Abstract:Atmospheric nitrogen (N) deposition and phosphorus (P) addition both can change soil bacterial and fungal community structure with a consequent impact on ecosystem functions. However, which factor plays an important role in regulating responses of bacterial and fungal community to N and P enrichments remains unclear. We conducted a manipulative experiment to simulate N and P inputs (10 g N · m −2 · yr −1 NH 4 NO 3 or 10 g P · m −2 · yr −1 NaH 2 PO 4 ) and compared their effects on soil bacterial and fungal species richness and community composition. The results showed that the addition of N significantly increased NH 4 + and Al 3+ by 99.6% and 57.4%, respectively, and consequently led to a decline in soil pH from 4.18 to 3.75 after a 5-year treatment. P addition increased Al 3+ and available P by 27.0% and 10-fold, respectively, but had no effect on soil pH. N addition significantly decreased bacterial species richness and Shannon index and resulted in a substantial shift of bacterial community composition, whereas P addition did not. Neither N nor P addition changed fungal species richness, Shannon index, and fungal community composition. A structural equation model showed that the shift in bacterial community composition was related to an increase in soil acid cations. The principal component scores of soil nutrients showed a significantly positive relationship with fungal community composition. Our results suggest that N and P additions affect soil bacterial and fungal communities in different ways in subtropical forest. These findings highlight how the diversity of microbial communities of subtropical forest soil will depend on future scenarios of anthropogenic N deposition and P enrichment, with a particular sensitivity of bacterial community to N addition.

Abstract:Nutrient addition to forest ecosystems significantly influences belowground microbial diversity, community structure, and ecosystem functioning. Nitrogen (N) addition in forests is common in China, especially in the southeast region. However, the influence of N addition on belowground soil microbial community diversity in subtropical forests remains unclear. In May 2018, we randomly selected 12 experimental plots in a Pinus taiwanensis forest within the Daiyun Mountain Nature Reserve, Fujian Province, China, and subjected them to N addition treatments for one year. We investigated the responses of the soil microbial communities and identified the major elements that influenced microbial community composition in the experimental plots. The present study included three N treatments, i.e., the control (CT), low N addition (LN, 40 kg N ha -1 yr -1 ), and high N addition (HN, 80 kg N ha -1 yr -1 ), and two depths, 0−10 cm (topsoil) and 10−20 cm (subsoil), which were all sampled in the growing season (May) of 2019. Soil microbial diversity and community composition in the topsoil and subsoil were investigated using high-throughput sequencing of bacterial 16S rDNA genes and fungal internal transcribed spacer sequences. According to our results, 1) soil dissolved organic carbon (DOC) significantly decreased after HN addition, and available nitrogen (AN) significantly declined after LN addition, 2) bacterial α-diversity in the subsoil significantly decreased with HN addition, which was affected significantly by the interaction between N addition and soil layer, and 3) soil DOC, rather than pH, was the dominant environmental factor influencing soil bacterial community composition, while AN and MBN were the best predictors of soil fungal community structure dynamics. Moreover, N addition influence both diversity and community composition of soil bacteria more than those of fungi in the subtropical forests. The results of the present study provide further evidence to support shifts in soil microbial community structure in acidic subtropical forests in response to increasing N deposition.

Abstract:Increasing nitrogen (N) deposition alters the availability of soil nutrients and is likely to intensify phosphorus (P) limitations, especially in P-limited tropical and subtropical forests. Soil microorganisms play vital roles in carbon (C) and nutrient cycling, but it is unclear whether and how much N and P imbalances affect the soil's microbial metabolism and mechanisms of nutrient limitations. In this study, a 3-year field experiment of N and P addition (control (CK), 100 kg N ha-1 yr-1 (N), 50 kg P ha-1 yr-1 (P), and NP) was set up to analyze the extracellular enzyme activities and stoichiometry characteristics of the top mineral soils in Chinese fir plantations with different stand ages (7, 20, and 33 years old). The results showed that the enzyme activities associated with the acquisition of C (β-1,4-glucosidase (BG) and β-d-cellobiohydrolase (CBH)) and P (acid phosphatases (APs)) in the N treatment were significantly higher than those in the CK treatment. Moreover, vector analysis revealed that both the vector's length and angle increased in stands of all ages, which indicated that N addition aggravated microbial C and P limitations. The P and NP treatments both significantly decreased the activity of AP and the enzymes' N:P ratio, thereby alleviating microbial P limitations, as revealed by the reduction in the vector's angle. Stand age was found to promote all enzymatic activities but had no obvious effects on the limitation of microbial metabolism with or without added nutrients in the soils under Chinese fir. Available N, Olsen-P, and pH were the main drivers of microbial metabolic limitations related to C nutrients. These results provide useful data for understanding the change in soil microbial activity in response to environmental changes, and suggest that P fertilization should be considered for management to improve productivity and C sequestration in Chinese fir plantation in the context of increased deposition of N.

Abstract:The activity and stoichiometry of soil extracellular enzyme can provide a good indication for changes in soil nutrient availability and microbial demands for nutrients. However, it remains unclear how would nitrogen (N) deposition affect nutrient limitation of microbes in subtropical forest soils. We conducted a 5 years N addition experiment in a subtropical Phyllostachys pubescens forest. The soil nutrients and enzyme activities associated with carbon (C), N, and phosphorus (P) cycles were measured. We also examined the nutrient distribution of microorganisms using enzyme stoichiometry and vector analysis. The results showed that N addition significantly decreased the contents of soil soluble organic C and available P and increased that of available N. Furthermore, N addition significantly decreased β-N-acetyl-glucosaminidase (NAG) activity and NAG/ microbial biomass carbon (MBC), and increased acid phosphatase (ACP) and ACP/MBC. The low and moderate N addition levels significantly increased enzyme C/P, vector length, and vector angle, but significantly decreased enzyme N/P. Results of redundancy analysis showed that the change in soil enzyme activity and enzymatic stoichiometry were mainly driven by soil available P content under N addition. In summary, N addition altered the microbial nutrient acquisition strategy, which increased nutrient allocation to P-acquiring enzyme production but reduced that to N-acquiring enzyme production. Moreover, N addition exacerbated the C and P limitation of soil microorganisms. Appropriate amount of P fertilizer could be applied to improve soil fertility of subtropical P. pubescens forest in the future.

Abstract:Nitrogen deposition affects many ecological processes in terrestrial ecosystem and is the main driving force for global forest carbon sequestration. The interaction between soil bacterial communities and dissolved organic matter (DOM) is the central link of soil carbon and other nutrient cycle. However, how nitrogen (N) addition changes the relationship between bacterial community composition and DOM components (BCC-DOM) and which key bacterial taxa affect DOM components is not clear. Here, we explored the changes in soil bacterial community composition and diversity, DOM quantity and quality, and the relationship between BCC-DOM in a subtropical Pinus taiwanensis forest. The results showed that short-term N addition changes the bacterial community composition and diversity and transitions the surface bacterial community in a eutrophic state. N addition also decreases the dissolved organic carbon (DOC) concentration but increases DOM humification degree and humus-like components (C1 and C2) in the topsoil. Network analysis revealed that the relationships between BCC-DOM are more complex after N addition and the negative associations were dominant. Random forest analysis showed that Chloroflexi (TK10, Ktedonobacteria, and JG37.AG.4) and Nitrospirae (Nitrospira) are key factors for predicting humus-like DOM. Protein-like DOM was mainly related to Firmicutes (Clostridia). Furthermore, structural equation models revealed that soil pH and DOC content do not determine the key bacterial taxa and DOM components, but total nitrogen content significantly affects these parameters. Short-term N addition reduces forest soil carbon storage, but improves the relationships between soil carbon substrates. Further, N addition helps to regulate forest soil bacterial communities and strengthen the interactions with DOM.

Abstract:Atmospheric nitrogen (N) deposition is predicted to increase, especially in the subtropics. However, the responses of soil microorganisms to long-term N addition at the molecular level in N-rich subtropical forests have not been clarified. A long-term nutrient addition experiment was conducted in a subtropical evergreen old-growth forest in China. The four treatments were: control, low N (50 kg N ha −1 a −1 ), high N (100 kg N ha −1 a −1 ), and combined N and phosphorus (P) (100 kg N ha −1 a −1 + 50 kg P ha −1 a −1 ). Metagenomic sequencing characterized diversity and composition of soil microbial communities and used to construct bacterial/fungal co-occurrence networks. Nutrient-treated soils were more acidic and had higher levels of dissolved organic carbon than controls. There were no significant differences in microbial diversity and community composition across treatments. The addition of nutrients increased the abundance of copiotrophic bacteria and potentially beneficial microorganisms (e.g., Gemmatimonadetes, Chaetomium , and Aureobasidium ). Low N addition increased microbiome network connectivity. Three rare fungi were identified as module hubs under nutrient addition, indicating that low abundance fungi were more sensitive to increased nutrients. The results indicate that the overall composition of microbial communities was stable but not static to long-term N addition. Our findings provide new insights that can aid predictions of the response of soil microbial communities to long-term N addition.

Abstract:Elevated atmospheric nitrogen (N) deposition potentially enhances the degree of phosphorus (P) limitation in tropical and subtropical forests. However, it remains elusive that how soil microorganisms deal with the N deposition-enhanced P limitation. We collected soils experienced 9 years of manipulative N input at various rates (0, 40, and 80 kg N ha −1 y −1 ) in an old-growth subtropical natural forest. We measured soil total and available carbon (C), N and P, microbial biomass C, N and P, enzyme activities involved in C, N and P acquisition, microbial community structure, as well as net N and P mineralization. Additionally, we calculated element use efficiency and evaluated microbial homeostasis index. Our findings revealed that N input increased microbial biomass C:P (MB C:P ) and N:P (MB N:P ) ratios. The homeostasis indexes of MB C:P and MB N:P were 0.68 and 0.75, respectively, indicating stoichiometric flexibility. Interestingly, MB C:P and MB N:P correlated significantly with the fungi:bacteria ratio (F:B), not with N and P use efficiencies, net N and P mineralization, and enzyme C:P (EEA C:P ) and N:P (EEA N:P ) ratios. Furthermore, EEA C:P and EEA N:P correlated positively with F:B but did not negatively correlate with the C:P and N:P ratios of available resources and microbial biomass. The effects of N deposition on MB C:P , MB N:P and EEA N:P became insignificant when including F:B as a covariate. These findings suggest that microbes flexibly adapted to the N deposition enhanced P limitation by changing microbial community structure, which not only alter microbial biomass C:N:P stoichiometry, but also the enzyme production strategy. In summary, our research advances our understanding of how soil microorganisms deal with the N deposition-enhanced soil P limitation in subtropical forests.

Abstract:Nutrient inputs to forest ecosystems significantly influence aboveground plant community structure and ecosystem functioning. However, our knowledge of the influence of nitrogen (N) and/or phosphorus (P) inputs on belowground microbial communities in subtropical forests is still unclear. In this study, we used quantitative polymerase chain reaction and Illumina Miseq sequencing of the bacterial 16S rRNA gene to investigate bacterial abundance, diversity, and community composition in a Chinese fir plantation. The fertilization regimes were as follows: untreated control (CK), P amendment (P), N amendment (N), and N with P amendment (NP). Additions of N decreased soil pH and bacterial 16S rRNA gene abundance by 3.95 (from 4.69 to 3.95) and 3.95 × 109 copies g-1 dry soil (from 9.27 × 109 to 3.95 × 109 g-1 dry soil), respectively. Bacterial richness and diversity decreased with N addition (N and NP) rather than only P input. Proteobacteria, Acidobacteria, and Actinobacteria were the major phylum across all treatments. Nitrogen addition increased the relative abundance of Proteobacteria and Actinobacteria by 42.0 and 10.5%, respectively, while it reduced that of Acidobacteria by 26.5%. Bacterial community structure in the CK and P treatments was different from that in the N and NP treatments upon principle coordinates analysis. Phosphorus addition did not significantly affect soil bacterial communities, and no interactions between N and P inputs on microbial traits were observed. Soil pH and mineral N availability appeared to have a cooperative effect on bacterial abundance and community structure, with soil pH being the key influencing factor by canonical correspondence analysis. These results indicate that inorganic N rather than P fertilization affected both bacterial abundance and community composition in subtropical forests.

Abstract:AbstractElevated nitrogen (N) deposition may aggravate phosphorus (P) deficiency in forests in the warm humid regions of China. To our knowledge, the interactive effects of long-term N deposition and P availability on soil microorganisms in tropical replanted forests remain unclear. We conducted an N and P manipulation experiment with four treatments: control, N addition (15 g N m−2·yr−1), P addition (15 g P m−2·yr−1) and N and P addition (15 + 15 g N and P m−2·yr−1, respectively) in disturbed (planted pine forest with recent harvests of understory vegetation and litter) and rehabilitated (planted with pine, but mixed with broadleaf returning by natural succession) forests in southern China. Nitrogen addition did not significantly affect soil microbial biomass, but significantly decreased the abundance of gram-negative bacteria PLFAs in both forest types. Microbial biomass increased significantly after P addition in the disturbed forest but not in the rehabilitated forest. No interactions between N and P additions on soil microorganisms were observed in either forest type. Our results suggest that microbial growth in replanted forests of southern China may be limited by P rather than by N and this P limitation may be greater in disturbed forests.

Abstract:Introduction Fungal communities are key players in the soil biogeochemical processes of forest ecosystems. Although it has been illustrated that soil fungi are susceptible to environmental changes, little is known about the interactive effects of nitrogen (N) and phosphorus (P) enrichment on the soil fungal functional guild. Materials and Methods Here, a 5‐year N and P addition manipulation experiment was conducted in two growing stages (i.e., tree ages 80 years) of a subtropical forest, in which soil fungal diversity and functional guilds were investigated using a multiple‐time sampling strategy. Results Fungal species richness and Shannon indices were significantly different across sampling times. Fungal community composition was significantly affected by both N and P addition but not by forest age. Old forest had a higher fungal network complexity than young forest, and fertilisation decreased soil fungal network complexity and generated looser and more random networks in comparison with the control. The community composition of symbiotrophic and saprotrophic fungi was significantly impacted by N and P addition and correlated with soil available P and total P contents and soil C:P and N:P. Conclusions Our findings highlight that soil P availability has a strong effect on soil fungal communities and their functional guild composition in the subtropical forest.
Long-Term Nitrogen Addition Exerts Minor Effects on Microbial Community but Alters Sensitive Microbial Species in a Subtropical Natural Forest

Abstract:Increasing nitrogen (N) deposition profoundly affects nutrient cycling in soil, thereby influencing forest ecosystem productivity and function. Soil microorganisms are integral in driving nutrient turnover; the changes in microbial communities in response to N deposition and the associated soil nutrient availability, especially of limited nutrients, are far from clear. To explore the changes in soil bacterial and fungal communities and their key environmental drivers under N deposition, we conducted a multilevel field N addition experiment in a Castanopsis carlesii natural forest. Soil properties and bacterial and fungal communities were investigated. There were no significant changes in alpha diversities (presented as Chao1 and Shannon's indexes) and beta diversities of bacteria and fungi among the three treatments. Consistently, the relative abundances of dominant bacterial phyla (i.e., Proteobacteria, Acidobacteria, Actinobacteria, Chloroflexi, and Planctomycetes) and fungal phyla (i.e., Basidiomycota, Ascomycota, and Rozellomycota) did not change following N addition. These results suggest that N deposition did not alter microbial community diversity and structure. In addition, the results of the Mantel test showed that soil pH, NO3−-N, dissolved organic N (DON), and total phosphorus (TP) predominantly influenced the community diversity and structure in bacteria, but not in fungi. Meanwhile, the relative abundance of some sensitive microbial genera, such as Bryobacter, Bradyrhizobium, Sorangium, and Archaeorhizomyces, were significantly decreased. These results indicate a decreased microbial ability for N fixation and P mobilization induced by N deposition. Moreover, there were significant relationships between Bryobacter, Bradyrhizobium, and Archaeorhizomyces and NO3−-N and available P (AP), suggesting that the responses of sensitive microbial groups to N deposition likely depend on the changes in available nutrients in soil, especially limited N or P. Collectively, 6 years of N addition had no significant influence on microbial communities, but some sensitive microbial groups were associated with N or P turnover. This finding emphasizes the critical roles of sensitive microbial species in meditating limited nutrient cycling in soil under climate change.

Abstract:Soil cyanobacteria in tropical forests is understudied despite its important role in soil biochemical process and plant growth. Under a nitrogen (N) deposition background in tropical forests, it is important to learn how soil cyanobacterial communities respond to N deposition and whether phosphorus (P) mediated this response. A fully two-factor (N and P additions) factorial design with four blocks (replicates), each including a 12 × 12 m plot per treatment (Control, +N, +P, and +NP) were established in a tropical secondary forest in 2009. In July of 2022, soil cyanobacteria at 0–10 cm and 10–20 cm depths in the experimental site were collected and analyzed using a metagenomic method. The impact of N and P additions on soil cyanobacteria remained consistent across the different soil depths, even though there was a significant contrast between the two layers. The effect of N addition on soil cyanobacteria did not significantly interact with P addition. N addition increased soil N availability and decreased soil pH but did not significantly affect the soil cyanobacterial community. In contrast, P addition increased soil P availability and soil pH, but decreased soil N availability and substantially changed the soil cyanobacterial community. P addition significantly decreased the abundance of soil cyanobacteria, especially abundant ones. P addition also increased cyanobacterial species richness and Shannon's diversity, which might be explained by the decline in dominant species and the emergence of new species as nestedness and indicator species analyses suggest. We concluded that (1) soil cyanobacteria in tropical forests exhibits a greater sensitivity to elevated P availability compared to N; (2) an increase in soil P supply may mitigate the advantage held by dominant species, thus facilitating the growth of other species and leading to alterations in the soil cyanobacterial community. This study improves our understanding on how soil cyanobacterial communities in tropical forest responds to N and P addition.

Abstract:Rhizosphere is a hot zone formed by root–microbial interaction, and microbial activities in this zone differ from those in bulk soil. Nitrogen (N) and phosphorus (P) inputs are able to change forest soil nutrient availability, affecting microbial communities and extracellular enzyme secretion. However, the impact of N and P additions on the structure and functions of rhizosphere microbial community in Chinese fir (Cunninghamia lanceolata (Lamb.) Hook) plantations are not yet clear. To reveal the impact of short-term N and P inputs on microbial community structure and functions in rhizosphere soil, soil physicochemical properties, phospholipid fatty acids, and seven hydrolytic enzyme activities were measured in Chinese fir rhizosphere soil after one year of nutrient addition. N addition reduced the rhizosphere's pH and increased ammonium N, but the rhizosphere's available N (AN) initially wentdown and then up along the P-addition gradient. The rhizosphere fungi:bacteria ratio showed a decline after N addition, while a concave peak change occurred as rhizosphere AN under P addition. Moreover, rhizosphere extracellular enzyme activities and microbial C limitation climbed markedly with N addition rates, while this also showed an obviously unimodal pattern along the P-addition gradient. P addition did not alleviate rhizosphere microbial P limitation. Our findings suggest inconsistent responses of rhizosphere microorganisms of Chinese fir soil to N and P additions. Rhizosphere N availability can regulate microbial community structure and extracellular enzymes by influencing microbial C limitation. The study provides more knowledge on microbial activities in rhizosphere soil of subtropical forests under global changes.

Phosphorus Addition Decreases Soil Fungal Richness and Alters Fungal Guilds in Two Tropical Forests

Abstract:Soil microbes regulate key functions of terrestrial ecosystems, but the effects and underlying mechanisms of elevated nutrient inputs on soil microbial diversity in tropical forests remain unclear. We evaluated the effects of seven-year nitrogen (N) and phosphorus (P) additions on soil microbial diversity in two tropical montane rainforests (P-limited primary forest and N-limited secondary forest). The results showed that soil bacterial richness was not sensitive to N or P addition, whereas soil fungal richness decreased under P and combined N and P additions. Furthermore, P inputs shifted soil fungal community composition: the relative abundance of mutualistic fungi increased and that of saprotrophic fungi decreased. The underlying mechanisms of these effects differed in the two forests. In the P-limited primary forest, the reduction in soil fungal richness was mainly related to a negative microbial interaction between microbial species and a stimulation in plant productivity. In the N-limited secondary forest, P inputs increased soil N and P imbalance, which caused a decline in the fungal richness. Overall, anthropogenic P enrichment could reduce soil fungal richness and alter fungal functional guilds in tropical forests, which will have divergent consequences on plant productivity and microbial functioning.

Abstract:Increased soil nitrogen (N) from atmospheric N deposition could change microbial communities and functions. However, the underlying mechanisms and whether soil phosphorus (P) status are responsible for these changes still have not been well explained. Here, we investigated the effects of N and P additions on soil bacterial and fungal communities and predicted their functional compositions in a temperate forest. We found that N addition significantly decreased soil bacterial diversity in the organic (O) horizon, but tended to increase bacterial diversity in the mineral (A) horizon soil. P addition alone did not significantly change soil bacterial diversity but mitigated the negative effect of N addition on bacterial diversity in the O horizon. Neither N addition nor P addition significantly influenced soil fungal diversity. Changes in soil microbial community composition under N and P additions were mainly due to the shifts in soil pH and NO3− contents. N addition can affect bacterial functional potentials, such as ureolysis, N fixation, respiration, decomposition of organic matter processes, and fungal guilds, such as pathogen, saprotroph, and mycorrhizal fungi, by which more C probably was lost in O horizon soil under increased N deposition. However, P addition can alleviate or switch the effects of increased N deposition on the microbial functional potentials in O horizon soil and may even be a benefit for more C sequestration in A horizon soil. Our results highlight the different responses of microorganisms to N and P additions between O and A horizons and provides an important insight for predicting the changes in forest C storage status under increasing N deposition in the future.

Abstract:Biological nitrogen (N) fixation is an important source of N in terrestrial ecosystems, but the response of soil microbial N fixation rate to N deposition in different forest ecosystems still remains uncertain. We conducted a field N addition experiment to simulate atmosphere N deposition in subtropical Pinus taiwanensis and Castanopsis faberi forests. We set up three levels of nitrogen addition using urea as the N source: 0 (control), 40 (low N), and 80 g N·hm-2·a-1(high N) to examine the chemical properties, microbial biomass C, enzyme activities, and nifH gene copies of top soils (0-10 cm). We also measured the microbial N fixation rate using the 15N labeling method. Results showed that N addition significantly reduced the soil microbial N fixation rate in the P. taiwanensis and C. faberi forests by 29%-33% and 10%-18%, respectively. Nitrogen addition significantly reduced N-acquiring enzyme (i.e., β-1, 4-N-acetylglucosaminidase) activity and nifH gene copies in both forest soils. There was a significant positive correlation between the microbial N fixation rate and soil dissolved organic C content in the P. taiwanensis forest, but a significant negative relationship between the rate of soil microbial nitrogen fixation and NH4+-N content in the C. faberi forest. Overall, soil microbial N fixation function in the P. taiwanensis forest was more sensitive to N addition than that in the C. faberi forest, and the factors affecting microbial N fixation varied between the two forest soils. The study could provide insights into the effects of N addition on biological N fixation in forest ecosystems, and a theoretical basis for forest management.

Abstract:Rapid increase of nitrogen (N) deposition could alter nutrient availability, leading to changes in soil microbial processes and ecosystem carbon and nutrient cycling. However, the effects of N deposition on soil microbes remain elusive in the tropical rainforests in Asia. Here, we conducted a 3-year N addition experiment with four treatments (0, 20, 50, and 100 kg N ha−1 year−1) in a primary and secondary tropical montane forest in Hainan Island, China, to explore the effects of elevated N availability on soil microbial community composition. We also conducted a phosphorus (P) treatment (50 kg P ha−1 year−1) and a N + P treatment (50 kg N ha−1 year−1 + 50 kg P ha−1 year−1) to examine potential P limitation driven by N deposition in highly weathered tropical soils, using a bar-coded pyrosequencing technique. The composition of soil bacterial communities differed dramatically between the primary and secondary forests, but not significant dissimilarity among the fertilization treatments. The community composition, phylogenetic diversity and phylotype richness were significantly correlated with soil pH, total organic C (TOC), and total N (TN), respectively. There were significant differences between the primary and secondary forest in pH, TOC, and TN, but not among the fertilization treatments. These results suggest that differences in soil nutrient status between the primary and secondary forests due to different successional stages rather than chronic N fertilization may be the major factor affecting soil bacterial composition in the tropical montane rainforests.

Abstract:AimsLong-term nitrogen (N) fertilization has been shown to profoundly affect the soil microorganisms and strongly result in several imbalances in element concentrations. The objective of this study was to examine links among the soil microorganisms, enzyme activities, and soil carbon (C), N, and phosphorus (P) stoichiometry in a subtropical Chinese fir (Cunninghamia lanceolata (Lamb.) Hook) plantation after continuous N fertilization for 13 years.MethodsThis study was performed in 25-year-old fir plantation along a fertilization gradient (0, 60, 120, and 240 kg N ha−1 yr.−1), designated as N0, N1, N2, and N3, respectively. Soil microbial properties, including the microbial community composition, as revealed by phospholipid fatty acids (PLFAs), and soil enzyme activities (i.e., sucrase, urease and catalase) were measured, and soil elemental stoichiometry was calculated based on soil C, N, and P concentrations. A redundancy analysis (RDA) was conducted to determine the relationship between soil C:N:P stoichiometry and soil microbial properties.ResultsCompared with the control (N0), N fertilization decreased the total PLFAs (−12.20%), bacteria (−14.33%), fungi (−12.97%), and actinomycetes (−17.11%) on average. Sucrase, urease and catalase activities were enhanced by low and middle levels of N (N1 and N2), but not with high level of N (N3). Long-term N fertilization decreased soil pH, C to N ratio (C/N), and C to P ratio (C/P), while increased soil C, N and N to P ratio (N/P). The RDA identified the first two axes of soil stoichiometry variation that explained 20.4% of the variation at the soil depth of 0–20 cm, 28.6% at 20–40 cm and 49.9% at 40–60 cm in PLFAs biomarkers and enzymes, respectively. Significant correlations between soil stoichiometry (soil N/P and C/P ratio) and soil microbial properties were found in this study.ConclusionsThese observations suggested that long-term N fertilization influenced soil microbial community composition and enzyme activities by changing the soil C/P and N/P ratios. Future studies are needed to consider the coupling relationships between soil microbial community composition, enzyme activities and elemental stoichiometry in different ecosystems under future climatic change.

Abstract:Large amounts of nitrogen (N) are loaded in tropical and subtropical regions due to anthropogenic activities. However, the long-term effect of this deposition on soil biodiversity and its ecological functions remains poorly understood. In this study, we established a 10-year-long field experiment examining the effect of simulated N deposition on soil bacterial diversity and litter decomposition processes in a subtropical Chinese fir forest. The N levels were 0 (control), 6, 12, and 24 g m(-2) yr(-1). Soil bacterial diversity was measured by high throughput sequencing analysis, while litter decomposition was investigated by a microcosm experiment with a reciprocal soil and litter design. Our long-term experimental results demonstrated that N addition significantly changed soil pH, NH4+ -N, available P and soil moisture; reduced soil bacterial diversity; and changed soil microbial community composition at the phyla and class levels. Meanwhile, the rate of litter decomposition decreased in treatments with high doses of N addition. The decreased soil pH and changes in understory plants were significantly correlated with the altered soil microbial diversity and community composition according to the structure equation model analysis. Our results showed that changes in soil microbial community composition (but not soil microbial diversity) mainly contributed to the decline in litter decomposition in higher doses of N addition. Our findings conclude that global change phenomena such as N deposition would depress both aboveand below-ground biodiversity and would strongly affect ecosystem functions and processes in subtropical forests.
