Abstract:Introduction The production of Eucalyptus , a principal economic tree genus in China, is faced with challenges related to soil phosphorus (P) limitations. In this study, we explore variations in phosphorus content, storage, and transformation in Eucalyptus forests. We hypothesize that mixed forests augment soil aggregate stability and P content and that microaggregates are pivotal in determining P differences between mixed and pure forests. Additionally, we posit that mixed forests foster P transformation, enhancing its efficacy in the soil. Current research on the distribution and transformation of soil total P (TP) and P fractions at the soil aggregate level is limited. Methods In this study, we selected soil from a Eucalyptus - Mytilaria laosensis Lecomte mixed forest, Eucalyptus - Erythrophleum fordii Oliv mixed forest, and pure Eucalyptus forest in Chongzuo County, Guangxi, China, as the research objects. Using a dry-sieving method, we divided the soil collected in situ from the 0–40 cm layer into aggregates of >2, 1–2, 0.25–1, and <0.25 mm particle sizes, measured the TP and P fractions (resin-extractable inorganic P, bicarbonate-extractable inorganic P, bicarbonate-extractable organic P, sodium hydroxide-extractable inorganic P, sodium hydroxide-extractable organic P, dilute hydrochloric acid-extractable P, concentrated hydrochloric acid extractable inorganic P, concentrated hydrochloric acid-extractable organic P and residue-P) in different aggregates, and used redundancy analysis and PLS SEM to reveal key factors affecting soil P accumulation and transformation. Results The results showed that compared to pure Eucalyptus forests, mixed Eucalyptus forests significantly enhanced the stability of soil aggregates and the content and storage of phosphorus, especially the Eucalyptus-Mytilaria laosensis mixed forest. The content of total soil phosphorus and its fractions decreased with increasing aggregate particle size, while the opposite trend was observed for stored P, with aggregates <0.25 mm being the main fraction influencing soil phosphorus accumulation. The transformation process of P fractions was primarily constrained by dissolution rates, mineralization rates, biological activity, including the action of microbes, fungi, and plant–root interactions, and other factors. Discussion Mixed forests increased the transformation of phosphorus in soil aggregates, effectivel enhancing the availability of soil phosphorus. In summary, this study provides important evidence for the systematic management of subtropical artificia Eucalyptus forests and the sustainable utilization of soil resources.

Abstract:Soil extracellular enzymatic activity (EEA) and extracellular enzymatic stoichiometry (EES) within aggregates indicate variations in soil-nutrient effectiveness and the nutrient requirements of microorganisms. However, the responses of soil EEA and EES after introducing N2-fixing tree species into Eucalyptus plantations are poorly understood. Therefore, we examined soils from a 15-year-old pure Eucalyptus urophylla plantation (PP) and mixed E. urophylla and Acacia mangium plantation (MP) based on the theory of EEA and EES at the aggregate scale. Aggregates were separated into four fractions using a dry-sieving procedure: >2, 1–2, 0.25–1, and <0.25 mm. We measured the EEA of soil carbon (C)-, nitrogen (N)-, and phosphorus (P)-acquiring enzymes, and examined potential factors (soil physicochemical properties, microbial biomass, and litterfall [LF]) that may influence EEA and EES. Significantly higher (p < 0.05) EEA levels in all aggregates were found in MP than in PP. The average natural logarithmic ratio of C-, N-, and P-acquiring enzyme activities in our study was 1.44:1.21:1, which deviated from the global mean ratio of 1:1:1 and implied that soil microbes were limited by C and N. Moreover, the enzyme C:N ratio (EC:N), C:P ratio (EC:P), and vector length (VL) were markedly lower (p < 0.05) in bulk soil and most aggregates in MP compared to PP, suggesting that C limitation was more serious in PP than in MP. Furthermore, while the vector angle (VA) of bulk soil and four aggregate sizes were all <45° in both the PP and the MP, they were markedly higher (p 2 mm aggregate in MP than in PP. This indicated that mixing N2-fixing species with Eucalyptus alleviated but did not eliminate N limitation. Our study also found that nitrate nitrogen (NO3−-N), total nitrogen (TN), and microbial biomass C:P ratio (MBC:MBP) were the main factors driving changes in EEA, while LF was a key factor controlling EES (p < 0.05). Overall, introducing N2-fixing species into the Eucalyptus plantation alleviated but did not eliminate C and N limitation. The results provide specific recommendations for soil-nutrient management in Eucalyptus plantations in subtropical China.

Abstract:Increasing nitrogen (N) deposition alters the availability of soil nutrients and is likely to intensify phosphorus (P) limitations, especially in P-limited tropical and subtropical forests. Soil microorganisms play vital roles in carbon (C) and nutrient cycling, but it is unclear whether and how much N and P imbalances affect the soil's microbial metabolism and mechanisms of nutrient limitations. In this study, a 3-year field experiment of N and P addition (control (CK), 100 kg N ha-1 yr-1 (N), 50 kg P ha-1 yr-1 (P), and NP) was set up to analyze the extracellular enzyme activities and stoichiometry characteristics of the top mineral soils in Chinese fir plantations with different stand ages (7, 20, and 33 years old). The results showed that the enzyme activities associated with the acquisition of C (β-1,4-glucosidase (BG) and β-d-cellobiohydrolase (CBH)) and P (acid phosphatases (APs)) in the N treatment were significantly higher than those in the CK treatment. Moreover, vector analysis revealed that both the vector's length and angle increased in stands of all ages, which indicated that N addition aggravated microbial C and P limitations. The P and NP treatments both significantly decreased the activity of AP and the enzymes' N:P ratio, thereby alleviating microbial P limitations, as revealed by the reduction in the vector's angle. Stand age was found to promote all enzymatic activities but had no obvious effects on the limitation of microbial metabolism with or without added nutrients in the soils under Chinese fir. Available N, Olsen-P, and pH were the main drivers of microbial metabolic limitations related to C nutrients. These results provide useful data for understanding the change in soil microbial activity in response to environmental changes, and suggest that P fertilization should be considered for management to improve productivity and C sequestration in Chinese fir plantation in the context of increased deposition of N.

Abstract:Background and aimsMicrobial communities and their associated enzyme activities affect the quantity and chemical quality of carbon in soil. We aimed to evaluate the biochemical mechanisms underlying how N2-fixing species influences soil organic carbon chemical composition through soil microbial functional groups and enzyme activities.MethodsWe examined the effects of N2-fixing species mixed with Eucalyptus on soil carbon storage, and the chemical composition of an 8-year-old pure Eucalyptus urophylla plantation (PP) and a mixed E.urophylla and Acacia mangium plantation (MP).ResultsThe soil carbon stock and recalcitrant carbon chemical component significantly increased in surface soil in MP. The total PLFAs and bacterial PLFAs increased by 29.1% and 27.0% in cool-dry season, while in the warm-wet season, the total PLFAs and bacterial PLFAs increased by 13.1% and 27.3%, respectively. However, the fungal PLFAs decreased significantly in warm-wet season in MP. The total activity of the cellulose-degrading enzyme β-glucosidase was significantly greater with mixed N2-fixing species in both dry-cool and wet-warm season. The increase in the Alk-C/O-Alk-C ratio and SOC was strongly associated with both C-acquisition activity and bacterial community.ConclusionsOur findings highlight the importance of N2-fixing species in regulating both soil microbial communities and their functioning in association with soil extracellular enzyme activities, which contribute to the increased soil carbon storage and recalcitrant carbon composition in Eucalyptus plantations.

Responses of Soil CO2 Emission and Tree Productivity to Nitrogen and Phosphorus Additions in a Nitrogen-Rich Subtropical Chinese Fir Plantation

Abstract:Nitrogen (N) and phosphorus (P) nutrients have been regularly applied to improve productivity in intensively managed and short-rotation forest plantations in subtropical China. Under the constraint of the national policy of "carbon neutrality", it is necessary to determine the rational fertilization options by considering both forest productivity and soil CO2 emissions. Past worldwide studies have shown varied responses of forest soil heterotrophic respiration and CO2 emissions to N and P additions. This study designed six treatments with N additions (high level: 15 g N/m2, HN), P (low: 5 g P/m2, LP; high: 15 g P/m2, HP), and their interactions (HNLP and HNHP) to explore the effects of N and P additions on soil CO2 emissions in a P-limited and N-rich Chinese fir plantation (Cunninghamia lanceolata), and we identified the underlying controls using the structural equation model (SEM). The results indicated that LP, HNLP, and HNHP treatments significantly increased soil CO2 emissions in the first four months after treatment and the effects leveled since then. The balance between N and P inputs affected the responses of soil CO2 emissions to P additions. A low P addition significantly increased tree productivity, but the promoting effect gradually declined and was no longer significant after 3 years. Other treatments did not significantly affect tree productivity. The SEM analysis revealed that the promoting effects of P additions on CO2 emission were mainly due to their effects on increasing soil water-soluble organic carbon content and reducing microbial biomass nitrogen content. Considering both soil respiration and tree productivity, this study suggested that LP treatment can effectively balance the N and P nutrients and, in the meantime, maintain relatively low greenhouse gas emissions; thus a low P application level is suggested for N-rich Chinese fir plantations.

Abstract:Elevated atmospheric nitrogen (N) deposition potentially enhances the degree of phosphorus (P) limitation in tropical and subtropical forests. However, it remains elusive that how soil microorganisms deal with the N deposition-enhanced P limitation. We collected soils experienced 9 years of manipulative N input at various rates (0, 40, and 80 kg N ha −1 y −1 ) in an old-growth subtropical natural forest. We measured soil total and available carbon (C), N and P, microbial biomass C, N and P, enzyme activities involved in C, N and P acquisition, microbial community structure, as well as net N and P mineralization. Additionally, we calculated element use efficiency and evaluated microbial homeostasis index. Our findings revealed that N input increased microbial biomass C:P (MB C:P ) and N:P (MB N:P ) ratios. The homeostasis indexes of MB C:P and MB N:P were 0.68 and 0.75, respectively, indicating stoichiometric flexibility. Interestingly, MB C:P and MB N:P correlated significantly with the fungi:bacteria ratio (F:B), not with N and P use efficiencies, net N and P mineralization, and enzyme C:P (EEA C:P ) and N:P (EEA N:P ) ratios. Furthermore, EEA C:P and EEA N:P correlated positively with F:B but did not negatively correlate with the C:P and N:P ratios of available resources and microbial biomass. The effects of N deposition on MB C:P , MB N:P and EEA N:P became insignificant when including F:B as a covariate. These findings suggest that microbes flexibly adapted to the N deposition enhanced P limitation by changing microbial community structure, which not only alter microbial biomass C:N:P stoichiometry, but also the enzyme production strategy. In summary, our research advances our understanding of how soil microorganisms deal with the N deposition-enhanced soil P limitation in subtropical forests.

Phosphorus uptake in four tree species under nitrogen addition in subtropical China

Abstract:Atmospheric N deposition is a serious problem in subtropical China where N is present in large amounts but P is deficient. Several studies hypothesized that N2 fixers can overcome phosphorus limitation by trading fixed N2 for soil phosphorus. In order to know whether N2 fixers could invest fixed N2 in extracellular phosphatase production and could stimulate arbuscular mycorrhizal fungi (AMF) to acquire soil P in N-rich subtropical China, an open-air greenhouse experiment was carried out. Two N2 fixers (Acacia mangium and Ormosia pinnata) and two non-N2 fixers (Schima superba and Pinus massoniana) were exposed to three levels of N addition: 5.6 kg ha-1 a-1 (ambient N), 15.6 kg ha-1 a-1 (middle N), and 20.6 kg ha-1 a-1 (high N). We found that the capacity of plants to acquire soil P in N-rich subtropical China is species specific. The higher P uptake rates were found for N2 fixers than non-N2 fixers under N addition, which were related to the greater soil APA and higher AMF (p < 0.01) in the soil of N2 fixers. However, with time, high N addition decreased more significant quantities of soil microbial phospholipid fatty acids (PLFAs) in the soil of N2 fixers than that of non-N2 fixers (p < 0.05). We conclude that N2 fixers have higher P uptake capacity than non-N2 fixers under ambient N deposition in subtropical China. However, continuing N deposition in the future might affect P uptake ability of N2 fixers as high N addition would decrease soil microbial PLFAs of N2 fixers.
The responses of carbon- and nitrogen-acquiring enzymes to nitrogen and phosphorus additions in two plantations in southern China

Abstract:In tropical plantations, nutrients such as nitrogen (N) or phosphorus (P) are often applied as management practices. However, the effects of such nutrient additions on topsoil C- and N-acquiring enzymes activities are unclear. In this study, the impacts of fertilization on β-1,4-glucosidase (BG), β-D-cellobiosidase (CBH), β-1,4-xylosidase (BX), β-1,4-N-acetyl-glucosaminidase (NAG), and leucine amino peptidase (LAP) enzymes activities from topsoil and litter layer of two tropical plantations (Acacia auriculiformis and Eucalyptus urophylla) were measured. The results showed that N addition had neutral impact on topsoil enzymes, while significantly elevating the activities of BG, CBH, BX, and NAG in the litter layer. P fertilization had no impacts except for an elevation of NAG in litter sample. There was no interactions found between N and P additions on these enzyme activities. The clearer impacts of N over P fertilization were unexpected because that the study site receives a high rate of atmospheric N deposition, and has low soil P availability. The impact of P fertilization on hydrolytic enzyme activities may be less important compared with that of N.

Abstract:Agricultural fertilization may change processes of elemental biogeochemical cycles and alter the ecological function. Ecoenzymatic stoichiometric feature plays a critical role in global soil carbon (C) metabolism, driving element cycles, and mediating atmospheric composition in response to agricultural nutrient management. Despite the importance on crop growth, the role of phosphorous (P) in compliance with eco-stoichiometry on soil C and nitrogen (N) sequestration in the paddy field remains poorly understood in the context of climate change. Here, we collected soil samples from a field experiment after 6 years of chemical P application at a gradient of 0 (P-0), 30 (P-30), 60 (P-60), and 90 (P-90) kg ha(-1) in order to evaluate the role of P on stoichiometric properties in terms of soil chemical, microbial biomass, and eco-enzyme activities as well as greenhouse gas (GHG: CO2, N2O and CH4) emissions. Continuous P input increased soil total organic C and N by 1.3-9.2% and 3%-13%, respectively. P input induced C and N limitations as indicated by the decreased ratio of C:P and N:P in the soil and microbial biomass. A synergistic mechanism among the ecoenzymatic stoichiometry, which regulated the ecological function of microbial C and N acquisition and were stoichiometrically related to P input, stimulated soil C and N sequestration in the paddy field. The lower emissions of N2O and CH4 under the higher P application (P-60 and P-90) in July and the insignificant difference in N2O emission in August compared to P-30; however, continuous P input enhanced CO2 fluxes for both samplings. There is a technical conflict for simultaneously regulating three types of GHGs in terms of the eco-stoichiometry mechanism under P fertilization. Thus, it is recommended that the P input in paddy fields not exceed 60 kg ha(-1) may maximize soil C sequestration, minimize P export, and guarantee grain yields.
Impact of native tree species introduction on soil nutrient and bacterial community in Eucalyptus plantations

Abstract:The introduction of native tree species can enhance soil ecological functions and mitigate the decline in soil status triggered by the high-intensity monoculture of Eucalyptus plantations, but the specific impact is still unclear. In this study, differences in the structural and function for soil bacterial communities were investigated between evergreen broad-leaved secondary forests, Eucalyptus pure plantations, and mixed plantations of Eucalyptus with two native species in Guangxi: Manglietia glauca and Mytilaria laosensis , respectively. Additionally, soil physicochemical properties and enzyme activities were determined. The results showed that pH, soil sucrase and peroxidase activities, and bacterial alpha-diversity indices were significantly higher in Eucalyptus mixed plantations with M. glauca and M. laosensis , compared to Eucalyptus pure plantations. Bacterial biomarkers ( Burkholderiales, Bradyrhizobium, and Actinobacteria ) with nitrogen fixation and phosphate solubilization functions were enriched in Eucalyptus with M. glauca and M. laosensis . Redundancy analysis showed that pH, effective boron (AB), and soil moisture were the main soil environmental factors driving changes in the structure of the soil bacterial community. Thus, we concluded that mixed plantations of Eucalyptus with M. glauca or M. laosensis have positive effects on mitigating soil acidification, increasing soil enzymatic activity, and improving soil ecological functions, which provides useful information for the development of sustainable management of Eucalyptus plantations.

Differential mechanisms underlying responses of soil bacterial and fungal communities to nitrogen and phosphorus inputs in a subtropical forest

Abstract:Atmospheric nitrogen (N) deposition and phosphorus (P) addition both can change soil bacterial and fungal community structure with a consequent impact on ecosystem functions. However, which factor plays an important role in regulating responses of bacterial and fungal community to N and P enrichments remains unclear. We conducted a manipulative experiment to simulate N and P inputs (10 g N · m −2 · yr −1 NH 4 NO 3 or 10 g P · m −2 · yr −1 NaH 2 PO 4 ) and compared their effects on soil bacterial and fungal species richness and community composition. The results showed that the addition of N significantly increased NH 4 + and Al 3+ by 99.6% and 57.4%, respectively, and consequently led to a decline in soil pH from 4.18 to 3.75 after a 5-year treatment. P addition increased Al 3+ and available P by 27.0% and 10-fold, respectively, but had no effect on soil pH. N addition significantly decreased bacterial species richness and Shannon index and resulted in a substantial shift of bacterial community composition, whereas P addition did not. Neither N nor P addition changed fungal species richness, Shannon index, and fungal community composition. A structural equation model showed that the shift in bacterial community composition was related to an increase in soil acid cations. The principal component scores of soil nutrients showed a significantly positive relationship with fungal community composition. Our results suggest that N and P additions affect soil bacterial and fungal communities in different ways in subtropical forest. These findings highlight how the diversity of microbial communities of subtropical forest soil will depend on future scenarios of anthropogenic N deposition and P enrichment, with a particular sensitivity of bacterial community to N addition.

Abstract:Increasing evidence has suggested that nitrogen (N) deposition has great effects on forest productivity and nutrient cycling, but how these effects are mediated by soil phosphorus (P) availability and tree species identity remains unclear. Here, a two-year factorial experiment was conducted to examine the short-term effects of N addition (100 kg N ha−1 year−1), P addition (50 kg P ha−1 year−1) and their interactions on tree growth and soil nutrient cycling of a Mongolian pine (Pinus sylvestris var. mongolica) plantation and a neighboring Simon poplar (Populus simonii) plantation in Northeast China. Results showed that N addition promoted tree growth (stem increment), reduced soil microbial biomass carbon (C) and N in the Mongolian pine plantation, but did not affect those in the Simon poplar plantation, and these N addition effects were not interacted by P addition. P addition significantly increased microbial biomass C and N in the Simon poplar plantation. N addition elevated soil phosphatase activities and reduced net N mineralization rate, and these N addition effects were alleviated by simultaneous P addition in both plantations. N addition decreased soil labile inorganic P concentration in both plantations and elevated labile organic P in the Mongolian pine plantation, but only the decrease in labile inorganic P with N addition in the Mongolian pine plantation was counteracted by P addition. N addition significantly reduced soil pH, with a greater extent in the Mongolian pine plantation than in the Simon poplar plantation. Moreover, soil pH response explained most of the microbial and nutrient transformation response. Above results indicated that short-term N addition impacts on tree growth and soil microbial biomass were more strongly influenced by tree species identity than by P addition, while N addition effects on soil N and P transformations were strongly influenced by P addition. Drought resistance of trees and acid buffering capacity of soils are key traits influencing the short-term N addition impacts on the growth of trees and microorganisms in this semiarid region.

Abstract:Background: Transforming coniferous plantation into broadleaved or mixed broadleaved-coniferous plantations is the tendency of forest management strategies in subtropical China. However, the effects of this conversion on soil phosphorus (P) cycling microbial functional genes are still unknown. Methods: Soil samples were collected from 0-20, 20-40, and 40-60 cm (topsoil, middle layer, and subsoil, respectively) under coniferous Pinus massoniana (PM), broadleaved Erythrophleum fordii (EF), and their mixed (PM/EF) plantation in subtropical China. Used metagenomic sequencing to examine the alterations of relative abundances and molecular ecological network structure of soil P-cycling functional genes after the conversion of plantations. Results: The composition of P-cycling genes in the topsoil of PM stand was significantly different from that of PM/EF and EF stands (p < 0.05), and total phosphorus (TP) was the main factor causing this difference. After transforming PM plantation into EF plantation, the relative abundances of P solubilization and mineralization genes significantly increased in the topsoil and middle layer with the decrease of soil TP content. The abundances of P-starvation response regulation genes also significantly increased in the subsoil (p < 0.05), which may have been influenced by soil organic carbon (SOC). The dominant genes in all soil layers under three plantations were phoR, glpP, gcd, ppk, and ppx. Transforming PM into EF plantation apparently increased gcd abundance in the topsoil (p < 0.05), with TP and NO3 --N being the main influencing factors. After transforming PM into PM/EF plantations, the molecular ecological network structure of P-cycling genes was more complex; moreover, the key genes in the network were modified with the transformation of PM plantation. Conclusion: Transforming PM into EF plantation mainly improved the phosphate solubilizing potential of microorganisms at topsoil, while transforming PM into PM/EF plantation may have enhanced structural stability of microbial P-cycling genes react to environmental changes.
Phosphorus availability regulates nitrogen fixation rate through a key diazotrophic assembly: Evidence from a subtropical Moso bamboo forest subjected to nitrogen application

Abstract:Biological N fixation (BNF) is an important N input process for terrestrial ecosystems. Long-term N application increases the availability of N, but may also lead to phosphorus (P) deficiency or an imbalance between N and P. Here, we performed a 5-year N application experiment in a subtropical Phyllostachys heterocycla forest in site and a P application experiment in vitro to investigate the effect of N application on the BNF rate and its regulatory factor. The BNF rate, nifH gene, free-living diazotrophic community composition and plant properties were measured. We found that N application suppressed the BNF rate and nifH gene abundance, whereas the BNF rate in soils with added P was significantly higher overall than that in soils without added P. Moreover, we identified a key diazotrophic assembly (Mod#2), primarily comprising Bradyrhizobium , Geobacter , Desulfovibrio , Anaeromyxobacter , and Pseudodesulfovibrio , which explained 77 % of the BNF rate variation. There was a significant positive correlation between the Mod#2 abundance and soil available P, and the random forest results showed that soil available P is the most important factor affecting the Mod#2 abundance. Our findings highlight the importance of soil P availability in regulating the activities of key diazotrophs, and thus increasing P supply may help to promote N accumulation and primary productivity through facilitating the BNF process in forest ecosystems.

Fertilization- and Irrigation-Modified Bacterial Community Composition and Stimulated Enzyme Activity of Eucalyptus Plantations Soil

Abstract:Irrigation and fertilization are essential management practices for increasing forest productivity. They also impact the soil ecosystem and the microbial population. In order to examine the soil bacterial community composition and structure in response to irrigation and fertilization in a Eucalyptus plantations, a total of 20 soil samples collected from Eucalyptus plantations were analyzed using high-throughput sequencing. Experimental treatments consisting of control (CK, no irrigation or fertilization), fertilization only (F), irrigation only (W), and irrigation and fertilization (WF). The results showed a positive correlation between soil enzyme activities (urease, cellulase, and chitinase) and fertilization treatments. These enzyme activities were also significantly correlated with the diversity of soil bacterial communities in Eucalyptus plantations.. Bacteria diversity was considerably increased under irrigation and fertilization (W, F, and WF) treatments when compared with the CK treatment. Additionally, the soil bacterial richness was increased in the Eucalyptus plantations soil under irrigation (W and WF) treatments. The Acidobacteria (38.92–47.9%), Proteobacteria (20.50–28.30%), and Chloroflexi (13.88–15.55%) were the predominant phyla found in the Eucalyptus plantations soil. Specifically, compared to the CK treatment, the relative abundance of Proteobacteria was considerably higher under the W, F, and WF treatments, while the relative abundance of Acidobacteria was considerably lower. The contents of total phosphorus, accessible potassium, and organic carbon in the soil were all positively associated with fertilization and irrigation treatments. Under the WF treatment, the abundance of bacteria associated with nitrogen and carbon metabolisms, enzyme activity, and soil nutrient contents showed an increase, indicating the positive impact of irrigation and fertilization on Eucalyptus plantations production. Collectively, these findings provide the scientific and managerial bases for improving the productivity of Eucalyptus plantations.

Abstract:Soil respiration ( R s ) is a major component of the global carbon (C) cycle and is influenced by the availability of nutrients such as phosphorus (P). However, the response of R s to P addition in P-limited subtropical forest ecosystems and the underlying mechanisms remain poorly understood. To address this, we conducted a P addition experiment (50 kg P ha −1 yr −1 ) in a subtropical Chinese fir ( Cunninghamia lanceolata ) plantation forest. We separated R s into heterotrophic respiration ( R h ), root respiration ( R r ), and mycorrhizal hyphal respiration (R m ), and quantified soil properties, microbial biomass (phospholipid fatty acid, PLFA), fungal community composition (ITS), and the activity of extracellular enzymes. Phosphorus addition significantly increased R s and R h , but decreased R r and did not influence R m . Further, P addition increased fungal, bacterial, and total PLFAs, and phenol oxidase activity. Conversely, P application decreased root biomass and did not alter the relative abundance of symbiotrophic fungi. Phosphorus enrichment therefore enhances soil C emissions by promoting organic matter decomposition by heterotrophic activity, rather than via increases in root or mycorrhizal respiration. This advances our mechanistic understanding of the relationship between fertility and soil respiration in subtropical forests, with implications for predicting soil C emissions under global change.

Abstract:Rhizosphere is a hot zone formed by root–microbial interaction, and microbial activities in this zone differ from those in bulk soil. Nitrogen (N) and phosphorus (P) inputs are able to change forest soil nutrient availability, affecting microbial communities and extracellular enzyme secretion. However, the impact of N and P additions on the structure and functions of rhizosphere microbial community in Chinese fir (Cunninghamia lanceolata (Lamb.) Hook) plantations are not yet clear. To reveal the impact of short-term N and P inputs on microbial community structure and functions in rhizosphere soil, soil physicochemical properties, phospholipid fatty acids, and seven hydrolytic enzyme activities were measured in Chinese fir rhizosphere soil after one year of nutrient addition. N addition reduced the rhizosphere's pH and increased ammonium N, but the rhizosphere's available N (AN) initially wentdown and then up along the P-addition gradient. The rhizosphere fungi:bacteria ratio showed a decline after N addition, while a concave peak change occurred as rhizosphere AN under P addition. Moreover, rhizosphere extracellular enzyme activities and microbial C limitation climbed markedly with N addition rates, while this also showed an obviously unimodal pattern along the P-addition gradient. P addition did not alleviate rhizosphere microbial P limitation. Our findings suggest inconsistent responses of rhizosphere microorganisms of Chinese fir soil to N and P additions. Rhizosphere N availability can regulate microbial community structure and extracellular enzymes by influencing microbial C limitation. The study provides more knowledge on microbial activities in rhizosphere soil of subtropical forests under global changes.

Abstract:The fungal community plays an important role in forest ecosystems via the provision of resources to plant nutrition and productivity. However, the ecology of the fungal network and its relationship with phosphorus (P) dynamics remain poorly understood in mixed forest plantations. Here, we analyzed the fungal community using the amplicon sequencing in plantations of pure Eucalyptus grandis, with (E + N) and without N fertilization (E), besides pure Acacia mangium (A), and in a consortium of E. grandis and A. mangium (E + A), at 27 and 39 months after planting. We analyzed chemical, physical and biochemical soil and litter attributes related to P cycling, and the fungal community structure to find out if mixed plantations can increase fungal connections and to identify their role in the P dynamics in the soil-litter system. Soil organic fraction (OF), phosphorus in OF, total-P and acid phosphatase activity were significantly higher in E + A and A treatments regardless of the sampling period. Total N and P, richness, and Shannon diversity of the fungi in the litter was significantly higher in the treatments E + A and A. The fungal community structure in litter differed between treatments and sampling periods, and E + A showed an intermediate structure between the two pure treatments (E) and (A). E + A correlated highly with P dynamics when evaluated by both Pearson and redundancy analyses, particularly in the litter layer. Co-occurrence networks of fungal taxa became simpler in pure E. grandis plantations, whereas mixed system (E + A) showed a more connected and complex network. Our findings provide novel evidence that mixed forest plantations promote positive responses in the fungal community connections, which are closely related to P availability in the system, prominently in the litter layer. This indicates that the litter layer represents a specific niche to improve nutrient cycling by fungi in mixed forest ecosystems.
