Abstract:Food safety problems attributed to foodborne pathogenic bacteria seriously endanger human health and cause substantial economic losses. Novel assays for rapid and sensitive identification of foodborne pathogenic bacteria are highly desired. In this study, a multicolor sensing system has been established for simultaneous determination of four foodborne bacteria by exploiting oxidase mimicking activity of aptamer-functionalized manganese dioxide-coated ferriferrous oxide (apt-Fe3O4/MnO2) nanocomposites and oxTMB etching of gold nanorods (AuNRs). Apt-Fe3O4/MnO2 nanocomposites were used as capture probes to recognize and capture specific bacteria. The captured bacteria blocked the catalytic sites of the magnetic conjugate, which inhibited the catalyzation of oxTMB and further reduced the etching of AuNRs. Consequently, the longitudinal shift of AuNRs decreased linearly with the increase of the concentration of bacteria ranging from 10 to 106 CFU mL-1. Instrumental detection limits for S. aureus, L. monocytogenes, E. coli O157:H7 and V. parahaemolyticus reached down to 1.3 CFU mL-1, 1.2 CFU mL-1, 1.3 CFU mL-1 and 1.4 CFU mL-1, respectively. And their visual detection limit was as low as 10 CFU mL-1. The whole detection process only needs 40 min, suggesting that this method is promising in on-site detection of bacteria.

Abstract:Staphylococcus aureus ( S. aureus ) is a widespread pathogenic bacterium, causing foodborne disease and posing threat to public safety. Thus, it is of great importance for rapid and effective determination of S. aureus . In this protocol, we synthesized a smart DNA-modified Au/CuMOF bioconjugate material (ssDNA-Au/CuMOF) with multiple functions of biorecognition, electrochemical and catalytical activity, and explored its application in dual-mode determination of S. aureus . Magnetic bead (MB) modified with aptamers (MB-Apt) was employed for specific S. aureus capture. The incubation of ssDNA-Au/CuMOF and MB-Apt with different concentration of S. aureus led to the difference in quantity of ssDNA-Au/CuMOF confined on MB or released in supernatant. After magnetic separation, dual-mode detection was realized by the electrochemical measurement and the material-catalyzed colorimetry. This method demonstrated a satisfied analytical performance with a broad linear range from 10 to 10 8 CFU mL −1 , a colorimetric minimum resolution as low as 48 CFU mL −1 , an electrochemical detection limit of 5 CFU mL −1 , and good selectivity in distinguishing target from non-target bacteria. Its application in detecting S. aureus in complex samples also showed good response, indicating the great potential for S. aureus detection in real samples and providing a methodological and technical support for food monitoring.

Aptamer-functionalized polydiacetylene biosensor for the detection of three foodborne pathogens

Abstract:Rapid, simple and sensitive screening of foodborne pathogens is of great significance to ensure food safety. In this study, an aptamer-functionalized polydiacetylene (Apta-PDA) biosensor was developed for the detection of E. coli O157:H7, S. typhimurium or V. parahaemolyticus. First, aptamers responding to the target bacteria were modified on the surface of magnetic beads by covalent binding to form MBs-oligonucleotide conjugates for bacterial enrichment. Then, an Apta-PDA biosensor was obtained by connecting the aptamers to the PDA nanovesicles using the carbodiimide method. Molecular recognition occurred in the presence of the target bacteria, whereby the aptamer folded into a sequence-defined unique structure, resulting in an MBs-Apta/bacteria/Apta-PDA sandwich structure. Due to the optical properties of PDA, the blue-red transition of the detection system could be observed by the naked eye and quantified by the colorimetric response percentage (CR%). Under optimized conditions, the detection limits of E. coli O157:H7, S. typhimurium and V. parahaemolyticus were 39, 60 and 60 CFU/ml, respectively, with a selectivity of 100% and a reaction time of 30 min. Compared with the gold standard method, the accuracy of the three target bacteria detection reached 98%, 97.5% and 97%, respectively, and the sensitivity and specificity were both greater than 90%. The entire detection process was rapid and easy to execute without any special equipment, making this technology particularly suitable for resource-poor laboratories or regions.

Abstract:An assembled-aptasensor based on Fe3O4@Au@Ag nanocomposites grafting onto the gold foil was prepared, which can be developed into a universal approach for sensitive and rapid detection of various pathogenic bacteria, such as Escherichia coli (E. coli), Salmonella typhimurium (S. typhimurium), Staphylococcus aureus (S. aureus), Listeria monocytogenes (L. monocytogenes), Pseudomonas aeruginosa (P. aeruginosa), and Shigella flexneri (S. flexneri). Firstly, the gold foil paper was modified with thiolated capture probe and SERS tag in proportion, and at the same time, the specific thiolated aptamer probe for corresponding pathogenic bacteria was fixed with Fe3O4@Au@Ag nanocomposites. An obvious Raman signal can be subsequently increased about 106 times by the external electromagnetic field enhancement at the “hot spots” caused by the hybridization of aptamer and capture probe. But in the presence of target pathogenic bacteria, Raman intensity will decrease as Fe3O4@Au@Ag nanocomposites are dissociated from gold foil. Thus, all of the concentrations of the six kinds of pathogenic bacteria both in PBS and liquorice extract showed an obvious negative linear correlation with the Raman intensity of SERS tag in the range of 10–107 CFU/mL with detection limits were all lower than 10 CFU/mL. And there was no significant difference between our method and the plate counting method. Besides, the assembled-aptasensor had superior specific recognition ability even in the mixed interfering bacteria. Our study showed that this assembled-aptasensor had good specific detection ability to a variety of foodborne pathogens based on magnetic field-assisted SERS technique, which can be used for rapid and sensitive detection of a variety of pathogens in complex substrates.Graphical abstract

Abstract:An aptamer-based sensitive method was developed here for detection of multiple foodbome pathogens in food matrix by surface-enhanced Raman scattering (SERS) technology. Polydimethylsiloxane (PDMS) film was first prepared and then coated with gold nanopartides (AuNP) to act as an active substrate for the enhancement of Raman scattering. The as-prepared Au-PDMS film was functionalized with specific pathogen aptamers (Apt) to capture the targets. In addition, aptamers functionalized AuNP integrated with Raman reporters (4-Mercaptobenzoic acid (4-MBA)/Nile blue A (NBA)) were fabricated as pathogen-specific SERS probes. In this scheme, pathogens were first captured by Apt-Au-PDMS film and then bind with SERS probes to allow the formation of a sandwich assay to complete the sensor module for the detection of multiple pathogens. With Vibrio parahaemolyticus and Salmonella typhimurium as model targets, this protocol can selectively detect 18 cfu/mL and 27 cfu/mL, respectively. Furthermore, this platform can be successfully applied to detect pathogens in seafood samples with recoveries ranging from 82.9% to 95.1%. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Given the frequent occurrence of multidrug-resistant bacteria (MDRB), which pose a severe threat towards public hygiene, it is important to construct facile and sensitive strategies for the detection of MDRB. We developed a fluorescent/colorimetric dual-mode biosensing platform for the analysis of MDRB by integrating the remarkable peroxidase-like activity of boric acid-decorated fluorescent framework hybrid (MOF@COF) nanozyme and enhanced bacterial affinity of DNA programmable multivalent aptamer scaffold. The efficiency and sensitivity of the proposed analysis approach were dramatically increased due to richer aptamers and high-performance MOF@COF nanozyme. Compared with commonly used single recognition methods, multivalent aptamers and bacteria-affinity boric acid groups on the nanozyme surface could identify multiple sites of MDRBs to improve site utilization for ultrasensitive biosensing. The platform realized selective detection of Acinetobacter baumannii (AB) and Pseudomonas aeruginosa (PA) in two modes with consistent results. In colorimetry and fluorescence modes, the linear ranges of AB were 10 2 -10 8 and 10–10 8 CFU/mL with corresponding detection limits of 12 and 2 CFU/mL, and the linear ranges of PA were 10 2 -10 8 and 10–10 8 CFU/mL with corresponding detection limits of 10 and 3 CFU/mL, respectively. More importantly, the biosensing platform could be applied for MDRB quantification in biological specimens, including human serum, cerebrospinal fluid, and urine with remarkable recoveries (96.8–105.0%) in both modes, indicating the bright prospect of the biosensing platform for MDRB determination in clinical diagnosis.

Abstract:Foodborne pathogens could result in serious health issues to public and thus some rapid analysis methods are urgent developed to monitor them. Herein, a dual-mode aptasensor based on electrochemical and colorimetric modes was constructed for the on-site detection of Vibrio parahaemolyticus (V.P) in shrimps as a model. A novel nanoprobe (@MXenes) was developed to generate both electrochemical and colorimetric signals by modifying phenylboronic acid and ferrocene on Pt-MXenes nanocomposite. The nanoprobe exhibited both features of peroxidase mimic and specific recognition. Simultaneously, a V.P aptamer-functionalized electrode interface was fabricated to firstly specifically capture V.P, then conjugated with @MXenes to form a sandwich complex. The complex could catalyze tetramethylbenzidine (TMB)-H2O2 to produce a visible signal, and generate electrochemical signal simultaneously. Under the optimal condition, the aptasensor could determine as low as 5 CFU mL−1 V.P by electrochemical mode and 30 CFU mL−1 V.P by colorimetric mode. These superior performances may be attributed to that the synergistic catalysis and conductive effects deriving from Pt and MXenes on the nanoprobe can significantly amplify the signals to improve detection sensitivity. In particular, the dual detection system could mutually verify to achieve high accuracy. The aptasensor could also be expanded to rapid analysis of other food-borne pathogens through changing the aptamer on the electrode.

Abstract:Cytotoxic activities of unsaturated fatty acids against lymphocytes were studied by demonstrating the inhibition of RNA synthetic activity of incubated lymphocytes. In general, the cytotoxic activity of fatty acids increases with decreasing number of carbon atoms and increasing number of double bonds. An empirical formula predicting the degree of impairment of lymphocytes by various fatty acids was proposed. Furthermore, protective effects of serum on cytotoxic actions of unsaturated fatty acids to lymphocytes were studied and the significance of these findings was discussed.

Abstract:Simultaneous detection of multiple foodborne pathogens is of great importance for ensuring food safety. Herein, we present a sensitive dual-channel electrochemical biosensor based on copper metal organic frameworks (CuMOF) and lead metal organic framework (PbMOF) for simultaneous detection of Salmonella typhimurium ( S. typhimurium ) and Listeria monocytogenes ( L. monocytogenes ). The MOF-based nanotags were prepared by functionalizing gold nanoparticles loaded CuMOF (Au@CuMOF) and PbMOF (Au@PbMOF) with signal DNA sequences 1 (sDNA1) and sDNA2, respectively. By selecting invA of S. typhimurium and inlA gene of L. monocytogenes as targe sequences, a sandwich-typed dual-channel biosensor was developed on glassy carbon electrodes (GCE) through hybridization reactions. The sensitive detection of S. typhimurium and L. monocytogenes was achieved by the direct differential pulse voltametric (DPV) signals of Cu 2+ and Pb 2+ . Under optimal conditions, channel 1 of the biosensor showed linear range for invA gene of S. typhimurium in 1 × 10 −14 −1 × 10 −8 M with low detection limit (LOD) of 3.42 × 10 −16 M ( S/N = 3), and channel 2 of the biosensor showed linear range for inlA gene of L. monocytogenes in 1 × 10 −13 −1 × 10 −8 M with LOD of 6.11 × 10 −15 M (S/N = 3). The dual-channel biosensor showed good selectivity which were used to detect S. typhimurium with linear range of 5−1.0 × 10 4 CFU mL −1 (LOD of 2.33 CFU mL −1 ), and L. monocytogenes with linear range of 10 − 1.0 × 10 4 CFU mL −1 (LOD of 6.61 CFU mL −1 ).

Abstract:Foodborne diseases caused by Salmonella enterica ( S. enterica ) and Staphylococcus aureus ( S. aureus ) significantly impact public health, underscoring the imperative for highly sensitive, rapid, and accurate detection technologies to ensure food safety and prevent human diseases. Nanomaterials hold great promise in the development of high-sensitivity transistor biosensors. In this work, field-effect transistor (FET) comprising high-purity carbon nanotubes (CNTs) were fabricated and modified with corresponding nucleic acid aptamers for the high-affinity and selective capture of S. enterica and S. aureus . The aptamer-functionalized CNT-FET biosensor demonstrated ultra-sensitive and rapid detection of these foodborne pathogens. Experimental results indicated that the biosensor could detect S. enterica at a limit of detection (LOD) as low as 1 CFU in PBS buffer, and S. aureus at an LOD of 1.2 CFUs, achieving single-cell level detection accuracy with exceptional specificity. The biosensor exhibited a rapid response time, completing single detections within 200 s. Even in the presence of interference from six complex food matrices, the biosensor maintained its ultra-sensitive (3.1 CFUs) and rapid response (within 200 s) characteristics for both pathogens. The developed aptamer-functionalized CNT-FET biosensor demonstrates a capability for low-cost, ultra-sensitive, label-free, and rapid detection of low-abundance S. enterica and S. aureus in both buffer solutions and complex environments. This innovation holds significant potential for applying this detection technology to on-site rapid testing scenarios, offering a promising solution to the pressing need for efficient and reliable pathogen monitoring in various settings.

Abstract:Foodborne pathogens, especially bacteria, are explicitly threatening public health worldwide. Biosensors represent advances in rapid diagnosis with high sensitivity and selectivity. However, multiplexed analysis and minimal pretreatment are still challenging. We fabricate a gold nanoparticle (Au NP)-amplified microcantilever array biosensor that is capable of determining ultralow concentrations of foodborne bacteria including Escherichia coli O157:H7, Vibrio parahaemolyticus, Salmonella, Staphylococcus aureus, Listeria monocytogenes, Shigella, etc. The method is much faster than using conventional tools without germiculturing and PCR amplification. The six pairs of ssDNA probes (ssDNA1 + ssDNA2 partially complementary to the target gene) that originated from the sequence analysis of the specific gene of the bacteria were developed and validated. The ssDNA1 probes were modified with -S-(CH2)6 at the 5′-end and ready to immobilize on the self-assembled monolayers (SAMs) of the sensing cantilevers in the array and couple with Au NPs, while 6-mercapto-1-hexanol SAM modification was carried out on the reference cantilevers to eliminate the interferences by detecting the deflection from the environment induced by non-specific interactions. For multianalyte sensing, the target gene sequence was captured by the ssDNA2-Au NPs in the solution, and then the Au NPs-ssDNA2-target complex was hybridized with ssNDA1 fixed on the beam of the cantilever sensor, which results in a secondary cascade amplification effect. Integrated with the enrichment of the Au NP platform and the microcantilever array sensor detection, multiple bacteria could be rapidly and accurately determined as low as 1–9 cells/mL, and the working ranges were three to four orders of magnitude. There was virtually no cross-reaction among the various probes with different species. As described herein, it holds great potential for rapid, multiplexed, and ultrasensitive detection in food, environment, clinical, and communal samples.

Abstract:Metal-organic framework (MOF) thin films with flexible nature and prominent qualities have opened doors to new technological applications in different fields. Herein, we propose an electrochemical biosensor for the dual detection of Staphylococcus aureus based on the electrodeposition of Cu metal-organic framework (Cu-MOF) thin films. The promising sensing layer with features of good electronic conductivity and enhanced electron-transfer property can not only identify S. aureus through specific micrococcal nucleases in the supernatant but also detect the pathogen directly via aptamer recognition. The dual analysis design ensures the accuracy of this method for S. aureus detection in the range of 7-7 x 106 cfu/mL with the limits of detection of 1.9 and 5.2 cfu/mL. Moreover, the analytical method validation confirmed that the biosensor could efficiently work in complex biological samples, showing good selectivity and specificity and great potential for clinical diagnosis. More importantly, the current proposed strategy is simple and easy to implement without the need for extra signaling elements, which is convenient for timely clinical detection.

Abstract:A signal amplification electrochemical biosensor chip was developed to integrate loop-mediated isothermal amplification (LAMP) based on in situ nucleic acid amplification and methyl blue (MB) serving as the hybridization redox indicator for sensitive and selective foodborne pathogen detection without a washing step. The electrochemical biosensor chip was designed by a screen-printed carbon electrode modified with gold nanoparticles (Au NPs) and covered with polydimethylsiloxane membrane to form a microcell. The primers of the target were immobilized on the Au NPs by covalent attachment for in situ amplification. The electroactive MB was used as the electrochemical signal reporter and embedded into the double-stranded DNA (dsDNA) amplicons generated by LAMP. Differential pulse voltammetry was introduced to survey the dsDNA hybridization with MB, which differentiates the specifically electrode-unbound and -bound labels without a washing step. Pyrene as the back-filling agent can further improve response signaling by reducing non-specific adsorption. This method is operationally simple, specific, and effective. The biosensor showed a detection linear range of 102-107 CFU mL-1 with the limit of detection of 17.7 CFU mL-1 within 40 min. This method showed promise for on-site testing of foodborne pathogens and could be integrated into an all-in-one device.

Abstract:Rapid detection of foodborne pathogens plays a more and more significant role in the field of public health. In this research, we developed a convenient electrochemical biosensor mediated by filtration-assisted pretreatment and multiple thermostatic enzymes (Multi-TEs) amplification system for the detection of Staphylococcus aureus ( S. aureus ) in complex matrices. Firstly, the designed filtration-assisted pretreatment could simultaneously collect and enrich target bacterium from various actual samples within 5 min. Then, the target bacterial genome template was reacted at 37 °C in Multi-TEs amplification system to obtain specific double stranded products. Finally, the specific double stranded products were fixed on the surface of the electrode and the electrochemical indicators (Methylene blue, MB) was combined for the quantitative one-step electrochemical analysis of S. aureus . This assay has great linear calibration curves with the concentration ranging from 10 1 CFU/mL to 10 6 CFU/mL. The limit of detections (LOD) was defined as 1.28 CFU/mL, which performance is superior to some commercial ELISA kits. Meanwhile, amazingly, it has excellent specificity for S. aureus against other foodborne pathogens and could be applied for S. aureus analysis in human serum samples with the great recoveries and associated relative standard deviations (RSDs). These features do make the ideal facile electrochemical analytical platform for on-site detection of S. aureus.

Abstract:The inability to integrate detection and disinfection hindered building a unified pathogen monitoring platform, risking secondary contamination. Herein, a novel "four - in - one" platform for monitoring foodborne Listeria monocytogenes (L. monocytogenes) was presented. The magnetic daptomycin - functionalized Fe3O4 (Dap/Fe3O4) could selectively bind to L. monocytogenes, enhancing detection accuracy. The separated bacteria were captured by aptamers - functionalized Fe - doped - silica nanoparticles (Apt/Fe@SiNPs) for tri - mode detection. Besides fluorescence, the Apt/Fe@SiNPs converted 3,3',5,5' - tetramethylbenzidine (TMB) to oxidized TMB (oxTMB) via peroxidase activity, allowing colorimetric and subsequent photothermal detection upon irradiation, as low as 2.06 CFU/mL. Magnetic - induced aggregation of Apt/Fe@SiNPs generated toxic hydroxyl radicals around L. monocytogenes, achieving ∼99.6% disinfection. Furthermore, the biofilm of L. monocytogenes was effectively inhibited by the action of hydroxyl radicals. The platform might offer a promising prospect to control L. monocytogenes in food industries.
