Abstract:Pirarubicin (THP), doxorubicin (DOX), cyclophosphamide (CTX), and vincristine (VCR) are widely used in the treatment of patients with non-Hodgkin's Lymphoma. Herein, a precise and sensitive method was developed for the determination of THP, DOX, CTX and VCR in human plasma by high-performance liquid-chromatography-tandem mass spectrometry (LC-MS/MS). Liquid-liquid extraction was applied to extract THP, DOX, CTX, VCR, and the internal standard (IS, Pioglitazone) in plasma. Agilent Eclipse XDB-C18 (3.0 mm × 100 mm) was utilized and chromatographic separation was obtained in eight minutes. Mobile phases were composed of methanol and buffer (10 mM ammonium formate containing 0.1% formic acid). The method was linear within the concentration range of 1-500 ng/mL for THP, 2-1000 ng/mL for DOX, 2.5-1250 ng/mL for CTX, and 3-1500 ng/mL for VCR. The intra- and inter-day precisions of QC samples were found to be below 9.31 and 13.66%, and accuracy ranged from -0.2 to 9.07%, respectively. THP, DOX, CTX, VCR and the internal standard were stable in several conditions. Finally, this method was successfully utilized to simultaneously determine THP, DOX, CTX and VCR in human plasma of 15 patients with non-Hodgkin's Lymphoma after intravenous administration. Finally, the method was successfully employed in the clinical determination of THP, DOX, CTX, and VCR in patients with non-Hodgkin lymphoma after administration of RCHOP (rituximab plus cyclophosphamide, doxorubicin, vincristine, and prednisone) regimens.
Quantification of paclitaxel in rat plasma by LC-MS/MS and pharmacokinetics of paclitaxel thermosensi-tive liposomes in rats

Abstract:OBJECTIVE: To develop and validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of paclitaxel in rat plasma and to study the pharmacokinetics of paclitaxel thermosensitive liposomes in rats. METHODS: The plasma was alkalized with sodium bicarbonate (pH 8.0) and extracted with tert-butyl methyl ether (TBME). Paclitaxel and IS(docetaxel) were separated on an Agilent Zorbax SB C18 column (2.1 mm × 50 mm, 3.5 μm) with the mobile phase consisting of methanol-0.5 mmol · L-1 ammonium acetate (70: 30, V/V) at a flow rate of 300 μL · min-1. The injection volume was 5 μL An Agilent 6410 A QQQ triple-quadrople mass spectrometer equipped with an electrospray ionization (ESI) source was used as detector and was operated in positive ion mode. Selected ion monitoring (SIM) was performed and the m/z of ions selected for quantitation were 876.3 → 308.1 (paclitaxel) and 830.2 → 549.2 (internal standard, docetaxel) . SD rats were injected paclitaxel thermosensitive liposomes and paclitaxel injection via tail vein at the dose of 7 mg · kg-1, respectively. Pharmacokinetic parameters were calculated. RESULTS: The chromatograms showed no endogenous interfering peaks in blank rat plasma. Each analysis was completed within 3.5 min. The linear range of paclitaxel in plasma was 2-1 000 ng · mL-1. The lower limit of quantification was 2 ng · mL-1. The intra-run and inter-run relative standard deviations (RSDs) were less than 15%. The differences of f1/2β, MRT, AUC and CL were significant between the two preparations. CONCLUSION: The method is sensitive, accurate, and convenient for the pharmacokinetic study of paclitaxel thermosensitive liposomes. Compared with paclitaxel injection, paclitaxel thermosensitive liposomes has a controlled release rate, which can alleviate the acute toxicity and improve the tolerance to a certain extent.

Abstract:A simple, rapid, and sensitive reversed-phase HPLC method was developed and validated for determination of paclitaxel (PTX) in plasma, various organs and tumor tissues of tumor-bearing mice. Tissue specimens of liver, kidneys, spleen, lungs, heart and tumor were separately homogenized in normal saline. Plasma or tissue homogenate (250 μl) containing PTX and internal standard (diazepam) were extracted by diethyl ether (6 ml). The separation was achieved on a μ-Bondapak C18 HPLC column using sodium acetate buffer solution (0.01 M)/acetonitrile (58/42 v/v) at pH 5 ± 0.1 and flow rate of 1.9 mL/min. The effluent was monitored at 227 nm and column temperature was adjusted at 58oC. The internal standard and PTX were eluted at 4.2 and 5.2 min, respectively and no interfering peaks were observed. Calibration curves were linear over the concentration range of 0.25-10 μg/ml of PTX in plasma and 0.3-20 μg/ml PTX in tissue homogenates with acceptable precision and accuracy (<15%). The mean recoveries of the drug after plasma extraction was 87.4% ± 3.6 while those of tissue homogenates ranged from 62.1± 4.5 to 75.5± 3.2 depending on the type of tissues studied. PTX was stable in samples with no evidence of degradation during 3 freeze–thaw cycles and 3 months storage at −70 °C. The developed HPLC method was applied to quantify PTX in the mouse plasma and tissues after intravenous administration of 10 mg equivalent PTX/Kg dose of PTX-loaded tocopherol succinate-chitosan-polyethylene glycol-folate (TS-CS-PEG-FA) micelles formulation or Anzatax® (Cremophor® EL- based formulation of PTX) to female Balb/c mice. Abstract:The prominent stromal compartment surrounds pancreatic ductal adenocarcinoma (PDAC) protects the tumor cells from chemo- or radio-therapy. We hypothesized that our nano formulation carried cyclopamine (CPA, stroma modulator) and paclitaxel (PTX, anti-tumor agent) could increase the permeation of PTX through the stromal compartment and improve the intratumoral delivery of PTX. In the present study a sensitive, reliable UPLC-MS/MS method was developed and validated to quantify PTX and CPA simultaneously in mouse whole blood, pancreas, liver and spleen samples. Docetaxel was used as the internal standard. The method demonstrated a linear range of 0.5-2000 ng/mL for whole blood and tissue homogenates for both PTX and CPA. Accuracy and precision of the assay were all within ±15 %. Matrix effects for both analytes were within 15%. Recoveries from whole blood; liver, spleen and pancreas homogenates were 92.7-105.2% for PTX and 72.8-99.7% for CPA. The stability was within ±15% in all test biomatrices. The validated method met the acceptance criteria according to FDA regulatory guidelines. The method was successfully applied to support pharmacokinetic and biodistribution study for PTX and CPA in mice biomatrices.

Abstract:A sensitive and selective liquid chromatographic–tandem mass spectrometric (LC–MS/MS) method for the determination of paclitaxel (Taxol) and its two major metabolites in human plasma has been developed. Samples were prepared after liquid–liquid extraction and analyzed on a C18 column interfaced with a Q-Trap tandem mass spectrometer. Positive electrospray ionization was employed as the ionization source. The mobile phase consisted of acetonitrile–water (0.05% formic acid) (65:35) at the flow rate of 0.25 mL/min. The analytes and internal standard docetaxel were both detected by use of multiple reaction monitoring mode. The method was linear in the concentration range of 0.5–500.0 ng/mL for paclitaxel, 6α-hydroxypaclitaxel and p-3′-hydroxypaclitaxel, respectively. The lower limit of quantification (LLOQ) was 0.5 ng/mL for paclitaxel, 6α-hydroxypaclitaxel and p-3′-hydroxypaclitaxel, respectively. The intra- and inter-day relative standard deviation across three validation runs over the entire concentration range was less than 8.18%. The accuracy determined at three concentrations was within ±10.8% in terms of relative error. The total run time was 7.0 min. This assay offers advantages in terms of expediency, and suitability for the analysis of paclitaxel and its metabolites in various biological fluids.

Abstract:Lysimachia capillipes Hemsl (Primulaceae), a folk medicinal plant in China, showed significant anti-tumor activities in vivo and in vitro. Capilliposide B (LC-B) and capilliposide C (LC-C) are the main bioactive components in this plant. To explore their tissue distribution, a reliable bioanalytical method for the quantification of LC-B, LC-C and their bioactive metabolite, capilliposide A (LC-A), in mouse tissues was developed and validated. In this study, the tissue distribution profiles of the three compounds were examined after intravenous administration of pure LC-B and oral administration of total saponins of L. capillipes Hemsl extract (LCE) for 10days. Method validation was conducted over the curve range 10.0-5000ng/mL for all three analytes in various tissue homogenates. The relative standard deviation of intra-day and inter-day precision of the QC samples was <14.7%, and the accuracy ranged from 85.9 to 114.0%. The results indicated that LC-B was rapidly and widely distributed throughout the whole body except for muscle following intravenous administration of LC-B. In addition, LC-A was only in liver, intestine, lung and stomach. After oral administration of LCE, LC-B and LC-C were distributed into various tissues. The highest levels were observed in stomach and intestine.
The Development of a Quantitative and Qualitative Method Based on UHPLC-QTOF MS/MS for Evaluation Paclitaxel–tetrandrine Interaction and Its Application to a Pharmacokinetic Study

Abstract:Paclitaxel is a broad-spectrum anti-cancer drug by targeting microtubulin. However, multidrug resistant (MDR) makes its clinical application more difficult and results in failure of chemotherapy. Tetrandrine as a potential multidrug resistant modulator could be combined with other anti-cancer drugs. In this study, ultra-performance liquid chromatography (UHPLC) combined with quadrupole time-of-flight mass spectrometry (QTOF) was applied to simultaneously qualitative and quantitative analysis of paclitaxel for the pharmacokinetic studies while combined with tetrandrine. This method was developed based on non-target screening mode IDA (Information Dependent Acquisition). As a result, the validated range was 0.25-64ng/ml (30µl plasma) for paclitaxel. Totally 33 metabolites of paclitaxel and tetrandine were identified in vivo and in vitro. The main metabolites of PTX were dose-dependent decreased with different amounts of tetrandine co-administration no matter in vivo and in vitro, the exposure of PTX increased in pharmacokinetic study. The verified method is sensitive accurate and effective for the simultaneous determination of paclitaxel and its metabolites in blood, urine and live microsome incubation samples and it was successfully applied to evaluate the pharmacokinetics and drug-drug interaction between paclitaxel and tetrandine. Furthermore, a biosensor technology, surface plasmon resonance (SPR) analysis was applied to preliminary evaluate the competitive protein binding of multiple components. The SPR analysis indicated that the affinity between 6-hydroxy-paclitaxel and micotubulin is similar to that between paclitaxel and micotubulin, and tetrandrine also does not form a competitive combination with paclitaxel. For human, 6-hydroxy-paclitaxel is the one of main metabolites of paclitaxel, so the results suggested that tetrandine has an influence on the metabolite of paclitaxel, but tetrandine and the main metabolites of PTX probably do not affect PTX's biological targeting, the effect of its pharmacological action needs to be further studied.

Abstract:ObjectiveTo establish a liquid chromatography-tandem mass spectrometry(LC-MS/MS)for the quantitative determination of plasma paclitaxel,and to evaluate its performance.Methods Plasma paclitaxel was determined quantitatively by LC-MS/MS. High performance liquid chromatography was used. The flow speed was 0.3 mL/min. Multiple reaction monitoring(MRM) was used for quantitative determination. The ion pairs of paclitaxel and PTX-D5 were 876.4>308.2 and 881.5>313.2,respectively. According to U.S. Food and Drug Administration(FDA) guideline about bioanalytical method validation,the general analytical performance validation was performed for linearity,detection limit,precision and accuracy.ResultsThe linearity was 10-1 000 ng/mL. The detection limit was 10 ng/mL. Within-run and between-run coefficients of variation(CV) were≤4.2% and≤8.9%,respectively. The accuracy was 88.7%-104.0%.Conclusions The performance of established LC-MS/MS meets evaluation standards,and the assay is accurate and precise for the determination of paclitaxel.

Abstract:A sensitive and selective method has been developed and validated for the quantitation of paclitaxel in human plasma by liquid chromatography-eleetrospray ionization mass spectrometry (LC-ESI-MS).Paclitaxel and norethindrone (used as internal standard,I.S.) were extracted from human plasma by a one-step liquid-liquid extraction with t-butyl methyl ether.Separation on a Zorbax SB-C18 column (100 mm×2.1 mm,3.5 μm,Agilent) was achieved by gradient elution with methanol and 0.2 mmol/L ammonium formate containing 0.1％ formic acid.The selected-ion monitoring (SIM) targeted ions of [M+Na]+ at m/z 876.5 for paclitaxel and [M+H](+) at m/z 299.4 for I.S.The assay was validated in the range of 1.0-400 ng/mL (r＞0.998) with LLOQ of 1.0 ng/mL.Intra- and inter-day precisions were all less than 9.0％,with accuracies of±6.8％.The method was successfully applied to evaluate the pharmacokinetics of paclitaxel liposome for injection in patients.

Abstract:The development of improved or targeted drugs that discriminate between normal and tumor tissues is the key therapeutic issue in cancer research. However, the development of an analytical method with a high accuracy and sensitivity to achieve quantitative assessment of the tumor targeting of anticancer drugs and even intratumor heterogeneous distribution of these drugs at the early stages of drug research and development is a major challenge. Mass spectrometry imaging is a label-free molecular imaging technique that provides spatial-temporal information on the distribution of drugs and metabolites in organisms, and its application in the field of pharmaceutical development is rapidly increasing.Methods: The study presented here accurately quantified the distribution of paclitaxel (PTX) and its prodrug (PTX-R) in whole-body animal sections based on the virtual calibration quantitative mass spectrometry imaging (VC-QMSI) method, which is label-free and does not require internal standards, and then applied this technique to evaluate the tumor targeting efficiency in three treatment groups—the PTX-injection treatment group, PTX-liposome treatment group and PTX-R treatment group—in nude mice bearing subcutaneous A549 xenograft tumors.Results: These results indicated that PTX was widely distributed in multiple organs throughout the dosed body in the PTX-injection group and the PTX-liposome group. Notably, in the PTX-R group, both the prodrug and metabolized PTX were mainly distributed in the tumor tissue, and this group showed a significant difference compared with the PTX-liposome group, the relative targeting efficiency of PTX-R group was increased approximately 50-fold, leading to substantially decreased systemic toxicities. In addition, PTX-R showed a significant and specific accumulation in the poorly differentiated intratumor area and necrotic area.Conclusion: This method was demonstrated to be a reliable, feasible and easy-to-implement strategy to quantitatively map the absorption, distribution, metabolism and excretion (ADME) of a drug in the whole-body and tissue microregions and could therefore evaluate the tumor-targeting efficiency of anticancer drugs to predict drug efficacy and safety and provide key insights into drug disposition and mechanisms of action and resistance. Thus, this strategy could significantly facilitate the design and optimization of drugs at the early stage of drug research and development.

An Improved LC-MS/MS Method for Determination of Docetaxel and Its Application to Population Pharmacokinetic Study in Chinese Cancer Patients.

Abstract:Owing to its unpredictable side effects and efficacy, the anticancer drug docetaxel requires improved characterisation of its pharmacokinetic profiles through population pharmacokinetic (PPK) studies. A sensitive and rugged liquid chromatography-tandem mass spectrometry method for the detection of docetaxel in human plasma was developed and optimised using paclitaxel as an internal standard (IS). The plasma samples underwent rapid extraction using Hybrid solid-phase extraction-protein precipitation. The analyte and IS were separated with an isocratic system on a Zorbax Eclipse Plus C18 column using water containing 0.05% acetic acid along with 20 μM sodium acetate and methanol (30/70, v/v) as the mobile phase. Quantification was performed with a triple quadrupole mass spectrometer through multiple reaction monitoring in positive mode, using the m/z 830.3→548.8 and m/z 876.3→307.7 transitions for docetaxel and paclitaxel, respectively. The range of the calibration curve was 1-500 ng/mL for docetaxel and the linear correlation coefficient was greater than 0.99. The accuracies ranged from -4.6% to 4.2%, and the precision was no higher than 7.0% for the analytes. No significant matrix effect was observed. Both docetaxel and the IS showed considerable recovery. This method was finally applied to the establishment of a PPK model to optimise the clinical use of docetaxel.

Abstract:One of the major obstacles to successful paclitaxel (PTX) chemotherapy is toxic side effects, which are often due to the conventional surfactants used, such as Cremophor EL. PTX is characterized by its hydrophobicity and insolubility, which limit its application in ovarian cancer therapy. The aim of this study was to develop Cremophor EL-free PTX-loaded methoxy poly(ethylene glycol)-block-(lactic-co-glycolic acid) copolymers (PLGA-mPEG) nanoparticles (NPs) using d-alpha-tocopheryl polyethylene glycol 1000 succinate (TPGS) as a novel emulsifier. The ability of nanoparticles loaded with paclitaxel (NP-PTX) to inhibit tumor growth was assessed in vitro and in vivo. The acute toxicity of NP-PTX was also evaluated in vivo. We found that paclitaxel was efficiently encapsulated into PLGA-mPEG NPs with a low concentration of TPGS as the emulsifier. The synthesized NP-PTX demonstrated the desired diameter of 80 nm as characterized by transmission electron microscopy. The NP-PTX also exhibited a sustained release of loaded PTX over 4 d with the same chemotherapeutic efficiency and reduced side effects. NP-PTX-treated cells showed slightly lower cytotoxic responses compared with those treated with free PTX at the same concentration. In vivo studies confirmed that NP-PTX significantly enhanced the median lethal dose of paclitaxel by 10-fold, and a similar effect on the inhibition of tumor growth was achieved in nude mice.

Abstract:Objective To prepare the thermosensitive liposomes containing paclitaxel(PTL)and study their anticancer effect.Methods Both PTL and paclitaxel liposomes(PL) were prepared by the thin-film method.The HPLC method was established to determine paclitaxel concentration in vivo and in vitro.The model of mice Lewis sarcoma was established and divided into four groups randomly:control,PTX,PL and PTL groups.Each group was administrated with drug and local hyperthermia for 30 min.The volume and weight of tumor were detected.Results The mean particle size of the liposomes ranged 70-140 nm and the drug entrapment efficiency was about 99%.The phase transition temperature of PTL was about 42℃.In vivo,the study of thermosensitive characteristics of PTL indicated that drug content in heated location was significantly higher than that of the unheated.The inhibition ratio of tumor in PTX,PL and PTL groups was 49.79%,57.40% and 75.66%,respectively.Conclusion Compared with PTX and PTL,PTL has an evident thermosensitive characteristic and increases the anticancer effect of paclitaxel remarkably when combined with hyperthermia.

In the present study, a rapid and specific HPLC-MS/MS method was developed and validated to simultaneously determine gefitinib, M387783, M523595, M537194 and M608236 in tumor tissues of H1975 human lung cancer xenografts of nude mice. The established HPLC-MS/MS method was validated for specificity, linearity, accuracy and precision, matrix effect and recovery, carryover and dilution integrity, and analyte stability. The standard curves were linear (r(2) >= 0.99) over the range of 0.5-100 ng/mL for M608236 and 1-200 ng/mL for gefitinib, M523595 and M537194 as well as M387783. The accuracy ranged from -8.35 to 6.03% relative error; and the precision was <15% relative standard deviation. Recoveries (87.74-99.96%) and matrix effects (86.60-106.40%) were satisfactory in the biological matrix examined. Stability studies showed that the analytes were stable during the assay procedure and storage. Finally, the validated method was successfully applied to study the pharmacokinetics profiles for gefitinib and its metabolites in nonsmall cell lung cancer (NSCLC) xenograft mouse tumors. Meanwhile, MTT assay showed that gefitinib had a more powerful inhibitory effect than its four major metabolites in H1975 NSCLC cells. This validated HPLC-MS/MS method may be applied to help understand the mechanisms of gefitinib resistance in EGFR-mutant nonsmall cell lung cancer.
Optimization of mobile phase composition in development of an LC-MS/MS method for quantitation of paclitaxel and docetaxel

Abstract:Objective: To investigate and compare the ionization behaviors of paclitaxel and docetaxel by exposing to various additives in two sampling ways: direct infusion or by liquid chromatograph into mass spectrometer. Methods: The working solutions of paclitaxel and docetaxel, mixed with different additives, were led into the electrospray ionization source, via a syringe pump or via the liquid chromatographic system, for detection and collection of mass spectra or chromatograms of the analytes.The influence of additives, with various sorts, concentrations and pH, in solutions or mobile phases on the ionization efficiency of paclitaxel and docetaxel were studied. Results: Similar patterns of mass spectra intensity variations were observed between these two sampling methods, in that the highest ionization efficiency and strongest signal intensity were obtained when the analytes were exposed to the additive as 1 mmol/L ammonium formate (pH=6.5) in both methods. Conclusions: Direct infusion of samples into mass spectrometer is an efficient, simple and reliable way to optimize the mobile phase composition in the development of liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for quantitation of paclitaxel and docetaxel.Based on these observations, a sensitive and reproducible LC-MS/MS method for the determination of paclitaxel in human plasma was successfully established and validated.
A Validated UHPLC-MS/MS Method for Determination of TQ-B3203 in Human Plasma and Its Application to a Pharmacokinetic Study in Chinese Patients with Advanced Solid Tumor

Abstract:TQ-B3203 is a new topoisomerase I inhibitor derived from camptothecin. In this paper, a simple and reliable ultra high-performance liquid chromatography-tandem mass spectrometric method was developed and validated for determination of TQ-B3203 in human plasma with TQ-B3203-d(8) used as the internal standard. Bis(p-nitrophenyl)phosphate (2 mol/L) was added to ensure the stability of TQ-B3203 in human plasma. Plasma samples were protein precipitated by methanol and processed samples were chromatographed on an AQUITY BEH C-8 column (50 x 2.1 mm, id 1.7 mu m) with acetonitrile and water (0.1% formic acid) as the mobile phase. The calibration curves showed good linearity (R >= 0.99) over the concentration range of 0.5-500 ng/mL. Within- and between-run precision were <= 5.8% and the accuracy was within the range of -8.3 to 14.0%. This method was further successfully applied to a pharmacokinetic study of TQ-B3203 in Chinese advanced solid cancer patients after administration of TQ-B3203 liposome injection.

Abstract:Vitamin-D analogues have emerged as potential stroma-modulating agents for the treatment of pancreatic ductal adenocarcinoma (PDAC). One such agent, calcipotriol (Cal) has shown significant activity in in vitro and in vivo models of pancreatic ductal adenocarcinoma. Attempts in our lab have been focused on establishing the therapeutic merits of co-formulating this agent with the chemotherapeutic drug paclitaxel (PTX) in animal models. Accurate and reliable quantifications of these agents is critical to successful pharmacokinetic/pharmacodynamic (PK/PD) projections from animals into humans. Herein, we developed a LC–MS/MS assay for measuring Cal and PTX in whole blood and plasma.A liquid-liquid analyte extraction procedure, using a mixture of water-MeOH (50:50, v/v) and hexane-dichloromethane- isopropyl alcohol (150:15:5, v/v/v) was used. Chromatographic separation was carried out on Kinetex C18 column (1.7 μm, 100 × 2.10 mm) under an isocratic elution at a flow rate of 0.4 mL/min with a total runtime of 3.5 min. The mobile phase was composed of ammonium acetate (pH 6.51; 5 mM)-methanol (15:85, v/v). The analytes were ionized by positive electrospray ionization using API 5500-Qtrap triple quadrupole mass spectrometer (Applied Biosystem/AB SCIEX). The linearity of calibration curves for both analytes were established at 0.5 (LLOQ)-500 ng/mL with correlation coefficients exceeding 0.99. Spiked whole blood and plasma samples were used as surrogates for matrix validation. For both analytes, the intra-day and inter-day accuracies were 90.5–105 % and 96.6–106 %, respectively, while the corresponding precisions were 3.09–10.7 % and 5.20–12.9 %. No carryover was observed for the analytes which also remained acceptably stable in the surrogate matrices under relevant conditions.The assay is robust, reliable and sensitive in rat whole blood and plasma. The analytes extraction procedure performs acceptably well in both matrices with high recoveries and minimal matrix effects. Additionally, only 20 μL of rat whole blood or plasma is required and the total run time per sample is 3.5 min.PK studies enabled by the assay revealed that when co-administered, PTX AUC0→∞ and Cmax increased while those of Cal decreased. This finding alerts a potential drug-drug interaction and warrants further investigation in studies using this combination regimen.

Abstract:European Journal of Mass Spectrometry, Ahead of Print. Present work describes the development of a liquid chromatography tandem mass spectrometry-based bioanalytical method for the reliable simultaneous quantification of docetaxel (DXL) and carvacrol (CVL) in the mice plasma. A rapid and sensitive bioanalytical method was developed and optimized in mice plasma using Paclitaxel as an internal standard. Validation of the bioanalytical method was performed according to the ICH M10 guideline covering the range of 9.62–1923.08 ng/mL in the mice plasma milleu at the low, mid, and high-quality control concentrations of 28.86 ng/mL, 961.54 ng/mL, and 1346.15 ng/mL, respectively for both the analytes. Validation parameters such as accuracy, precision, carryover-test, matrix effect, and reinjection reproducibility were carried out and were found in limits. Stability studies (Benchtop, autosampler, freeze-thaw, and long-term) were performed and found to be within limits. The developed bioanalytical method was found to be suitable for the simultaneous quantification of DXL and CVL in the mice plasma.
