Abstract:Unprecedented levels of nitrogen (N) have entered terrestrial ecosystems over the past century, which substantially influences the carbon (C) exchange between the atmosphere and biosphere. Temperature and moisture are generally regarded as the major controllers over the N effects on ecosystem C uptake and release. N-phosphorous (P) stoichiometry regulates the growth and metabolisms of plants and soil organisms, thereby affecting many ecosystem C processes. However, it remains unclear how the N-induced shift in the plant N:P ratio affects ecosystem production and C fluxes and its relative importance. We conducted a field manipulative experiment with eight N addition levels in a Tibetan alpine steppe and assessed the influences of N on aboveground net primary production (ANPP), gross ecosystem productivity (GEP), ecosystem respiration (ER), and net ecosystem exchange (NEE); we used linear mixed-effects models to further determine the relative contributions of various factors to the N-induced changes in these parameters. Our results showed that the ANPP, GEP, ER, and NEE all exhibited nonlinear responses to increasing N additions. Further analysis demonstrated that the plant N:P ratio played a dominate role in shaping these C exchange processes. There was a positive relationship between the N-induced changes in ANPP (ΔANPP) and the plant N:P ratio (ΔN:P), whereas the ΔGEP, ΔER, and ΔNEE exhibited quadratic correlations with the ΔN:P. In contrast, soil temperature and moisture were only secondary predictors for the changes in ecosystem production and C fluxes along the N addition gradient. These findings highlight the importance of plant N:P ratio in regulating ecosystem C exchange, which is crucial for improving our understanding of C cycles under the scenarios of global N enrichment.

Abstract:Nitrogen (N) and phosphorus (P) have significant effects on soil microbial community diversity, composition, and function. Also, trees of different life stages have different fertilization requirements. In this study, we designed three N additions and three P levels (5 years of experimental treatment) at two Metasequoia glyptostroboides plantations of different ages (young, 6 years old; middle mature, 24 years old) to understand how different addition levels of N and P affect the soil microbiome. Here, the N fertilization of M. glyptostroboides plantation land (5 years of experimental treatment) significantly enriched microbes (e.g., Lysobacter, Luteimonas, and Rhodanobacter) involved in nitrification, denitrification, and P-starvation response regulation, which might further lead to the decreasing in alpha diversity (especially in 6YMP soil). The P addition could impact the genes involved in inorganic P-solubilization and organic P-mineralization by increasing soil AP and TP. Moreover, the functional differences in the soil microbiomes were identified between the 6YMP and 24YMP soil. This study provides valuable information that improves our understanding on the effects of N and P input on the belowground soil microbial community and functional characteristics in plantations of different stand ages.

Long-term Nitrogen Addition Increases Denitrification Potential and Functional Gene Abundance and Changes Denitrifying Communities in Acidic Tea Plantation Soil

Abstract:The response of soil denitrification to nitrogen (N) addition in the acidic and perennial agriculture systems and its underlying mechanisms remain poorly understood. Therefore, a long-term (12 years) field trial was conducted to explore the effects of different N application rates on the soil denitrification potential (DP), functional genes, and denitrifying microbial communities of a tea plantation. The study found that N application to the soil significantly increased the DP and the absolute abundance of denitrifying genes, such as narG, nirK, norB, and nosZ. The diversity of denitrifying communities (genus level) significantly decreased with increasing N rates. Moreover, the denitrifying communities composition significantly differed among the soils with different rates of N fertilization. Further variance partitioning analysis (VPA) revealed that the soil (39.04%) and pruned litter (32.53%) properties largely contributed to the variation in the denitrifying communities. Dissolved organic carbon (DOC) and soil pH, pruned litter's total crude fiber (TCF) content and total polyphenols to total N ratio (TP/TN), and narG and nirK abundance significantly (VIP >1.0) influenced the DP. Finally, partial least squares path modeling (PLS-PM) revealed that N addition indirectly affected the DP by changing specific soil and pruned litter properties and functional gene abundance. Thus, the findings suggest that tea plantation is a major source of N2O emissions that significantly enhance under N application and provide theoretical support for N fertilizer management in an acidic tea plantation system.

Abstract:Increasing evidence has suggested that nitrogen (N) deposition has great effects on forest productivity and nutrient cycling, but how these effects are mediated by soil phosphorus (P) availability and tree species identity remains unclear. Here, a two-year factorial experiment was conducted to examine the short-term effects of N addition (100 kg N ha−1 year−1), P addition (50 kg P ha−1 year−1) and their interactions on tree growth and soil nutrient cycling of a Mongolian pine (Pinus sylvestris var. mongolica) plantation and a neighboring Simon poplar (Populus simonii) plantation in Northeast China. Results showed that N addition promoted tree growth (stem increment), reduced soil microbial biomass carbon (C) and N in the Mongolian pine plantation, but did not affect those in the Simon poplar plantation, and these N addition effects were not interacted by P addition. P addition significantly increased microbial biomass C and N in the Simon poplar plantation. N addition elevated soil phosphatase activities and reduced net N mineralization rate, and these N addition effects were alleviated by simultaneous P addition in both plantations. N addition decreased soil labile inorganic P concentration in both plantations and elevated labile organic P in the Mongolian pine plantation, but only the decrease in labile inorganic P with N addition in the Mongolian pine plantation was counteracted by P addition. N addition significantly reduced soil pH, with a greater extent in the Mongolian pine plantation than in the Simon poplar plantation. Moreover, soil pH response explained most of the microbial and nutrient transformation response. Above results indicated that short-term N addition impacts on tree growth and soil microbial biomass were more strongly influenced by tree species identity than by P addition, while N addition effects on soil N and P transformations were strongly influenced by P addition. Drought resistance of trees and acid buffering capacity of soils are key traits influencing the short-term N addition impacts on the growth of trees and microorganisms in this semiarid region.

Abstract:Increasing nitrogen (N) deposition alters the availability of soil nutrients and is likely to intensify phosphorus (P) limitations, especially in P-limited tropical and subtropical forests. Soil microorganisms play vital roles in carbon (C) and nutrient cycling, but it is unclear whether and how much N and P imbalances affect the soil's microbial metabolism and mechanisms of nutrient limitations. In this study, a 3-year field experiment of N and P addition (control (CK), 100 kg N ha-1 yr-1 (N), 50 kg P ha-1 yr-1 (P), and NP) was set up to analyze the extracellular enzyme activities and stoichiometry characteristics of the top mineral soils in Chinese fir plantations with different stand ages (7, 20, and 33 years old). The results showed that the enzyme activities associated with the acquisition of C (β-1,4-glucosidase (BG) and β-d-cellobiohydrolase (CBH)) and P (acid phosphatases (APs)) in the N treatment were significantly higher than those in the CK treatment. Moreover, vector analysis revealed that both the vector's length and angle increased in stands of all ages, which indicated that N addition aggravated microbial C and P limitations. The P and NP treatments both significantly decreased the activity of AP and the enzymes' N:P ratio, thereby alleviating microbial P limitations, as revealed by the reduction in the vector's angle. Stand age was found to promote all enzymatic activities but had no obvious effects on the limitation of microbial metabolism with or without added nutrients in the soils under Chinese fir. Available N, Olsen-P, and pH were the main drivers of microbial metabolic limitations related to C nutrients. These results provide useful data for understanding the change in soil microbial activity in response to environmental changes, and suggest that P fertilization should be considered for management to improve productivity and C sequestration in Chinese fir plantation in the context of increased deposition of N.

Abstract:To predict the effects of nitrogen deposition on nitrogen-mineralizing enzyme activity and soil microbial community structure in artificial temperate forests in northern China, we studied the soil properties, nitrogen-mineralizing enzyme activity, and microbial community structure in the soil of a Korean pine plantation in which different concentrations (0, 20, 40, 80 kg N ha−1 year−1) of ammonium nitrate were applied for 5 consecutive years. The results showed that nitrogen addition at different concentrations did not significantly affect the soil pH. High nitrogen addition (80 kg N ha−1 year−1) significantly increased the soil organic matter, ammonium nitrogen, and nitrate nitrogen content in the Korean pine plantation, and ammonium nitrogen was the key factor that influenced the soil fungal community structure. The urease activity under the moderate nitrogen addition treatment (40 kg N ha−1 year−1) was significantly lower than that under the control (0 kg N ha−1 year−1), and the protease activity in the three treatments was also significantly lower than that in the control. There was no significant correlation between microbial community structure and the four mineralizing enzymes. After nitrogen addition at different concentrations, the Simpson and Shannon indexes of soil bacteria decreased significantly under low nitrogen addition (20 kg N ha−1 year−1), but the α-diversity index of soil fungi did not show significant differences under nitrogen addition. The microbial community composition was significantly changed by the different treatments. PLS-DA analysis showed that Tardiphaga was an important genus that made the greatest contribution to the differences in bacterial community composition among treatments, as was Taeniolella for fungal community composition. The low level of nitrogen addition inhibited nitrogen mineralization in the Korean pine plantation by reducing the relative abundances of Nitrosomonadaceae and Betaproteobacteriales and by reducing the abundances of symbiotrophic fungi. Berkelbacteria and Polyporales were bacteria and fungi, respectively, that changed significantly under the high nitrogen addition treatment (80 kg N ha−1 year−1). This study provides more data to support predictions of the changes in nitrogen-mineralizing enzyme activity and microbial community structure in artificial temperate forest soils in response to increased nitrogen deposition.

Abstract:Nitrogen (N) deposition and its ecological effects on forest ecosystems have received global attention. Plantations play an important role in mitigating climate change through assimilating atmospheric CO2. However, the mechanisms by which increasing N additions affect net ecosystem production (NEP) of plantations remain poorly understood. A field experiment was initialized in May 2009, which incorporated additions of four rates of N (control (no N addition), low-N (5 g N m(-2) yr(-1)), medium-N (10 g N m(-2) yr(-1)), and high-N (15 g N m(-2) yr(-1))) at the Saihanba Forestry Center, Hebei Province, northern China, a locality that contains the largest area of plantations in China. Net primary production (NPP), soil respiration, and its autotrophic and heterotrophic components were measured. Plant tissue carbon (C) and N concentrations (including foliage, litter, and fine roots), microbial biomass, microbial community composition, extracellular enzyme activities, and soil pH were also measured. N addition significantly increased NPP, which was associated with increased litter N concentrations. Autotrophic respiration (AR) increased but heterotrophic respiration (HR) decreased in the high N compared with the medium N plots, although the HR in high and medium N plots did not significantly differ from that in the control. The increased AR may derive from mycorrhizal respiration and rhizospheric microbial respiration, not live root respiration, because fine root biomass and N concentrations showed no significant differences. Although the HR was significantly suppressed in the high-N plots, soil microbial biomass, composition, or activity of extracellular enzymes were not significantly changed. Reduced pH with fertilization also could not explain the pattern of HR. The reduction of HR may be related to altered microbial C use efficiency. NEP was significantly enhanced by N addition, from 149 to 426.6 g C m(-2) yr(-1). Short-term N addition may significantly enhance the role of plantations as an important C sink.

Abstract:AbstractElevated nitrogen (N) deposition may aggravate phosphorus (P) deficiency in forests in the warm humid regions of China. To our knowledge, the interactive effects of long-term N deposition and P availability on soil microorganisms in tropical replanted forests remain unclear. We conducted an N and P manipulation experiment with four treatments: control, N addition (15 g N m−2·yr−1), P addition (15 g P m−2·yr−1) and N and P addition (15 + 15 g N and P m−2·yr−1, respectively) in disturbed (planted pine forest with recent harvests of understory vegetation and litter) and rehabilitated (planted with pine, but mixed with broadleaf returning by natural succession) forests in southern China. Nitrogen addition did not significantly affect soil microbial biomass, but significantly decreased the abundance of gram-negative bacteria PLFAs in both forest types. Microbial biomass increased significantly after P addition in the disturbed forest but not in the rehabilitated forest. No interactions between N and P additions on soil microorganisms were observed in either forest type. Our results suggest that microbial growth in replanted forests of southern China may be limited by P rather than by N and this P limitation may be greater in disturbed forests.

Abstract:We examined the effects of nitrogen addition (0, 30, 60, and 90 kg N·hm-2·a-1) to soil microbial biomass, enzyme activities, and nutrient contents of the Pinus massoniana-Quercus variabilis mixed plantations in the Three Gorges Reservoir Area, with the aim to provide a theoretical basis for predicting soil carbon dynamics under the background of continuously increasing atmospheric nitrogen deposition in this area. The results showed that nitrogen addition at all levels led to a significant increase of the contents of organic carbon, total nitrogen, microbial biomass carbon (MBC), microbial biomass nitrogen (MBN), and microbial biomass phosphorus (MBP) in the forest soil, while a decrease of soil pH-value, and no significant effect on the total phosphorus content. Nitrogen addition increased the activities of β-1-4 glucosidase (BG), cellobiose hydrolase (CB), acid phosphatase (AP), N-acetylglucosaminosidase (NAG) and peroxidase (POD), while inhibited that of polyphenol oxidase (PPO). There was a significant seasonal variation in soil oxidase activities, in which the peroxidase activity was higher in May and August, and the polyphenol oxidase activity was the highest in August. Soil enzyme activities were significantly correlated with soil moisture and the contents of soil nutrients, MBC, MBN, and MBP. The variation of soil enzyme activities was caused by the comprehensive effects of multiple factors. The redundancy analysis (RDA) showed that the contents of total soil nitrogen and MBC were the main environmental factors driving soil enzyme activities. The continuous increase of atmosphere nitrogen deposition would lead to soil acidification and promote the turnover of soil organic carbon and nutrient cycling in the Pinus massoniana-Quercus variabilis mixed plantations of the study area.

Abstract:Atmospheric nitrogen (N) deposition is among the main manifestations of global change and has profoundly affected forest biogeochemical cycles. However, the threshold of N deposition to soil nutrient contents and enzyme activities has rarely been studied in a forest. In this study, we explored the effects of N deposition on soil nutrients and enzyme activities in a Larix principis-rupprechtii plantation on the northern Yanshan Mountain through multigradient N addition experiments (0, 5, 10, 20, 40, 80, and 160 kg N ha −1 year −1 ) after fertilization for 2 years. Compared with the controls, N addition first led to a decrease in soil NH 4+ -N and NO 3- -N, which then increased significantly. N addition had no significant effects on other soil nutrients. N addition overall elevated soil β-glucosidase activity. N application of >40 kg N ha −1 year −1 significantly reduced soil leucine aminopeptidase activity but had no significant effects on soil acid phosphatase, N-acetyl-β-D-glucosidase, and urease activities. N addition increased the overall stoichiometry ratio of EEA C:N and EEA C:P, but EEA N:P started decreasing after N application of 40 kg N ha −1 year −1 . The ratios of C, N, and P acquisition activities changed from 1:1.2:1 under the control conditions to 1:1.1:1 under the N application of 160 kg N ha −1 year −1 . N addition increased the overall vector length and had no significant effects on the vector angle. Correlation and redundancy analyses revealed that N addition-induced change in available soil N was the main factor affecting soil enzyme activity and stoichiometry. In general, different enzyme activities had different sensitivities to N addition. Moderate N addition or atmospheric N deposition (e.g., <40 kg N ha −1 year −1 ) had beneficial effects on soil nutrient cycling and microorganisms in a Larix principis-rupprechtii plantation.

Abstract:Atmospheric nitrogen (N) deposition and phosphorus (P) addition both can change soil bacterial and fungal community structure with a consequent impact on ecosystem functions. However, which factor plays an important role in regulating responses of bacterial and fungal community to N and P enrichments remains unclear. We conducted a manipulative experiment to simulate N and P inputs (10 g N · m −2 · yr −1 NH 4 NO 3 or 10 g P · m −2 · yr −1 NaH 2 PO 4 ) and compared their effects on soil bacterial and fungal species richness and community composition. The results showed that the addition of N significantly increased NH 4 + and Al 3+ by 99.6% and 57.4%, respectively, and consequently led to a decline in soil pH from 4.18 to 3.75 after a 5-year treatment. P addition increased Al 3+ and available P by 27.0% and 10-fold, respectively, but had no effect on soil pH. N addition significantly decreased bacterial species richness and Shannon index and resulted in a substantial shift of bacterial community composition, whereas P addition did not. Neither N nor P addition changed fungal species richness, Shannon index, and fungal community composition. A structural equation model showed that the shift in bacterial community composition was related to an increase in soil acid cations. The principal component scores of soil nutrients showed a significantly positive relationship with fungal community composition. Our results suggest that N and P additions affect soil bacterial and fungal communities in different ways in subtropical forest. These findings highlight how the diversity of microbial communities of subtropical forest soil will depend on future scenarios of anthropogenic N deposition and P enrichment, with a particular sensitivity of bacterial community to N addition.

Abstract:Nitrogen (N) deposition has a profound influence on forest soil carbon (C) and N pools, but there was no consensus on the responses of different C and N components in different forest types. In this study, a two-year simulated N deposition experiment with four levels of N (NH4NO3)-addition treatments (0, 50, 100, and 150 kg N/hm(2).a) were conducted in Larix gmelinii (LG) and Quercus mongolica (QM) plantation in Northeast China, in order to investigate the C and N pool dynamics under continuously enhanced N deposition. Soil organic carbon (SOC), soil total N (STN) and their active components (readily oxidizable C, ROC; dissolved organic C, DOC; microbial biomass C, MBC, dissolved organic N, DON; microbial biomass N, MBN) of the forest soil were measured monthly from May to October 2017. C and N contents in LG were observed higher than in QM. N addition had no effect on SOC and STN of LG, but significantly increased SOC and STN of QM at low N addition level. Low N addition generally raised active C components (ROC, DOC, and MBC) in both plantations, whereas high N addition did not significantly affect these components, or even decreased ROC in LG soil. Low N addition also increased STN and MBN of QM, while no significant change in STN and MBN of LG was observed. DON was directly affected by N addition and increased significantly with elevated N addition levels. The results indicated that N addition, especially of low rate, might enhance the C sequestration capacity of the forest soils and mitigate climate change.

Abstract:The plant nutrient acquisition strategies are diverse, such as root nutrient acquisition and leaf nutrient resorption, playing important roles in driving soil processes, vegetation performance as well as ecosystem nutrient cycling. However, it is still in a debate whether there is a synergy or tradeoff between above- and below-ground nutrient acquisition strategy under nitrogen (N) and phosphorus (P) addition, or with stand age. Herein, this study investigated the responses of root-soil accumulation factor ( RSAF ) and leaf nutrient resorption efficiency ( NuRE ) to long-term N and P fertilization, and further explored the trade-off between them in Metasequoia glyptostroboides plantations with different stand age. Results showed that under N fertilization in young plantations, leaf N resorption efficiency ( NRE ) increased, and root-soil accumulation factor for P ( RSAF-P ) decreased. For young forests under P fertilization, the NRE increased whereas RSAF-P decreased. For middle-aged forests under P fertilization, the NRE and leaf P resorption efficiency ( PRE ) increased and the RSAF-P decreased. Under P fertilization in young and middle-aged plantations, PRE had a significant positive correlation with RSAF-P . Under N fertilization in young plantations, NRE was significantly positive correlated with root-soil accumulation factor for N ( RSAF-N ). The covariance-based structural equation modeling (CB-SEM) analysis indicated that stand age had positive effects on PRE whether under N or P fertilization, as well as on RSAF-P under N fertilization, whereas had no effects on the NRE or RSAF-N. Overall, our results can shed light on the nutrient acquisition strategies of M. glyptostroboides plantations under future environmental changes and the results could be applied to the nutrient management practices.

Abstract:Elevated atmospheric nitrogen (N) deposition potentially enhances the degree of phosphorus (P) limitation in tropical and subtropical forests. However, it remains elusive that how soil microorganisms deal with the N deposition-enhanced P limitation. We collected soils experienced 9 years of manipulative N input at various rates (0, 40, and 80 kg N ha −1 y −1 ) in an old-growth subtropical natural forest. We measured soil total and available carbon (C), N and P, microbial biomass C, N and P, enzyme activities involved in C, N and P acquisition, microbial community structure, as well as net N and P mineralization. Additionally, we calculated element use efficiency and evaluated microbial homeostasis index. Our findings revealed that N input increased microbial biomass C:P (MB C:P ) and N:P (MB N:P ) ratios. The homeostasis indexes of MB C:P and MB N:P were 0.68 and 0.75, respectively, indicating stoichiometric flexibility. Interestingly, MB C:P and MB N:P correlated significantly with the fungi:bacteria ratio (F:B), not with N and P use efficiencies, net N and P mineralization, and enzyme C:P (EEA C:P ) and N:P (EEA N:P ) ratios. Furthermore, EEA C:P and EEA N:P correlated positively with F:B but did not negatively correlate with the C:P and N:P ratios of available resources and microbial biomass. The effects of N deposition on MB C:P , MB N:P and EEA N:P became insignificant when including F:B as a covariate. These findings suggest that microbes flexibly adapted to the N deposition enhanced P limitation by changing microbial community structure, which not only alter microbial biomass C:N:P stoichiometry, but also the enzyme production strategy. In summary, our research advances our understanding of how soil microorganisms deal with the N deposition-enhanced soil P limitation in subtropical forests.

Abstract:Our findings provide a novel line of evidence that long‐term N deposition did not increase plant P demand in a N‐saturated mature tropical forest. The underlying mechanism is that plants did not increase N uptakes therefore nor increase P uptakes (a stable leaf N: P stoichiometry) in an already N‐saturated ecosystem. Different N addition rates regulated soil P transformation via microbial community transition. These findings help improve the understanding of plant P acquisition and modelling of biogeochemical N–P cycling and vegetation productivity in N‐rich forest ecosystems, particularly considering the fact that chronic N deposition may likely lead to soil N richness and even saturation of many forests in the future. Nitrogen (N) deposition usually increases plant tissue N concentrations and thus phosphorus (P) demand in young and/or N‐limited forests, but the N deposition effect on plant P demand has rarely been assessed in N‐saturated forests. Impacts of 18‐year external N additions (Control: 0, Low N: 50, Moderate N:100 and High N: 150 kg N ha−1 year−1) on leaf P of four plant life‐forms (tree, shrub, herb and liana), P fractions of bulk and rhizosphere soils were examined in a N‐saturated mature tropical forest in southern China. Leaf N, P and N: P ratios of all plant life‐forms remained stable under three N additions. Among soil P fractions, moderate labile organic P increased by 25%–33% across three N additions; and soil total P was increased by 11.76% under Low N, and 8.87% under High N, compared with the control. The PLS‐PM results showed that path coefficient of microbial community to available P significantly increased and of inorganic P to available P significantly decreased under N additions than control. N additions improved soil P availability through microbe‐mediated P transformation: Low N significantly increased soil microbial taxonomic diversity, and a higher microbial diversity could enlarge the sources of nutrient acquisition and stimulate decomposition of recalcitrant organic matters; while High N significantly decreased soil microbial taxonomic diversity, the remaining microorganisms that were screened by N‐rich environments had the characteristics of resisting the N addition effects and maintained efficient P acquisition. Abstract:Nutrient inputs to forest ecosystems significantly influence aboveground plant community structure and ecosystem functioning. However, our knowledge of the influence of nitrogen (N) and/or phosphorus (P) inputs on belowground microbial communities in subtropical forests is still unclear. In this study, we used quantitative polymerase chain reaction and Illumina Miseq sequencing of the bacterial 16S rRNA gene to investigate bacterial abundance, diversity, and community composition in a Chinese fir plantation. The fertilization regimes were as follows: untreated control (CK), P amendment (P), N amendment (N), and N with P amendment (NP). Additions of N decreased soil pH and bacterial 16S rRNA gene abundance by 3.95 (from 4.69 to 3.95) and 3.95 × 109 copies g-1 dry soil (from 9.27 × 109 to 3.95 × 109 g-1 dry soil), respectively. Bacterial richness and diversity decreased with N addition (N and NP) rather than only P input. Proteobacteria, Acidobacteria, and Actinobacteria were the major phylum across all treatments. Nitrogen addition increased the relative abundance of Proteobacteria and Actinobacteria by 42.0 and 10.5%, respectively, while it reduced that of Acidobacteria by 26.5%. Bacterial community structure in the CK and P treatments was different from that in the N and NP treatments upon principle coordinates analysis. Phosphorus addition did not significantly affect soil bacterial communities, and no interactions between N and P inputs on microbial traits were observed. Soil pH and mineral N availability appeared to have a cooperative effect on bacterial abundance and community structure, with soil pH being the key influencing factor by canonical correspondence analysis. These results indicate that inorganic N rather than P fertilization affected both bacterial abundance and community composition in subtropical forests.
