Abstract:New biosensors based on electrochemical impedance spectroscopy (EIS) were developed for detection of Escherichia coli O157:H7 through the surface immobilization of monoclonal antibody onto indium tin oxide (ITO) electrode. The biosensor could detect the target bacteria with a detection limit of 4×10 3 CFU/mL. A linear response in the electron-transfer resistance for the concentration of E.coli cells was found between 4×10 3 and 4×10 6CFU/mL. The biosensor characterizes with high sensitivity, simple operation, enabling relatively rapid and reliable analysis. The result proved the prospect of EIS biosensors on application to biomedical and environmental detection of E.coli O157:H7.

Abstract:Immunobiosensors were developed for detection of Escherichia coli O157:H7 based on the surface immobilization of monoclone antibodies onto indium tin oxide (ITO) electrodes. The immobilization of antibodies onto ITO chips was carried out by silanization. The effects of epoxysilane monolayer, the antibody layer on the electrochemical properties of the electrode, and the combined target bacteria were analyzed through cyclic voltammetry and electrochemical impedance spectroscopy. By using Randles model as the equivalent circuit, the concentration of the target bacteria can be quantitatively analyzed in terms of the change of electron transfer resistance. The biosensor could detect the target bacteria with a detection limit of 4times103 CFU/mL. A linear response was found between 4times1034times106 CFU/mL. This biosensor was characterized with high sensitivity, excellent selectivity, short detection time and easy operation. It has a promising application in clinical laboratory diagnoses, environmental detection and food safety

Abstract:The two-dimensional porphyrin-based covalent organic framework (denoted by Tph-TDC-COF) was used as the sensitive layerto build an aptamer-based electrochemical sensor for the detection of Escherichia coli (E.coli). Tph-TDC-COF produced with 5,10,15,20-tetrakis(4-aminophenyl)-21H, 23H-porphine (Tph) and [2,2′-bithiophene]-2,5′-dicarbaldehyde (TDC) as building blocks exhibited a highly conjugated structure, outstanding conductivity, large specific surface area, and strong bioaffinity towards aptamers. The adoption of Tph-TDC-COF-modified electrode resulted in improved sensing performance and increased anchoring affinity toward the E.coli-targeted aptamer. Under optimal conditions, the Tph-TDC-COF-based electrochemical aptasensor demonstrated an extremely low detection limit of 0.17 CFU mL−1 for E.coli detection within a linear range of 10 to 1 × 108 CFU mL−1, accompanied by good stability, excellent reproducibility and regeneration ability, and wide practical applications. The current electrochemical aptasensing technique has the potential to be extended to detect different foodborne bacteria using specific aptamer, therefore widening the application of COFs in biosensing and food safety fields.

Abstract:Foodborne diseases caused by Salmonella enterica ( S. enterica ) and Staphylococcus aureus ( S. aureus ) significantly impact public health, underscoring the imperative for highly sensitive, rapid, and accurate detection technologies to ensure food safety and prevent human diseases. Nanomaterials hold great promise in the development of high-sensitivity transistor biosensors. In this work, field-effect transistor (FET) comprising high-purity carbon nanotubes (CNTs) were fabricated and modified with corresponding nucleic acid aptamers for the high-affinity and selective capture of S. enterica and S. aureus . The aptamer-functionalized CNT-FET biosensor demonstrated ultra-sensitive and rapid detection of these foodborne pathogens. Experimental results indicated that the biosensor could detect S. enterica at a limit of detection (LOD) as low as 1 CFU in PBS buffer, and S. aureus at an LOD of 1.2 CFUs, achieving single-cell level detection accuracy with exceptional specificity. The biosensor exhibited a rapid response time, completing single detections within 200 s. Even in the presence of interference from six complex food matrices, the biosensor maintained its ultra-sensitive (3.1 CFUs) and rapid response (within 200 s) characteristics for both pathogens. The developed aptamer-functionalized CNT-FET biosensor demonstrates a capability for low-cost, ultra-sensitive, label-free, and rapid detection of low-abundance S. enterica and S. aureus in both buffer solutions and complex environments. This innovation holds significant potential for applying this detection technology to on-site rapid testing scenarios, offering a promising solution to the pressing need for efficient and reliable pathogen monitoring in various settings.

Abstract:Escherichia coli O157:H7 is a predominant foodborne pathogen with severe pathogenicity, leading to increasing attention given to rapid and sensitive detection. Herein, we propose an impedance biosensor using new kinds of screen-printed interdigitated microelectrodes (SPIMs) and wheat germ agglutinin (WGA) for signal amplification to detect E. coli O157:H7 with high sensitivity and time-efficiency. The SPIMs integrate the high sensitivity and short response time of the interdigitated electrodes and the low cost of the screen-printed electrodes. Self-assembling of bi-functional 3-dithiobis-(sulfosuccinimidyl-propionate) (DTSP) on the SPIMs was investigated and was proved to be able to improve adsorption quantity and stability of biomaterials. WGA was further adopted to enhance the signal taking advantage of the abundant lectin-binding sites on the bacteria surface. The immunosensor exhibited a detection limit of 102 cfu·mL(-1), with a linear detection range from 10(2) to 10(7) cfu·mL(-1) (r2 = 0.98). The total detection time was less than 1 h, showing its comparable sensitivity and rapid response. Furthermore, the low cost of one SPIM significantly reduced the detection cost of the biosensor. The biosensor may have great promise in food safety analysis and lead to a portable biosensing system for routine monitoring of foodborne pathogens.

Abstract:Eating food contaminated by foodborne pathogens can lead to illness. The development of electrochemical sensors for pathogen detection has received widespread attention. However, the analytical performance of electrochemical sensors is inevitably affected by the non-specific adsorption of molecules in the sample. Moreover, the external signal probes might be affected by the complex components in the sample accompanied with signal suppression. This work presents an electrochemical aptasensor for Salmonella typhimurium detection based on the self-signal of poly-xanthurenic acid and the antifouling ability of chondroitin sulfate. The detection time was 60 min. The linear range was from 101 to 107 CFU/mL, and the detection limit was 3 CFU/mL. The biosensors presented good repeatability and storage stability. And the biosensors has been successfully applied in milk and orange juice. This strategy is expected to be applied in the design of other antifouling biosensors, to achieve rapid detection of pathogens and ensure food safety.

Abstract:The foodborne pathogens have caused many life-threatening diseases. Herein, a new electrochemical platform was developed for detection, using 3D walking machine and enzyme-free toehold-mediated strand displacement. High-variable region of 16S rDNA fragment of Escherichia coli ( E. coli ) was used as the target to verify the approach. In presence of target, Exonuclease III (Exo III)-powered hairpin-loaded Au nanoparticles () was aroused, which emerged large amounts of 3D toehold-loaded AuNPs () walking machine. The "legs" of the walking machine hybridized with hairpin2 (HP2) tracks probe modified on surface of electrode and exposed toehold region of HP2 probe. The toehold-mediated strand displacement triggered rolling of AuNPs along the electrode, through the interaction of hairpin3-F c (HP3-F c ) probe with toehold region of HP2 probe. Thus large amounts of F c were closed to the electrode and led to significant current responses for E. coli detection. The method could continuously recycle low-concentration targets. Furthermore, due to the increase of "legs" and decrease of the derailment of leg DNA, which was beneficial to shorten detection time and amplify response. The detection limits (LOD) was 20 CFU/mL, the detection time was less than 1.5 h. This method was promising to be applied in early diagnosis of foodborne pathogen.

Abstract:Foodborne pathogens are harmful to human health because they can contaminate food and induce diseases. To efficiently distinguish and determine foodborne bacteria, an ultrasensitive point-of-care electrochemical biosensor was designed for 16S rRNA detection by coupling a signal amplification strategy with MoS2-based nanoprobes. Gold nanoparticles and thionine co-functionalized molybdenum disulfide (MoS2) nanocomposites (MoS2-Thi-AuNPs) were used to construct nanoprobes, which can efficiently monitor the detection process and amplify the detection signal. In the presence of Escherichia coli (E. coli) 16S rRNA, a classical sandwiched DNA structure was formed on the surface of a hierarchical flower-like gold nanostructure-decorated screen-printed carbon electrode (HFGN-SPCE), generating an obvious electrochemical signal from Thi. Under optimal conditions, this designed electrochemical biosensor showed a wide dynamic range (0-1.0 × 106 fM), low detection limit (2.8 fM), high selectivity and accepted stability for E. coli 16S rRNA detection in ideal buffers. Moreover, this biosensor can efficiently determine 16S rRNA in milk samples and 10 CFU mL-1 bacterial lysate. All experimental results suggested that this biosensor has a promising application in the detection of foodborne pathogens.

Abstract:Rapid detection of foodborne pathogens plays a more and more significant role in the field of public health. In this research, we developed a convenient electrochemical biosensor mediated by filtration-assisted pretreatment and multiple thermostatic enzymes (Multi-TEs) amplification system for the detection of Staphylococcus aureus ( S. aureus ) in complex matrices. Firstly, the designed filtration-assisted pretreatment could simultaneously collect and enrich target bacterium from various actual samples within 5 min. Then, the target bacterial genome template was reacted at 37 °C in Multi-TEs amplification system to obtain specific double stranded products. Finally, the specific double stranded products were fixed on the surface of the electrode and the electrochemical indicators (Methylene blue, MB) was combined for the quantitative one-step electrochemical analysis of S. aureus . This assay has great linear calibration curves with the concentration ranging from 10 1 CFU/mL to 10 6 CFU/mL. The limit of detections (LOD) was defined as 1.28 CFU/mL, which performance is superior to some commercial ELISA kits. Meanwhile, amazingly, it has excellent specificity for S. aureus against other foodborne pathogens and could be applied for S. aureus analysis in human serum samples with the great recoveries and associated relative standard deviations (RSDs). These features do make the ideal facile electrochemical analytical platform for on-site detection of S. aureus.

Abstract:A signal amplification electrochemical biosensor chip was developed to integrate loop-mediated isothermal amplification (LAMP) based on in situ nucleic acid amplification and methyl blue (MB) serving as the hybridization redox indicator for sensitive and selective foodborne pathogen detection without a washing step. The electrochemical biosensor chip was designed by a screen-printed carbon electrode modified with gold nanoparticles (Au NPs) and covered with polydimethylsiloxane membrane to form a microcell. The primers of the target were immobilized on the Au NPs by covalent attachment for in situ amplification. The electroactive MB was used as the electrochemical signal reporter and embedded into the double-stranded DNA (dsDNA) amplicons generated by LAMP. Differential pulse voltammetry was introduced to survey the dsDNA hybridization with MB, which differentiates the specifically electrode-unbound and -bound labels without a washing step. Pyrene as the back-filling agent can further improve response signaling by reducing non-specific adsorption. This method is operationally simple, specific, and effective. The biosensor showed a detection linear range of 102-107 CFU mL-1 with the limit of detection of 17.7 CFU mL-1 within 40 min. This method showed promise for on-site testing of foodborne pathogens and could be integrated into an all-in-one device.
Aptamer-functionalized polydiacetylene biosensor for the detection of three foodborne pathogens

Abstract:Rapid, simple and sensitive screening of foodborne pathogens is of great significance to ensure food safety. In this study, an aptamer-functionalized polydiacetylene (Apta-PDA) biosensor was developed for the detection of E. coli O157:H7, S. typhimurium or V. parahaemolyticus. First, aptamers responding to the target bacteria were modified on the surface of magnetic beads by covalent binding to form MBs-oligonucleotide conjugates for bacterial enrichment. Then, an Apta-PDA biosensor was obtained by connecting the aptamers to the PDA nanovesicles using the carbodiimide method. Molecular recognition occurred in the presence of the target bacteria, whereby the aptamer folded into a sequence-defined unique structure, resulting in an MBs-Apta/bacteria/Apta-PDA sandwich structure. Due to the optical properties of PDA, the blue-red transition of the detection system could be observed by the naked eye and quantified by the colorimetric response percentage (CR%). Under optimized conditions, the detection limits of E. coli O157:H7, S. typhimurium and V. parahaemolyticus were 39, 60 and 60 CFU/ml, respectively, with a selectivity of 100% and a reaction time of 30 min. Compared with the gold standard method, the accuracy of the three target bacteria detection reached 98%, 97.5% and 97%, respectively, and the sensitivity and specificity were both greater than 90%. The entire detection process was rapid and easy to execute without any special equipment, making this technology particularly suitable for resource-poor laboratories or regions.

Abstract:Staphylococcus aureus (S. aureus) is one of the most common foodborne pathogens worldwide, which poses a great threat to public health. It is of utmost importance to develop rapid, simple, and sensitive methods for the determination of S. aureus. A signal-on photoelectrochemical (PEC) aptasensor is constructed herein based on titanium carbide (Ti3C2Tx)-Au nanobipyramids (NBPs)/ZnO nanoarrays (NRs). The reliability and capability of the PEC aptasensor make it suitable for the sensitive and selective determination of S. aureus. First, the electrostatically self-assembled Ti3C2Tx-Au NBP nanomaterial was coated on the ZnO NR surface by a spin-coating method. On the one hand, Ti3C2Tx-Au NBPs can broaden the spectral absorption of ZnO NRs, resulting in Ti3C2Tx-Au NBPs/ZnO NR composites that exhibit a wide range of absorption from the ultraviolet to the infrared region. On the other hand, Ti3C2Tx can reduce the agglomeration of nanoparticles, while Au NBPs can effectively fix the aptamer through the Au-S bond. Specifically, the experimental results show that when S. aureus is present, the Au NBPs-aptamer-S. aureus complex is shed from the electrode surface, altering the interfacial electron transfer model and reducing the steric hindrance. Consequently, an amplified photocurrent signal for the quantitative determination of S. aureus is obtained. Under optimal experimental conditions, a linear correlation is observed between the current response of the aptasensor and the logarithm of the S. aureus concentration (ranging from 1.0 to 1.0 × 106 CFU/mL), with an impressive detection limit as low as 0.5 CFU/mL. Furthermore, the aptasensor has been successfully employed for the detection of S. aureus in milk, with the recovery of 93.0%-99.0%. Hence, this research offers a novel approach for the detection of foodborne pathogens and other noxious substances.
A dual-mode aptasensor for foodborne pathogens detection using Pt, phenylboric acid and ferrocene modified Ti3C2 MXenes nanoprobe

Abstract:Foodborne pathogens could result in serious health issues to public and thus some rapid analysis methods are urgent developed to monitor them. Herein, a dual-mode aptasensor based on electrochemical and colorimetric modes was constructed for the on-site detection of Vibrio parahaemolyticus (V.P) in shrimps as a model. A novel nanoprobe (@MXenes) was developed to generate both electrochemical and colorimetric signals by modifying phenylboronic acid and ferrocene on Pt-MXenes nanocomposite. The nanoprobe exhibited both features of peroxidase mimic and specific recognition. Simultaneously, a V.P aptamer-functionalized electrode interface was fabricated to firstly specifically capture V.P, then conjugated with @MXenes to form a sandwich complex. The complex could catalyze tetramethylbenzidine (TMB)-H2O2 to produce a visible signal, and generate electrochemical signal simultaneously. Under the optimal condition, the aptasensor could determine as low as 5 CFU mL−1 V.P by electrochemical mode and 30 CFU mL−1 V.P by colorimetric mode. These superior performances may be attributed to that the synergistic catalysis and conductive effects deriving from Pt and MXenes on the nanoprobe can significantly amplify the signals to improve detection sensitivity. In particular, the dual detection system could mutually verify to achieve high accuracy. The aptasensor could also be expanded to rapid analysis of other food-borne pathogens through changing the aptamer on the electrode.

Abstract:Foodborne pathogens can cause serious food safety problems. Therefore, developing fast, sensitive, and economic strategies for the determination of foodborne pathogens have become of great significance for the food safety and clinical diagnosis of foodborne diseases. Compared with the traditional methods with time-consuming, inefficient, and expensiveness, the electrochemical biosensors have the advantages of simplicity, rapidity, high sensitivity, miniaturization, and low cost, which are more conducive to rapid detection of foodborne pathogens. For the past few years, carbon nanomaterials have been frequently used to construct electrochemical-based foodborne pathogens biosensors attribute to their high conductivity, stability, and biocompatibility. Herein, this review summarizes the challenges and opportunities faced by the application of electrochemical biosensors based on carbon nanomaterials for monitoring foodborne pathogens in the recent years (2015–2020), which will open up new horizons for their development in foodborne pathogens detection.

Abstract:Based on the principle of antigen-antibody and microbial immunosensor using TiO2 nanowire bundle as sensitive material,a rapid food pathogen detection integrated system is designed and realized.The impedance detection circuit adopts a highly integrated chip AD5933 to measure TiO2 nanowire bundle impedance variation with method of electrochemical impedance spectroscopy.E.coli are used as an experimental sample for testing the performance of detection integrated system.The system can rapid detect the E.coli in food in one hour,and the detection limit can reach 4.5×102 cfu/mL.The immunosensor of system shows excellent reproducibility between frequency of 1 000 Hz~10 000 Hz.
Nanozyme-Mediated Catalytic Signal Amplification for Microfluidic Biosensing of Foodborne Bacteria.

Abstract:Early detection of foodborne bacteria is urgently needed to ensure food quality and to avoid the outbreak of foodborne bacterial diseases. Here, a kind of metal-organic framework (Zr-MOF) modified with Pt nanoparticles (Pt-PCN-224) was designed as a peroxidase-like signal amplifier for microfluidic biosensing of foodborne bacteria. Taking Escherichia coli (E. coli) O157:H7 as a model, a linear range from 2.93 × 102 to 2.93 × 108 CFU/mL and a limit of detection of 2 CFU/mL were obtained. The whole detection procedure was integrated into a single microfluidic chip. Water, milk, and cabbage samples were successfully detected, showing consistency with the results of the standard culture method. Recoveries were in the range from 90 to 110% in spiked testing. The proposed microfluidic biosensor realized the specific and sensitive detection of E. coli O157:H7 within 1 h, implying broad prospects of MOF with biomimetic enzyme activities for biosensing.

Abstract:Metal-organic framework (MOF) thin films with flexible nature and prominent qualities have opened doors to new technological applications in different fields. Herein, we propose an electrochemical biosensor for the dual detection of Staphylococcus aureus based on the electrodeposition of Cu metal-organic framework (Cu-MOF) thin films. The promising sensing layer with features of good electronic conductivity and enhanced electron-transfer property can not only identify S. aureus through specific micrococcal nucleases in the supernatant but also detect the pathogen directly via aptamer recognition. The dual analysis design ensures the accuracy of this method for S. aureus detection in the range of 7-7 x 106 cfu/mL with the limits of detection of 1.9 and 5.2 cfu/mL. Moreover, the analytical method validation confirmed that the biosensor could efficiently work in complex biological samples, showing good selectivity and specificity and great potential for clinical diagnosis. More importantly, the current proposed strategy is simple and easy to implement without the need for extra signaling elements, which is convenient for timely clinical detection.

Abstract:Background: As a critical topic of international concern, food safety has received great attention in recent years. The hazardous substances (such as antibiotics, heavy metal ions, food additives, and foodborne bacteria) in foodstuffs would cause threat to human health and economic losses in food industry. Despite of high sensitivity, accuracy, and reliability of conventional techniques for analysis of food contaminants, they often require complicated apparatus, well-trained personalized operation, and laborious and time-consuming procedure. In this regard, new sensing strategies for convenient, fast, and sensitive detection of food contaminants should be developed for food safety. Scope and approach: Metal-organic frameworks (MOFs), as a large category of porous crystalline materials, could be used as efficient platforms for constructing diverse chemosensors and biosensors, for their high porosity, adjustable compositions or structures, and good stability. A variety of MOFs, MOFs-based composites, and MOFsbased derivatives show excellent fluorescence (FL), chemical functionality, and strong bioaffinity toward probes (DNA, aptamers, or antibodies), exhibiting great potentials as FL emitters, electrode materials, or platforms of biosensors for selective and sensitive detection of hazard analytes in foodstuffs. By coupling with different determination techniques such as FL, electrochemical (EC), photoelectrochemical (PEC) or surface-enhanced Raman spectroscopy methods, MOFs-based materials have shown promising applications for detecting diverse analytes. Furthermore, the current challenges and future developments of MOFs-based materials for analysis of food contaminants have been discussed. Key findings and conclusions: Although some reviews on the applications of MOFs in food packing and food safety have been documented, this comprehensive review will provide new insights to the construction of chemosensors and biosensors with MOFs-based materials for determination of food contaminants toward food safety monitoring.
