Abstract:The increasing demand for rapid and sensitive roadside identification of trace illegal drugs drives the development of high-performance miniature mass spectrometric instrumentation and methodology. Here, we report a synchronized flash-thermal-desorption purging and ion injection (SFTDPI) method to increase the sensitive and rapid screening of volatile and nonvolatile illegal drugs for miniature ion trap mass spectrometry (ITMS). The flash-thermal desorption could reach 290 degrees C in 2.5 s, which could achieve efficient vaporization of nonvolatile noscapine with boiling point at 565 degrees C. ITMS using discontinuous atmospheric pressure interface (DAPI) has an ion utilization ratio of less than 1%, the synchronized purging for flash-thermal desorption and ion injection with DAPI could accurately control the time interval along with the desorption, gas purging, ionization and ion injection, and the sample utilization ratio increases more than five times. The miniature SFTDPI-ITMS presents good performance: (1) more than 60 times improvement in sensitivity was achieved compared to the previously reported thermal-desorption acetone-assisted photoionization ion trap mass spectrometer for nonvolatile drugs, and the minimum detectable quantity reaches 50 pg for fentanyl. (2) Ten kinds of mixing drugs with boiling point difference of 300 degrees C can be simultaneously identified within 3 s under a single analysis. SFTDPI-ITMS was deployed at the roadside checkpoint of Sino-Burmese border; fentanyl in the captured encapsulated powder and the suspected opiate had been successfully identified.

Abstract:Due to endogenous existence and hazardous properties, the quantification of fatty and aromatic aldehydes in food matrices is indispensable to food safety. Current analytical methodologies for aldehydes in food suffer from several drawbacks (typically complicated pretreatment, long analysis time and low analytical throughput, low sensitivity, etc.). In this work, a highly sensitive, high-throughput and sample pretreatment-free method for aldehyde analysis was developed to address the above problems, where in situ extraction and derivatization of aldehydes with hydrazines to hydrazones were integrated into reactive paper spray ionization mass spectrometry (reactive PSI-MS) considering weakly ionizable properties of aldehydes. Four aldehydes, formaldehyde, acetaldehyde, propionaldehyde and benzaldehyde were reacted with dansyl hydrazine in 1.5 % v/v HAc acidified methanol on the qualitative fast-speed filter paper for 5 min, and then a voltage (+4 kV) was applied to initiate paper spray for mass spectrometry detection with internal calibration by dansyl amide. Once the completion of MS analysis, the glass plate was moved to introduce the second triangular tip towards the MS inlet (5 mm apart) for next trial, achieving high-throughput reactive PSI-MS analysis (60 h(-1)). The four aldehydes showed good linearity in the concentration range from 2 to 150 mu M with low detection limits of 0.03-0.15 mu M. The method was applied to direct quantification of four aldehydes in 15 kinds of foods such as vegetables, fruits and meat products. The proposed method has the advantages of simple operation, high efficiency and high sensitivity, making it attractive to routine analysis of aldehydes in food, environmental and biological samples.
Rapid Screening of Illegally Added Drugs in Functional Food Using a Miniature Ion Trap Mass Spectrometer

Abstract:With the expansion of the functional food market, the qualification assessment of these products has become a major challenge, and efficient analytical tools are urgently needed. Here, a miniature mass spectrometer (MS) with self-aspiration capillary electrospray ionization (SACESI) source and ion trap analyzer was developed for rapid screening of various illegally added drugs in functional foods. No chromatographic separation was required, but a simplified two-step pretreatment method was developed to reduce the operational procedures and time consumption of the entire analysis. SACESI source uses capillary action to drive solution injection, which utilizes a simple structure and convenient operation to constitute a kind of disposable MS detection solution. To achieve accurate and automatic identification, an intelligent recognition algorithm with steps of spectrum preprocessing, characteristic peak matching, and support vector machine learning was constructed. The relative accuracy of rapid screening of 31 suspicious drugs in various samples is up to 99.78%. It achieves 100% correct identification for the 55 batches of actual samples captured by on-site inspection, which demonstrates the feasibility of the proposed analytical system and strategy in food safety applications.

Abstract:The present study reports on a streamlined analytical platform for rapid, straightforward, and on-site sample analysis using a miniature mass spectrometer with adequate tandem mass spectrometry (MS/MS) capability. An “all-in-one” workflow is developed combining accelerated air-assisted in-syringe extraction, sorbent-packed membrane clean-up, and needle spray ionization with an in-house built syringe assembly. A custom-made metal needle with a sharp, thin, and conical tip of micrometer-scale served as an integral component of the syringe assembly. This needle was capable of both aspirating a high-speed airflow for a highly efficient in-syringe extraction and performing stable electrospray for direct miniature mass spectrometry analysis. As a proof-of-concept demonstration, the developed protocol was implemented for the identification of 40 adulterated cosmetic ingredients and validated in terms of sensitivity, linearity, matrix effect, and recovery. A custom expandable mass spectral library was established in-house for smart suspect screening. Compound search and identification were conducted following a two-stage strategy using the full-scan MS and MS/MS data. The methodology demonstrates excellent potential for outside-the-laboratory chemical analysis by reducing the high time and labor costs associated with traditional sample preparation and chromatographic separation procedures, which would meet product quality and safety inspection requirements for on-site compliance check and regulatory enforcement at the testing sites such as supermarkets, department stores, wholesale markets, and roadside stalls.
Quick and High-Throughput Quantification of Β-Agonists Residues in Animal-Derived Foods Using Enzymatic Probe Sonication Coupled with Automatic Solid-Phase Extraction

Abstract:A high demand exists in veterinary drug residual analysis for rapid, automatic and high-throughput analytical techniques that produce data simultaneous and faster. Here, we describe a combined automated solid-phase extraction (SPE) and enzymatic probe sonication (EPS), subsequently ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) for the quick extraction, automated clean-up as well as simultaneous quantification of 22 β-agonists residues in animal-derived foods. Enhanced by the ultrasonic probe, only 2 min was needed for exhaustively extraction of β-agonists from foods of animal-origin; whereas traditional enzymatic digestion methods require hours or even days, which making it more appropriate for high-throughput biomonitoring. Moreover, the clean-up and pre-concentration procedures were conducted on the automatic SPE, which allowed 36 samples being performed simultaneously within 30 min. The mean recoveries for the target analytes in the fortified samples varied from 80.8% and 104.5% with the intra-day relative standard deviation (RSD) lower than 8.7%.
Simultaneous Determination of Multi-Component Mycotoxin Contaminants in Foods and Feeds by Ultra-Performance Liquid Chromatography Tandem Mass Spectrometry.

Abstract:The present study developed an improved analytical method for the simultaneous quantification of 17 kinds of Aspergillus, Fusarium and Penicillium mycotoxin contaminants in foods and feeds by ultra-performance liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry (UPLC–MS/MS) under the multiple reaction monitoring (MRM) mode, and especially focused on the optimization of extraction, clean-up, UPLC separation and MS/MS parameters of analytes. Homogenized samples were sequentially extracted by 84% (v/v) of acetonitrile aqueous solution with the selected internal standard (zearalanone) spiking, SPE clean-up with Mycosep 226 Aflazon+ Multifunctional cartridges, filtration, concentration and secondary filtration. Using double sample injection method, the analytes were separated by UPLC BEH C18 column (100mm×2.1mm I.D., 1.7μm), and eluted with ammonium acetate/methanol and aqueous ammonia/methanol for the ESI+ and ESI− analysis, respectively. The 10 positive ions and 7 negative ions of mycotoxins were separated by gradient elution with the retention time of 6.5 and 4min, respectively. The LOQ of selected analytes ranged from 0.01 to 0.70μgkg−1, which was lower than the criteria of EU, USA and other countries on the determination of the minimum limiting level of various mycotoxins in foods including baby foods and feed stuffs. Meanwhile, high correlation coefficients (r2>0.99) of 17 mycotoxins were obtained within their respective linear ranges (0.05–20μgkg−1 for 10 positive ions and 0.5–50μgkg−1 for 7 negative ions) and reasonable recoveries (70.6–119.0%) of them were also demonstrated in different spiked levels. This quantitative method has many advantages including simple pretreatment, rapid determination and high sensitivity, which could be applied to the determination and quantification of multi-component mycotoxin contaminants in complex matrixes. Meanwhile, the method successfully fulfilled the minimum limiting level requests from various countries.

Abstract:This article reports on the development of an atmospheric pressure filtering electrospray ionization (FESI) source - composed of a syringe, a filter, and the ESI source - for the determination of furazolidone, amoxicillin, nitrofurantoin, and nitrofural, common antibiotics. Spiked food samples were also analyzed; the analytes were placed in the filter head, and the flow rate of the extraction solution from the syringe pump was adjusted. Using a mixture of oleic acid and antibiotics, hydrophilic and hydrophobic filters were compared, demonstrating the advantage of FESI in reducing the background and noise for the mixture. The device simplifies not only the experimental procedure but also the sample pretreatment process. Additionally, the limits of detection of the antibiotics were determined to be 1 mg L-1 for furazolidone, amoxicillin, and nitrofurantoin and 2 mg L-1 for nitrofural. The linear regression coefficients (R2) of the antibiotic standard solutions were all >= 0.99. Furthermore, the spiked recovery rates of pork, chicken, and egg samples ranged from 73.3% to 107.5%, with relative standard deviations (RSDs) from 1.1% to 15.6%. This study uncovers the potential capabilities of ion trap mass spectrometry (ITMS) for the on-site detection of antibiotics and offers a new method for ensuring food safety.

Abstract:Direct mass spectrometry analysis of untreated samples of volumes as low as 0.2 µL were achieved using fast extraction and nanoESI (electrospray ionization) in a combined fashion. The analytes in dried samples on paper substrates were extracted by organic solvent in a nanoESI tube and ionized with a high voltage applied for generating a spray. The ionization source produced stable signals for different atmospheric pressure interfaces of triple quadrupole instruments. Analysis time more than 20 minutes were available with 10 µL solvent consumed for the entire analysis process. The performance in qualitative and quantitative analysis was characterized with a wide variety of samples. Limits of detection as low as 0.1 ng/mL (corresponding to an absolute amount of 0.05 pg) were obtained for analysis of atrazine in river water, thiabendazole in orange homogenate, and methamphetamine in blood.

Abstract:led some experts to recommend that patients starting a weight loss program be provided a choice between empirically supported dietary approaches. For example, a Mediterranean diet tends to be one high in vegetables and fruits and low in fat diet, whereas a diet high in protein, such as the South Beach Diet, is consistent with a low-carbohydrate diet, yet both may be effective. Other types of diet have also been investigated relative to their ability to contribute to weight loss. Vegetarian diets have been found to be effective for weight management, but effects could be deleterious when energy restriction is included (Farmer, 2014). In their systematic review, Mytton, Nnoaham, Eyles, Scarborough, and Ni Mhurchu (2014) found that increased consumption of fruits and vegetables without decreasing intake of other foods did not contribute to weight gain and, in some instances, could be infl uential in weight maintenance. A randomized control trial comparing weight loss outcomes of the Mediterranean diet (Med diet) with MyPyramid in a sample of postpartum breastfeeding women found both diets resulted in weight loss during the postpartum period with weight loss change ranging from -2.31 ± 3.42 (p < 0.001) for participants in the Med diet group to -3.11 ± 3.35 (p < .001) for those in the MyPyramid group (StendellHollis, Thompson, West, Wertheim, & Thomson, 2013). Abstract:Rapid analysis of metal ions and organic compounds in strong acidic solutions is of sustainable interest in multiple disciplines. However, complicated and time-consuming pretreatments are always required for MS analysis of the compounds in strong acidic solutions. Otherwise, it will result in a weak signal and cause serious damage to the mass spectrometer. Herein, a simple method inherited from nano-ESI MS was developed for rapid analysis of strong acidic solutions. Nanoliter (nL) strong acidic solution was first loaded in the nano-ESI emitter, followed by evaporation to remove the H+ and leave the analytes on the wall of the nano-ESI emitter. The evaporation process can be completed within 1 min because of the extremely tiny volume (≤ 1 nL) of the loaded solution. Then, the dried analytes on the wall of the nano-ESI emitter were redissolved by loading a new solvent, followed by nano-ESI MS analysis. By using this method, metal ions and organic compounds in the strong acidic solution can be detected with low sample consumption (1 nL), high speed (< 2 min/sample), high sensitivity (limit of detection = 0.2 μg/L), and high accuracy (> 90%). Proof-of-concept applications of the present method have been successfully achieved for the analysis of gastric juice (pH of the sample = 1), monitoring reaction catalyzed by strong acid (pH of the system = 0), and micro-area analysis of ores (pH of the extraction solvent = 0), showing great application potential in multiple fields.

A Small Footprint and Robust Interface for Solid Phase Microextraction and Mass Spectrometry Based on Vibrating Sharp-Edge Spray Ionization

Abstract:Combining solid phase microextraction (SPME) and mass spectrometry (MS) analysis has become increasingly important to many bioanalytical, environmental, and forensic applications due to its simplicity, rapid analysis, and capability of reducing matrix effects for complex samples. To further promote the adoption of SPME-MS based analysis and expand its application scope calls for efficient and convenient interfaces that couple the SPME sample handling with the efficient analyte ionization for MS. Here, we report a novel interface that integrates both the desorption and the ionization steps in one device based on the capillary vibrating sharp-edge spray ionization (cVSSI) method. We demonstrated that the cVSSI is capable of nebulizing liquid samples in a pulled-tip glass capillary with a battery powered function generator. The cVSSI device allows the insertion of a SPME probe into the spray capillary for desorption and then direct nebulization of the desorption solvent in situ. With the integrated interface, we have demonstrated rapid MS analysis of drug compounds from serum samples. Quantitative determination of various drug compounds including metoprolol, pindolol, acebutolol, oxprenolol, capecitabine, and irinotecan was achieved with good linearity (<i>R</i><sup>2</sup> = 0.97-0.99) and limit of detection ranging from 0.25 to 0.59 ng/mL without using a high voltage source. Only 3.5 μL of desorption solvent and 3 min desorption time were needed for the present method. Overall, we demonstrated a portable SPME-MS interface featuring high sensitivity, short analysis time, small footprint, and low cost, which makes it an attractive method for many applications requiring sample cleanup including drug compound monitoring, environmental sample analysis, and forensic sample analysis.

Abstract:Pesticides residues in foodstuffs are longstanding of great concern to consumers and governments, thus reliable evaluation techniques for these residues are necessary to ensure food safety. Emerging ambient ionization mass spectrometry (AIMS), a transformative technology in the field of analytical chemistry, is becoming a promising and solid evaluation technology due to its advantages of direct, real-time and in-situ ionization on samples without complex pretreatments. To provide useful guidance on the evaluation techniques in the field of food safety, we offered a comprehensive review on the AIMS technology and introduced their novel applications for the analysis of residual pesticides in foodstuffs under different testing scenarios (i.e., quantitative, screening, imaging, high-throughput detection and rapid on-site analysis). Meanwhile, the creative combination of AIMS with high-resolution mass analyzer (e.g., orbitrap and time-of-flight) was fundamentally mentioned based on recent studies about the detection and evaluation of multi-residual pesticides between 2015 and 2021. Finally, the technical challenges and prospects associated with AIMS operation in food industry were discussed.

Abstract:RATIONALE: Extractive electrospray ionization mass spectrometry (EESI-MS) was invented as a typical ambient mass spectrometry method (AMS) and has been used for analyzing complex liquid samples. Here, we designed a Venturi effect-based self-aspiration sampling device and applied it to the EESI-MS for high-throughput analysis of liquid sample.METHODS: A special concentric nebulizer was designed and employed to produce a suction force for the direct aspiration of liquid samples, followed by ionization and detection. This sample aspiration process was explained and optimized using computational fluid dynamics (CFD) analysis. Experiment data were recorded to exhibit the sensitivity, memory effect, inter-day reproducibility, throughput, and applicability of the self-aspiration sampling EESI-MS.RESULTS: The limit of detection (LOD) of this method was determined as 4.5 x 10(-10) g/mL (S/N = 3) for caffeine, and the sample throughput and relative standard deviation (RSD) for full scan mode can reach 0.67 samples/s and 4.76%, respectively. Even for MS/MS mode, the frequency can still be kept at 0.4 samples/s (RSD = 4.71%). Inter-day RSD examined in 1 week was below 10% for the signal of characteristic fragment ions of reserpine. Moreover, based on this method, the amount of caffeine in instant coffee was determined as 4.7%. This device was also proven to be suitable for the protein/peptide analysis.CONCLUSIONS: These experiment results, especially the amazing results on sample throughput and inter-day RSD, suggest that we provide a valuable device which can be used for the direct high-throughput qualitative/quantitative mass spectrometry analysis of real liquid samples in ambient. Abstract:A high-throughput method for rapid screening of active ingredients in drugs has been developed with mass spectrometry coupled to a low-temperature plasma (LTP) probe ion source. Without sample preparation or pretreatment, the active ingredients of 11 types of commercial pharmaceuticals, including hormones, antipyretic analgesics, cardiovascular, digestant, neuro-psychotherapeutic, diuretic, antithyroid, sulfa anti-inflammatory, antiparastic, sedative-hypnotics, and antibacterial, were directly desorbed/ionized and detected by a linear ion trap mass spectrometry (MS). The structures of these ingredients were elucidated by tandem MS. The analysis of 18 methyltestosterone tablets could be accomplished within 1.9 min, which allows fast detection with a speed of approximate 600 samples within 1 h. This work demonstrated that LTP probe ion source combined with MS is a high-throughput method for screening of pharmaceuticals and potentially applied to on-line quality control in pharmaceutical industry. Rapid characterization of complex viscous samples at molecular levels by neutral desorption extractive electrospray ionization mass spectrometry

Abstract:In this protocol, the sample (which could be a bulk or heterogeneous fluid, or a greasy surface) is treated with a neutral desorption (ND) sampling gas beam, and the resulting analyte mixtures are directly characterized by extractive electrospray ionization mass spectrometry (EESI-MS). The ND device can be specifically constructed such that the sampling gas beam is bubbled through the liquid sample (microjet sampling) or directed to impact the sample surface (e.g., for the analysis of a material like cheese). The ND-EESI-MS analysis process requires no sample pretreatment because it can tolerate an extremely complex matrix. ND-EESI-MS allows real-time, online chemical profiling of highly viscous samples under ambient conditions. Both volatile and nonvolatile analytes from viscous samples can easily be detected and quantified by ND-EESI-MS, thereby providing an MS-based analytical platform for multiple disciplines (e.g., for the food industry, for drug discovery, and for the biological and life sciences). Here we describe the ND-EESI-MS protocol for viscous sample analysis, including the experimental design, equipment setup, reagent preparation, data acquisition and analysis steps. The data collection process takes <1 min per sample, although the time required for the whole procedure, which largely depends on the experimental preparation processes, might be considerably longer.

Herein, a novel ionization technique called induced self-aspiration-electrospray ionization (ISA-ESI) was developed based on self- aspiration sampling and capacitive induction. The sample solution polarized in a strong electric field was pulsed drawn into a capillary that was connected to a subambient chamber. The sample solution with polarized ions forms a charged liquid column, which can initiate an electrospray when reaching the capillary outlet. In addition to the self-aspiration ability, the use of a constant high voltage supply and no electrical contact with the solution can also simplify the sampling and ionization operation, enabling a convenient ESI mass spectrometry analysis. The developed ISA-ESI source has been used for multidimensional monitoring of chemical reactions as well as liquid extraction surface analysis of plant tissues. It was expected that this special ionization method could be extended to automated high-throughput ESI-MS analysis.
Direct determination of glyphosate, aminomethylphosphonic acid and glufosinate in food samples with ion chromatography coupled to electrospray ionization tandem mass spectrometry

Abstract:A fast and reliable method for the direct determination of the herbicide glyphosate, its major degradation product aminomethylphosphonic acid (AMPA) and glufosinate is presented for a variety of food matrices. The Quick Polar Pesticides in food of Plant Origin method (QuPPe-PO-Method) was used for extraction without further preconcentration or clean-up steps involving e.g. solid phase extraction (SPE). The method makes use of a commercially available high performance liquid chromatograph coupled to a tandem mass spectrometer with electrospray ionization (LC-ESI-MS/MS) - as present in many laboratories - equipped with an ion chromatography (IC)-column using an MS-compatible eluent made of 0.8% formic acid in water. Due to the absence of time-consuming clean-up procedures, strong matrix effects (ME) of up to 91% for AMPA in grapefruit can be observed, when comparing its sensitivity to that obtained for solvent-based standards. The limits of detection (LODs) were determined for the sample matrices apple, mushrooms, grapefruit, linseed, red lentils and wheat and they were found to be in the range of 0.09 to 0.8, 0.04 to 1 and 0.2 to 2 µg/kg for glyphosate, AMPA and glufosinate, respectively. For the same matrices the validation was carried out according to SANTE guidelines for different commodity groups by spiking them up prior to extraction to concentrations ranging from 10 to 400 µg/kg for matrices with high water content and from 10 to 800 µg/kg for matrices with low water content. When using solvent-based calibration under the use of isotopically labelled internal standards (ILIS) the recoveries were found to range from 84% to 120% and the relative standard deviations (RSD) range between 1% and 19% for glyphosate, AMPA and glufosinate at all fortification levels for all matrices investigated. Accordingly, the method was successfully introduced in our laboratory with limits of quantification (LOQs) of 10 µg/kg for glyphosate, AMPA and glufosinate in samples from SANTE commodity groups 1, 2, 4a and 5. The reliability and robustness of the method are demonstrated by showing a recovery control chart obtained for glyphosate in randomly selected samples from different commodity groups. Therefore, the samples were spiked up with 10 µg/kg of glyphosate during routine analysis, whereby all recoveries were found to be in the range between 70 and 120%.

Abstract:Electrospray ionization (ESI) has become a powerful tool for the analysis of biomolecules by mass spectrometry (MS). The process of ESI is difficult to control, and side reactions such as electrochemical reactions can occur during the ESI process because of the high voltages applied. Herein, a novel on-demand MS analysis method was developed based on discontinuous ion injection-induced ESI on a miniature MS system. Highly efficient ionization was enabled under low voltages (<300 V) using a discontinuous atmospheric pressure interface. On-demand ionization showed comparable sensitivity with regular nanoESI for the analyses of a series of compounds. It was found to be softer than regular ESI or nanoESI methods for ionization of proteins such as myoglobin and cytochrome C. As the ionization finished as soon as the interface was closed, the sample consumption was observed to reduce significantly for MS analysis, allowing single-cell analysis with multiple MS and MS/MS measurements.

Abstract:Dielectric barrier discharge ionization is increasingly used for rapid detection in ambient mass spectrometry, although more often for gaseous and highly volatile samples than for solids and liquids. In this project, we present a rapid and sensitive method for detecting mycotoxins and demonstrate its capability for the detection of aflatoxin B1, zearalenone, and ochratoxin A in food samples. Our method is based on thermal desorption coupled to dielectric barrier discharge ionization mass spectrometry (TD-DBDI-MS), which we show generates minimal interferences and produces almost exclusively molecular ions. We detected mycotoxins in various food samples, including corn, peanuts, millet, and rice. Our method has a linear dynamic range of 1 μg/kg to 100 μg/kg for all three mycotoxins and a limit of detection (LOD) of 0.31 μg/kg, 0.28 μg/kg and 0.43 μg/kg, respectively. It is simple, rapid, reduces the pretreatment steps and has significant potential for practical applications.
