Abstract:Pirarubicin (THP), doxorubicin (DOX), cyclophosphamide (CTX), and vincristine (VCR) are widely used in the treatment of patients with non-Hodgkin's Lymphoma. Herein, a precise and sensitive method was developed for the determination of THP, DOX, CTX and VCR in human plasma by high-performance liquid-chromatography-tandem mass spectrometry (LC-MS/MS). Liquid-liquid extraction was applied to extract THP, DOX, CTX, VCR, and the internal standard (IS, Pioglitazone) in plasma. Agilent Eclipse XDB-C18 (3.0 mm × 100 mm) was utilized and chromatographic separation was obtained in eight minutes. Mobile phases were composed of methanol and buffer (10 mM ammonium formate containing 0.1% formic acid). The method was linear within the concentration range of 1-500 ng/mL for THP, 2-1000 ng/mL for DOX, 2.5-1250 ng/mL for CTX, and 3-1500 ng/mL for VCR. The intra- and inter-day precisions of QC samples were found to be below 9.31 and 13.66%, and accuracy ranged from -0.2 to 9.07%, respectively. THP, DOX, CTX, VCR and the internal standard were stable in several conditions. Finally, this method was successfully utilized to simultaneously determine THP, DOX, CTX and VCR in human plasma of 15 patients with non-Hodgkin's Lymphoma after intravenous administration. Finally, the method was successfully employed in the clinical determination of THP, DOX, CTX, and VCR in patients with non-Hodgkin lymphoma after administration of RCHOP (rituximab plus cyclophosphamide, doxorubicin, vincristine, and prednisone) regimens.

Abstract:A sensitive and selective liquid chromatographic–tandem mass spectrometric (LC–MS/MS) method for the determination of paclitaxel (Taxol) and its two major metabolites in human plasma has been developed. Samples were prepared after liquid–liquid extraction and analyzed on a C18 column interfaced with a Q-Trap tandem mass spectrometer. Positive electrospray ionization was employed as the ionization source. The mobile phase consisted of acetonitrile–water (0.05% formic acid) (65:35) at the flow rate of 0.25 mL/min. The analytes and internal standard docetaxel were both detected by use of multiple reaction monitoring mode. The method was linear in the concentration range of 0.5–500.0 ng/mL for paclitaxel, 6α-hydroxypaclitaxel and p-3′-hydroxypaclitaxel, respectively. The lower limit of quantification (LLOQ) was 0.5 ng/mL for paclitaxel, 6α-hydroxypaclitaxel and p-3′-hydroxypaclitaxel, respectively. The intra- and inter-day relative standard deviation across three validation runs over the entire concentration range was less than 8.18%. The accuracy determined at three concentrations was within ±10.8% in terms of relative error. The total run time was 7.0 min. This assay offers advantages in terms of expediency, and suitability for the analysis of paclitaxel and its metabolites in various biological fluids.
Quantification of paclitaxel in rat plasma by LC-MS/MS and pharmacokinetics of paclitaxel thermosensi-tive liposomes in rats

Abstract:OBJECTIVE: To develop and validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of paclitaxel in rat plasma and to study the pharmacokinetics of paclitaxel thermosensitive liposomes in rats. METHODS: The plasma was alkalized with sodium bicarbonate (pH 8.0) and extracted with tert-butyl methyl ether (TBME). Paclitaxel and IS(docetaxel) were separated on an Agilent Zorbax SB C18 column (2.1 mm × 50 mm, 3.5 μm) with the mobile phase consisting of methanol-0.5 mmol · L-1 ammonium acetate (70: 30, V/V) at a flow rate of 300 μL · min-1. The injection volume was 5 μL An Agilent 6410 A QQQ triple-quadrople mass spectrometer equipped with an electrospray ionization (ESI) source was used as detector and was operated in positive ion mode. Selected ion monitoring (SIM) was performed and the m/z of ions selected for quantitation were 876.3 → 308.1 (paclitaxel) and 830.2 → 549.2 (internal standard, docetaxel) . SD rats were injected paclitaxel thermosensitive liposomes and paclitaxel injection via tail vein at the dose of 7 mg · kg-1, respectively. Pharmacokinetic parameters were calculated. RESULTS: The chromatograms showed no endogenous interfering peaks in blank rat plasma. Each analysis was completed within 3.5 min. The linear range of paclitaxel in plasma was 2-1 000 ng · mL-1. The lower limit of quantification was 2 ng · mL-1. The intra-run and inter-run relative standard deviations (RSDs) were less than 15%. The differences of f1/2β, MRT, AUC and CL were significant between the two preparations. CONCLUSION: The method is sensitive, accurate, and convenient for the pharmacokinetic study of paclitaxel thermosensitive liposomes. Compared with paclitaxel injection, paclitaxel thermosensitive liposomes has a controlled release rate, which can alleviate the acute toxicity and improve the tolerance to a certain extent.

Abstract:ObjectiveTo establish a liquid chromatography-tandem mass spectrometry(LC-MS/MS)for the quantitative determination of plasma paclitaxel,and to evaluate its performance.Methods Plasma paclitaxel was determined quantitatively by LC-MS/MS. High performance liquid chromatography was used. The flow speed was 0.3 mL/min. Multiple reaction monitoring(MRM) was used for quantitative determination. The ion pairs of paclitaxel and PTX-D5 were 876.4>308.2 and 881.5>313.2,respectively. According to U.S. Food and Drug Administration(FDA) guideline about bioanalytical method validation,the general analytical performance validation was performed for linearity,detection limit,precision and accuracy.ResultsThe linearity was 10-1 000 ng/mL. The detection limit was 10 ng/mL. Within-run and between-run coefficients of variation(CV) were≤4.2% and≤8.9%,respectively. The accuracy was 88.7%-104.0%.Conclusions The performance of established LC-MS/MS meets evaluation standards,and the assay is accurate and precise for the determination of paclitaxel.

Abstract:A sensitive and selective method has been developed and validated for the quantitation of paclitaxel in human plasma by liquid chromatography-eleetrospray ionization mass spectrometry (LC-ESI-MS).Paclitaxel and norethindrone (used as internal standard,I.S.) were extracted from human plasma by a one-step liquid-liquid extraction with t-butyl methyl ether.Separation on a Zorbax SB-C18 column (100 mm×2.1 mm,3.5 μm,Agilent) was achieved by gradient elution with methanol and 0.2 mmol/L ammonium formate containing 0.1％ formic acid.The selected-ion monitoring (SIM) targeted ions of [M+Na]+ at m/z 876.5 for paclitaxel and [M+H](+) at m/z 299.4 for I.S.The assay was validated in the range of 1.0-400 ng/mL (r＞0.998) with LLOQ of 1.0 ng/mL.Intra- and inter-day precisions were all less than 9.0％,with accuracies of±6.8％.The method was successfully applied to evaluate the pharmacokinetics of paclitaxel liposome for injection in patients.

Abstract:A simple, rapid, and sensitive reversed-phase HPLC method was developed and validated for determination of paclitaxel (PTX) in plasma, various organs and tumor tissues of tumor-bearing mice. Tissue specimens of liver, kidneys, spleen, lungs, heart and tumor were separately homogenized in normal saline. Plasma or tissue homogenate (250 μl) containing PTX and internal standard (diazepam) were extracted by diethyl ether (6 ml). The separation was achieved on a μ-Bondapak C18 HPLC column using sodium acetate buffer solution (0.01 M)/acetonitrile (58/42 v/v) at pH 5 ± 0.1 and flow rate of 1.9 mL/min. The effluent was monitored at 227 nm and column temperature was adjusted at 58oC. The internal standard and PTX were eluted at 4.2 and 5.2 min, respectively and no interfering peaks were observed. Calibration curves were linear over the concentration range of 0.25-10 μg/ml of PTX in plasma and 0.3-20 μg/ml PTX in tissue homogenates with acceptable precision and accuracy (<15%). The mean recoveries of the drug after plasma extraction was 87.4% ± 3.6 while those of tissue homogenates ranged from 62.1± 4.5 to 75.5± 3.2 depending on the type of tissues studied. PTX was stable in samples with no evidence of degradation during 3 freeze–thaw cycles and 3 months storage at −70 °C. The developed HPLC method was applied to quantify PTX in the mouse plasma and tissues after intravenous administration of 10 mg equivalent PTX/Kg dose of PTX-loaded tocopherol succinate-chitosan-polyethylene glycol-folate (TS-CS-PEG-FA) micelles formulation or Anzatax® (Cremophor® EL- based formulation of PTX) to female Balb/c mice. Abstract:Owing to its unpredictable side effects and efficacy, the anticancer drug docetaxel requires improved characterisation of its pharmacokinetic profiles through population pharmacokinetic (PPK) studies. A sensitive and rugged liquid chromatography-tandem mass spectrometry method for the detection of docetaxel in human plasma was developed and optimised using paclitaxel as an internal standard (IS). The plasma samples underwent rapid extraction using Hybrid solid-phase extraction-protein precipitation. The analyte and IS were separated with an isocratic system on a Zorbax Eclipse Plus C18 column using water containing 0.05% acetic acid along with 20 μM sodium acetate and methanol (30/70, v/v) as the mobile phase. Quantification was performed with a triple quadrupole mass spectrometer through multiple reaction monitoring in positive mode, using the m/z 830.3→548.8 and m/z 876.3→307.7 transitions for docetaxel and paclitaxel, respectively. The range of the calibration curve was 1-500 ng/mL for docetaxel and the linear correlation coefficient was greater than 0.99. The accuracies ranged from -4.6% to 4.2%, and the precision was no higher than 7.0% for the analytes. No significant matrix effect was observed. Both docetaxel and the IS showed considerable recovery. This method was finally applied to the establishment of a PPK model to optimise the clinical use of docetaxel.

Abstract:European Journal of Mass Spectrometry, Ahead of Print. Present work describes the development of a liquid chromatography tandem mass spectrometry-based bioanalytical method for the reliable simultaneous quantification of docetaxel (DXL) and carvacrol (CVL) in the mice plasma. A rapid and sensitive bioanalytical method was developed and optimized in mice plasma using Paclitaxel as an internal standard. Validation of the bioanalytical method was performed according to the ICH M10 guideline covering the range of 9.62–1923.08 ng/mL in the mice plasma milleu at the low, mid, and high-quality control concentrations of 28.86 ng/mL, 961.54 ng/mL, and 1346.15 ng/mL, respectively for both the analytes. Validation parameters such as accuracy, precision, carryover-test, matrix effect, and reinjection reproducibility were carried out and were found in limits. Stability studies (Benchtop, autosampler, freeze-thaw, and long-term) were performed and found to be within limits. The developed bioanalytical method was found to be suitable for the simultaneous quantification of DXL and CVL in the mice plasma.

Abstract:The prominent stromal compartment surrounds pancreatic ductal adenocarcinoma (PDAC) protects the tumor cells from chemo- or radio-therapy. We hypothesized that our nano formulation carried cyclopamine (CPA, stroma modulator) and paclitaxel (PTX, anti-tumor agent) could increase the permeation of PTX through the stromal compartment and improve the intratumoral delivery of PTX. In the present study a sensitive, reliable UPLC-MS/MS method was developed and validated to quantify PTX and CPA simultaneously in mouse whole blood, pancreas, liver and spleen samples. Docetaxel was used as the internal standard. The method demonstrated a linear range of 0.5-2000 ng/mL for whole blood and tissue homogenates for both PTX and CPA. Accuracy and precision of the assay were all within ±15 %. Matrix effects for both analytes were within 15%. Recoveries from whole blood; liver, spleen and pancreas homogenates were 92.7-105.2% for PTX and 72.8-99.7% for CPA. The stability was within ±15% in all test biomatrices. The validated method met the acceptance criteria according to FDA regulatory guidelines. The method was successfully applied to support pharmacokinetic and biodistribution study for PTX and CPA in mice biomatrices.

Abstract:Paclitaxel is a broad-spectrum anti-cancer drug by targeting microtubulin. However, multidrug resistant (MDR) makes its clinical application more difficult and results in failure of chemotherapy. Tetrandrine as a potential multidrug resistant modulator could be combined with other anti-cancer drugs. In this study, ultra-performance liquid chromatography (UHPLC) combined with quadrupole time-of-flight mass spectrometry (QTOF) was applied to simultaneously qualitative and quantitative analysis of paclitaxel for the pharmacokinetic studies while combined with tetrandrine. This method was developed based on non-target screening mode IDA (Information Dependent Acquisition). As a result, the validated range was 0.25-64ng/ml (30µl plasma) for paclitaxel. Totally 33 metabolites of paclitaxel and tetrandine were identified in vivo and in vitro. The main metabolites of PTX were dose-dependent decreased with different amounts of tetrandine co-administration no matter in vivo and in vitro, the exposure of PTX increased in pharmacokinetic study. The verified method is sensitive accurate and effective for the simultaneous determination of paclitaxel and its metabolites in blood, urine and live microsome incubation samples and it was successfully applied to evaluate the pharmacokinetics and drug-drug interaction between paclitaxel and tetrandine. Furthermore, a biosensor technology, surface plasmon resonance (SPR) analysis was applied to preliminary evaluate the competitive protein binding of multiple components. The SPR analysis indicated that the affinity between 6-hydroxy-paclitaxel and micotubulin is similar to that between paclitaxel and micotubulin, and tetrandrine also does not form a competitive combination with paclitaxel. For human, 6-hydroxy-paclitaxel is the one of main metabolites of paclitaxel, so the results suggested that tetrandine has an influence on the metabolite of paclitaxel, but tetrandine and the main metabolites of PTX probably do not affect PTX's biological targeting, the effect of its pharmacological action needs to be further studied.
A RP-HPLC method for determination of paclitaxel in its solid dispersion

Abstract:Aim To develop and validate a RP-HPLC method for the analysis of paclitaxel in a solid dispersion. Methods Paclitaxel and the internal standard norethisterone were separated using a Phenomenex ODS 3 column and monitored at a wavelength at 227 nm. The isocratic mobile phase consisting of methanol-acetonitrile-water (40:30:30, V/V) was pumped at a flow-rate of 1.0 mL·min-1. The dissolution studies were performed according to published studies. Results Under these chromatographic conditions, the calibration curve was linear in the range of 4-40 μg·mL-1 with the correlation coefficient of 0.9999. The mean recovery was 98.42 % (RSD = 1.19 %). At the 60 min time point, the dissolution of paclitaxel from the solid dispersion was nearly 100 %, however, the original form of paclitaxel was about 30 %. Conclusion The method was proven to be specific, accurate and precise for determining the dissolution of paclitaxel from solid dispersion.

Abstract:A simple and accurate reversed‐phase high performance liquid chromatography method was developed and validated for the simultaneous determination of dexamethasone, dexamethasone 21‐acetate, and paclitaxel in simulated biological matrix. Triamcinolone Acetonide was used as the internal standard (IS). A one step liquid–liquid extraction with ethyl acetate was used for sample cleanup. A gradient elution on a C−18 column with acetonitrile and 0.1% o‐phosphoric acid in water was employed to get a cleanup baseline and efficient separation of the three chemical entities. Dexamethasone and dexamethasone 21‐acetate were observed at 254 nm and paclitaxel was observed at 228 nm. The method was highly selective and the calibration curves were linear over the range of 0.25–10.0 µg/mL with a 12 min chromatographic run time. The assay method was validated in terms of accuracy, precision, absolute recovery, freeze‐thaw stability, bench top stability, and reinjection reproducibility. The lower limit of quantitation was 0.25 µg/mL for all the analytes. The intra and inter‐batch precision and accuracy were found to be well within acceptable limits (<15%). The assay is simple, reliable, and fast and is deemed suitable for application to in vitro release rate studies of these drugs from implantable devices, as well as to their in vivo pharmacokinetic studies.

Development and validation of a novel high-performance liquid chromatography (HPLC) method for the detection of related substances of pralsetinib, a new anti-lung cancer drug

Abstract:Background: Pralsetinib, a targeted inhibitor of the RET enzyme, plays a critical role in the treatment of adult patients with locally advanced or metastatic non-small cell lung cancer (NSCLC) characterized by RET gene fusion mutations following platinum-based chemotherapy. Nevertheless, impurities resulting from the manufacturing and degradation of pralsetinib have the potential to impact its therapeutic effectiveness and safety profile. Methods: To address this issue, a liquid chromatography method was developed and validated for the specific identification of pralsetinib and its related impurities. The separation of pralsetinib and its related impurities was achieved via a Waters X Bridge C18 column with dimensions of 4.6 mm × 250 mm and a particle size of 5 μm. Mobile phase A was composed of 20 mmol/L potassium dihydrogen phosphate (KH2PO4) and acetonitrile (ACN) at a volume ratio of 19:1, while mobile phase B consisted solely of ACN, utilizing a gradient elution technique. Detection was performed at a wavelength of 260 nm, with an injection volume of 10 μL and a flow rate of 1.0 mL/min. Results: The chromatographic method established in this study was validated according to the ICH Q2 (R1) guidelines. The method demonstrated excellent linearity over a specific concentration range (imp-A: 0.035-10.21 μg/mL; imp-B: 0.09-10.16 μg/mL; imp-C: 0.15-10.19 μg/mL; pralsetinib: 0.04-10.32 μg/mL). Additionally, the method possesses high sensitivity, with detection limits for impurities A, B, C, and pralsetinib of 0.01, 0.03, 0.015, and 0.013 μg/mL, respectively, and quantification limits of 0.035, 0.09, 0.05, and 0.04 μg/mL, respectively. In terms of specificity, stability, repeatability, accuracy, and robustness, the method met the validation acceptance criteria. Overall, the chromatographic technique established in this study can effectively separate pralsetinib and its impurities, providing reliable assurance for the accurate detection and quantification of impurities. Conclusion: The chromatographic method developed in this study can be utilized for the detection of pralsetinib and its impurities, offering a crucial reference for research on the quality of pralsetinib.

Abstract:Objective: To investigate and compare the ionization behaviors of paclitaxel and docetaxel by exposing to various additives in two sampling ways: direct infusion or by liquid chromatograph into mass spectrometer. Methods: The working solutions of paclitaxel and docetaxel, mixed with different additives, were led into the electrospray ionization source, via a syringe pump or via the liquid chromatographic system, for detection and collection of mass spectra or chromatograms of the analytes.The influence of additives, with various sorts, concentrations and pH, in solutions or mobile phases on the ionization efficiency of paclitaxel and docetaxel were studied. Results: Similar patterns of mass spectra intensity variations were observed between these two sampling methods, in that the highest ionization efficiency and strongest signal intensity were obtained when the analytes were exposed to the additive as 1 mmol/L ammonium formate (pH=6.5) in both methods. Conclusions: Direct infusion of samples into mass spectrometer is an efficient, simple and reliable way to optimize the mobile phase composition in the development of liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for quantitation of paclitaxel and docetaxel.Based on these observations, a sensitive and reproducible LC-MS/MS method for the determination of paclitaxel in human plasma was successfully established and validated.
A Simple Eco-Friendly HPLC-PDA Method for the Simultaneous Determination of Paclitaxel and Seliciclib in Plasma Samples for Assessing Their Pharmacodynamics and Pharmacokinetics in Combination Therapy for Uterine Sarcoma

Abstract:Background/Objectives: Uterine sarcoma, a rare cancer originating in the smooth muscle of the uterus, exhibits high rates of recurrence and metastasis. It represents one of the most challenging types of cancer due to its chemorefractory nature, showing little response to conventional chemotherapy methods and displaying a relative survival rate of 30-40%. A potentially promising approach for treating uterine sarcoma involves combination therapy with paclitaxel (PAC), a microtubule-targeting agent, and seliciclib (SEL), a cyclin-dependent kinase inhibitor. SEL has been identified as a drug that can enhance the effectiveness of PAC through synergistic effects. To further refine this treatment strategy, an efficient analytical tool capable of simultaneously measuring the concentrations of PAC and SEL in blood plasma is needed. This tool would make it easier to study the pharmacokinetic interactions of potential drugs and assist in monitoring therapy when administering this combination treatment. Regrettably, a method meeting these specific requirements has not been documented in the existing literature. Methods: This article introduces the first HPLC technique employing a PDA detector to concurrently measure PAC and SEL levels in plasma. The methodology underwent validation in accordance with the ICH standards for validating bioanalytical methods. Results: The method exhibited linearity in the concentrations ranging from 0.8 to 100 µg mL-1 for both PAC and SEL. The limits of quantification were determined and found to be 1.34 and 1.25 µg mL-1 for PAC and SEL, respectively. All the other validation criteria conformed to the ICH validation standards. The HPLC-PDA method was successfully employed to quantify both PAC and SEL in plasma samples with a high level of reliability (in terms of accuracy and precision). The eco-friendliness of the approach was verified using three thorough assessments. This technique serves as a valuable asset in establishing the correct dosage and administration schedule for the combined treatment involving PAC and SEL, ensuring the desired therapeutic effects and safety in managing uterine sarcoma. Conclusions: The proposed HPLC-PDA method is the first reliable and eco-friendly method developed to simultaneously determine PAC and SEL in high-throughput plasma samples in clinical laboratories.
An Oligonucleotide Microarray for Detection of Cytochrome P450 Enzyme Gene Polymorphisms Related to Paclitaxel Metabolism

Abstract:Objective To develop an accurate,rapid,high throughout genotyping method based on oligonucleotide microarray for cytochrome P450 gene polymorphisms related to paclitaxel metabolism.Methods The mutant points of 2C8 * 3,3A4 * 18 and 3A5 * 3C from cytochrome P450 gene were regarded as targets.Based on the sequences in the GenBank,the wild-type and mutant-type probes were specially designed for each mutant point.PCR primers were located in the both sides of mutant point,and furthermore the fragments of PCR products were less than 200 bp.Each type of standard plasmids was constructed.Thus,all the olignucleotide probes were modified with 3'amino-group,and the reverse primers were labeled with fluorescein (Cy3).The probes were immobilized onto certain glass slides.The specific fragments of three genes were amplified and then hybridized with oligonucleotide microarray.The results were analyzed by using certain software.Finally this assay was applied to detect 50 clinical blood specimens.Results When PCR products from standard plasmids were hybridized with DNA microarray,the corresponding probes produced positive signals.Meanwhile,the non-specific hybridization signals did not appear.The results of clinical specimens showed that the mutant rate of CYP2C8 * 3 was 2%.The point of CYP3A4 * 18 for all the clinical specimens was wild-type and the mutant rate of CYP3A5 * 3C was 62%. Meanwhile,the results from detecting 50 clinical blood specimens using oligonucleotide microarray were the same as sequencing analysis.Conclusions Oligonucleotide microarray is a reliable and accurate genotyping assay for cytochrome P450 2C8 * 3.3A4 * 18 and 3A5 * 3C polymorphisms related to paclitaxel simultaneously.This genotyping assay is a high-throughout method for guiding personalized therapy and analyzing metabolism of paclitaxel in vivo.

Abstract:A method of high-performance liquid chromatography–tandem mass spectrometry (LC–MS-MS) has been developed for the trace analysis of paclitaxel and other six taxoids in three Taxus species including Taxus cuspidata, Taxus media and Taxus chinensis var. mairiei. Seven taxoids were separated using a gradient mode on an Eclipse XDB-C18 column (4.6×150mm; i.d., 5μm) at 20°C. The compound separations were detected by an API 3000 mass spectrometer equipped with a TurboIonSpray™ interface. The compounds were detected using electrospray ionization (ESI) in positive-ion mode and quantified by multiple-reaction monitoring (MRM) mode using the transition mass of m/z 567.4→445.4, 609.3→549.5, 944.9→286.4, 812.6→286.1, 832.8→264.1, 854.4→286.1, and 812.6→286.1 for 10-DAB III, baccatin III, 7-xyl-10-DAT, 10-DAT, cephalomannine, paclitaxel, and 7-epi-10-DAT, respectively. The ranges of limit of quantitation (LOQ) for taxoids were 32, 14, 26, 14, 20, 32, and 26ng/mL for 10-DAB III, baccatin III, 7-xyl-10-DAT, 10-DAT, cephalomannine, paclitaxel, and 7-epi-10-DAT, respectively. Linearity was confirmed over the whole calibration range (0.07–45, 0.058–37.5, 0.058–37.5, 0.056–36.3, 0.053–33.8, 0.057–37.5, and 0.06–38.8μg/mL for 10-DAB III, baccatin III, 7-xyl-10-DAT, 10-DAT, cephalomannine, paclitaxel, and 7-epi-10-DAT, respectively) with coefficients higher than 0.9903. The inter- and intra-day precision of taxoids ranged from 2.86% to 6.31% for retention time and ranged from 3.91% to 7.33% for peak area. The recovery rates of this method were higher than 94.32% for 10-DAB III, 94.68% for baccatin III, 93.65% for 7-xyl-10-DAT, 93.29% for 10-DAT, 92.91% for cephalomannine, 93.41% for paclitaxel, and 93.06% for 7-epi-10-DAT, respectively.

Abstract:Aim & Objective: This study evaluates the potential of combining paclitaxel (PTX) and bortezomib (BTZ) for breast cancer therapy. Materials & Methods: The nanoformulation was optimized via Box-Behnken Design (BBD), with method validation adhering to US-FDA guidelines. Results: Multiple reaction monitoring transitions for PTX, BTZ and internal standard were m/z 855.80→286.60, 366.80→226.00 and 179.80→110.00, respectively. Elution done on C18 Luna column with 0.1% FA in MeOH:10 mM ammonium acetate. The size of nanoformulation was 133.9 ± 1.97 nm, PDI 0.19 ± 0.01 and zeta potential -19.20 ± 1.36 mV. Pharmacokinetics showed higher Cmax for PTX-BTZ-NE (313.75 ± 10.71 ng/ml PTX, 11.92 ± 0.53 ng/ml BTZ) versus free PTX-BTZ (104 ± 13.06 ng/ml PTX, 1.9 ± 0.08 ng/ml BTZ). Conclusion: Future findings will contribute to the treatment of breast cancer using PTX and BTZ.
