Abstract:Unprecedented levels of nitrogen (N) have entered terrestrial ecosystems over the past century, which substantially influences the carbon (C) exchange between the atmosphere and biosphere. Temperature and moisture are generally regarded as the major controllers over the N effects on ecosystem C uptake and release. N-phosphorous (P) stoichiometry regulates the growth and metabolisms of plants and soil organisms, thereby affecting many ecosystem C processes. However, it remains unclear how the N-induced shift in the plant N:P ratio affects ecosystem production and C fluxes and its relative importance. We conducted a field manipulative experiment with eight N addition levels in a Tibetan alpine steppe and assessed the influences of N on aboveground net primary production (ANPP), gross ecosystem productivity (GEP), ecosystem respiration (ER), and net ecosystem exchange (NEE); we used linear mixed-effects models to further determine the relative contributions of various factors to the N-induced changes in these parameters. Our results showed that the ANPP, GEP, ER, and NEE all exhibited nonlinear responses to increasing N additions. Further analysis demonstrated that the plant N:P ratio played a dominate role in shaping these C exchange processes. There was a positive relationship between the N-induced changes in ANPP (ΔANPP) and the plant N:P ratio (ΔN:P), whereas the ΔGEP, ΔER, and ΔNEE exhibited quadratic correlations with the ΔN:P. In contrast, soil temperature and moisture were only secondary predictors for the changes in ecosystem production and C fluxes along the N addition gradient. These findings highlight the importance of plant N:P ratio in regulating ecosystem C exchange, which is crucial for improving our understanding of C cycles under the scenarios of global N enrichment.

Abstract:Increases in soil available nitrogen (N) influence N-cycle gene abundances and emission of nitrous oxide (N 2 O), which is primarily due to N-induced soil acidification in forest. Moreover, the extent of microbial-N saturation could control microbial activity and N 2 O emission. The contributions of N-induced alterations of microbial-N saturation and N-cycle gene abundances to N 2 O emission have rarely been quantified. Here, the mechanism underlying N 2 O emission under N additions (three chemical forms of N, i.e., NO 3 − -N, NH 4 + -N and NH 4 NO 3 –N, and each at two rates, 50 and 150 kg N ha −1 year −1 , respectively) spanning 2011–2021 was investigated in a temperate forest in Beijing. Results showed N 2 O emissions increased at both low and high N rates of all the three forms compared with control during the whole experiment. However, N 2 O emissions were lower in high rate of NH 4 NO 3 –N and NH 4 + -N treatments than the corresponding low N rates in the recent three years. Effects of N on microbial-N saturation and abundances of N-cycle genes were dependent on the N rate and form as well as experimental time. Specifically, negative effects of N on N-cycle gene abundances and positive effects of N on microbial-N saturation were demonstrated in high N rate treatments, particularly with NH 4 + addition during 2019–2021. Such effects were associated with soil acidification. A hump-backed trend between microbial-N saturation and N 2 O emissions was observed, suggesting N 2 O emissions decreased with increase of the microbial-N saturation. Furthermore, N-induced decreases in N-cycle gene abundances restrained N 2 O emissions. In particular, the nitrification process, dominated by ammonia-oxidize archaea, is critical to determination of N 2 O emissions in response to the N addition in the temperate forest. We confirmed N addition promoted soil microbial-N saturation and reduced N-cycle gene abundances, which restrained the continuous increase in N 2 O emissions. It is important for understanding the forest-N-microbe nexus under climate change.

Abstract:Introduction: Continuous nitrogen deposition increases the nitrogen content of terrestrial ecosystem and affects the geochemical cycle of soil nitrogen. Forest-grassland ecotone is the interface area of forest and grassland and is sensitive to global climate change. However, the structure composition and diversity of soil microbial communities and their relationship with soil environmental factors at increasing nitrogen deposition have not been sufficiently studied in forest-grassland ecotone. Methods: In this study, experiments were carried out with four nitrogen addition treatments (0 kgN·hm −2 ·a −1 , 10 kgN·hm −2 ·a −1 , 20 kgN·hm −2 ·a −1 and 40 kgN·hm −2 ·a −1 ) to simulate nitrogen deposition in a forest-grassland ecotone in northwest Liaoning Province, China. High-throughput sequencing and qPCR technologies were used to analyze the composition, structure, and diversity characteristics of the soil microbial communities under different levels of nitrogen addition. Results and discussion: The results showed that soil pH decreased significantly at increasing nitrogen concentrations, and the total nitrogen and ammonium nitrogen contents first increased and then decreased, which were significantly higher in the N10 treatment than in other treatments (N:0.32 ~ 0.48 g/kg; NH 4 + -N: 11.54 ~ 13 mg/kg). With the increase in nitrogen concentration, the net nitrogen mineralization, nitrification, and ammoniation rates decreased. The addition of nitrogen had no significant effect on the diversity and structure of the fungal community, while the diversity of the bacterial community decreased significantly at increasing nitrogen concentrations. Ascomycetes and Actinomycetes were the dominant fungal and bacterial phyla, respectively. The relative abundance of Ascomycetes was negatively correlated with total nitrogen content, while that of Actinomycetes was positively correlated with soil pH. The fungal community diversity was significantly negatively correlated with nitrate nitrogen, while the diversity of the bacterial community was significantly positively correlated with soil pH. No significant differences in the abundance of functional genes related to soil nitrogen transformations under the different treatments were observed. Overall, the distribution pattern and driving factors were different in soil microbial communities in a forest-grassland ecotone in northwest Liaoning. Our study enriches research content related to factors that affect the forest-grassland ecotone.

Abstract:Investigating the response of soil microbial communities to nitrogen (N) deposition is critical to understanding biogeochemical processes and the sustainable development of forests. However, whether and to what extent different forms of N deposition affect soil microbial communities in temperate forests is not fully clear. In this work, a field experiment with three years of simulated nitrogen deposition was conducted in temperate forests. The glycine and urea were chosen as organic nitrogen (ON) source, while NH4NO3 was chosen as inorganic nitrogen (IN) source. Different ratios of ON to IN (CK = 0:0, Mix-1 = 10:0, Mix-2 = 7:3, Mix-3 = 5:5, Mix-4 = 3:7, Mix-5 = 0:10) were mixed and then used with equal total amounts of 10 kg·N·ha−1·a−1. We determined soil microbial diversity and community composition for bacteria and fungi (16S rRNA and ITS), and soil parameters. Different forms of N addition significantly changed the soil bacterial and fungal communities. Mixed N sources had a positive effect on soil bacterial diversity and a negative effect on fungal diversity. Bacterial and fungal community structures were significantly separated under different forms of N addition. Soil pH was the main factor affecting the change in fungal community structure, while bacterial community structure was mainly controlled by STN. We also found that Proteobacteria, Acidobacteriota, Basidiomycota and Ascomycota were the most abundant phyla, regardless of the form of N addition. RDA showed that C/P and NH4+ were the main factors driving the change in bacterial community composition, and C/P, pH and C/N were the main factors driving the change in fungal community composition. Our results indicate that different components of N deposition need to be considered when studying the effects of N deposition on soil microorganisms in terrestrial ecosystems.

Abstract:Increasing nitrogen (N) deposition profoundly affects nutrient cycling in soil, thereby influencing forest ecosystem productivity and function. Soil microorganisms are integral in driving nutrient turnover; the changes in microbial communities in response to N deposition and the associated soil nutrient availability, especially of limited nutrients, are far from clear. To explore the changes in soil bacterial and fungal communities and their key environmental drivers under N deposition, we conducted a multilevel field N addition experiment in a Castanopsis carlesii natural forest. Soil properties and bacterial and fungal communities were investigated. There were no significant changes in alpha diversities (presented as Chao1 and Shannon's indexes) and beta diversities of bacteria and fungi among the three treatments. Consistently, the relative abundances of dominant bacterial phyla (i.e., Proteobacteria, Acidobacteria, Actinobacteria, Chloroflexi, and Planctomycetes) and fungal phyla (i.e., Basidiomycota, Ascomycota, and Rozellomycota) did not change following N addition. These results suggest that N deposition did not alter microbial community diversity and structure. In addition, the results of the Mantel test showed that soil pH, NO3−-N, dissolved organic N (DON), and total phosphorus (TP) predominantly influenced the community diversity and structure in bacteria, but not in fungi. Meanwhile, the relative abundance of some sensitive microbial genera, such as Bryobacter, Bradyrhizobium, Sorangium, and Archaeorhizomyces, were significantly decreased. These results indicate a decreased microbial ability for N fixation and P mobilization induced by N deposition. Moreover, there were significant relationships between Bryobacter, Bradyrhizobium, and Archaeorhizomyces and NO3−-N and available P (AP), suggesting that the responses of sensitive microbial groups to N deposition likely depend on the changes in available nutrients in soil, especially limited N or P. Collectively, 6 years of N addition had no significant influence on microbial communities, but some sensitive microbial groups were associated with N or P turnover. This finding emphasizes the critical roles of sensitive microbial species in meditating limited nutrient cycling in soil under climate change.

Response of gene abundance of ammonia-oxidizing microorganisms and denitrifying microorganisms to nitrogen and phosphorus addition in subtropical forest.

Abstract:We conducted a nitrogen (N) and phosphorus (P) addition experiment in Qianjiangyuan National Park in 2015, to investigate the response of ammonia-oxidizing microorganisms and denitrifying microorganisms. There were four treatments, including N addition (N), P addition (P), NP, and control (CK). Soil samples were collected in April (wet season) and November (dry season) of 2021. The abundance of amoA gene of ammonia-oxidizing microorganisms (i.e., ammonia-oxidizing archaea, AOA; ammonia-oxidizing bacteria, AOB; comammox) and denitrifying microbial genes (i.e., nirS, nirK, and nosZ) were determined using quantitative PCR approach. The results showed that soil pH was significantly decreased by long-term N addition, while soil ammonium and nitrate contents were significantly increased. Soil available P and total P contents were significantly increased with the long-term P addition. The addition of N (N and NP treatments) significantly increased the abundance of AOB-amoA gene in both seasons, and reached the highest in the N treatment around 8.30×107 copies·g-1 dry soil. The abundance of AOA-amoA gene was significantly higher in the NP treatment than that in CK, with the highest value around 1.17×109 copies·g-1 dry soil. There was no significant difference in N-related gene abundances between two seasons except for the abundance of comammox-amoA. Nitrogen addition exerted significant effect on the abundance of AOB-amoA, nirK and nosZ genes, especially in wet season. Phosphorus addition exerted significant effect on the abundance of AOA-amoA and AOB-amoA genes in both seasons, but did not affect denitrifying gene abundances. Soil pH, ammonium, nitrate, available P, and soil water contents were the main factors affecting the abundance of soil N-related functional genes. In summary, the response of soil ammonia-oxidizing microorganisms and denitrifying microorganisms was more sensitive to N addition than to P addition. These findings shed new light for evaluating soil nutrient availability as well as their response mechanism to global change in subtropical forests.

The active functional microbes contribute differently to soil nitrification and denitrification potential under long-term fertilizer regimes in North-East China

Abstract:Nitrogen (N) cycling microorganisms mediate soil nitrogen transformation processes, thereby affecting agricultural production and environment quality. However, it is not fully understood how active N-cycling microbial community in soil respond to long-term fertilization, as well as which microorganisms regulate soil nitrogen cycling in agricultural ecosystem. Here, we collected the soils from different depths and seasons at a 29-year fertilization experimental field (organic/chemical fertilizer), and investigated the transcriptions of N-cycling functional genes and their contribution to potential nitrification and denitrification. We found that long-term fertilization exerted significant impacts on the transcript abundances of nitrifiers (AOA amo A, AOB amo A and hao ) and denitrifiers ( nar G and nos Z), which was also notably influenced by season variation. The transcriptions of AOA amo A, hao , and nar G genes were lowest in autumn, and AOB amo A and nos Z transcript abundances were highest in autumn. Compared to no fertilization, soil potential nitrification rate (PNR) was reduced in fertilization treatments, while soil potential denitrification rate (PDR) was significantly enhanced in organic combined chemical fertilizer treatment. Both PNR and PDR were highest in 0–20 cm among the tested soil depths. Path model indicated active nitrifiers and denitrifiers had significant impact on soil PNR and PDR, respectively. The transcriptions of AOA amo A and nxr genes were significantly correlated with soil PNR (Pearson correlation, r > 0.174 , p < 0.05 ). Significant correlation of nap A and nos Z transcriptions with soil PDR (Pearson correlation, r > 0.234 , p < 0.05 ) was also revealed. Random forest analysis showed that SOC content and soil pH were the important factors explaining the total variance of active nitrifers and denitrifiers, respectively. Taken together, long-term fertilization regimes reduced soil PNR and enhanced PDR, which could be attributed to the different responses of active N-cycling microorganisms to soil environment variations. This work provides new insight into the nitrogen cycle, particularly microbial indicators in nitrification and denitrification of long-term fertilized agricultural ecosystems.

Abstract:Atmospheric nitrogen (N) deposition has a significant influence on soil organic carbon (SOC) accumulation in forest ecosystems. Microbial residues, as by-products of microbial anabolism, account for a significant fraction of soil C pools. However, how N deposition affects the accumulation of soil microbial residues in different forest biomes remains unclear. Here, we investigated the effects of six/seven-year N additions on microbial residues (amino sugar biomarkers) in eight forests from tropical to boreal zone in eastern China. Our results showed a minor change in the soil microbial residue concentrations but a significant change in the contribution of microbial residue-C to SOC after N addition. The contribution of fungal residue-C to SOC decreased under low N addition (50 kg N ha(-1) yr(-1)) in the tropical secondary forest (-19%), but increased under high N addition (100 kg N ha(-1) yr(-1)) in the temperate Korean pine mixed forest (+21%). The contribution of bacterial residue-C to SOC increased under the high N addition in the subtropical Castanopsis carlesii forest (+26%) and under the low N addition in the temperate birch forest (+38%), respectively. The responses of microbial residue-C in SOC to N addition depended on the changes in soil total N concentration and fungi to bacteria ratio under N addition and climate. Taken together, these findings provide the experimental evidence that N addition diversely regulates the formation and composition of microbial-derived C in SOC in forest ecosystems. C:N:P stoichiometry responses to 10 years of nitrogen addition differ across soil components and plant organs in a subtropical Pleioblastus amarus forest

Abstract:How stoichiometry in different ecosystem components responds to long-term nitrogen (N) addition is crucial for understanding within-ecosystem biogeochemistry cycling processes in the context of global change. To explore the effects of long-term N addition on nutrient stoichiometry in soil and plant components in forest ecosystem, a 10-year N addition experiment using ammonium nitrate (NH4NO3) was conducted in a bamboo forest in the Rainy Zone of West China, where the background N deposition is the highest in the world. Four N treatment levels (+0, +50, +150, +300 kg N ha−1 yr−1) (CK, LN, MN, HN) were applied monthly since November 2007, and then, the C:N:P stoichiometry of soil, microbial biomass, and enzymes in rhizosphere soil and bulk soil, and plant organs were measured. N addition decreased the stoichiometry of C:N:P of soil, microbial biomass, and enzymes. Soil C:N:P change under N addition treatments was stronger in bulk soil, while C:N:P changes for microbial biomass and enzyme activity were significant in rhizosphere soil. N addition significantly decreased TOC in bulk soil. Changes in MBC:MBN:MBP in rhizosphere and bulk soil were mainly caused by MBN and MBP, and MBP performance was consistent with that of AP. The main variable leading to the change of enzyme C:N:P in rhizosphere soil was BG and AP, and in bulk soil was LAP + NAG activity. Plant root C:P and N:P increased with N addition, while those for leaves and twigs did not. N addition significantly reduced the pH of both rhizosphere and bulk soils. These results suggest that the stoichiometry responses of rhizosphere and bulk soils were different due to the influence of plant roots. Soil acidification, enhanced aluminum toxicity potential, decreased root biomass and enhanced microbial P limitation caused by N addition were the important mechanisms that promoted stoichiometry changes in this ecosystem. Under the chronic input of N deposition, the stoichiometry between plant and soil evolved in different directions, which may lead to the decoupling of plants from soils.

Abstract:Anthropogenic nitrogen (N) inputs substantially influence the N cycle in agricultural ecosystems. However, the potential links among various environmental factors, nitrogen functional genes, and transformation rates under N fertilization remain poorly understood. Here, we conducted a five-year field experiment and collected 54 soil samples from three 0-4 m boreholes across different treatments: control, N-addition (nitrogen fertilizer) and NPK-addition (combined application of nitrogen, phosphorus and potassium fertilizers) treatments. Our results revealed pronounced variations in soil physiochemical parameters, metal concentrations and antibiotic levels under both N and NPK treatments. These alternations induced significant shifts in bacterial and fungal communities, altered NFG abundance and composition, and greatly enhanced rates of nitrate reduction processes. Notably, nutrients, antibiotics and bacteria exerted a more pronounced influence on NFGs and nitrate reduction under N treatment, whereas nutrients, metals, bacteria and fungi had a significant impact under NPK treatment. Furthermore, we established multidimensional correlations between nitrate reduction gene profiles and the activity rates under N and NPK treatments, contrasting with the absence of significant relationships in the control treatment. These findings shed light on the intricate relationships between microbial genetics and ecosystem functions in agricultural ecosystem, which is of significance for predicting and managing metabolic processes effectively.
Divergent Responses Of Soil Carbon And Nitrogen Pools To Short-Term Nitrogen Addition Between Two Plantations In Northeast China

Abstract:Nitrogen (N) deposition has a profound influence on forest soil carbon (C) and N pools, but there was no consensus on the responses of different C and N components in different forest types. In this study, a two-year simulated N deposition experiment with four levels of N (NH4NO3)-addition treatments (0, 50, 100, and 150 kg N/hm(2).a) were conducted in Larix gmelinii (LG) and Quercus mongolica (QM) plantation in Northeast China, in order to investigate the C and N pool dynamics under continuously enhanced N deposition. Soil organic carbon (SOC), soil total N (STN) and their active components (readily oxidizable C, ROC; dissolved organic C, DOC; microbial biomass C, MBC, dissolved organic N, DON; microbial biomass N, MBN) of the forest soil were measured monthly from May to October 2017. C and N contents in LG were observed higher than in QM. N addition had no effect on SOC and STN of LG, but significantly increased SOC and STN of QM at low N addition level. Low N addition generally raised active C components (ROC, DOC, and MBC) in both plantations, whereas high N addition did not significantly affect these components, or even decreased ROC in LG soil. Low N addition also increased STN and MBN of QM, while no significant change in STN and MBN of LG was observed. DON was directly affected by N addition and increased significantly with elevated N addition levels. The results indicated that N addition, especially of low rate, might enhance the C sequestration capacity of the forest soils and mitigate climate change.

Abstract:Increasing atmospheric nitrogen (N) deposition has substantially affected carbon (C) and nutrient cycling in forest ecosystems. However, the responses of different soil organic carbon (SOC) fractions with different turnover rates to N addition are highly divergent, and the underlying mechanisms remain elusive. In this study, we explored the responses of surface soil (0-10 cm) characteristics and microbial communities to six years of experimental N addition (0, 50, 100 and 150 kg N ha(-1) yr(-1)) in a subtropical evergreen broadleaf forest in southern China. Our results showed that N addition led to significant soil acidification (pH from 5.3 to 4.9). Microbial biomass carbon and total microbial, bacterial and fungal abundance (phospholipid fatty acid, PLFA) were reduced by N addition, but extracellular enzymes involved in C, N and phosphorus (P) cycling were not responsive to N addition. Soil extractable Ca2+ concentration was depleted by N addition, while other extractable cations (Fe3+, Al3+, Mg2+, K+, Na+) were not affected. Moreover, N addition did not significantly change the C and N concentration of bulk soil. We further separated the bulk soil into particulate organic matter (>53 mu m, POM) and mineral-associated organic matter (<53 mu m, MAOM) fractions by wet sieving. Interestingly, C in the POM fraction was significantly increased by N addition, while C in the MAOM fraction was depleted by N addition. Correlation analysis and structural equation modeling results suggested that N addition may suppress microbial decomposition of plant inputs and thus lead to accumulation of C in the POM fraction, while it may reduce the mineral sorption of microbial necromass and products and thus cause depletion of C in the MAOM fraction. Taken together, our results highlighted the vulnerability of soil C in the stable MAOM fraction to N addition, and emphasized the role of soil metals (particularly extractable Ca2+) and pH in controlling soil C storage under N addition.

Abstract:Atmospheric nitrogen (N) deposition and phosphorus (P) addition both can change soil bacterial and fungal community structure with a consequent impact on ecosystem functions. However, which factor plays an important role in regulating responses of bacterial and fungal community to N and P enrichments remains unclear. We conducted a manipulative experiment to simulate N and P inputs (10 g N · m −2 · yr −1 NH 4 NO 3 or 10 g P · m −2 · yr −1 NaH 2 PO 4 ) and compared their effects on soil bacterial and fungal species richness and community composition. The results showed that the addition of N significantly increased NH 4 + and Al 3+ by 99.6% and 57.4%, respectively, and consequently led to a decline in soil pH from 4.18 to 3.75 after a 5-year treatment. P addition increased Al 3+ and available P by 27.0% and 10-fold, respectively, but had no effect on soil pH. N addition significantly decreased bacterial species richness and Shannon index and resulted in a substantial shift of bacterial community composition, whereas P addition did not. Neither N nor P addition changed fungal species richness, Shannon index, and fungal community composition. A structural equation model showed that the shift in bacterial community composition was related to an increase in soil acid cations. The principal component scores of soil nutrients showed a significantly positive relationship with fungal community composition. Our results suggest that N and P additions affect soil bacterial and fungal communities in different ways in subtropical forest. These findings highlight how the diversity of microbial communities of subtropical forest soil will depend on future scenarios of anthropogenic N deposition and P enrichment, with a particular sensitivity of bacterial community to N addition.

Abstract:Increasing atmospheric nitrogen (N) deposition has profound effects on carbon (C) cycling in forest ecosystems. As an important part of belowground C dynamics, soil respiration is potentially affected by changing N availability. However, the responses of total soil respiration (RST) and its three components, soil respiration derived from plant roots (RSR), root-free soil (RSS) and the litter layer (RSL), to such N enrichment remains poorly understood. To assess the effects of N enrichment on soil respiration components, three levels of N addition, namely low (LN, 50 kg N ha−1 year−1), medium (MN, 100 kg N ha−1 year−1) and high (HN, 150 kg N ha−1 year−1), were conducted over five growing seasons from 2011 to 2015 in a temperate Chinese pine (Pinus tabulaeformis) forest in northern China. A control plot without N addition (CK) was also established. The five-year mean annual rate of RST was 2.18 ± 0.43 μmol m−2 s−1, and the contributions of RSR, RSS and RSL were 8.8 ± 3.1%, 82.2 ± 4.5% and 9.0 ± 5.5%, respectively. Compared with CK, RST was significantly increased by 16.5% in the HN plots, but not in the LN or MN treatments. RSS was significantly decreased by 18.1%, 26.6% and 18.4% in the LN, MN and HN plots, respectively, due to the reduction of both microbial biomass carbon (MBC) and enzyme activity. In contrast, RSR was increased by more than twice under the MN treatment, which promoted root growth and activity (higher fine root biomass and N concentration). A significant elevation in RSL was only detected in the HN plots, where the increased litter input enhanced litter decomposition and hence RSL. Our findings clearly demonstrated that N addition of different intensities had different effects on soil components. In particular, the above- and belowground components of heterotrophic respiration, RSL and RSR, showed contrasting responses to high level addition of N. Thus, we highlight that the response of soil respiration components to N addition should be examined individually. Our results may contribute to a better understanding of soil respiration dynamics under future N scenarios, and have important implications in forest management.
Long-term Nitrogen Addition Modifies Microbial Composition and Functions for Slow Carbon Cycling and Increased Sequestration in Tropical Forest Soil.

Abstract:Nitrogen (N) deposition is a component of global change that has considerable impact on belowground carbon (C) dynamics. Plant growth stimulation and alterations of fungal community composition and functions are the main mechanisms driving soil C gains following N deposition in N-limited temperate forests. In N-rich tropical forests, however, N deposition generally has minor effects on plant growth; consequently, C storage in soil may strongly depend on the microbial processes that drive litter and soil organic matter decomposition. Here, we investigated how microbial functions in old-growth tropical forest soil responded to 13 years of N addition at four rates: 0 (Control), 50 (Low-N), 100 (Medium-N), and 150 (High-N) kg N ha(-1) year(-1). Soil organic carbon (SOC) content increased under High-N, corresponding to a 33% decrease in CO2 efflux, and reductions in relative abundances of bacteria as well as genes responsible for cellulose and chitin degradation. A 113% increase in N2O emission was positively correlated with soil acidification and an increase in the relative abundances of denitrification genes (narG and norB). Soil acidification induced by N addition decreased available P concentrations, and was associated with reductions in the relative abundance of phytase. The decreased relative abundance of bacteria and key functional gene groups for C degradation were related to slower SOC decomposition, indicating the key mechanisms driving SOC accumulation in the tropical forest soil subjected to High-N addition. However, changes in microbial functional groups associated with N and P cycling led to coincidentally large increases in N2O emissions, and exacerbated soil P deficiency. These two factors partially offset the perceived beneficial effects of N addition on SOC storage in tropical forest soils. These findings suggest a potential to incorporate microbial community and functions into Earth system models considering their effects on greenhouse gas emission, biogeochemical processes, and biodiversity of tropical ecosystems.

Abstract:To understand the effects of nitrogen deposition on element cycling and nutrient limitation status in forest ecosystems, we examined the effects of nitrogen deposition on the stoichiometric characteristics of forest soil-microbial-extracellular enzymes in Pinus yunnanensis forest. We conducted a field experiment with control (CK, 0 g N·m-2·a-1), low nitrogen (LN, 10 g N·m-2·a-1), medium nitrogen (MN, 20 g N·m-2·a-1) and high nitrogen (HN, 25 g N·m-2·a-1) since 2019. We collected soil samples (0-5 cm, 5-10 cm and 10-20 cm) at September 2022, and measured the contents of soil organic, total nitrogen, total phosphorus, microbial biomass carbon, nitrogen and phosphorus (MBC, MBN, MBP) and the activities of C, N, and P acquisition enzymes. The results showed that nitrogen deposition significantly reduced soil organic content, C:N and C:P by 6.9%-29.8%, 7.6%-45.2% and 6.5%-28.6%, and increased soil total N content and N:P by 10.0%-45.0% and 19.0%-46.0%, respectively. Nitrogen addition did not affect soil total P content. Except for soil C:N and C:P, soil nutrient content and stoichiometric ratio were highest in 0-5 cm soil layer. MN and HN treatments significantly decreased MBN by 11.0%-12.7%. MBC, MBP, and their stoichiometry did not change significantly under nitrogen deposition. Soil microbial nutrient content in 0-5 cm soil layer was significantly higher than that in other soil layers. Nitrogen deposition significantly decreased the activities of cellobiose hydrolase and leucine aminopeptidase (decreased by 14.5%-16.2% and 48.7%-66.3%). HN treatment promoted β-1,4-glucosidase activity (increased by 68.0%), but inhibited soil enzyme stoichiometric carbon to nitrogen ratio and nitrogen to phosphorus ratio (decreased by 95.4% and 88.4%). LN and MN treatment promoted β-1,4-N-acetylglucosaminidase activity (increased by 68.3%-116.6%), but inhibited enzyme stoichiometric carbon to phosphorus ratio (decreased by 14.9%-29.4%). Alkaline phosphatase activity had no significant change. Soil enzyme activities were significantly decreased with increasing soil depth. Soil total N and total P and microbial nutrients were negatively correlated with vector angle (representing microbial nitrogen or phosphorus limitation), while vector length (representing microbial carbon limitation) was consistently significantly positively correlated with vector angle, suggesting the synergistic promotion between microbial carbon limitation and phosphorus limitation. Nitrogen deposition gradually shifted to phosphorus limitation while alleviating microbial nitrogen limitation in P. yunnanensis forest. In addition, microbial activities in this region was limited by C availability, and the relationship between microbial C and P limitation was proportional.

Abstract:Atmospheric nitrogen (N) deposition is among the main manifestations of global change and has profoundly affected forest biogeochemical cycles. However, the threshold of N deposition to soil nutrient contents and enzyme activities has rarely been studied in a forest. In this study, we explored the effects of N deposition on soil nutrients and enzyme activities in a Larix principis-rupprechtii plantation on the northern Yanshan Mountain through multigradient N addition experiments (0, 5, 10, 20, 40, 80, and 160 kg N ha −1 year −1 ) after fertilization for 2 years. Compared with the controls, N addition first led to a decrease in soil NH 4+ -N and NO 3- -N, which then increased significantly. N addition had no significant effects on other soil nutrients. N addition overall elevated soil β-glucosidase activity. N application of >40 kg N ha −1 year −1 significantly reduced soil leucine aminopeptidase activity but had no significant effects on soil acid phosphatase, N-acetyl-β-D-glucosidase, and urease activities. N addition increased the overall stoichiometry ratio of EEA C:N and EEA C:P, but EEA N:P started decreasing after N application of 40 kg N ha −1 year −1 . The ratios of C, N, and P acquisition activities changed from 1:1.2:1 under the control conditions to 1:1.1:1 under the N application of 160 kg N ha −1 year −1 . N addition increased the overall vector length and had no significant effects on the vector angle. Correlation and redundancy analyses revealed that N addition-induced change in available soil N was the main factor affecting soil enzyme activity and stoichiometry. In general, different enzyme activities had different sensitivities to N addition. Moderate N addition or atmospheric N deposition (e.g., <40 kg N ha −1 year −1 ) had beneficial effects on soil nutrient cycling and microorganisms in a Larix principis-rupprechtii plantation.

Abstract:Nitrogen (N) deposition is a worldwide issue caused by human activity. Long-term deposition of N strongly influences plant productivity and community composition. However, it is still unclear how the microbial community responds to long-term N addition in a desert ecosystem. Therefore, a long-term experiment was conducted in the Gurbantonggut Desert in northwestern China in 2015. Four N addition rates, 0 (CK), 5 (N1), 20 (N2), and 80 (N3) kg N ha −1 yr. −1 , were tested and the soil was sampled after 6 years of N addition. High-throughput sequencing (HTS) was used to analyze the soil microbial composition. The HTS results showed that N addition had no significant effect on the bacterial α-diversity and β-diversity ( p > 0.05) but significantly reduced the archaeal β-diversity ( p < 0.05). The fungal Chao1 and ACE indexes in the N2 treatment increased by 24.10 and 26.07%, respectively. In addition, N addition affected the bacterial and fungal community structures. For example, compared to CK, the relative abundance of Actinobacteria increased by 17.80%, and the relative abundance of Bacteroidetes was reduced by 44.46% under N3 treatment. Additionally, N addition also changed the bacterial and fungal community functions. The N3 treatment showed increased relative abundance of nitrate-reducing bacteria (27.06% higher than CK). The relative abundance of symbiotrophic fungi was increased in the N1 treatment (253.11% higher than CK). SOC and NH 4 + -N could explain 62% of the changes in the fungal community function. N addition can directly affect the bacterial community function or indirectly through NO 3 − -N. These results suggest that different microbial groups may have various responses to N addition. Compared with bacteria and fungi, the effect of N addition was less on the archaeal community. Meanwhile, N-mediated changes of the soil properties play an essential role in changes in the microbial community. The results in the present study provided a reliable basis for an understanding of how the microbial community in a desert ecosystem adapts to long-term N deposition.

Abstract:Nitrogen (N) deposition is a global environmental issue that can have significant impacts on the community structure and function in ecosystems. Fungi play a key role in soil biogeochemical cycles and their community structures are tightly linked to the health and productivity of forest ecosystems. Based on high-throughput sequencing and ergosterol extraction, we examined the changes in community structure, composition, and biomass of soil ectomycorrhizal (ECM) and saprophytic (SAP) fungi in 0–10 cm soil layer after 8 years of continuous N addition and their driving factors in a temperate Korean pine plantation in northeast China. Our results showed that N addition increased fungal community richness, with the highest richness and Chao1 index under the low N treatment (LN: 20 kg N ha −1 yr −1 ). Based on the FUN Guild database, we found that the relative abundance of ECM and SAP fungi increased first and then decreased with increasing N deposition concentration. The molecular ecological network analysis showed that the interaction between ECM and SAP fungi was enhanced by N addition, and the interaction was mainly positive in the ECM fungal network. N addition increased fungal biomass, and the total fungal biomass (TFB) was the highest under the MN treatment (6.05 ± 0.3 mg g −1 ). Overall, we concluded that N addition changed soil biochemical parameters, increased fungal activity, and enhanced functional fungal interactions in the Korean pine plantation over an 8-year simulated N addition. We need to consider the effects of complex soil conditions on soil fungi and emphasize the importance of regulating soil fungal community structure and biomass for managing forest ecosystems. These findings could deepen our understanding of the effects of increased N deposition on soil fungi in temperate forests in northern China, which can provide the theoretical basis for reducing the effects of increased N deposition on forest soil.
