibers with ca. 10 nm resolution, the highest so far. Optical sub-wavelength resolution in hyperspectral mapping of absorbance and molecular orientation were carried out for comparison at IR wavelengths of 2–10 μm using synchrotron radiation. A reliable distinction of transmission changes by only 1–2% as the anisotropy of amide bands was obtained from nanometer-thin slices of silk.

Abstract:We have studied the ultrafast saturation behavior of a high-density well-aligned single-walled carbon nanotubes saturable absorber (HDWA-SWCNT SA), obtained by a high-pressure and high-temperature treatment of commercially available single-wall carbon nanotubes (SWCNTs) and related it to femtosecond erbium-doped fiber laser performance. We have observed the polarization dependence of a nonlinear optical saturation, along with a low saturation energy level of <1 fJ, limited to the detector threshold used, and the ultrafast response time of <250 fs, while the modulation depth was approximately 12%. We have obtained the generation of ultrashort stretched pulses with a low mode-locking launching threshold of ~100 mW and an average output power of 12.5 mW in an erbium-doped ring laser with the hybrid mode-locking of a VDVA-SWNT SA in combination with the effects of nonlinear polarization evolution. Dechirped pulses with a duration of 180 fs were generated, with a repetition rate of about 42.22 MHz. The average output power standard deviation was about 0.06% RMS during 3 h of measurement.

Abstract:Silk fibers are one of the most attractive natural materials that exhibit enchanting luster and superior mechanical properties. Exploring the unique hierarchical architecture structures of natural silk fibers is the basis to understanding these unique properties. Here, we report observations of 3.1 +/- 0.8 nm silk nanofibrils and 3.7 +/- 0.9 A silk molecule chains exfoliated from natural silkworm silk fibers. Interestingly, the individual nanofibrils and protein chains show periodic diameters fluctuating along their axes. We further find that the thicker regions are relatively softer and the thinner regions are stiffer, and these can be assigned to alternatively distributed alpha-helix and beta-sheet domains, respectively. Based on these observations, we proposed a refined structure model of natural silk, which ranges from the molecular level to the fiber scale. These findings provide new opportunities to understand and exploit the unique structure-property relationships found in natural silk fibers, including inspiring the design of new artificial materials.

Abstract:This paper demonstrates a possible new microfiber bio near field lens that uses minor ampullate spider silk,spun from the Nephila edulis spider, to create a real time image of a surface using near field optical techniques. The microfiber bio lens is the world's first natural superlens created by exploring biological materials. The resolution of the surface image overcomes the diffraction limit, with the ability to resolve patterns at 100 nm under a standard white light source in reflection mode. This resolution offers further developments in superlens technology and paves the way for new bio optics.

Abstract:We present an experimental set up and the calculation approach for the measureme n t of the spectral phase of femtosecond laser pulse by using a spectral phase int erferometry for direct electric reconstruction (SPIDER) technique in this paper, the method for determining crucial parameters as Ωand τwas given. W ith the femtosecond laser pulse from the Ti:sapphire laser, we obtained the phase of the laser spectrum by using the SPIDER, the temporal profile of the original laser pulse was deduced based on the result. It reveals the FWHM width is 107fs, which is in good agreement with the result measured directly by using an autocorrelat or.

Abstract:Nanofibrils play a pivotal role in spider silk and are responsible for many of the impressive properties of this unique natural material. However, little is known about the internal structure of these protein fibrils. We carry out polarized Raman and polarized Fourier-transform infrared spectroscopies on native spider silk nanofibrils and determine the concentrations of six distinct protein secondary structures, including β-sheets, and two types of helical structures, for which we also determine orientation distributions. Our advancements in peak assignments are in full agreement with the published silk vibrational spectroscopy literature. We further corroborate our findings with X-ray diffraction and magic-angle spinning nuclear magnetic resonance experiments. Based on the latter and on polypeptide Raman spectra, we assess the role of key amino acids in different secondary structures. For the recluse spider we develop a highly detailed structural model, featuring seven levels of structural hierarchy. The approaches we develop are directly applicable to other proteinaceous materials.

Abstract:Multiple foci and a long filament are observed when we focus a femtosecond laser pulse into a fused-silica sample. The dependences of the intensity distribution of the plasma luminescence on the pulse energy and the numerical aperture (NA) of the focusing objective are investigated. Multiple foci are observed when NA of </=0.65 . A long filament tail is formed instead of multiple foci when the NA is 0.85. A physical image of femtoseond pulse propagation is given by a model based on the nonlinear Schrödinger equation.

Abstract:A new atomic force microscopy (AFM)-based technique, single-molecule force spectroscopy (SMFS), was used to study the nanomechanics of Bombyx mori silk fibroin. Three types of force-extension curves were found in the system. A modified freely jointed chain (MFJC) model can fit two of the three types well, but the fit parameters are different. The third type of force curve, in which there exists a plateau, cannot be modeled by MFJC. These results may show that there are three kinds of conformations in the silk fibroin system and SMFS can "distinguish" different conformations of the polymer chains.

Abstract:Irradiation of solid surfaces with intense ultrashort laser pulses represents a unique way of depositing energy into materials. It allows to realize states of extreme electronic excitation and/or very high temperature and pressure, and to drive materials close to and beyond fundamental stability limits. As a consequence, structural changes and phase transitions often occur along unusual pathways and under strongly non-equilibrium conditions. Due to the inherent multiscale nature - both temporally and spatially - of these irreversible processes their direct experimental observation requires techniques that combine high temporal resolution with the appropriate spatial resolution and the capability to obtain good quality data on a single pulse/event basis. In this respect fourth generation light sources, namely short wavelength, short pulse free electron lasers (FELs) are offering new and fascinating possibilities. As an example, this chapter will discuss the results of scattering experiments carried at the FLASH free electron laser at DESY (Hamburg, Germany), which allowed us to resolve laser-induced structure formation at surfaces on the nanometer to sub-micron length scale and in temporal regimes ranging from picoseconds to several nanoseconds with sub-picosecond resolution.

Abstract:We demonstrate a fiber-based femtosecond laser source, which can deliver 37 MHz, similar to 100 fs laser pulses tunable from 920 nm to 1030 nm with up to 10 nJ pulse energy. Pumped by an ultrafast Yb-doped fiber laser, a short piece of tapered photonic crystal fiber with lower dispersion was used to generate self-phase-modulation filtered for the tunable pulses. We believe that this new type of high-energy ultrafast fiber laser source can be an ideal solution for the two-photon fluorescence microscopy applications.

Abstract:We report a second-order nonlinear nanoprobe for characterizing ultrafast optical near fields. The proposed nanoprobe comprises second harmonic nanocrystals attached to a carbon nanotube, which is in turn attached to a silica fiber taper. We demonstrate in situ pulse characterization directly in the air core of a photonic crystal fiber. Further, it is shown that nanoprobes containing a single nanocrystal in the tip of the nanotube can be fabricated by auxiliary focused ion beam nanomilling. These results indicate that the proposed nanoprobe can open an avenue for probing the evolution of ultrafast optical fields in complex three-dimensional micro-or nanostructures. Abstract:Synchrotron FTIR (S-FTIR) microspectroscopy was used to monitor both protein secondary structures (conformations) and their orientations in single cocoon silk fibers of the Chinese Tussah silk moth ( Antheraea pernyi ). In addition, to understand further the relationship between structure and properties of single silk fibers, we studied the changes of orientation and content of different secondary structures in single A. pernyi silk fibers when subjected to different strains. The results showed that the content and orientation of β-sheet was almost unchanged for strains from 0 to 0.3. However, the orientation of α-helix and random coil improved progressively with increasing strain, with a parallel decrease in α-helix content and an increase in random coil. This clearly indicates that most of the deformation upon stretching of the single fiber is due to the change of orientation in the amorphous regions coupled with a conversion of some of the α-helix to random coil. These observations provide an explanation for the supercontraction behavior of certain animal silks and are likely to facilitate understanding and optimization of postdrawing used in the conjunction with the wet-spinning of silk fibers from regenerated silk solutions. Thus, our work demonstrates the power of S-FTIR microspectroscopy for studying biopolymers.
