Abstract:Bin blending is one of the main steps in pharmaceutical production processes. Commercial-scale production of expensive products typically does not allow to perform a large number of experiments in order to optimize the process. Alternatively, Discrete Element Method (DEM) simulations can be used to evaluate the powder behavior (flow and blending pattern) during blending, identify the risks (e.g., segregation), and provide solutions to mitigate them. In this work, DEM simulations are used to investigate the blending of two granulated powders in commercial-scale cone and cylindrical (hoop) blenders. The DEM contact model parameters were calibrated based on the experimental compression and ring shear tests for both granulated powders to mimic the bulk powder behavior in the simulations. The model's output was compared to the experiments in one of the blending cases. The blending efficiency in the cone blenders was evaluated considering the fill levels, the presence of baffles, the rotating directions, the filling order, and the bin sizes. Furthermore, for the hoop blenders, the effects of blender's angle, rotation speed, and filling order were addressed. The main findings of the work were that, in cone blenders, the blending can be improved by introducing baffles and changing in the rotational direction frequently. In hoop blenders, blending can be improved by increasing the inclination angle from the horizontal plane and the rotational speed.

Abstract:Compared with the original Discrete Element Method, the coarse-grained DEM significantly improves simulation efficiency. In CG DEM, the coarse-grained ratio is defined as the diameter of the coarse-grained particle divided by the diameter of the original particle. When the upper limit of coarse-grained ratio is used, the simulation is the most efficient. The upper value for simulating mixing process of spherical fuel particles was determined using a multi-level methodology. The adequacy of the upper value of the coarse-grained ratio was assessed by comparing the mixing characteristics and kinetic prop-erties of the coarse-grained particle systems with those of the original particle systems in a rotating drum with blades. The impact of rotation speed and particle number on the upper value was examined and dis-cussed. It was observed that the kinetic energy increases with an increase in the coarse-grained ratio, lim-iting the efficiency improvement. To ensure kinetic energy conservation, a scaling law for the restitution coefficient was proposed. Combined with the Reduced Particle Stiffness method, the simulation time of the coarse-grained case is only 1/3030 that of the original case (with a particle number of 2.3 million) when the upper limit of the coarse-grained ratio was used (12.0).(c) 2023 Published by Elsevier B.V. on behalf of The Society of Powder Technology Japan. Abstract:An efficient Discrete Element Method (DEM) modeling methodology for mixing of solid flavor particles in a rotating drum is presented. Machine learning and optimization algorithms are combined with geometric scaling and particle size distribution trimming to produce several orders of magnitude reduction in computational expense relative to the traditional approach of modeling at the physical system scale and performing purely simulation-based model calibration. The validity of the methodology is evaluated by comparing DEM model results for the evolution of the mixture uniformity with a laboratory scale drum and good agreement is observed. The limitations of the approach with respect to the amount of data required and the degree of geometric scaling and particle size trimming that is possible are also discussed. The approach developed could serve as an example for future simulation studies on fine particles in various processes that are relevant to the food, ingredient and pharmaceutical industry.

Abstract:Continuous Direct Compaction (CDC) has emerged as a promising route towards producing solid dosage forms while reducing material, development time and energy consumption. Understanding the response of powder processing unit operations, especially blenders, is crucial. There is a substantial body of work around how lubrication via batch blender operation affects tablet critical quality attributes such as hardness and tensile strength. But, aside from being batch operations, the design of these blenders is such that they operate with low-shear, low-intensity mixing at Froude number values significantly below 0.4 (Froude number Fr being the dimensionless ratio of inertial to gravitational forces). The present work explores the performance of a mini-blender which has a fundamentally different mode of operation (static vessel with rotating blades around a mixing shaft as opposed to rotating vessel with no mixing shaft). This difference allows a substantially wider operating range in terms of speed and shear (and Fr values). The present work evaluates how its performance compares to other blenders studied in the literature. Tablet compaction data from blends produced at various intensities and regimes of mixing in the mini-blender follow a common trajectory. Model equations from literature are suitably modified by inclusion of the Froude number Fr , but only for situations where the Froude number was sufficiently high (1 < Fr ). The results suggest that although a similar lubrication extent plateau is eventually reached it is the intensity of mixing ( i.e. captured using the Froude number as a surrogate) which is important for the lubrication dynamics in the mini-blender, next to the number of revolutions. The degree of fill or headspace, on the other hand, is only crucial to the performance of common batch blenders. Testing using alternative formulations shows the same common trend across mixing intensities, suggesting the validity of the approach to capture lubrication dynamics for this system.

Abstract:We present a design strategy of in-line mixers for 3D printing and an evaluation protocol that comprehensively characterizes the performance of hydrodynamics, reagent mixing, heat exchange, and reaction behavior. For demonstration, we engineer a split-and-recombination (SAR)-type mixer with stationary oblique baffles which effectively blends two streams in the laminar flow regime, particularly at low and intermediate Reynolds number. The 3D printing technique that opens up new design freedom and enables rapid model evaluation and optimization is low-cost and easy to implement in the laboratory. Simulations of hydrodynamics, heat transfer, and mass transfer including reactions form a solid foundation of mixer evaluation. Experimental results of the Villermaux-Dushman reaction and pressure measurement validate the numerical model and illustrate the difference between the designed SAR mixer and a conventional T-mixer. In addition, superior mixing performance of the SAR mixer is confirmed by the mixing intensity simulations, as compared with T-/helical mixers. The presented model yields new insights into the scalability and applicability of the 3D-printed integrated reactor.
On the Applicability of the Coarse Grained Coupled CFD-DEM Model to Predict the Heat Transfer During the Fluidized Bed Drying of Pharmaceutical Granules

Abstract:Fluidized bed dryer often used in the pharmaceutical industry for drying of wet granules. Coupled computational fluid dynamics (CFD) - discrete element method (DEM) is frequently used to model the drying process because of its ability to obtain the relevant information at the particle level. However, it becomes almost impossible to model the industrial scale fluidized bed dryer using the coupled CFD-DEM method because of the presence of large number of particles [Formula: see text]. To reduce the number of particles to be tracked in the simulation, coarse grained coupled CFD-DEM method was developed by researchers where a certain number of particles of the original system was represented by a relatively bigger particle in the coarse-grained system. The appropriate scaling of the particle-particle and particle-fluid interaction forces is necessary to make sure that the dynamics of the coarse-grained particles/parcels accurately represent the dynamics of the original particles. The coarse-graining of the drying process of pharmaceutical granules during fluidization needs systematic coarse-graining of the momentum, heat, and solvent vapor transfer process. A coarse grained coupled CFD-DEM method was used to model the momentum and heat transfer during the fluidization of pharmaceutical granules. It was shown that the heat transfer during the fluidization of large number of particles could be predicted by simulating a smaller number of bigger particles with appropriate scaling of particle-particle heat and momentum transfer, and particle-fluid heat and momentum transfer at significantly smaller computational time. This model can be further extended by including a coarse-grained moisture transport model in future.

Abstract:An investigation on the predictive capabilities of Discrete Element Method simulations of a powder mixing process in a laboratory scale paddle blade mixer is presented. The visco-elasto-plastic frictional adhesive DEM contact model of Thakur et al. (2014) was used to represent the cohesive behaviour of an aluminosilicate powder in which the model parameters were determined using experimental flow energy measurements from the FT4 powder rheometer. DEM simulations of the mixing process using the contact model parameters were evaluated against the experimental measurements of the mixing rate. The results demonstrated that whilst the DEM model is capable of reproducing the FT4 flow energy of the powder to an excellent agreement, the simulations of the mixing process produced a qualitative agreement on the trend of the mixing rate in the experiments for both dry and wet powders. The mixing was under-predicted in the simulations, suggesting that flow energy measurements alone may not be sufficient for the optimization of a DEM model of powder mixing.

Abstract:This paper is the second in a series of two that describes the application of discrete element method (DEM) and reduced order modeling to predict the effect of disturbances in the concentration of drug substance at the inlet of a continuous powder mixer on the concentration of the drug substance at the outlet of the mixer. In the companion publication, small-scale material characterization tests, a careful DEM parameter calibration and DEM simulations of the manufacturing process were used to develop a reliable RTD models. In the current work, the same calibration workflow was employed to evaluate the predictive ability of the resulting reduced-order model for an extended design space. DEM simulations were extrapolated using a relay race method and the cumulative RTD was accurately parameterized using the n-CSTR model. By performing experiments and simulations, a calibrated DEM model predicted the response of a continuous powder mixer to step changes in the inlet concentration of an API. Thus, carefully calibrated DEM models was used to guide and reduce experimental work and to establish an adequate control strategy. In addition, a further reduction in the computational effort was obtained by using the relay race method to extrapolate results. The predicted RTD curves were then parameterized to develop reduced order models and used to simulate the process in a matter of seconds. Overall, a control strategy evaluation tool based on high-fidelity DEM simulations was developed using material-sparing small-scale characterization tests.
An Experimentally Validated Coarse-Grain DEM Study of Monodisperse Granular Mixing

Abstract:Considering being applied to industrial applications with billions of particles, the discrete element method (DEM) is still limited by the computational competence of computers, regardless of the advances in computer hardware or parallel programming techniques. In this paper, therefore, a coarse-grain (CG) discrete element method is adopted to deal with the immense number of particles in granular mixing, which has the potential to gain considerable computational advantage for industry scale DEM simulations. The experiment results as well as the simulation results prove the validity of taking place of smaller particles by larger particles with sacrificing a little detail in a drum flow. The simulation results also indicate that the rotation speed of the drum nearly has no effect on the application of the CG-DEM. The mixing tendency as well as the flow pattern of particles in the CG system is quite similar to the original system. In addition, the simulation results of an industrial application show that the CG-DEM is applicable to the double-screw conical mixer. The granular mixing in the double-screw conical mixer can be predicted by the CG-DEM, and the prediction of device power consumption is accurate by correcting the friction coefficient based on the theory of the CG-DEM, and the prediction of the wear rate of the device is broadly accurate after being corrected by a correction factor. Using CG-DEM can efficiently save computational cost with cutting back on the level of detail and accuracy.

Abstract:Discrete element method (DEM) is an effective approach to investigate non-spherical granular systems that are widely encountered in nature, industry, and our daily life. To describe the complex shapes of non-spherical particles, different particle shape representation approaches have been developed. In this paper, comparative study of modeling tablets using multi-sphere approach, multi-super-ellipsoid approach, and polyhedron approach is done to investigate the effect of different approaches. The accuracy of these approaches is testified by experiments and corresponding simulations of the packing, heap forming, and the flow in a rotating drum with two kinds of biconvex tablets firstly. Then the simulations of the flow of two kinds of biconvex tablets in a horizontal rotating drum are carried out by DEM. The simulation results are compared to discuss the effect of different approaches, which include the repose angle, the tablet orientation, the axial movement and mixing, the contact number, the percentage of the tablets reaching the free surface of the granular bed, and the computational efficiency. The results show that the simulations using multi-super-ellipsoid approach and polyhedron approach have some differences with those using multi-sphere approach.

Abstract:In continuous powder handling processes, precise and consistent feeding is crucial for ensuring the quality of the final product. The intermixing effect caused by agitators, which alters the powder's bulk density, flow rate, and flow patterns, plays a significant role in this process, yet it is often overlooked. This study combines discrete element method (DEM) modeling and experiments using a commercial-scale feeder to propose a Digital Twin (DT) framework. The DEM model accurately captures key flow features, such as bypass trajectories, stagnant zones, and preferential flow patterns, while providing quantitative predictions for the feed factor and zones prone to material accumulation. Scenario analysis is performed to identify the most favorable operating ranges of the screw-agitator ratio and screw speed, considering the cohesive properties of the powder. The study demonstrates that powders with poor flow characteristics require tighter operational constraints, as the screw-agitator ratio is susceptible to variations in mass feed rate. This contribution highlights the importance of selecting an appropriate screw-agitator ratio instead of maintaining a fixed value. Properly choosing this ratio helps determine an optimal operation window, which aims to achieve a minimum agitation level needed to induce unhindered flow and reduce variability in the mass flow rate.
Dry Powder Mixing Is Feasible in Continuous Twin Screw Extruder: Towards Lean Extrusion Process for Oral Solid Dosage Manufacturing

Abstract:Using discrete element method (DEM) modeling and near-infrared (NIR) spectroscopy, the feasibility of powder mixing in the initial pre-melting zones of a twin screw extruder using two independent feeders was studied. Previous work in the pharmaceutical and food industry has focused on mixing when materials are melted or on material homogeneity at the extruder's output. Depending on the formulation, ensuring a fully blended formulation prior to melting may be desired. Experiments were conducted using a Coperion ZSK-18 extruder to evaluate if blend uniformity can be achieved by exploring screw configuration, screw speed, and powder feed rate. As powder exited the extruder and deposited on a conveyor belt, an in-line NIR spectrophotometer measured spectra of material. Chemometric-based models predicted unknown concentrations to evaluate if blend uniformity was achieved. Using the EDEM software, Hertz-Mindlin contact model, and dimensions of the extruder, DEM simulations complemented the experimental work. The DEM computational models provided understanding of mixing patterns inside the extruder at particle scale and helped select the screw configuration before doing experimentation. The simulations showed good axial mixing for all the screw configurations studied, while good cross (radial) mixing was only observed for the screw configuration with 90-degree kneading elements. Therefore, the screw configuration with two 90-degree kneading elements was chosen for the experimental study. The RTD profiles when using a screw configuration with only conveying screw elements are comparable to a plug flow reactor (PFR), while the profiles when using kneading elements are more comparable to an ideal continuous stirred tank reactor (CSTR). For the screw configuration with 90 degrees kneading elements, the mean residence time (MRT) decreases with an increase in the screw speed. Experimental NIR spectra showed that concentrations can be predicted with an error of 2%. It was demonstrated that the twin screw extruder can provide proper dry powder mixing of two powder feed streams based on a unit dose scale, enabling continuous powder mixing prior to the melting zone in the extruder for the formulation studied with a cohesive API. This setup may also work for other types of formulations. These studies can help in developing lean hot melt as well as wet extrusion/granulation processes using twin screw extruders for the continuous manufacturing of oral solid dosage products.
Semicontinuous Blending of Pharmaceutical Ingredients and the Impact of Process Parameters on the Blending Performance of an Integrated Feeder Blender Operating Semicontinuously

Abstract:The pharmaceutical industry is looking for new and innovative ways of manufacturing to improve product quality and reduce process complexity. In manufacturing oral solid dosage products, blending is a crucial step in ensuring the homogeneity of active pharmaceutical ingredients (APIs) in the final product. Currently, batch and continuous blending are the two commonly used modes for blending in the industry. However, these methods have limitations in terms of blending time, manual intervention, and flexibility in handling multiple ingredients. To address these limitations, this study aims to explore the feasibility and benefits of using a semicontinuous blending mode in the pharmaceutical industry. A case study is conducted using a binary blend of microcrystalline cellulose and acetaminophen to compare the performance of the semicontinuous mode of blending with the batch and continuous blending modes. The results show that the semicontinuous blending setup can produce blends with good blend uniformity and homogeneity and that the output can be used for both batch and continuous downstream operations. The effect of variation in the three most important process parameters, impeller rotation per minute, blending time, and fill level on the blend uniformity, is also investigated. The semicontinuous blending mode had a higher line rate of 12.5 kg/hour than a similarly sized batch blender at 3.6 kg/hour and less than that of a continuous blender. The benefits of the new blending mode include reduced blending time, minimal manual intervention, flexibility in blending multiple ingredients, easier scale-up, and a smaller footprint. Overall, this study highlights the relative advantages of using this new semicontinuous blending mode in pharmaceutical manufacturing and its potential as a good alternative to the existing blending modes. The semicontinuous mode is well placed between the batch blending and continuous blending mode, with many benefits over the former mode and performance comparable to the latter continuous mode.

Abstract:Process simulation using mathematical modeling tools is becoming more common in the pharmaceutical industry. A mechanistic model is a mathematical modeling tool that can enhance process understanding, reduce experimentation cost and improve product quality. A commonly used mechanistic modeling approach for powder is the discrete element method (DEM). Most pharmaceutical materials have powder or granular material. Therefore, DEM might be widely applied in the pharmaceutical industry. This review focused on the basic elements of DEM and its implementations in pharmaceutical manufacturing simulation. Contact models and input parameters are essential elements in DEM simulation. Contact models computed contact forces acting on the particle-particle and particle-geometry interactions. Input parameters were divided into two types—material properties and interaction parameters. Various calibration methods were presented to define the interaction parameters of pharmaceutical materials. Several applications of DEM simulation in pharmaceutical manufacturing processes, such as milling, blending, granulation and coating, were categorized and summarized. Based on this review, DEM simulation might provide a systematic process understanding and process control to ensure the quality of a drug product.

Investigating the effects of material properties on the mixing dynamics of cohesive particles in a twin screw mixer using a discrete element method approach

Abstract:A discrete element method (DEM) model was developed for a co-rotating twin-screw mixer (TSM) consisting of conveying and kneading elements. This model was used to simulate the mixing of a cohesive binary system of particles (P1 and P2). The particles were calibrated using angle of repose and dynamic yield strength simulations. Properties of P1 were kept constant throughout the study while properties of P2 were varied. The TSM model was used to study the influence of particle size, density, and flowability of P2 on process outputs such as powder holdup, mean residence time (MRT), and degree of mixing along the length of the TSM. All the three particle properties were varied at three-factor levels with flowability ranging from good flow (GF), poor flow (PF), and very very poor flow (VVPF) based on Carr’s classification. The model provides fundamental insights into the differences in mixing in the conveying sections of the mixer for materials with different flowability. Due to the co-rotating nature of the screws, the majority of the material in the conveying sections was carried on one side of the TSM for GF and PF particles, while both the screws carried VVPF particles. The total steady-state holdup and the mean residence time (MRT) of particles inside the TSM were heavily influenced by particle flowability compared to the other two tested particle parameters. The holdup in the conveying compartments ranged from 11%–15% of the total holdup, while the kneading compartment comprised 40% of the total holdup in the mixer. The simulations found that VVPF particles mixed the fastest and that the GF particles had the highest tendency to demix. The model was also used to suggest and test other screw design configurations to improve mixing. A screw design with kneading elements closer to the mixer outlet resulted in better mixing of GF particles. A screw design consisting only of conveying elements achieved the same degree of mixing as the original screw design for VVPF particles.

Abstract:We perform experiments and discrete element simulations on the dosing of cohesive granular materials in a simplified geometry. The setup is a simplified canister box where the powder is dosed out of the box through the action of a constant-pitch screw feeder connected to a motor. A dose consists of a rotation step followed by a period of rest before the next dosage. From the experiments, we report on the operational performance of the dosing process through a variation of dosage time, coil pitch and initial powder mass. We find that the dosed mass shows an increasing linear dependence on the dosage time and rotation speed. In contrast, the mass output from the canister is not directly proportional to an increase/decrease in the number coils. By calibrating the interparticle friction and cohesion, we show that DEM simulation can quantitatively reproduce the experimental findings for smaller masses but also overestimate arching and blockage. With appropriate homogenization tools, further insights into microstructure and macroscopic fields can be obtained. This work shows that particle scaling and the adaptation of particle properties is a viable approach to overcome the untreatable number of particles inherent in experiments with fine, cohesive powders and opens the gateway to simulating their flow in more complex geometries.

Abstract:In this work, a high-fidelity digital twin was developed to support the design and testing of control strategies for drug product manufacturing via direct compression. The high-fidelity digital twin platform was based on typical pharmaceutical equipment, materials, and direct compression continuous processes. The paper describes in detail the material characterization, the Discrete Element Method (DEM) model and the DEM model parameter calibration approach and provides a comparison of the system's response to the experimental results for stepwise changes in the API concentration at the mixer inlet. A calibration method for a cohesive DEM contact model parameter estimation was introduced. To assure a correct prediction for a wide range of processes, the calibration approach contained four characterization experiments using different stress states and different measurement principles, namely the bulk density test, compression with elastic recovery, the shear cell, and the rotating drum. To demonstrate the sensitivity of the DEM contact parameters to the process response, two powder characterization data sets with different powder flowability were applied. The results showed that the calibration method could differentiate between the different material batches of the same blend and that small-scale material characterization tests could be used to predict the residence time distribution in a continuous manufacturing process.

Abstract:Process intensification involves the miniaturization of equipment while retaining process throughput and performance. The pharmaceutical industry can benefit from this approach especially during drug product development, where the availability of active pharmaceutical ingredients (API) is often limited. It reduces the need for process scale up, as equipment used during product development and commercial production is identical. However, applications of process intensification for processing pharmaceutical powders are limited so far. Here we show that semi-continuous mini-blending can be utilized for process intensification of blending of API and excipients. Uniform blending at commercially relevant throughputs was achieved through mini-blends with a volume of less than ten liters. Our results demonstrate that blending speed, cycle time and blender fill level can be optimized without compromising blending performance. Acceptable blend uniformity is obtained over a broad range of operating parameters, by choosing the right excipients. The optimized throughput of the mini-blending process is in line with the desired throughput of a commercial Continuous Direct Compression (CDC) process.

Abstract:Bulk material handling equipment can be numerically studied using the DEM-MBD method, where the particulate dynamics are solved by DEM (Discrete Element Method) and the motion characteristics of the mechanical components are modeled by MBD (Multi-Body Dynamics). In this work, a two-way coupling framework is proposed for the co-simulation of our self-developed DEM solver (DEMSLab) with the commercial MBD software (RecurDyn) using a parallel staggered coupling approach. To verify the proposed coupling method, the simulation result of a torsional spring damper system is compared with the corresponding analytical solution. After that, to assess the feasibility of this method in industrial-scale applications, a series of simulations involving a wheel loader is carried out. Different simulation conditions are adopted to reveal their impact on computational efficiency and simulation accuracy. These conditions include the mesh representation of the equipment geometry, the MBD time step size, and the coupling interval. An improved mesh simplification algorithm is proposed and used to reasonably simplify the surface mesh of the wheel loader, thus lowering the computational intensity of particle-wall interactions in DEM. The comparison results indicate that the computational time can be largely saved while the simulation accuracy remains considerably high. The MBD step size and coupling interval are crucial for numerical stability and should be selected carefully.

Abstract:Abstract One of the key challenges in the implementation of discrete element method (DEM) to model powder’s flow is the appropriate selection of material parameters, where empirical approaches are mostly applied. The aim of this study is to develop an alternative systematic numerical approach that can efficiently and accurately predict the influence of different DEM parameters on various sought macroscopic responses, where, accordingly, model validation based on experimental data is applied. Therefore, design of experiment and multivariate regression analysis, using an optimized quadratic D-optimal design model and new analysis tools, i.e., adjusted response and Pareto graphs, are applied. A special focus is laid on the impact of six DEM microscopic input parameters (i.e., coefficients of static and rolling friction, coefficient of restitution, particle size, Young’s modulus and cohesion energy density) on five macroscopic output responses (i.e., angle of repose, porosity, mass flow rate, translational kinetic energy and computation time) using angle of repose tests applied to free-flowing and cohesive powders. The underlying analyses and tests show, for instance, the substantial impact of the rolling friction coefficient and the minor role of the static friction coefficient or the particle size on the angle of repose in cohesive powders. In addition, in both powders, the porosity parameter is highly influenced by the static and rolling friction coefficients.
